Relationships among Students' Understanding of Genetics Topics, Meaningful Learning Orientation, and Reasoning Ability (생물학습에서 중학생들의 학습 성향, 논리적 사고력과 학업 성취도와의 관계 분석)

  • The purpose of this study was to examine the relationship among an achievement, a meaningful learning orientation and a reasoning ability. 149 third grade middle school students were tested. The achievement test was designed to measure students' interrelated understanding of genetics. A modified LPQ(Learning Process Questionnaires) was used to measure students' meaningful learning orientation. Students' reasoning ability were identified by the short version GALT(Group Assessment of Logical Thinking). Correlations between different variables were examined. Regression analyses were conducted to examine the predictive influence of meaningful learning orientation and reasoning ability on the achievement of students. And ANCOVA was used to identify the interaction of these variables on students' achievement. Students did not understand well enough the concepts of genetics. Meaningful learning orientation indicated a significant gender difference. Girls tend to do more meaningful learning than boys(p<.05). Many students(48.76%) were at the transitional cognitive level. Results of correlations indicated that students' attainment of meaningful understanding was significantly and positively related with a meaningful learning orientation and a reasoning ability. But there was no significant correlation between students' meaningful learning orientation and reasoning ability. Regression analyses indicated that learning orientation and reasoning ability were able to predict the achievement of students. They predicted better on solving genetics problem than understanding genetics problem. Results of ANCOVA showed that the test scores of genetics were significantly different according to not only learning orientation levels but also cognitive levels. But, there was no interaction between learning orientation and cognitive levels. Within the transitional and formal cognitive level, the meaningful learners performed significantly better than the rote learners on the test of genetics.

Cultivation of Cauliflower Mushroom (Sparassis crispa) Using Coniferous Sawdust-based Media with Barley Flours (보릿가루가 첨가된 침엽수 톱밥을 이용한 꽃송이버섯 재배)

  • Cauliflower mushroom (Sparassis crispa) is an edible mushroom that shows remarkably high contents of $1,3-{\beta}-D-glucan$ compared to other edible mushrooms. The mushroom is known to give high antitumor and immunologic activities, thus the mushroom is recently cultivated in Japan and Korea. However, cultivation methods are being kept in secret or under patents by some companies with complicated procedures. This study was conducted to solve the problem by providing a simple method for the cultivation of cauliflower mushroom. We could produce the mushroom using sawdust-based media of Larix leptolepis, Pinus densiflora and P. koraiensis with addition of barley flours and sugar. The ratio of the sawdust : barley flours : sugar used for this study was 80:20:3 in dry-weight base, and the water content was adjusted as 65%. The productivity was somewhat different among the species of sawdust media and the strains of Sparassis crispa for the cultivation, and the mushroom production reached up to 177g from 650g base(productivity : 27%). Thus, this medium is strongly recommended as a practical cultivation method for cauliflower mushroom. By the way, further study for the determination of adequate concentration of barley flour and the selection of proper strain for each sawdust species is needed.

Cellular viability and genetic expression of human gingival fibroblasts to zirconia with enamel matrix derivative ($Emdogain^{(R)}$)

