Measurement of 1,4-dioxane in surface water by headspace GC-MS (GC-MS에 의한 지표수에서 1,4-Dioxane의 측정)

  • 1,4-Dioxane was classified as a Group 2B carcinogen by the International Agency for Research on Cancer. The compound was measured in surface water with a headspace gas chromatographic mass spectrometric detection. A 5 mL water sample was placed in a 10 mL headspace vial and saturated with NaCl, and the solution was spiked with 1,4-dioxane-d8 as an internal standard and sealed with a cap. Water samples were collected from twenty-two basins of Gum-River on June and September 2012, respectively. As a result, 1,4-dioxane was detected in the concentration range of $0.49-43.0{\mu}g/L$ (mean $2.0{\mu}g/L$) in the frequency of about 30% in surface water samples.

A Review of the Optimum Feeding Rates and Feeding Frequency in Korean Rockfish Sebastes schlegeli Reared at Seven Different Water Temperatures

  • Seven feeding trials were conducted to determine the effects of feeding rate and frequency in the Korean rockfish Sebastes schlegeli at seven different water temperatures. Two feeding-rate experiments for 5-g Korean rockfish at $17^{\circ}C$ and $20^{\circ}C$; three feeding rate experiments for 16-g Korean rockfish at $16^{\circ}C$ and $24^{\circ}C$; and finally, two feeding frequency experiments for 93- and 100-g at $15^{\circ}C$ and $20^{\circ}C$ were conducted. Twenty fish averaging $5.5{\pm}0.2$ g ($mean{\pm}SD$) were randomly distributed into 18 indoor tanks containing 40-L seawater from a semi-recirculation system. Fish were fed a commercial diet for 4 weeks at water temperatures of $17^{\circ}C$ and $20^{\circ}C$. Two feeding trials were conducted to determine the optimum feeding frequency in growing Korean rockfish reared at temperatures of $15^{\circ}C$ and $19^{\circ}C$. Broken line regression analysis of weight gain (WG) indicated that the optimum feeding rates of 5-g growing Korean rockfish were 4.48% (BW/day; BW, body weight) at $17^{\circ}C$ and 4.83% (BW/day) at $20^{\circ}C$. Broken line regression analysis of WG showed optimum feeding rates of 16-g juvenile Korean rockfish of 3.41% (BW/day) at $16^{\circ}C$, 3.75% (BW/day) at $20^{\circ}C$, and 3.34% (BW/day) at $24^{\circ}C$. The biological performance, results, along with morphological indices and serological characteristics, suggest that a feeding frequency of 1 meal/day was optimal to improve WG in growing Korean rockfish grown from 93 to 133 g at a water temperature of $15^{\circ}C$, and 100 to 132 g at $19^{\circ}C$.

Occurrence of Perchlorate in Drinking Water in Korea (국내 주요 정수장 수돗물의 퍼클로레이트 검출현황)

  • Perchlorate is an unregulated contaminant but recently many cases of perchlorate contamination have been reported in the US. In Japan, also, a couple of contamination incidents caused by perchlorate occurred. Against this backdrop, investigation on perchlorate was urgent for Korea. Accordingly, perchlorate investigation in tap water and river water was conducted one time a week for seven consecutive weeks at major water sources of Korea including Han River, Guem River, Yeong-san River, and Nak-dong River. Perchlorate was not found at Han River, Guem River, and Yeong-san River. However, all the 4 tap water samples in Nak-dong River recorded high level of perchlorate. Among others, NT1 located at Daegu posted the highest value of $22.3{\mu}g/L$. This level is lower than $24.5{\mu}g/L$, the threshold recommended by US EPA. Still, perchlorate contamination in drinking water can deal a heavy blow to nerve development of infants and children by causing iodine deficiency. At the 1st and 2nd investigations, perchlorate concentration reached $18.7{\sim}95.6{\mu}g/L$ and $4.0{\sim}25.6{\mu}g/L$ respectively, both of which are relatively higher. The high perchlorate concentration in Nak-dong River was possible because of the waste water discharged from LCD manufacturing factory which was located at NS3 in Gumi, Korea. Perchlorate concentration of waste water from the factory was $730{\sim}1,858{\mu}g/L$.

The Effect of Salt Contents in Diet and Water on Performance and Physiological Changes in Broiler Chicks (염분 수준이 육계 병아리의 생산성과 생리적인 변화에 미치는 영향)

  • To investigate the effects of salt contents in diet and water on the growth of broiler chicks, 288 heads of Hubbards strain chicks, were fed with com-soybean meal diets containing 0, 0.25, 0.5, 0.75 1.0, 2.0, 3.0, or 4.0% salt from 1d to l4d posthatching. And 180 chicks were also supplemented with salt to their water at 0, 0.1, 0.2, 0.5 or 1.0%. Feed intake and weight gain tended to decrease as the salt levels in diets increased and those were the lowest in 4% group as 481 g and 168 g, however, mortality was the highest in 4% group as 75.0%. As for the salt levels in water, feed intake and weight gain were low as 427 g and 162 g in 1% group, while mortality was 77.8%. Salt poisoning chicks were shown with thirsty, appetite deficiency, rising impossibility, sleepiness, or convulsions, and in autopsy, were shown with ascites, hydropericardium, cardiac hypertrophy, hemorrhage of eyelid, hemorrhage of trachea, hemorrhage of intestine, etc. Conclusively, salt content above 4% in diet or 1% in water could reduce the performance in broiler chicks.

