T-cell Mediated Immunity in Pulmonary and Extrapulmonary Tuberculosis (폐 및 폐외결핵환자에서의 T 림프구 매개성 면역기능의 변화에 관한 연구)

  • Background: T-cell mediated cellular immunity has been suggested as an important mechanism in mycobacterial infection and imbalance between helper/inducer and suppressor/cytotoxic T-cell has been suggested as an important immunological abnormality in the pathogenesis of tuberculosis in human. Method: To determine whether there is any difference in T-cell mediated immunity in the pathogenesis of pulmonary and extra pulmonary tuberculosis, total numbers of WBC&lymphocytes were counted and helper/inducer and suppressor/cytotoxic cells were calculated by flow cytometry. Blastogenesis after stimulation with Concanavalin-A, Phytohemagglutinin and PPD were measured by $^3H$-thymidine uptake. PPD skin test was performed as an in vivo test. Results: 1)There was no significant difference in the size of PPD skin test between pulmonary and extrapulmonary tuberculosis groups. 2)Number of total lymphocytes significantly decreased in tuberculosis patients compared with healthy control group. But there was no significant difference between pulmonary and extrapulmonary tuberculosis groups. 3) Number of HLA-DR and Interleukin-2 receptor (+) cells were significantly increased in tuberculosis patients. But there was no significant difference between pulmonary and extra pulmonary tuberculosis groups. 4) There was no significant difference in the numbers of WBC, $T_3$, $T_4$ and $T_8$ lymphocytes and $T_4/T_8$ ratio between tuberculosis patients and healthy controls. 5) There was no significant difference in the blastogenesis after stimulation with specific and non-specific blastogens between tuberculosis patients and healthy controls. 6) The percentage and absolute number of $T_4$ lymphocyte were significantly correlated with the size of PPD skin test. (r=0.689 and 0.598). Conclusion: From these results, it is concluded that there was no difference in T-cell mediated immunity between pulmonary and extra pulmonary tuberculosis group. But, because it is suspected that there might be some difference in the role of T-cell mediated immunity in the pathogenesis of pulmonary and extra pulmonary tuberculosis or even among the extrapulmonary tuberculosis patients, further studies would be required.

The Characteristics of 7-zone-diagnosis on Postpartum Depression (산후우울군과 비우울군의 7구역 진단기 특성연구)

  • Objectives: Since early developed measurement of Postpartum depression(PPD) doesn't reflect physical symptoms, we need to find out how PPD and related physical disorders are expressed in functional medical methods. The study was conducted to find out characteristics of 7 Zone diagnosis and postpartum physical symptoms on PPD. Methods: To 216 postpartum women who did normal delivery, we measured 7-zone-diagnosis Factor AA's height on 5th day and 15th day postpartum. And we surveyed EPDS and postpartum physical symptoms question on 15th day postpartum. Subjects were divided into normal and depression group by EPDS score. And we compared Factor AA's height between two groups and Factor AA's height change in each group. Results: In depression group, Factor AA's height was higher than normal group on 5th day and 15th day postpartum. In both groups Factor AA's height was decreased on 15th day compared to 5th day postpartum. In normal group 2, 6, 7 zone were significantly decreased and in depression group only 6 zone was significantly decreased. Conclusions: These results demonstrate that postpartum depression group has low recuperative power. And these can be judged by Factor AA's height change.

The Activity and Utilization of Urease Inhibitors (요소분해효소 억제물질의 작용과 응용에 관한 연구)

  • Urea, the major N source of world agriculture involves a serious urea-N loss through NH$_3$volatilization. Approaches to decrease N loss include using urease inhibitors in view of the environmental protection and the increase of urea-N efficiency. The purpose of laboratory researches was toassess the potential value of urease inhibitors to increase urea-N efficiency in soil and Kentucky blue-grass(Poa Pratensis L.) turf. The activity of urease inhibitors Phenyiphosphorodiamjdate(ppD) and N-(n-butyl) thiophosphoric triamjde(NBPT) measured to break-down ammonia volatilization. The soil and turf used in this project were from the fairway in one of the Korean gof course. The researches were carried out for two weeks to measure the urease activities on urea hydrolysis under four temperatures (10~ 40$^{\circ}C$) and for one week on turfgrass using forced-draft system. Results indicated that Urea-N involves considerable loss through gaseous NH$_3$ by urease activities in plant-soil systems. Urease inhibitors PPD and NBPT have potential value for increasing N use efficiency by reduing NH$_3$ volatilization. NBPT deserves futher evaluation as fertilizer amendment than PPD use of urea in turf industries.

