Multi-Immunogenic Outer Membrane Vesicles Derived from a MsbB-Deficient Salmonella enterica Serovar Typhimurium Mutant

  • To develop low endotoxic and multi-immunogenic outer membrane vesicles (OMVs), a deletion mutant of the msbB gene in Salmonella enterica serovar Typhimurium (S. Typhimurium) was used as a source of low endotoxic OMV, and an expression vector of the canine parvovirus (CPV) VP2 epitope fused to the bacterial OmpA protein was constructed and transformed into the Salmonella ${\Delta}msbB$ mutant. In a lethality test, BALB/c mice injected intraperitoneally with the Salmonella ${\Delta}msbB$ mutant survived for 7 days, whereas mice injected intraperitoneally with the wild type survived for 3 days. Moreover, all mice inoculated orally with the ${\Delta}msbB$ mutant survived for 30 days, but 80% of mice inoculated orally with the wild type survived. The OmpA::CPV VP2 epitope fusion protein was expressed successfully and associated with the outer membrane and OMV fractions from the mutant S. Typhimurium transformed with the fusion protein-expressing vector. In immunogenicity tests, sera obtained from the mice immunized with either the Salmonella msbB mutant or its OMVs containing the OmpA::CPV VP2 epitope showed bactericidal activities against wild-type S. Typhimurium and contained specific antibodies to the CPV VP2 epitope. In the hemagglutination inhibition (HI) assay as a measurement of CPV-neutralizing activity in the immune sera, there was an 8-fold increase of HI titer in the OMV-immunized group compared with the control. These results suggested that the CPV-neutralizing antibody response was raised by immunization with OMV containing the OmpA::CPV VP2 epitope, as well as the protective immune response against S. Typhimurium in BALB/c mice.

  • Digital Radiography (DR) has improved a quality of resolution based on a wide dynamic range, high detective quantum efficiency (DQE), and modulation transfer function (MTF), compared with film/screen(F/s). Unlike expectation that a low level of radiation can be used in examination, high level of signal to noise ratio(SNR) due to over-exposure caused increase of exposed dose to patients. Also, the auto exposure control (AEC) using Kilovolage(kVp) in F/S can cause over-exposure. Hence, in this study, we proposed a proper method for using DR, in which effect of tubing Kilovolage on device's image, DR MTF measurement with changes of tubing current (mA), and the quantitative evaluation of skull phantom captured images' PSNR were evaluated. Changes of contrast with tubing Kilovolage can be improved by retouching, and MTF changes according to tubing current(1.41~1.39 lp/mm in 50% area, and 3.19~2.8 lp/mm in 10% area) does not influence on resolution of image. As a result, high tubing Kilovoltage, and tubing current will be suitable to use of DR.

APPLICATION OF A DUAL-ENERGY MONOCHROMATIC XRAY CT ALGORITHM TO POLYCHROMATIC X-RAY CT: A FEASIBILITY STUDY

  • In this study, a simple post-reconstruction dual-energy computed tomography (CT) method is proposed. A dual-energy CT algorithm for monochromatic x-rays was adopted and applied to the dual-energy CT of polychromatic x-rays by assigning a representative mono-energy. The accuracy of algorithm implementation was tested with mathematical phantoms. To test the sensitivity of this algorithm to the inaccuracy of representative energy value in energy values, a simulation study was performed with mathematical phantom. To represent a polychromatic x-ray energy spectrum with a single-energy, mean energy and equivalent energy were used, and the results were compared. The feasibility of the proposed method was experimentally tested with two different micro-CTs and a test phantom made of polymethyl methacrylate (PMMA), water, and graphite. The dual-energy calculations were carried out with CT images of all possible energy pairs among 40, 50, 60, 70, and 80 kVp. The effective atomic number and the electron density values obtained from the proposed method were compared with theoretical values. The results showed that, except the errors in the effective atomic number of graphite, most of the errors were less than 10 % for both CT scanners, and for the combination of 60 kVp and 70 kVp, errors less than 6.0 % could be achieved with a Polaris 90 CT. The proposed method shows simplicity of calibration, demonstrating its practicality and feasibility for use with a general polychromatic CT.

