Solubilization of Insoluble Phosphates by Penicillium sp. GL-101 Isolated from Soil (토양에서 분리한 Penicillium sp. GL-101에 의한 난용성 인산염의 가용화)

  • Phosphate solubilizing microorganisms (1,000 bacteria and 200 fungi) were isolated from soil around Kyungnam and Kyungbook regions using potato dextrose agar-calcium phosphate medium. A fungus with the greatest phosphate solubilizing activity was selected and identified to Penicillium sp. GL-101, based on the morphological characteristics of conidiophore and conidia; flask shape of phialide, simple branching type of conidiophore, and columnar shape of conidial head, in malt extract agar and potato dextrose agar media. The optimum temperature and initial pH to solubilize rock phosphate in potato dextrose broth-rock phosphate medium were $25^{\circ}C$ and pH 7.5, respectively. In these optimum conditions, phosphate solubilizing activities of Penicillium sp. GL-101 against four types of insoluble phosphate: tricalcium-phosphate, aluminium phosphate, hydroxyapatite and rock phosphate, were quantitatively determined. As results, this fungus highly discharged free phosphates to the culture broth with the concentrations of 1,152 ppm against tricalcium-phosphate, 565 ppm against rock phosphate, 292 ppm against aluminium phosphate, and 217 ppm against hydroxyapatite, respectively.

A Study on Nutrient Intakes, Glycemic Index, and Glycemic Load according to Obesity Index in Elementary School Students (남녀 초등학생의 비만도에 따른 영양소 섭취 및 Glycemic Index, Glycemic Load에 관한 연구)

  • The purpose of this study was to evaluate nutrients intakes, glycemic index (GI), glycemic load (GL) according to obesity index in elementary school students. The study subjects included 229 elementary school students (boys=108, girls=121) who were divided into 3 groups consisting of an underweight group (obesity index<-10%, n=58), a normal weight group (10%${\leq}$ obesity index<10%, n=130) and an overweight group (obesity index${\geq}$10%, n=41) by their obesity index. The nutrient and food intakes data obtained by a 3-day food record were analyzed. Daily dietary GI and GL values were calculated from the 3-day food record. The average age of the subjects was 11.9 years. The mean daily energy intake was 2,186.8 kcal in the underweight group, 2,123.5 kcal in the normal weight group, and 2,174.2 kcal in the overweight group. The intakes of calcium and animal calcium per 1,000 kcal in the overweight group were significantly lower than in the underweight and normal weight groups (p<0.01, p<0.05), and fruit, egg and milk intakes in the overweight group were lower than those in the underweight group (p<0.05, p<0.05, p<0.05). The mean daily dietary GI of the underweight, normal weight, and overweight groups were 67.7, 68.4 and 69.5, respectively (p<0.05). The mean daily dietary GL of the underweight, normal weight, and overweight groups were 212.8, 208.1 and 213.3, respectively. The major food source of dietary GI and GL in the three groups was rice. Other major food sources of dietary GI were croquettes, hand-rolled noddle soups, instant noddles, milk, and rice cake. Dietary GI was not significantly correlated with weight, obesity or body mass index, when adjusted for energy, carbohydrate, and dietary fiber. However, GL adjusted to energy, carbohydrate and dietary fiber tended to correlate with obesity index (r=0.126, p=0.059). These results suggest that dietary GI and GL have possibility affecting obesity-related indicators in elementary school students.

Studies on the Composition of Lipid Class and Fatty Acid of Korean Black Soybean (한국 재래 검점콩의 지방질 조성에 관한 연구)

  • Total lipids of Korean black Soybean were extracted, purified and fractionated into three lipid classes (Neutral lipid, NL; glycolipid, GL;phospholipid, PL) and the composition of lipid class and fatty acid were studied. Total lipids of black soybean consisted of 88.5% NL, 2.5% GL and-9.0% PL. In the NL, triglycerides were predominant(80.4%) and also sterol esters, hydrocarbons, diglycerides, free fatty acids and sterols were identified. The major component of GL were monogalactosyl diglycerides and esterified steryl glycosides, and then cerebrosides, steryl glycosides, digalactosyl diglycerides were also found. As major component of the PL, phosphatidyl ethanolamine and phosphatidyl choline were observed, other compounds such as phosphatidyl inositol, phosphatidyl serine and lysophosphatidyl choline were also determined. The major fatty acids in the NL and GL were linoleic acid, oleic acid and palmitic acid, however, PL contained higher relative content of palmitic acid and lower level of oleic acid compared with those of NL and GL.

