Induction of Laccase from Wood-Rotting Fungi with 2,5-Xylidine (2,5-Xylidine을 이용한 목재부후균으로부터 Laccase 효소의 유도)

  • Some white-rot fungi, screened at the Laboratory of Forest Products Microbiological Chemistry, Chungbuk National University were cultured and added the inducer of laccase enzyme, 2,5-xylidine. The fungi named by CB-13, CB-20, CB-99, CB-100 and CB-123 strains showed positive results in the decolorization of aromatic compounds, carminic acid and Rhemazol brilliant blue R. Concerned to the inducing effect of 2,5-xylidine on laccase activity, CB-20, CB-100 and CB-123 strains showed very high activity by addition of 2,5-xylidine, whilst CB-13, CB-99 and CB-124 strains produced relatively high laccase enzymes, regardless of inducer addition. There were no any laccase activities on CB-25, CB-64 and CB-139, even in addition of inducer. It is confirmed that some screened fungi have decolorizing ability on aromatic compounds, carminic acid and Rhemazol brilliant blue R. Also, the addition of inducer, 2,5-xylidine, has increased the activity of laccase enzyme which is secreted from some white-rot fungi.

  • Slag-CB is widely used in various fields that require groundwater control. It is a type of CB where a portion of the cement mixed with CB is replaced with GGBS. In general, Slag-CB has the advantage of long-term improvement in compressive strength, permeability, durability, and chemical resistance as the GGBS replacement ratio increases. However, there are problems such as decreased flexibility and resistance to deformation of the cut-off walls, as well as brittleness upon failure. To address these problems, some quality standards recommend designing Slag-CB with lower strength, which makes it challenging to apply high-strength Slag-CB with a high GGBS replacement ratio in the field.In this study, we aimed to improve the flexibility and resistance to deformation of Slag-CB to prevent brittle failure and improve the field applicability of Slag-CB. To achieve this, we evaluated the compressive behavior of nylon fiber-reinforced Slag-CB and proposed measures for enhancing the flexibility and resistance to deformation of Slag-CB.

Optimization of Post-Activation Systems to Improve the Embryonic Development in Porcine Parthenogenesis and Somatic Cell Nuclear Transfer

  • This study was conducted to establish the optimal chemical post-activation conditions in porcine embryonic development after parthenogenesis (PA) and somatic cell nuclear transfer (SCNT) using 4 different chemical compositions (cytochalasin B (CB), cyclohexamide (CHX), demecolcine (DC), 6-dimethylaminopurine (DMAP). Porcine embryos were produced by PA and SCNT and then, cultured for post-activation with CB ($7.5{\mu}g/mL$), CB ($7.5{\mu}g/mL$) + CHX ($10{\mu}g/mL$), CB ($7.5{\mu}g/mL$) +DC ($0.4{\mu}g/mL$), and CB ($7.5{\mu}g/mL$) + DMAP (2 mM). In PA embryonic development, cleavage rates have been significantly higher in CB group (94.7%) and CB+DMAP group (94.1%) than that of CB+CHX and CB+DC group (88.1 and 84.3%, respectively). There have been no significant differences in blastocyst formation rates among the four groups. In cell number of blastocyst was shown in CB group (42.3%) significantly higher than CB+CHX and CB+DC group (40.6 and 40.6%, respectively). In SCNT embryonic development, CB+DMAP group (89.7%) significant differences were found on embryo cleavage rates when compared with other three groups. Blastocyst formation rates in CB+DMAP group (26.9%) were significantly higher when compared with CB, CB+CHX, and CB+DC groups (25.5, 20.2, and 22.1%, respectively). In blastocyst cell number, CB+DMAP group (41.4%) was found higher significant difference compared with other three groups. Additionally, we have investigated survivin expression in early development stages of porcine SCNT embryos for more confirmation. Our results establish that CB group and CB+DMAP group for 4 h during post-activation improves pre-implantation improvement of PA and SCNT embryos.

RGS3 Suppresses cAMP Response Element (CRE) Activity Mediated by CB2 Cannabinoid Receptor in HEK293 Cells (캐너비노이드 수용체 CB2의 신호전달작용에 미치는 RGS3의 억제적 효과)

  • RGS proteins have been identified as negative regulators of G protein signalling pathways and attenuate the activity of GPCR receptors. However, information on the regulatory effects of RGS proteins in the activity of cannabinoid receptors is limited. In this study, the role of RGS proteins on the signal transduction of the CB2 cannabinoid receptor was investigated in HEK293 cells co-transfected with CB2-receptors and plasmids encoding RGS2, RGS3, RGS4 and RGS5. Treatment of cells with WIN55, 212-2, a CB2 receptor agonist, inhibited forskolin-induced cAMP response element (CRE) activity in CB2-transfected HEK293 (CB2-HEK293) cells. This inhibitory effect of WIN 55, 212-2 on CRE activity was reversed by co-transfection of CB2-HEK293 cells with RGS3, but not with RGS2, RGS4 and RGS5. However, endogenous RGS3 protein knocked down by a small interfering siRNA targeting RGS3 gene enhanced inhibition of forskolin induced CRE activity via agonist induced CB2 receptor signal transduction. These results indicate the functional role of endogenous RGS protein in cannabinoid signaling pathways and define receptor-selective roles of endogenous RGS3 in modulating CRE transcriptional responses to agonist induced CB2 receptor activity.

