족제비고사리 전엽체의 기내배양 및 기외 포자체 형성에 미치는 제요인 (Several Factors Affecting on In vitro Culture of Prothallus and Ex Vitro Sporophyte Formation from Prothallus of Dyropteris varia (L.) O. Kuntze)

  • 본 연구는 족제비고사리의 대량번식법을 개발하기 위하여 전엽체의 기내배양 및 기외 포자체 형성의 적정 조건을 구하고자 실시되었다. 기내에서 전엽체 증식은 MS배지의 무기물 조건에서 가장 활발하였고, $NH_4^+:NO_3^-$의 농도비가 10:50mM이고, 2%의 sucrose가 첨가된 배지에서 생장이 향상되었다. 전엽체 생장에 적합한 배지의 pH는 5.8이었으며, 적정 agar 농도는 $0.6{\sim}0.8%$로 조사되었다. 배지에 첨가된 생장조절물질(NA, IAA, 2,4-D, BAP, kinetin 및 2ip)은 대부분 농도가 높아질수록 전엽체의 생장을 더욱 저해하는 것으로 나타났다. 기내에서 증식된 전엽체를 토양 별로 기외이식한 결과, 상토 단용구에서 가장 많은 포자체가 형성되었으며, $GA_3\;100{\mu}M$에서 3시간 동안 침지처리 한 전엽체로부터 포자체의 형성이 현저히 촉진되었다.

경남 일부 지역 직장인과 대학생의 아침식사 식습관에 대한 연구 (Study on Breakfast Habits of Workers and College Students in Gyeongnam Area)

  • This study was conducted to investigate and compare the breakfast habits of workers (189 males and 182 females) and college students (217 males and 225 females) living in Gyeongnam (Masan, Jinhae, Changwon) based upon gender in each group. Most college students were in their twenties and not married while workers were in their twenties, thirties, and forties. Age distribution was even and 58,0% of them were married. Exactly 77% of the subjects were middle class, and 35.1% answered that their monthly allowances were less than 200,000 won. Exactly 36.5% of the subjects had breakfast regularly, and workers had breakfast more regularly than college students (p<.05). Subjects chose lack of time as their biggest reason for skipping breakfast (41.7%), and workers better understood that having breakfast is important (p<.001). Exactly 62.7% of the subjects displayed symptoms of skipping breakfast, with college students more frequently susceptible than workers (p<.001). Subjects' most frequent symptom was decreased concentration (40.6%). Exactly 67.0% of the subjects ate bap, guk, and mitbanchan as breakfast; the ratio was higher for college students' compared to workers and female students compared to male students. When preparing breakfast, 35.3% of the subjects considered digestible food first. Exactly 54.7% of the subjects preferred bap for breakfast, and college students preferred it more than the workers. Subjects ate kimbap the most when eating breakfast outside of the home; 62.1% of them emphasized convenience and 36.7% answered 2,100-3,000 won is proper price.

켄터키 블루그래스의 종자유래의 캘러스로부터 식물체 재분화 (Plant Regeneration from Seed-derived Callus in Kentucky Bluegrass(Poa pratensis L.))

  • 켄터키 블루그래스의 종자배양에서 캘러스 형성 및 식물체 재분화 체계를 확립하기 위하여 배지조성 및 배양조건 등에 따른 배양효율의 차이를 'Kenblue'를 공시품종으로 사용하여 수행한 실험의 결과를 요약하면 다음과 같다. 성숙종자로부터 캘러스 유도를 위한 생장조절제로는 2 mg/L 2,4-D와 0.2 mg/L BAP 조합이 가장 효과적이었으며, 유도된 캘러스로부터 식물체 재분화를 위해서는 1 mg/L NAA와 5mg/L kinetin 조합이 가장 효과적이었다. 캘러스 유도시 암상태에서 배양하는 것이 약한 광 상태에서 배양하는 것보다 효과적이었으며, 캘러스 유도와 식물체 재분화에 있어 기본배지로 N6 배지나 B5 배지보다 MS 배지를 사용하는 것이 식물체 재분화에 효과적이었다. 배지 내에 첨가되는 탄소원으로는 maltose가 sucrose 보다 효과적이었다.

