Characterization of CTL Clones Specific for Single Antigen, H60 Minor Histocompatibility Antigen

  • Background: Disparities of Minor H antigens can induce graft rejection after MHC-matched transplantation. H60 has been characterized as a dominant antigen expressed on hematopoietic cells and considered to be an ideal model antigen for study on graft-versus-leukemia effect. Methods: Splenocytes from C57BL/6 mice immunized with H60 congenic splenocytes were used for establishment of H60-specific CTL clones. Then the clones were characterized for proliferation capacity and cytotoxicity after stimulation with H60. Clone #14, #15, and #23 were tested for the TCR binding avidity to H60-peptide/$H-2K^b$ and analyzed for TCR sequences. Results: H60-specific CTL clones showed different levels of proliferation capacity and cytotoxic activity to H60-stimulation. Clones #14, #15, and #23 showed high proliferation activity, high cytotoxicity, and low activities on both aspects, respectively, and have TCRs with different binding avidities to H60-peptide/$H-2K^b$ with $t_{1/2}$ values of 4.87, 6.92, and 13.03 minutes, respectively. The TCR usages were $V{\alpha}12D-3-01+J{\alpha}11-01$ and $V{\beta}12-1-01+D{\beta}1-01+J2-7-01$ for clone #14, $V{\alpha}13D-1-02+J{\alpha}34-02$ and $V{\beta}13-1-02+D{\beta}2-01+J{\beta}2-7-01$ for clone #15, and $V{\alpha}16D+J{\alpha}45-01$ and $V{\beta}12-1-01+D{\beta}1-01+J{\beta}2-5-01$ for clone #23. Conclusion: The results will be useful for modeling GVL and generation TCR transgenic mouse.

  • 본 논문에서는 BPhen(Bathophenanthroline)과 BPhen에 Cs가 도핑 된 interlayer를 $C_{60}$활성층과 Al전극사이에 주입해 $C_{60}$을 기반으로 한 유기박막트랜지스터 (OTFTs)를 제작하여 전기적 특성을 향상시켰다. BPhen층을 증착하면 유기물과 금속층계면의 표면 거칠기가 낮아져 결과적으로 성능이 향상되는 것을 알 수 있었다. 또한 BPhen에 Cs가 도핑 된 interlayer를 co-evaporation을 이용하여 주입하였을 경우는 Contact resistance가 감소하였다. 이러한 $C_{60}$을 기반으로 한 BPhen에 Cs가 도핑 된 interlayer를 삽입한 유기박막트랜지스터는 향후 n형 유기박막트랜지스터를 제작하는데 있어서 적용이 될 것이라고 기대된다.

2-Chloromethyl-1-Dihydroxyphosphinylpyrrolidine (2C-1DPP)에 의한 백혈병 세포주 HL-60의 분화유도 효과 (Effect of 2-Chloromethyl-1-Dihydroxyphosphinylpyrrolidine(2C-1DPP) on Differentiation Induction of Human Leukemia HL-60 Cells)

  • We have examined the induction of HL-60 cell differentiation by treatment of 2-chloromethyl-1-dihydroxyphosphinyl pyrrolidine(2C-1DPP), which is derivative of piperidine and pyrrolidine by ${\alpha}-phosphoramidoakylation$ reaction. It was observed that HL-60 cell proliferation was dose- and time-dependently inhibited by treatment with 2C-1DPP. 2C-1DPP treatment caused a significant change in NBT reduction and enhanced ATRA-induced NBT reduction. Treatment of 2C-1DPP to HL-60 cells increased only CD11b expression in the cells, and also increased markedly G0/G1 stage arrest of HL-60 cells. These results can suggest that 2C-1DPP induced the differentiation of HL-60 cells to granulocytes lineage and enhanced ATRA-induced differentiation. Moreover, DNA expression levels of p27 were up-regulated during 2C-1DPP-dependent HL-60 cell differentiation. Our results suggest that 2C-1DPP have potential as a therapeutic agent in human leukemia.