The Evaluation of Dose Reduction and Quality of Images According to 80 kVp of Scan Mode Change in Pediatric Chest CT (소아 흉부 CT 검사에서 관전압 80 kVp 조건으로 스캔 모드별 방사선량 감소와 화질 평가)

  • To evaluate the usefulness of pediatric chest CT scans by comparing the dose, examination time, and image quality by applying Helical mode, High-pitch mode, and Volume Axial mode to minimize the radiation exposure and obtain high diagnostic value. Revolution (GE Healthcare, Wisconsin USA) was used to divide PBU-70 phantom into Helical mode, High-pitch mode, and Volume Axial mode. After acquiring images, ROI is set for each image, heart, bone, lung, and back-ground air, and the average value is obtained by measuring CT number (HU) and noise (SD). SNR and CNR were measured and compared with DLP values provided directly by the equipment. Determining statistical significance Statistical analysis was performed using ONE-WAY-ANAOVA using SPSS 21.0. In this experiment, it was possible to inspect at a short time without deterioration of image quality with the lowest dose when using volume axial mode. Although the detector coverage of 16 cm is limited to all pediatric chest CT scans, it is recommended to be actively used in pediatric patients, and further study is needed to apply other test sites in volume axial mode.

Effects of Iterative Reconstruction Algorithm, Automatic Exposure Control on Image Quality, and Radiation Dose: Phantom Experiments with Coronary CT Angiography Protocols (반복적 재구성 알고리즘과 관전류 자동 노출 조정 기법의 CT 영상 화질과 선량에 미치는 영향: 관상동맥 CT 조영 영상 프로토콜 기반의 팬텀 실험)

  • In this study, we investigated the effects of an iterative reconstruction algorithm and an automatic exposure control (AEC) technique on image quality and radiation dose through phantom experiments with coronary computed tomography (CT) angiography protocols. We scanned the AAPM CT performance phantom using 320 multi-detector-row CT. At the tube voltages of 80, 100, and 120 kVp, the scanning was repeated with two settings of the AEC technique, i.e., with the target standard deviations (SD) values of 33 (the higher tube current) and 44 (the lower tube current). The scanned projection data were reconstructed also in two ways, with the filtered back projection (FBP) and with the iterative reconstruction technique (AIDR-3D). The image quality was evaluated quantitatively with the noise standard deviation, modulation transfer function, and the contrast to noise ratio (CNR). More specifically, we analyzed the influences of selection of a tube voltage and a reconstruction algorithm on tube current modulation and consequently on radiation dose. Reduction of image noise by the iterative reconstruction algorithm compared with the FBP was revealed eminently, especially with the lower tube current protocols, i.e., it was decreased by 46% and 38%, when the AEC was established with the lower dose (the target SD=44) and the higher dose (the target SD=33), respectively. As a side effect of iterative reconstruction, the spatial resolution was decreased by a degree that could not mar the remarkable gains in terms of noise reduction. Consequently, if coronary CT angiogprahy is scanned and reconstructed using both the automatic exposure control and iterative reconstruction techniques, it is anticipated that, in comparison with a conventional acquisition method, image noise can be reduced significantly with slight decrease in spatial resolution, implying clinical advantages of radiation dose reduction, still being faithful to the ALARA principle.

Stationary and Moving Computed Radiography Grids : Comparative Observer's Perception (Computed Radiography에서 고정형 그리드와 이동형 그리드 영상의 인식률 비교)

  • This study assessed the degradation of image quality caused by grid artifacts and $moir{\acute{e}}$ pattern artifacts in a stationary grid, and the degradation of image quality caused by cut off artifacts in a moving grid. X-ray images were acquired in a stationary grid and a moving grid with X-ray exposure conditions of 100 cm, 80 kVp, and 30 mA using a CDRAD phantom and a 24 cm thickness acrylic phantom. Observer's perception of X-ray imaging using CDRAD Analyzer was mean 49.36, standard deviation 3.76, maximum 55.56, and minimum 38.67 in the stationary grid, and 47.04, 12.69, 55.56, and 20.89, respectively, in the moving grid. The stationary grid was superior to the moving grid in terms of the mean and standard deviation of observer's perception.