  • This study analyze indoor thermal comfort properties such as PMV, PPD and fluid variables when the cooling loads are light, occupant and ventilation. There are 6 cases to study for the indoor unit installation conditions and ventilation rates. Numerical method is used to study the indoor environmental properties and experimental study is adapted to analyze reaching time by variable cooling load conditions.

Effects of Ginsenoside Metabolites on GABAA Receptor-Mediated Ion Currents

  • In a previous report, we demonstrated that ginsenoside Rc, one of major ginsenosides from Panax ginseng, enhances ${\gamma}$-aminobutyric acid (GABA) $receptor_A$ ($GABA_A$)-mediated ion channel currents. However, little is known about the effects of ginsenoside metabolites on $GABA_A$ receptor channel activity. The present study investigated the effects of ginsenoside metabolites on human recombinant $GABA_A$ receptor (${\alpha}_1{\beta}_1{\gamma}_{2s}$) channel activity expressed in Xenopus oocytes using a two-electrode voltage clamp technique. M4, a metabolite of protopanaxatriol ginsenosides, more potently inhibited the GABA-induced inward peak current ($I_{GABA}$) than protopanaxadiol (PPD), a metabolite of PPD ginsenosides. The effect of M4 and PPD on $I_{GABA}$ was both concentration-dependent and reversible. The half-inhibitory concentration ($IC_{50}$) values of M4 and PPD were 17.1${\pm}$2.2 and 23.1${\pm}$8.6 ${\mu}M$, respectively. The inhibition of $I_{GABA}$ by M4 and PPD was voltage-independent and non-competitive. This study implies that the regulation of $GABA_A$ receptor channel activity by ginsenoside metabolites differs from that of ginsenosides.

A Qualitive Research of N2, O2 Permeation Property in PMDA/ MDA- Phenylene Diamine Copolyimide (PMDA/MDA-Phenylene diamine 계열의 공중합체막에서의 산소, 질소 투과 특성의 정성적 고찰)

  • Copolyimide membranes of different chemical structure based on pyromellitic dianhydride (PMDA)/methylendianiline(MDA) were prepared by varying their chemical compositions with adding meta-phenylendiamine (MPD), para- phenylendiamine (PPD), 2,4,6- trimethyl-1,3-phenylenediamine(TriMeMPD) as a co-monomer. The $N_2$ and $O_2$ permeation properties are qualitatively correlated to specific free volume and intersegmental distance of membrane. The partial replacement of MDA with MPD or PPD caused in the PMDA/MDA based membranes increase in density, and decrease in free volume, d-spacing, consequently resulted in decreased permeability coefficient. In the case of TriMeMPD, opposite results were observed. In all membranes, the permeability coefficients were pressure independent, and membranes which have high permeability coefficient showed low $N_2/O_2$ ideal separation factor as an usual manner. The permeability coefficient also increased with temperature and $N_2/O_2$ ideal separation factor decreased. The increase ratio of the $N_2$ permeability coefficient was larger than that of $O_2$.

A Study on the Pore Pressure Dissipation Test of the Piezocone (피에오콘의 간극수압 소산시험에 관한 연구)

  • A degree of consolidation at any time can be evaluated by using cone penetration test after soil improvement. In this case, after stopping the penetration of a piezocone, pore pressure dissipation(PPD) best is carried out until the pore pressure remains constant. Since the hydraulic conductivity of soft ground is very small, it takes very long time to finish the PPD test. This research is performed to develop a method overcoming this problem of the PPD test and reducing the test time. The analyses are carried out in the following ways : an equilibrium pore pressure can be determined by using pore pressure measured in the middle of the test, which is predicted by hyperbolic, Asaoka and Hoshino methods. And this equilibrium pore pressure is compared with the one measured in a test of long duration. As a result of the study, it is found that Hoshino method is the best way to predict the equilibrium pore pressure in a teat of short duration. And it is proposed as a methodology to fond a minimal time in which we can get an equilibrium pore pressure.

