Metabolomic Analysis of Ethyl Acetate and Methanol Extracts of Blueberry (Ethyl Acetate와 Methanol을 이용한 블루베리 추출물 대사체 분석)

  • Metabolite profiling of blueberry (cultivar "Spartan") was performed by extraction using different solvents, methanol and ethyl acetate, through metabolomic analysis using LC-MS/MS. Unsupervised classification method (PCA) and supervised prediction model (OPLS-DA) provided good categorization of metabolites according to the extraction solvents. Metabolites of the anthocyanin family, including delphinidin hexoside, delphinidin, 5-O-feruloylquinic acid, malvidin hexoside, malvidin-3-arabinoside, petunidin-3-arabinoside, and petunidin hexoside, were mainly detected in methanol fractions, whereas those of the flavonoid family, including chlorogenic acid, chlorogenic acid dimer, 6,8-di-C-arabinopyranosyl-luteolin, and luteolin were successfully prepared in the ethyl acetate fraction. Thus, metabolomic analysis of blueberry extracts allows for the simple profiling of whole and distinctive metabolites for future applications.

Comparison of Dioxin Analytical Results Between SP-2331 and DB-5MS Capillary Columns in the HRGC/HRMS Analysis (HRGC/HRMS에 SP-2331 및 DB-5MS 컬럼 사용시 다이옥신 결과의 비교)

  • In this study, the separation of toxic dioxin 2,3,7,8-congeners by DB-5MS and SP-2331 GC columns which are widely used in HRGC/HRMS analysis was examined. Through the dioxin analysis of column performance check standard solution and fly ash sample, the isomer specific separation of 2,3,7,8-substituted dioxins from tetra to hexa-isomers on DB-5MS and SP-2331 columns were studied. The effect of I-TEQ value by these columns was also studied. The total concentrations of toxic dioxins for the column performance check standard solution were 508.4 ng/mL analyzed by DB-5MS and 515.8 ng/mL analyzed by SP-2331, respectively. The I-TEQ value obtained by both columns was shown to be almost equivalent for the column performance check standard solution and fly ash sample.

Effect of MS Medium Strength and Nitrogen Concentration on Bulblet Formation and Growth of Lilium In Vitro (MS 배지 내 무기물 농도 및 질소 농도가 나리의 자구 형성과 비대에 미치는 영향)

  • To investigate the effects of MS medium strength and nitrogen concentration on bulblet formation from bulblet scale segment culture and bulblet growth from bulblet of Lilium oriental hybrid 'Casa Blanca', asiatic hybrid 'Mona', and longiflorum hybrid 'Hinomoto', 0.5∼2.0 strength of MS salts and 30∼120 mM nitrogen concentrations of MS medium were examined in vitro. The number of bulblets from bulb scale segment was favored in the strength of 0.5∼1.0 strength of MS salts or 30 mM total nitrogen concentration of MS medium in three cultivars. But the growth of bulblets formed in vitro was promoted in the 2 strength of MS medium or hight concentration of total nitrogen of MS medium up to 120 mM in three experimented cultivars.

  • Management science/operations research (MS/OR) educational software packages are widely used at the present time. Those software packages are expected to help students understand MS/OR techniques better. However, MS/OR educational software packages are often used as computational tools to obtain model solutions efficiently rather than as the tutoring software packages. Several possible reasons for the lack of effective tutoring capacity in MS/OR educational software packages are identified in this paper. The authors believe that the deficiency of tutoring capacity in those software is mainly due to technological limitations (computers and artificial intelligence) and the MS/OR professionals' perception about those software packages. Given technological limitations, feasible design and development approaches are provided to improve the tutoring effectiveness of MS/OR educational software packages.

Determination of Carbendazim in Commercial Agricultural Products by LC-MS/MS (LC-MS/MS를 이용한 농산물 중 카벤다짐 분석)

  • A rapid and precise method using LC-MS/MS was developed for carbendazim analysis in agricultural products. This compound was extracted with acetonitrile from agricultural products and cleaned up by solid-phase extraction procedure. The limit of detection and quantification were 0.001 mg/kg and 0.004 mg/kg, respectively. The mean recoveries and precision from 4 agricultural products, soybean sprout and mungbean sprout were in the range of 83.3-86.4% and 0.2-3.0% spiked at 1.0 mg/kg and those were in the range of 77.3-90.1% and 1.3-3.8% spiked at 0.02 mg/kg. The present method is faster and more precise compared with the multi-residue method of Korean Food Code. Therefore, we conclude that this method is suitable for carbendazim determination in a wide range of agricultural products.