Effects of Survivability of Frozen Porcine Embryos by Different Concentrations and Exposed Times of Cytochalasin-B before Vitrification (돼지 배아의 유리화 동결 시 Cytochalasin B의 농도와 처리 시간에 의한 효과)

  • The purpose of this study was to evaluate the effects of Cytochalasin B treatment on the survivability The different concentration and exposed times of CB of porcine embryos frozen by vitrification were observed. The results were summarized as follows ; 1. The survivability rates of the porcine embryos treated CB(60.5%) were significantly higher than non-treated(32.8%)(p<0.1). However the recovered with normal morphology rates(84.2%) were no significantly different than non-treated(81.9%). 2. We observed by different concentration of CB treatment, the group of CB treated with 7.5 $\mu\textrm{g}$/$m\ell$ were significantly showed a higher of normal morphology(44/45; 98%) and viability rate(33/45;73%) than other groups(morphology: 65∼70%, viability: 15∼74%)(p<0.05). While the control was lower as 70% of morphology and 38% of viability. 3. We examined exposed time of in-vitro CB treated embryos, the group of more than 20 minutes exposed were significantly were observed a higher rates of normal morphology and viability(p<0.05). And the group of 13 minutes, 14,000 rpm centrifuged(64%) were significantly higher than non-centrifuged(36%). Survivability of porcine embryos were improved in CB treated group. It suggested that 7.5 $\mu\textrm{g}$/$m\ell$ concentration of CB treatment, 20 minutes exposed times and 13 minutes, 14000 rpm centrifuged prior to vitrification improve normal morphology and survivability rates.

Fluorescence Enhancement of 7-Diethylamino-4-methylcoumarin by Noncovalent Dipolar Interactions with Cucurbiturils

  • We have investigated the complex forming behavior of cucurbit[6]urils(CB6) and cucurbit[7]urils(CB7) with 7-diethylamino-4-methylcoumarin(C460) in water. The electronic absorption maximum of C460 shows bathochromic shift with the addition of CB7 and fluorescence intensity is greatly increased, while CB6 has no noticeable effects on the spectroscopic properties of C460. It is noted that CB7 interacts more strongly with C460 than CB6 does. Fluorescence lifetime also significantly increased for the CB7 complex, which is attributed to reduced polarity surrounding C460 and/or C460 being in a restricted environment. The stoichiometry for the complex formation determined from the fluorescence titration measurement indicates that 2:1 complex in which two CB7 molecules bind to C460 is formed. Thus, two step equilibrium processes are suggested for the complex formation and the binding constants are estimated. The semi-empirical electronic structures calculations indicate that C460 is not included in the CB7 cavity but interacts noncovalently with the portal carbonyls of CB7.

  • This Analysis was conducted for 13 toxic-PCBs having TEF value among 209 PCBs isomers in sediment by isotope dilution HRGC/HRMS method. From the result, the recovery of surrogate standard was in the range of 71~99%. The concentration range for 13 toxic-PCBs in sediment was found to be 0.84~2.49 ng/g, among them the concentration levels of 2,2',3,3',4,4',5-HpCB(IUPAC No. 170) and 2,2',3,4,4',5,5'-HpCB(IUPAC No. 180) represented almost 50% of total concentrarion and that of 2,3,4,4,5-PeCB(IUPAC No. 114) showed over 10%. The TEQ concentration levelwas in the range of 0.38~2.63 pg-TEQ/g and 3,3',4,4',5-PeCB(IUPAC No. 126) concentration represented over 50% of total TEQ concentration.

Selective Homocysteine Assay with Cucurbit[7]uril by pH Regulation

  • We report the effect of pH on the supramolecular complexation of two biothiols, viz., homocysteine (Hcy) and cysteine (Cys), with cucurbit[7]uril (CB[7]). Under basic pH conditions, Cys did not complex with CB[7], whereas Hcy efficiently complexed with CB[7], as confirmed by 1H NMR spectroscopy and Ellman's reagent (5,5'-dithio-bis(2-nitrobenzoic acid), DTNB) assay. 1H NMR and Raman spectroscopic studies revealed that, in the absence of CB[7], Hcy auto-oxidized slowly (~36 h) to homocystine (HSSH) under basic pH conditions. However, the rate of Hcy oxidation increased by up to 150 fold in the presence of CB[7], as suggested by the DTNB assay. Thus, supramolecular complexation under basic pH conditions led to the formation of a HSSH-CB[7] complex, and not Hcy-CB[7]. The results indicate that Hcy is rapidly oxidized to HSSH under the catalysis of CB[7], which acts as a reaction chamber, in basic pH conditions. Our studies suggest that Hcy concentration, a risk factor for cardiovascular disease, can be selectively and more easily quantified by supramolecular complexation with CB [7].

  • This study demonstrates an effective one-pot regioselective synthesis of 3,7-diarylpyrazolo [1,5-${\alpha}$]pyrimidines. Moreover, the derivatives obtained were effective CB1R antagonists. These pyrazolo [1,5-${\alpha}$]pyrimidines with diaryl groups at the 3,7-positions are potential pharmacophores for CB1R candidates.

Anchoring Strength Characteristics by the Washing Process after Rubbing on the Polyimide Layers

  • The polar anchoring strength and surface ordering in a NLC, 4-n-pentyl-4'-cyanobiphenyl (5CB), on a rubbed polyimide (PI) surface containing trifluoromethyl moiety were studied. The large extrapolation length de of 5CB for washing process of water was measured at RS=114 mm. The polar anchoring energy of 5CB on the rubbed PI surface is decreased by the washing process. Also, the polar anchoring energy of 5CB increases with the rubbing strength on the PI surface. The surface ordering of 5CB for all washing processes is smaller than the non-washing process; it is attributed to the washing process.