Biosynthesis of rare 20(R)-protopanaxadiol/protopanaxatriol type ginsenosides through Escherichia coli engineered with uridine diphosphate glycosyltransferase genes

  • Background: Ginsenosides are known as the principal pharmacological active constituents in Panax medicinal plants such as Asian ginseng, American ginseng, and Notoginseng. Some ginsenosides, especially the 20(R) isomers, are found in trace amounts in natural sources and are difficult to chemically synthesize. The present study provides an approach to produce such trace ginsenosides applying biotransformation through Escherichia coli modified with relevant genes. Methods: Seven uridine diphosphate glycosyltransferase (UGT) genes originating from Panax notoginseng, Medicago sativa, and Bacillus subtilis were synthesized or cloned and constructed into pETM6, an ePathBrick vector, which were then introduced into E. coli BL21star (DE3) separately. 20(R)-Protopanaxadiol (PPD), 20(R)-protopanaxatriol (PPT), and 20(R)-type ginsenosides were used as substrates for biotransformation with recombinant E. coli modified with those UGT genes. Results: E. coli engineered with $GT95^{syn}$ selectively transfers a glucose moiety to the C20 hydroxyl of 20(R)-PPD and 20(R)-PPT to produce 20(R)-CK and 20(R)-F1, respectively. GTK1- and GTC1-modified E. coli glycosylated the C3-OH of 20(R)-PPD to form 20(R)-Rh2. Moreover, E. coli containing $p2GT95^{syn}K1$, a recreated two-step glycosylation pathway via the ePathBrich, implemented the successive glycosylation at C20-OH and C3-OH of 20(R)-PPD and yielded 20(R)-F2 in the biotransformation broth. Conclusion: This study demonstrates that rare 20(R)-ginsenosides can be produced through E. coli engineered with UTG genes.

Determination of THM(trihalomethane) in Rain by using Solid Phase Micro-Elctraction(SPME) Fiber Assembly (고상 미량 추출 장치(SPME, solid phase micro-extraction device)를 이용한 물 중의 THM(trihalomethane) 분석)

  • SPME deuce was applied to determine the THM in an aqueous solution. The 6 kinds of THM was quantitatively detenuned by using GC-ECD which has the sample eutracted on the SPME fiber from an aqueous solution for 10 min. The THM components were well separated from $CHCl_3$ to the last $CHBr_3$ UHh 13 mons at the condition. 6 kinds of the volatile halogenated organic compounds: $CHCl_3$, $CHBrCl_2$, $CHBrtCl_2$, $CHCl_3$, $C_2Cl$. and $CHBr_3$, showed well defirled calibration graph with good llnearlty from a few ppb level up to several tens of pub concentration. $CHBr_2Cl$ and $C_2C1_4$ were detected from a few samples among the 10 of river samples. CHCl3, however, was detected In 4 sea water samples with the highest of 10 ppd among the pouuted 6 positions. Trace level of $CHBr_2Cl$ and few pub level of $CHBr_3$ were also detected at the other two sample stations. Most of the 13 rain water samples collected from 6 sampling stations were contained ppd level of $CHCl_3$, and also $CHBr_2Cl$, and C_2Cl_4$ were only detected at trace level at a few rain samples among them. We could recognize the fact that our Ut and water enoronment has already been contaminated by certain volatile halogenated organic compounds through this study.

The effect of flap operation and metronidazole gel combined therapy on the treatment of the juvenile periodontitis (치은박리소파술시 metronidazole gel 병용이 유년성치주염 치료에 미치는 영향)

  • The aim of the study was to investigate the effect of surgical therapy combined with the usage of metronidazole gel in the treatment of juvenile periodontitis by comparing clinical indices of flap operation along with application of metronidazole gel and flap operation only. Comparing clinical indices of the baseline, 3 months after surgery, 6 months after surgery statistically, the results are as follows; 1. Bleeding on probing (BOP) was decreased significantly both in the experimental group and the control group at 3 months after surgery (p<0.05). BOP increased 0.9% in the control group 6 months after surgery, while in the experimental group, decreased 4.7% (p>0.05). However, there was no statistically significant difference in these groups (p>0.05). 2. Pocket probing depth (PPD) was decreased significantly both in the experimental group and the control group at 3 months after surgery (p<0.05). PPD increased 0.lmm in the control group 6 months after surgery, while in the experimental group, no increase of the depth could be observed (p>0.05). However, there was no statistically significant and difference in these groups (p>0.05). 3. Loss of attachment level(LOA) was decreased significantly both in the experimental group and the control group at 3 months after surgery (p<0.05). LOA increased 0.2mm in the control group 6 months after surgery, while in the experimental group, increased 0.3mm However, there was no statistically significant difference in these groups (p>0.05). In conclusion, flap operation was effective on the treatment of juvenile periodontitis. However, combined therapy of metronidazole gel could not give rise to any significant adjunctive effect on the treatment outcome.