  • 이탈리안 라이그라수(Lolium muliflorum Lam.)의 원형질체 분리 및 생존성에 영향을 미치는 몇가지 요인들에 대하여 조사한 바 다음과 같은 결과를 얻었다. 성숙배. 미숙배 및 유종에 있어서 분화력이 높이 캘러스 유기는 4^{\circ}C.$에서 7일간 저온처리와 기본배지에 $AgNO_3$ 5mg/을 첨가한 것이 효과적이었다. 캘러스로부터의 재분화는 BAP 0.2mg/l와 2,4-D 2mg/l의 조합에서 가장 좋았다. 유종에서 유기되 현탁배양 세포를 액체배지에서 5일 간 진탕배양한 후 효소(4% cellulase R10, 2% macerozyme, 2% pectinase)와 삼투압 조절제인 0.6M mannitol을 첨가한 용액에서 생존율이 높은 원형질체를 얻었다. 그리고원형질체는 KM8P배지의 agarose 고체배지에서 헤포분열과 미세포괴가 이루어졌다.

서울지역 주민의 연령.성별에 따른 식사행동 및 식품인식도 비교 (Dietary behaviors and food perception of Koreans living in Seoul by age and gender)

  • Korean food habits, dietary behaviors and perception of food were compared according to gender and age. The subjects were composed of 274 males and 315 females. According to age, the age group of 10s was 26.3%, 20s 24.6%, 30s 16.8%, 40s 18.7%, and 50s and over 13.6%, respectively. The majority (65.2%) of all age groups except the 20s took meals 3 times a day and the ratio 20s eating twice a day was higher than other age groups. All age groups took cooked rice(bap) two or three times a day and the 20s group took smaller amount than other age groups did. The younger age groups of 10s and 20s took larger amount of processed food. The older age groups appeared to have higher frequency of eating fish, bean, vegetable, Kimchi, laver.brown seaweed, soybean oil, sesame oil, coffee, tea, and rice cake(ddhuk). The younger groups have the higher frequency of eating ramyeon, corn flake, milk, milk-processed food, butterㆍmargarine, ham.sausage, carbonated drink candyㆍchocolate, hamburger, and pizza. All age groups perceived cooked rice(bap), meat, fish, egg, vegetable, fruit, and Kimchi to be the delicious, excellent and healthy food. According to the above results, age groups of 30s and over had relatively better food habits compared with those of younger groups. On the other hand, the groups of 10s and 20s had meals irregularly, showed a high ratio of skipping meals, and took processed food much. These results raised a concern of their nutritional imbalance in the future. Therefore, the correct nutritional education must be conducted to make these age groups have a healthy food habits.

산업체 급식에서 제공되는 식사의 염분 함량에 관한 연구 (A Study on the Salt Contents in Foods Served by Institutional Foodservice Establishment)

  • This study was conducted to determine the salt-intake from meals that were served by an institutional foodservice establishment for a month. Meals (176) were classified into 12 food groups by CAN Pro 3.0. These consisted of bap 6, mandu 5, guk 29, jjigae 9, jjim 4, gui 8, jeon 5, bokeum 25, jorim 16, fried 7, namul 49, pickled 12. The average salt content of 100 g of each menu item was significantly higher in the order of pickled>jorim>bokeum>gui>namul>jeon>jjigae>jjim>fried. The average salt and sodium contents of 1 potion of menu item was significantly higher in the order of jjigae>mandu>guk>bap>jorim>pickled>bokeum>gui>fried>jjim>gimchi>namul>jeon. The average salt contents of kim-chijjigae (6.05 g), eggjorim (7.01 g), cooked namul (2.78 g) and salted pepperleaves (4.67 g) was higher than that of the other menu items served frequently in the establishment. The average intake of salt and sodium was 23.48 g (sodium 9,310 mg) a day, 7.35 g (sodium 2,960 mg) with breakfast, 7.55 g (sodium 2,990 mg) with lunch and 8.33 g (sodium 3,300 mg) with dinner. The daily salt intake was slightly higher than that of DRIs (Dietary Reference Intakes).