  • 본 연구에서는 무설탕 무지방의 저칼로리 케익에서 다량으로 첨가되는 유화제의 양을 감소시키기 위하여, 다양한 종류의 유화제를 1.5온 혹은 15% 첨가수준에서 가수전처리 하거나, 전처리 하지 않은 경우가 저칼로리 케익에 미치는 영향을 조사하였다. 물리 화학적인 특성 및 관능검사에 사용된 유화제는 Vanall$^{R}$, sorbitan me-nostearate, polysorbate 60, distilled monoglyceride, su-crose ester F10, SE F7O 및 SE F160이다. 반죽에서 낮은 비중을 나타내고 안정된 분산 체계를 유지하여 케익 volume이 커진 경우는 고수준으로 전처리된 SMS, MG, SE Fl60 및 F70가 첨가된 케익군과 저수준으로 전처리된 SE Fl6O, F7O 및 PS 60가 첨가된 케익군이다. 전처리하여 저수준으로 SE Fl60, F70 및 PS 60을 첨가하여 케익을 만들 경우 부드러운 케익이 되며, 고수준으로 전처리하지 않고 VanallR 을 첨가한 케익과 비슷한 부피를 지닌 케익을 만들었다.

다중 섬 유전자 알고리즘 기반 A60 급 격벽 관통 관의 방화설계에 대한 이산변수 근사최적화 (Approximate Optimization with Discrete Variables of Fire Resistance Design of A60 Class Bulkhead Penetration Piece Based on Multi-island Genetic Algorithm)

  • A60 class bulkhead penetration piece is a fire resistance system installed on a bulkhead compartment to protect lives and to prevent flame diffusion in a fire accident on a ship and offshore plant. This study focuses on the approximate optimization of the fire resistance design of the A60 class bulkhead penetration piece using a multi-island genetic algorithm. Transient heat transfer analysis was performed to evaluate the fire resistance design of the A60 class bulkhead penetration piece. For approximate optimization, the bulkhead penetration piece length, diameter, material type, and insulation density were considered discrete design variables; moreover, temperature, cost, and productivity were considered constraint functions. The approximate optimum design problem based on the meta-model was formulated by determining the discrete design variables by minimizing the weight of the A60 class bulkhead penetration piece subject to the constraint functions. The meta-models used for the approximate optimization were the Kriging model, response surface method, and radial basis function-based neural network. The results from the approximate optimization were compared to the actual results of the analysis to determine approximate accuracy. We conclude that the radial basis function-based neural network among the meta-models used in the approximate optimization generates the most accurate optimum design results for the fire resistance design of the A60 class bulkhead penetration piece.

전환장애 증상 중 우울감 감소를 위한 통합심리학적 치료 프로그램 개발 및 효과성 검증: 60대 노년 여성 대상 (The Development and Effectiveness Verification of Integrative Psychotherapy Program for Relieving Depression among the Symptoms of Conversion Disorder : For Elderly Women in their 60s)

  • 본 연구는 전환장애 증상 중 우울감을 경험하는 60대 여성 22명을 대상으로 2021년 1월26일부터 2월9일까지 실험집단 대상의 연구를 시행하였다. 60대 노년여성의 우울감 감소와 신체증상 완화를 위해 통합심리학적 치료 프로그램인 집단상담 프로그램을 개발하고, 연구 대상자들의 사전-사후 변화를 비교 분석하여 효과성을 검증하고자 하였다. 이를 위하여 22명의 60대 여성을 대상으로 한국형 노인 우울 검사(K-GDS), 증상해석 질문지(SIQ), Hamilton 우울 평가척도 설문지, PHQ-9 및 대인관계 척도(RCS)를 실시하였다. 이후 사전-사후 대응 표본 t 검정, 상관분석과 다중 회귀 분석을 수행하였다. 연구 결과, 본 연구가 개발한 집단상담 프로그램을 통하여 60대 여성의 우울감 감소, 신체증상 완화, 부정적 정서 및 부정적 신념 감소, 대인관계에 긍정적인 변화가 나타나 그 효과를 실증적으로 입증하였다. 마지막으로, 본 연구의 의의와 한계를 논의하였다.

Synthesis of SnO2-Mn-C60 Nanocomposites and Their Photocatalytic Activity for Degradation of Organic Dyes

  • Nanocomposites based on $SnO_2-Mn$ were synthesized by the reaction of tin (II) chloride dihydrate and manganese (II) chloride tetrahydrate at a molar ratio of 10:1 in the presence of ammonium hydroxide at $80^{\circ}C$. The $SnO_2-Mn$ nanocomposites were stirred with fullerene [$C_{60}$] in a mass ratio of 2:1 in tetrahydrofuran to prepare $SnO_2-Mn-C_{60}$ nanocomposites; these nanocomposites were obtained upon heating the mixture of $SnO_2-Mn$ nanocomposites and fullerene [$C_{60}$] in an electric furnace at $700^{\circ}C$ for 2 h. The synthesized $SnO_2-Mn-C_{60}$ nanocomposites were confirmed through various characterization methods such as X-ray diffraction and scanning electron microscopy. The photocatalytic activities of the $SnO_2-Mn-C_{60}$ nanocomposites were demonstrated by the degradation of the organic dyes BG, MB, MO, and RhB under 254 nm irradiation and evaluated using UV-Vis spectrophotometry.