Effect of Acid Catalyst Kinds on the Pore Structural Characteristics of Water Glass based Silica Aerogel (산 촉매가 물유리 기반 실리카 에어로겔의 기공구조에 미치는 영향)

  • Water glass is much cheaper than silicon alkoxide, so it has advantage for commercialization. A condensation by acid catalyst makes considerable effect about the properties of water glass based silica aerogel among many factors in silica aerogel process. The pore structural properties of water glass based silica aerogel such as specific surface area and pore size distribution have been investigated through the changes in the amount and the kinds of acid catalyst. It has been confirmed that water glass based silica aerogel is affected by various conditions of catalyst in the condensation reaction such as the kind, concentration, and the amount of mole of acid catalyst on the properties of final products. Especially, it is checked that the effect of mole of acid is more prominent than that of concentration. In the case for conventional method with introducing 4M HCl in condensation step, the silica aerogel could be synthesized which has $394m^2/g$ of specific surface area, 2.20 cc/g of pore volume, 22.3 nm of average pore size, and 92.53% of porosity. On the other hand, when 4M sulfuric acid was used with 73 mmol at the condensation step of water glass based silica aerogel, the pore structural characteristics of water based silica aerogel showed better properties than the case of using HCl, for example, specific surface area was measured as $516m^2/g$, and pore volume, average pore diameter, and porosity were obtained as 3.10 cc/g, 24.1 nm, and 96.1%, respectively.

Study on the Physicochemistrical Property of the Clean Active Water Depositting on the Ceramic Stone by the Mineral-Oriental medicine (광물성 한약재 세라믹스톤 심적수의 이화학적인 특성에 대한 연구)

  • studied on the physicochemistrical property of the clean active water depositting on the ceramic stone by the mineral-Oriental medicine. The results are as follows. The cluster of the clean active water is observed 53-66.5㎐ for contral water. The clean active water is observed 7.2 ph and 1.3330 refractive index for contral water. In the chemical experimental test by ICP, the clean active water is observed 102℃ in boiling point and 4.8㎎/100g V.E for 0㎎g/100g of contral water. In the antibiotic experimentation, an anion, the dimintion index of bacilli, radiation degree and radiate energy of the clean active water increased for contral water. And in the deodorization experimentation, the clean active water is observed 95% deodorization index.

Analysis of Physiological Activity and Cytotoxicity of Fermented and Hot Water Extracts Using Residues after Onion Harvest (양파 수확 후 잔재물을 이용한 발효 및 열수 추출물의 생리활성과 세포독성 분석)

  • In order to utilize the residue that is thrown away after an onion harvest, we analyzed the physiological activity and cytotoxicity of fermented and hot water extracts of the residue. The pH of the extracts were all acidic, and organic matter content was 0.75% in the fermented extract and four times more than 0.19% in the hot water extract. The contents of nitrogen, phosphoric acid, calcium, and magnesium components, except for the potassium component among macroelements, were higher in the fermented extract than in the hot water extract. The content of iron and silicon among the micro-elements was also higher in the fermented extract than in the hot water extract. In addition, the content of boron was higher in the hot water extract than in the fermented extract. The total polyphenol contents of the fermented and hot water extracts were $16.2{\pm}3.3mg{\cdot}g^{-1}$ and $14.6{\pm}1.4mg{\cdot}g^{-1}$, respectively, which was $1.6mg{\cdot}g^{-1}$ higher in the fermented extract than in the hot water extract. However, the total flavonoid contents of the fermented and hot water extracts were $0.1{\pm}0.1mg{\cdot}g^{-1}$ and $4.8{\pm}0.7mg{\cdot}g^{-1}$, respectively, which was $4.7mg{\cdot}g^{-1}$ higher in the hot water extract than in the fermented extract. DPPH and ABTS radical scavenging ability for antioxidant activity were higher in the hot water extract than the fermented extract. The cytotoxicity of the extract using MTT assay showed cell viability of 101.6% and 97.9% in the fermented and hot water extracts, respectively. It was confirmed that there was no cytotoxicity in either extract.