  • Kinetin(4.7$\times$10\ulcornerM) and 6-benzylaminopurine (2.2$\times$10\ulcornerM) were found to increase ca. 1.5-fold putrescice content in corn grown in medium containing kinetin and 6-benzylaminopurine(6-BAP) for 3days whereas kinetin was found to decrase ca. 30% spermidine and spermine, respectively. KCI (3$\times$10\ulcornerM) was found to decrease more than 50% putrescine content. After germination, ornithine decarboxylase activity was observed to increase constantly whereas arginine decarboxylase activity remained constant, suggesting involvement in putrescine biosynthesis. 6-benzylaminopurine was shown to increase more activities of arginine ornithine decarboxylase than kinetin when they were added to medium.

우리나라 외식 식품 중 밥류와 국(탕) 및 찌개류의 베타카로틴과 레티놀 함량 분석 연구 (β-Carotene and Retinol Contents in Bap, Guk (Tang) and Jjigae of Eat-out Korean Foods)

  • 본 연구는 국가 식품영양성분 자료구축을 위하여 국내에서 유통되는 외식식품의 베타카로틴과 레티놀의 함량을 조사하고자 하였다. 조사된 시료는 강원도, 경기도, 경상도, 서울, 전라도, 충청도의 6개 지역에서 수거한 밥류(26종)와 국(탕) 및 찌개류(21종)를 대상으로 진행하였다. 조사된 모든 시료의 베타카로틴의 함량은 레티놀의 함량에 비하여 높게 분석되었으며, 레티놀의 함량은 대부분의 시료에서 0.000 ${\mu}g/100g$에 가까운 값을 나타내었다. 밥류 중 조사된 8종의 김밥류의 베타카로틴 함량은 100.496~754.869 ${\mu}g/100g$의 수준에서 분석되었으며, 일반 김밥류에 비하여 삼각김밥류의 베타카로틴과 레티놀의 함량이 다소 낮게 나타났다. 볶음밥류의 베타카로틴 함량은 28.933~404.338 ${\mu}g/100g$의 수준에서, 레티놀 함량은 2.679~33.970 ${\mu}g/100g$의 수준에서 분석되었다. 초밥류의 경우 베타카로틴 함량이 0.000~41.234 ${\mu}g/100g$으로, 레티놀 함량이 0.000~31.783 ${\mu}g/100g$으로 나타나 조사된 밥류 중에서 전반적으로 낮은 범위의 베타카로틴과 레티놀 함량을 나타내었다. 비빔밥의 베타카로틴과 레티놀 함량은 각각 200.091~489.867과 7.271~23.880 ${\mu}g/100g$의 범위에서 나타났다. 한편 국(탕) 및 찌개류의 베타카로틴 함량은 추어탕에서 346.261~843.947 ${\mu}g/100g$으로 가장 높은 것으로 확인되었다. 찌개류의 경우 41.670~352.604 ${\mu}g/100g$의 범위에서, 국류는 7.886~211.932 ${\mu}g/100g$의 범위에서 분석되었다. 다음으로 경상도를 제외한 대부분의 지역에서 동태찌개에서의 레티놀 함량이 가장 높은 것으로 나타났으며, 전라도(26.446 ${\mu}g/100g$), 강원도(82.322 ${\mu}g/100g$), 경기도(131.708 ${\mu}g/100g$), 서울(182.480 ${\mu}g/100g$), 충청도(189.112 ${\mu}g/100g$)의 순으로 분석되었다. 도가니탕, 곰탕, 내장탕, 추어탕, 콩나물해장국에서는 레티놀이 검출되지 않았다.