Mass production of potato microtubers by bioreactor culture (바이오리액터 배양기에 의한 감자소괴경 대량 생산)

  • Highest increase of biomass was observed when tissue-cultured potato (Solanum tuberosum L. cv. Chubaek) shoots were cultured in a liquid medium containing 1/3 MS solution in a 18 L bioreactor, as compared to 1/4 and 1/2 MS solution. The medium containing 1/4 MS solution showed higher increase of shoot biomass than one containing 1/2 MS solution. Potato microtubers were formed when the medium was exchanged with the medium for microtuber formation and incubated under dark condition. The microtubers were observed first at some axillary buds one week after incubation under dark condition and then at most of the axillary buds by the end of 3 weeks. The 1.5 MS liquid medium and $20^{\circ}C$ were optimal conditions. By the end of 6 weeks, more 1,000 microtubers were formed in the 18 L bioreactor. Then, greened microtubers were harvested after one week culture under light condition.

Quantitative Analysis of Brain Metabolite Spectrum Depending on the Concentration of the Contrast Media in Phantom (팬텀 내 조영제 농도에 따른 뇌 대사물질 Spectrum의 정량분석)

  • Quantitative analysis of MR spectrum depending on mole concentration of the contrast media in cereberal metabolite phantom was performed. PRESS pulse sequence was used to obtain MR spectrum at 3.0T MRI system (Archieva, Philips Healthcare, Best, Netherland), and the phantom contains brain metabolites such as N-Acetyl Asparatate (NAA), Choline (Cho), Creatine (Cr) and Lactate (Lac). In this study, optimization of MRS PRESS pulse sequency depending on the concentration of contrast media (0, 0.1 and $0.3mmol/{\ell}$) was evaluated for various repetition time(TR; 1500, 1700 and 2000 ms). In control (cotrast-media-free) group, NAA and Cho signals were the highest at TR 2000 ms than at 1700 and 1500 ms. Cr had the highest peak signal at TR 1500 ms. When concentration of contrast media was $0.1mmol/{\ell}$, the metabolites were increased NAA 73%, Cho 249%, Cr 37% at TR 1700 ms compared with other TR, and also signal increased at $0.3mmol/{\ell}$, In $0.5mmol/{\ell}$ of contrast agent, cerebral metabolite peaks reduced, especially when TR 1500 ms and 2000 ms they decreased below those of control group. The ratio of metabolite peaks such as NAA/Cr and Cho/Cr decreased as the concentration of the contrast agent increased from 0.1 to $0.5mmol/{\ell}$. Authors found that the optimization of PRESS sequence for 0.3T MRS was as follows: low density of contrast agent ($0.1mmol/{\ell}$ and $0.3mmol/{\ell}$) made the highest signal intensity, while high density of contrast agent reveals the least reduction of signal intensity at 1700 ms. In conclusion, authors believe that it is helpful to reduce TR for acquiring maximum signal intensity.

Detection of Diarrhetic Shellfish Poisons by LC-MS/MS (설사성 패류독의 LC-MS/MS에 의한 분석)

  • Diarrhetic shellfish poisons (DSP) such as okadaic acid (OA), dinophysistoxin-1 (DTX1), pectenotoxin-1(PTX1), PTX2, PTX6 and yessotoxin (YTX) were determined simultaneously by LC-MS/MS and mouse bioassay in the shellfishes (oyster, mussel, Washington purple clam, ark shell, scallop and short necked clam) collected at Tongyeong, from March to September, 2006. Oyster and mussel were found to contain DSP (0.05${\sim}$0.1 MU/g) in March by mouse bioassay; however, no DSP components were detected on the LC-MS/MS. Also, a small amount of DTX1 (0.05 ${\mu}g/g$) in mussel (June) and OA (0.01${\sim}$0.02 ${\mu}g/g$) in 5 species of shellfishes(August) were determined by LC-MS/MS.

  • Adventitious shoot buds, without an intervening callus phase, were induced from stem explants of Trichosanthes kirilowii on MS medium supplemented with 2 mg/L BA. The culture medium, carbon source as well as plant growth regulators were found to influence further shoot multiplication and eventual shoot growth. A maximum number of shoots was obtained on a MS medium containing 0.5 mg/L BA and 3% sucrose. Shoot produced in vitro were rooted on half-strength phytagelled 1/2MS medium prior to transfer to green house condition.

Evaluation of the inhibitory effect of Gynostemma pentaphyllum extracts on CYP450 enzyme activities using LC-MS/MS

  • Gynostemma pentaphyllum (Thunb.) Makino extract, a natural product with a history of traditional use, has gained attention for its potential health benefits. This study aimed to investigate its effects on key cytochrome P450 (CYP) enzymes using LC-MS/MS. Human liver microsomes and cDNA-expressed CYP2C8, CYP2C9, CYP2C19, and CYP3A4 supersomes were employed. Enzyme activity was assessed based on the formation of CYP-specific marker metabolites. The resulting data showed that the extract exhibited inhibitory effects on CYP2C8, CYP2C9, CYP2C19, and CYP3A4. Thus, G. pentaphyllum extract may influence the pharmacokinetics of drugs metabolized by CYP2C8, CYP2C9, CYP2C19, and CYP3A4. These findings emphasize the importance of considering potential herb-drug interactions when incorporating this extract into therapeutic regimens or dietary supplements.