Preparation and Characterization of Gel Food for Elderly (고령자용 겔상식품의 제조 및 특성연구)

  • Model gel food samples of yanggaeng (Y) were prepared for the elderly with various gelling agents, including agar (AG), low acyl gellan (GL), ${\kappa}$-carrageenan (CA), locust bean gum (LB), glucomannan (GM), and xanthan gum (XA) in different combinations as follows (in 1:1 ratio): LB+CA, GM+XA, and GM+CA. The quality characteristics of the different combinations were compared. The results revealed that water loss was highest for Y-GL, whereas there was no significant difference among the other samples. Y-GL showed the highest values for lightness in color, whereas Y-AG showed the lowest. Regarding textural properties, Y-LB+CA had the highest hardness value, whereas Y-GL had the lowest; the hardness of Y was related to the cross-section of the added gel. Finally, Y-GM+XA exhibited the highest score in overall acceptability in the sensory test by elderly, indicating that the preferable texture by elderly is slightly chewy, but not adhesive.

Treatment with Extracellular Vesicles from Giardia lamblia Alleviates Dextran Sulfate Sodium-Induced Colitis in C57BL/6 Mice

  • Inflammatory bowel disease (IBD) is a chronic and recurrent illness of the gastrointestinal tract. Treatment of IBD traditionally involves the use of aminosalicylic acid and steroids, while these drugs has been associated with untoward effects and refractoriness. The absence of effective treatment regimen against IBD has led to the exploration of new targets. Parasites are promising as an alternative therapy for IBD. Recent studies have highlighted the use of parasite-derived substances, such as excretory secretory products, extracellular vesicles (EVs), and exosomes, for the treatment of IBD. In this report, we examined whether EVs secreted by Giardia lamblia could prevent colitis in a mouse model. G. lamblia EVs (GlEVs) were prepared from in vitro cultures of Giardia trophozoites. Clinical signs, microscopic colon tissue inflammation, and cytokine expression levels were detected to assess the effect of GlEV treatment on dextran sulfate sodium (DSS)-induced experimental murine colitis. The administration of GlEVs prior to DSS challenge reduced the expression levels of pro-inflammatory cytokines, including tumor necrosis factor alpha, interleukin 1 beta, and interferon gamma. Our results indicate that GlEV can exert preventive effects and possess therapeutic properties against DSS-induced colitis.

Evaluation of Methane Generation Rate Constant(k) by Estimating Greenhouse Gas Emission in Small Scale Landfill (소규모 매립지에 대한 메탄발생속도상수(k) 산출 및 온실가스 발생량 평가)

  • In this study, greenhouse gas emission for small scale landfill (H and Y landfill) was investigated to deduce special the methane generation rate constant(k). To achieve the purpose, the data of physical composition was collected and amount of LFG emission was calculated by using FOD method suggested in 2006 IPCC GL. Also, amount of LFG emission was directly measured in the active landfill sites. By comparing the results, the methane generation rate constant(k), which was used as input variable in FOD method suggested in 2006 IPCC GL, was deduced. From the results on the physical composition, it was shown that the ranges of DOC per year in H (1997~2011) and Y (1994~2011) landfill sites were 13.16 %~23.79 % ($16.52{\pm}3.84%$) and 7.24 %~34.67 % ($14.56{\pm}7.30%$), respectively. The DOC results showed the differences with the suggested values (= 18 %) in 2006 IPCC GL. The average values of methane generation rate constant(k) from each landfill site were $0.0413yr^{-1}$ and $0.0117yr^{-1}$. The results of methane generation rate constant(k) was shown big difference with 2006 IPCC GL defualt value (k = 0.09). It was confirmed that calculation results of greenhouse gas emission using default value in 2006 IPCC GL show excessive output.

Diacylglycerol Production by Enzymatic Glycerolysis of Soybean Oil. (대두유에서 글리세롤리시스 반응을 이용한 디글리세리드의 효소적 생산)

  • Diglyceride (DG) was prepared by reaction of soybean oil and glycerol in the presence of lipase. The initial rate of DG production was greatly affected by the amount of lipase. However the DG content at equilibrium was hardly affected by the amount of lipase added to the reaction mixture. The initial rate of FFA formation was highly affected by the moisture content between 0.5 and 2.3%, but at higher water content (3.3-5.2%), there was a small increase in the rate. And DG content at equilibrium slowly increased with the increase of the water content in glycerol up to 4.4%. However, there was a sharp decrease in DG content at higher water content (5.2-6.4%) due to higher free fatty acid production. The highest yield of DC was obtained at the temperature ranges of 30-5$0^{\circ}C$. The final yield of DG was not dependent on the glycerol (GL) to triglyceride (TG) molar ratio. However, at the molar ratio of 0.75:1 (GL/TG), the enzyme-catalyzed reaction was highly efficient and utilized all the glycerol. In optimized conditions for glycerolysis a yield of approximately 45% DG was obtained. 66% of total DG was 1,3-DG.