An Efficient In vitro Propagation of Zanthoxylum piperitum DC.

  • A protocol is described for rapid multiplication of Zanthoxylum piperitum DC. (Rutaceae), an important aromatic and medicinal plant, through shoot-tip explant cultures. Murashige and Skoog (MS) medium supplemented with various concentrations of N-6-benzyladenine (BA), N-6-benzylaminopurine (BAP) and thidiazuron (TDZ), in single or in combination with ${\alpha}-naphthaleneacetic$ acid (NAA), was used to determine the rate of shoot proliferation. N-6-benzyladenine (BA) used at 0.5mg/l, was the most effective in initiating multiple shoot proliferation at the rate of 23 microshoots per shoot-tip explants after 40 days of culture. Shoot multiplication increased 1.2-fold in each successive subculture. Induction of rooting (98%) was achieved by transferring the shoots to the same basal medium containing 2 mg/l indole-3-butyric acid (IBA). Plantlets went through a hardening phase in a controlled growth chamber, prior to in vivo transfer. These results represented that possible application for the mass production of plantlets through in vitro culture system of Zanthoxylum piperitum DC.

감염 근관에서 혐기성 배양법과 간접 면역 형광법 및 DNA 프로브법에 의한 Porphyromonas endodontalis의 검출에 관한 비교 연구 (COMPARATIVE STUDY ON THE DETECTION OF PORPHYROMONAS ENDODONTALIS BY ANAEROBIC CULTURE, IIF AND DNA PROBE METHOD IN INFECTED ROOT CANALS)

  • There are many advantages when using IIF and DNA probe methods over anaerobic culture method in that they are time-and effort-saving, more precise and more sensitive. Furthermore, in IIF and DNA probe methods, the detection is possible only with small amount of bacteria, the quantitative analysis is possible, and the cell viability is not necessary. The purpose of this study is to observe the incidence of P.endodontalis by carrying out anaerobic culture, IIF and colony lift using DNA probe method respectively, and to compare these 3 methods in terms of effectiveness and sensitivity in order to identify the most effective detection method. 30 teeth with at least one clinical symptoms, with single canal, and with pulp necrosis were sampled. For sampling bacteria, access cavity was prepared after disinfecting tooth and its surroundings. Then the paper point was inserted up to the periapical area, leave there for a while, and finally it was placed into PRAS Ringer's sol. and PBS sol. In anaerobic culture method, P.endodontalis was identified by biochemical tests after subculturing black and brown colonies which were produced after 7 days of incubation on BAP and Brucella BAP in anaerobic chamber. To identify P.endodontalis in IIF method, species-specific polyclonal rabbit-antisera of P.endodontalis(ATCC 35406) was reacted with sampled PBS sol. dispensed onto glass slide, and then P.endodontalis was examined by phase contrast microscopy after incubating with Goat anti-rabbit lgG conjugated to Fluorescein isothiocyanate. For colony lift using DNA probe method, membranes were laid over colonies on the surface of BAP and were hybridized with cloned DNA probe of P.endodontalis. The existence of P.endodontalis was then identified by the methods of chemiluminescent detection and color metric detection. Black colony was found in 11 teeth out of 30 teeth and P.endodontalis was detected in 6 teeth (20 %) by anaerobic culture method, 16 teeth (53 %) by IIF method, and 7 teeth (23 %) by DNA probe method. IIF method is significantly better in detecting P.endodontalis than DNA probe method and anaerobic culture method. There was no significant differences between DNA probe method and anaerobic culture method. There was significant correlation between the formation of black colony and the existence of P.endodontalis. The probability of detecting P.endodontalis when black colony being present is 2.89 times higher than when not being present. There was significant relationship between the foul odor of clinical symptoms and P.endodontalis. The sensitivity of existing P.endodontalis when foul odor being present was 93.75 %, while the specificity of not existing P.endodontalis when foul odor not being present was 28.57 %. These results suggested that the probes of P.endodontalis will be used to decide the method and prognosis in endodontic treatments.