정상사람림프구와 HL-60 cell에서 목향의 세포독성과 백혈병세포 분화효과에 관한 연구 (Cytotoxicity in HL-60 cells and human lymphocytes and effect of leukemia cell differentiation induced by Saussureae Radix extract)

  • Objectives : This study was focused to investigate the toxicity of Saussurea lappa (SL) extracts in HL-60 cells and human lymphocytes. We also examined the differentiation effect of SL against leukemia cells. Methods : For examining the toxicity of SL, cytokinesis-block micronucleus (CBMN) assay and single cell gel eletrophoresis (SCGE) assay were used in present study. The cell differentiation effect of SL was evaluated by nitroblue tetrazolium (NBT) reduction assay. Results : The inhibition of cell growth in HL-60 cells was observed in a dose-dependant manner after SL treatment for 24 h. According to SCGE assay, HL-60 cells treated with SL increased DNA damage at $10{\mu}g/m{\ell}$, while DNA damage was induced by 0.1, 1, $10{\mu}g/m{\ell}$ concentration of SL in human lymphocytes. Our results indicated that SL have no genotoxic effect in HL-60 cells and human lymphocytes. Additionally, the differentiation effect was induced in $1{\mu}g/m{\ell}$ SL-treated HL-60 cells. Conclusions : From above results it is suggested that SL could be beneficial for the preparation of the useful agent for treating leukemia.

  • [ $C_{60}$ ] molecule, the Buckminsterfullerene, has generated great interest because of its unique molecular structure and of superconductivity exhibited in its alkali-doped solids. We have investigated the molecular stacking and crystal structure of $C_{60}$ thin crystals formed on amorphous carbon film. The $C_{60}$ powder which was chromatographically purified was dissolved in benzene. The thin crystals of $C_{60}$ were observed with a 300 keV transmission electron microscope. Electron diffraction analysis and direct imaging of its molecular stacking were carried out. It was found from this work that the molecules of $10.0{\AA}$ diameter are arrayed hexagonally on substrate surface and $8.7{\AA}$ lattice planes are quite often found in several types of ED patterns, which can never be explained with a fcc model. Therefore the structure of $C_{60}$ thin crystals is hcp, although we cannot fully exclude the possibility of co-existence of hcp and fcc.

인체 혈액암세포주(HL-60)에서 (-)-epigallocatechin-3-gallate에 의한 Aapoptosis 유도 (Induction of Apoptosis by (-)-epigallocatechin-3-gallate in HL-60 Cells)

  • (-)-Epigallocatechin-3-gallate (EGCG) is a polyphenolic compound found in peen tea leaves, and has been known to be one of the most potent catechin species which inhibits cell growth most possibly through an apoptotic cell death. We investigated the apoptotic activity of (-)-EGCG on the human myeloid leukemia cell line, HL-60. Our results of MTT test indicated that (-)-EGCG had a significant antiproliferation effect in HL-60 cells with $IC_{50}$/ (50% inhibition concentration) value of 65 $\mu$M. Giemsa statining of HL-60 cells treated with (-)-EGCG (100 $\mu$M) for 6hrs showed a typical apoptosis-specific morphological change including shrinkage of the cytoplasm, membrane blobbing and compaction of the nuclear chromatin. The DNA fragmentation was observed from the agarose gel electrophoresis of cells treated with (-)-EGCG for 3hrs or longer, and was progressed to a greater degree as treatment time increases. Treatment of the cells with (-)-EGCG (100 $\mu$M) resulted in a rapid release of mitochondrial cytochrome c into the cytosol, and a subsequent cleavage of caspase-3 to an active form in a treatment-time dependent manner. (-)-EGCG (100 $\mu$M) also stimulated proteolytic cleavage of poly-(ADP-ribose) polymerase (PARP) to an active form in HL-60 cells. Tlken together, (-)-EGCG appears to induce the apoptosis in human myeloid leukemia cells via a caspase-dependent pathway. These results suggest the possible application of (-)-EGCG, the major active compound in green tea, as an antiproliferative agent for cancer prevention.