Determination of the Effective Energy of X-Ray Beam Using Optically Stimulated Luminescent nanoDot Dosimeters (광자극형광나노닷선량계를 사용한 X선 빔의 유효에너지 결정)

  • The purpose of this study is to determine the effective energy of a polyenegetic X-ray beam. The half value layer(HVL) of aluminum for 80 kVp X-ray beam was measured by using optically stimulated luminescent nanoDot dosimeters(OSLnDs). The linear attenuation coefficient(${\mu}$) was calculated using the measured HVL. And the mass attenuation coefficient(${\mu}/{\rho}$) was obtained by dividing the linear attenuation coefficient by the density(${\rho}$) of aluminum. The effective energy($E_{eff}$) of the obtained mass attenuation coefficient was determined using data of the X-ray mass attenuation coefficients for photon energies of aluminum given by National Institute of Standards and Technology(NIST). As a result, the HVL value is 2.262 mmAl. The ${\mu}$ value is $3.06cm^{-1}$. The ${\mu}/{\rho}$ value is $1.114cm^2/g$. And the $E_{eff}$ value was determined at 29.79 keV.

Leakage and Scattered Radiation from X-ray Unit in Radiography (영상의학과 엑스선 발생장치의 누설 및 산란선량 측정)

  • The purposed of this study were measured the radiation exposure of patients and workers by generators, and the protection state for radiation facilities. The subject of the study by X-ray generators in university hospitals of capital area, we measured the maximum irradiation condition of 80 kVp, 200 mA, 0.1 second in the control entrance, control room window, entrance of radiography, adjacent site. The leakage dose per week was which the control entrance was 0.11 mR/week, control room window was 0.15 mR/week, entrance of radiography was 0.12 mR/week and adjacent site was 0.06 mR/week with X-ray unit the mean And the leakage mean dose was 0.11 mR/week. Diagnostic X-ray tubes must ensure that the leakage radiation in the maximum leakage dose in week emitted by the tube outside the useful beam does not exceed certain levels provided by standards.

Analysis of CT Image Quality Change according to Clinical Application Shielding Materials (임상 적용 차폐물질에 따른 선량 및 CT 화질 변화 분석)

  • Among brain CT scan conditions including the lens, the tube voltage was changed to 80, 100, and 120 kVp and applied. The change in dose was analyzed using lead, lead goggles and barium sulfate silicon shielding materials, and the degree of influence of the shielding materials on image quality was compared and analyzed by applying the SNR, CNR, and SSIM index analysis methods. As a result, it was analyzed that although the dose was reduced by applying all shielding materials, the difference in dose reduction was not large (P > 0.05). In addition, as for the change in image quality due to the application of the shielding material, SNR and CNR were the highest when lead goggles were applied, and the structural similarity was measured to be the best as it was closest to the reference value of 1 in SSIM analysis. Therefore, based on the results of this study, it is thought that if more diverse shielding materials and clinical test results are derived and applied, it will be helpful for the clinical application criteria in the case of shielding utilization inspection.

The Assessment of Scattered Ray According to the Beam Thickness of Z-axis in MDCT(Multi Detector Computed Tomography) (MDCT(다배열검출기 전산화단층촬영장치)에서 Z-축의 빔 두께에 따른 산란선의 평가)

  • The purpose of this study is to measure scattered ray which is occurred except for Z-axis range of the detector in MDCT's iso-center and present the basic data about the standard for reduction of scattered ray. The development of MDCT brings out the enlargement of beam thickness to the patient's Z-axis, which distributes to the increase in exposure dose according to the rise of scattered ray. Also MDCT brings out the increase of scattered ray about 4times more than SDCT. To evaluate scattered ray according to the change of beam thickness on MDCT, we measured scattered ray of MDCT's Z-axis beam thickness by using one 16-slice CTs and two 64-slice CTs. We used the ionization chamber 60ml 2026C as the equipment of measurement. In our results, we found out that the change of scattered ray according to the beam thickness in the same kVp has increase of scattered ray. Secondly we found out the increase of scattered ray according to the increase of kVp. Lastly we found out the decrease of scattered ray according to the increase of the distance from the ionization chamber.