  • Glucose isomerase (E.C.5.3.1.5) which reversibly catalyzes reaction between D-glucose and D-fructose was demonstrated in cell free extracts of alkalophilic Streptomyces sp. B-2 isolated from soil The optimum temperature, pH, and pH stability were 6$0^{\circ}C$, 10.5, and 7.8, respectively. The production of Gl in xylose and yeast extract was higher than that of other carbon source and nitrogen source. The Gl production was affected by Co2+ and Mg2).

Characteristics of Squid Viscera Oil (오징어 내장의 지방질조성)

  • The oil content and composition of squid visera were determined to obtain data for utilization of this by-product. There was no significant difference in the glycolipid (GL) and phospholipid (PL) content in Illex argentinus and Todarodes pacificus, but neutral lipid (NL) was different (p<0.05). The viscera oil of I. argentinus contained $30.50\%$ total lipid which consisted of $96.24\%$ NL, $2.63\%$ GL, $2.37\%$ PL, and contained $644mg\%$ cholesterol. The viscera oil of T. pacificus contained $30.20\%$ total lipid which consisted of $94.82\%$ NL, $2.85\%$ GL, $2.34\%$ PL, and contained $1,224\;mg\%$ cholesterol. The NL, GL and PL of viscera oil in I. argentinus mainly consist of triglyceride $(44.01\%)$, esterified steryl glycosides $(58.95\%)$ and phosphatidyl cholines $(32.36\%)$, respertively. Those of viscera oil in T. pacificus mainly consist of triglyceride $(39.63\%)$, monogalactosyl diglycerides $(51.67\%)$ and phosphatidyl cholines $(31.98\%)$, respectively. The major fatty acids of the viscera oil of I. argentinus and T. pacificus were C16 : 0, $C16\;:\;0,\;C18\;:\;1\omega9,\;C20\;:\;4\omega6,\;C20\;:\;5\omega3,\;C22\;:\;6\omega3$. In Illex argentinus, the fatty acids of NL mainly were $C16\;:\;0,\;C18\;:\;1\omega9,\;C20\;:\;4\omega6,\;C20\;:\;5\omega3,\;C22\;:\;6\omega3$. PL were $C16\;:\;1\omega7,\;C20\;:\;5\omega3,\;C22\;:\;6\omega3$ and GL were $C18\;:\;1\omega9,\;C20\;:\;5\omega3,\;22\;:\;6\omega3$. The major fatty acids of NL in T. pacificus were $C16\;:\;0,\;C18\;:\;1\omega9,\;C20\;:\;4\omega6,\;C20\;:\;5\omega3,\;C22\;:\;6\omega3$, PL were $C16\;:\;1\omega7,\;C20\;:\;5\omega3,\;C22\;:\;6\omega3$, and GL were $C18\;:\;1\omega9,\;C20\;:\;5\omega3,\;C22\;:\;6\omega3$.

Carcass Characteristics and Meat Quality of Swamp Buffaloes (Bubalus bubalis) Fattened at Different Feeding Intensities

  • Twenty-four male 1-year old swamp buffaloes (Bubalus bubalis) were randomly allocated to 4 groups. One group grazed on guinea grass (GG) and another on guinea grass and the legume Stylosanthes guianensis (GL). The other two groups were kept in pens and fed freshly cut guinea grass and concentrate at an amount of 1.5% (GC1.5) and 2.0% (GC2.0) of body weight, respectively. The effect of the different feeding intensities on carcass characteristics and meat quality were assessed. The mean body weight at slaughter was 398 (${\pm}16$) kg. Average daily gain was higher in concentrate-supplemented groups (570 and 540 g/d in GC1.5 and GC2.0, respectively) when compared to GG (316 g/d) and GL (354 g/d) (p<0.01). Likewise, the warm carcass weight was higher in GC1.5 and GC2.0 compared to GG and GL. Dressing percentage was 48.1% and 49.5% in GC1.5 and GC2.0 in comparison to 42.9% and 44.8% observed in GG and GL, respectively. Meat of Longissimus throracis from GC1.5 and GC2.0 was redder in color (p<0.01), while water holding capacity (drip and thawing loss) was improved in pasture-fed groups (p<0.05). Protein and fat content of Longissimus thoracis was higher in animals supplemented with concentrate (p<0.01), as was cholesterol content (p<0.05), whereas PUFA:SFA ratio was higher and n-6/n-3 ratio lower (p<0.01) in pasture-fed buffaloes. Results of the present study showed that the supplementation of pasture with concentrate enhances the growth and carcass characteristics of swamp buffaloes expressed in superior dressing percentage, better muscling, and redder meat with a higher content of protein and fat, whereas animals grazing only on pasture had a more favorable fatty acid profile and water holding capacity. In conclusion, the supplementation of concentrate at a rate of about 1.5% of body weight is recommended to improve the performance and carcass quality of buffaloes.