Study on the antioxidant, anti-inflammatory and Preservative features Water Chestnut (능실의 항산화, 항염증 및 방부 효과 연구)

  • The purpose of this study was to assess the antioxidant, anti-inflammatory and preservative effects of Water Chestnut from 70% ethanol extracts. The toxicity of extracts from Water Chestnut investigated using the RAW 264.7 cell showed higher than 90% of cell survival rate. The total content of polyphenol ethanol extract was $353.1{\pm}5.6mg/g$, while the total content of flavonoid was $26.2{\pm}1.4mg/g$. With a concentration level of $1{\sim}1000 ({\mu}g/m{\ell})$ ethanol extract of Water Chestnut the range of removal of 1, 1-diphenyl-2-picryl-hydrazyl (DPPH) radicals was $17.0{\pm}2.8%{\sim}88.6{\pm}0.6%$ respectively and the range of removal of 2,2'-azinobis-(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) radicals was also $2.3{\pm}0.8%{\sim}93.9{\pm}0.2%$ respectively. There were decreases in reactive oxygen species(ROS) generations ethanol extracts of Water Chestnut 1, 10, $100({\mu}g/m{\ell})$ and significance decrease at $100{\mu}g/m{\ell}$ (p<.01). As a result of measuring the Nitric Oxide(NO) generation amount of Water Chestnut extract 1, 10, $100({\mu}g/m{\ell})$ concentration exhibited significant (p<.001, p<.01) decreases. For the anti-bacterial features using a paper disk and the preservative features using petri film, no significance was found and therefore water chestnut extracts had not anti-bacterial or preservative features.

Mineral Contents of Hot Water Extracts and Shell of Shellfishes from Western Coast of Korea (서해안 조개류의 껍질과 열수 추출물의 무기질 함량)

  • Yields of shells, appearance of the shellfishes, mineral contents of the shells and their hot water extracts were determined in six shellfishes such as corb shell(CS). short neck clam(SNC). taste clam(TC), ark shell(AS). top shell(TS) and oyster(OY) from the western coast of Korea. Yields of shells in shellfishes were 70.I~80.5% but yields of TC was the lowest as 40.7%. The highest among weight of the shell the shellfishes was the TS(26.2g) and the lowest weight was TC(5.6g) Colors of CS, SNC, TC. AS. TS and OY were yellowish brown, brown, black. greenish brown and gray. respectively. Ca content of the shell of shellfishes was 36.23~38.78% and the content of K and Na were 0.23~4.54% and 1.48~l.59%, respectively. Contents of Na, Mg, Fe. Mn, Zn, Cu. P and S were in the range of 0.01~0.21%. It also contained heavy metals, such as Pb(1.90~7.75 ppm), Cd(0.5~4.50 ppm), As (1.40~4.30 ppm), Se (0.2~l.50 ppm). Cr(1.00~8.30 ppm) and Hg(0.002~8.2 ppm), Ca content in hot water extracts of shell of shellfish was the highest in TC(2,448 mg/100 g), and the lowest in SNC(115 mg/100 g). K content in TC extracts was the highest with the levels of 952 mg/100 g. P content of TC and SNC were 201 and 0.36 mg/100 g, respectively. The contents of Pb, As, Se and Cr were the highest in the extracts of TC and were 110. 40. 90, 20 $\mu$g/100 g, respectively. But Cd was not detected in the extracts of SNC.

Repeated Dose 4-Week Oral-Treatment for DRF Toxicity Test of HMC05 in Sprague-Dawley Rats (HMC05의 Sprague-Dawley 흰쥐를 이용한 4주 반복 경구투여 DRF 독성시험)

  • Objectives: HMCO5 is an extract obtained from 8 different herbal mixtures. We undertook a safety evaluation of HMCO5 for a dose range finding (DRF) toxicity test in specific pathogen free (SPF) Sprague-Dawley (SD) male and female rats. Methods: The male and female rats were divided into 4 groups, respectively; G(0), treated with distilled water: G(1), treated with 222 mg/kg HMC05: G(2), treated with 667 mg/kg HMC05, and G(3), treated with 2,000 mg/kg HMC05; HMC05 was administered orally for 4 weeks. The safety evaluation examined clinical signs, mortality, body weight, food consumption, water consumption, ophthalmic findings, urinalysis, hematological values, absolute & relative organ weights, and necropsy findings during the tests. Results: There were no changes in clinical signs, mortality, body weight, food consumption, water consumption, and ophthalmic findings examined during the test periods. In serum biochemical values, triglyceride was increased in male group G(3) and Na$^+$ decreased significantly in male groups G(2), G(3) and G(4). In male group G(4), spleen weight decreased relatively and increases of absolute & relative left ovary weights were found. In addition, an adhesion of liver to diaphragm was found in male group G(2). However, we could not find any dose-interrelationships in these changes. Conclusions: These results indicate that HMC05 extract did not show any toxicity in the DRF toxicity study. Therefore, it suggests that establishment of 1,000, 333 and 111 mg/kg dosages are moderate in a repeated dose 26-week oral toxicity study of HMC05.