  • PURPOSE. The objective of this study was to investigate the biologic effects of enamel matrix derivative (EMD) with different concentrations on cell viability and the genetic expression of human gingival fibroblasts (HGF) to zirconia surfaces. MATERIALS AND METHODS. Immortalized human gingival fibroblasts (HGF) were cultured (1) without EMD, (2) with EMD $25{\mu}g/mL$, and (3) with EMD $100{\mu}g/mL$ on zirconia discs. MTT assay was performed to evaluate the cell proliferation activity and SEM was carried out to examine the cellular morphology and attachment. The mRNA expression of collagen type I, osteopontin, fibronectin, and TGF-${\beta}1$ was evaluated with the real-time polymerase chain reaction (RT-PCR). RESULTS. From MTT assay, HGF showed more proliferation in EMD $25{\mu}g/mL$ group than control and EMD $100{\mu}g/mL$ group (P<.05). HGFs showed more flattened cellular morphology on the experimental groups than on the control group after 4h culture and more cellular attachments were observed on EMD $25{\mu}g/mL$ group and EMD $100{\mu}g/mL$ group after 24h culture. After 48h of culture, cellular attachment was similar in all groups. The mRNA expression of type I collagen increased in a concentration dependent manner. The genetic expression of osteopontin, fibronectin, and TGF-${\beta}1$ was increased at EMD $100{\mu}g/mL$. However, the mRNA expression of proteins associated with cellular attachment was decreased at EMD $25{\mu}g/mL$. CONCLUSION. Through this short term culture of HGF on zirconium discs, we conclude that EMD affects the proliferation, attachment, and cell morphology of HGF cells. Also, EMD stimulates production of extracellular matrix collagen, osteopontin, and TGF-${\beta}1$ in high concentration levels. CLINICAL RELEVANCE. With the use of EMD, protective barrier between attached gingiva and transmucosal zirconia abutment may be enhanced leading to final esthetic results with implants.

  • The "tang[tɑ:ŋ]" in Korean pronounciation means the beverage made of boiled medicinal herbs. The"Jeho-tang", the name of Drink in this abstract, is described in a variety of medical books including the "Dongeui Bogam" as being effective for illness from the summer heat in promoting digestion, curing the heatstroke and bringing it to a halt. The Drink was used as the Royal gifts granted to retainers and royal families on the Day of Dano-festival on the fifth of the fifth month of the year according to the lunar calendar, the items of encouragement for those who worked hard in sacrifices, and the awards for students of Confucianism who were proficient in their learning. The Jeho-tang used in this study was scientifically cooked again after a long time in history through looking at the methods written in the documents concerned with the Drink such as the "Dongeui Bogam" and the "Taste of Korea". In preparation of the medicinal herbs for the Drink, the powder of thinner than 30 mesh of the "Prunus mume", which is a species of Asian plum in the family of Rosaceae, and those of 50 mesh of the "Santalum album", which is the fragrant wood of trees in the genus Santalum and the "Amomum Xanthioides", which is produced in Vietnam and is the name of a kind of herb medicines, being very effective in the desease caused from heatstrare, were used. The sugar concentration of the honey boiled down long time at low heat was $82.43^{\circ}Bx$. When cooking in a double boiler, the inner part of the liquid for the Drink was kept at $80^{\circ}C$ for 12 hours to make it finished in a state of ointment. In the general composition of the finished Jeho-tang, the moisture content was 24.4%, 1.3% of crude fat, 1.4% crude protein and 0.7% ash, along with pH3.2. The acceptance on the whole was come out to be the highest in the sample diluted with the drinking water of 7-fold of the Jeho-tang, indicating that the 7-fold's addition of water was optimum level for drinking. In the Drink cooked by a vaccum pressure extractor for herb medicine, which was developed to improve the art of cooking, the longer the time of pressure was, the less the heterogeneous feeling at tongue was and the more the glossiness of the Drink was. The Jeho-tang cooked under pressure for 7 hours received an excellent evaluation in its acceptability in every way.

Effects of Lycii Fructus on the Ovariectomized Osteoporosis of Rats (구기자(枸杞子)가 난소적출(卵巢摘出) 흰쥐의 골조직대사(骨組織代謝)에 미치는 영향(影響))

  • Objectives: Osteoporosis is one of the most common metabolic bone disorders in the menopausal stage, a disease signified by decrease of bone mass. The purpose of this study was to learn how Lycii Fructus influences SD rats with osteoporosis caused by the removal of the ovaries. Methods: After the removal of the ovaries from 24 female Sprague-Dawley rats, they were divided into a normal group, a control group, and a Lycii Fructus (25mg/100g) administration group. After 8 weeks, the rats were sacrificed and their weights, albumin, AST, ALT, ALP, total-cholesterol, triglyceride, P, Ca, T4, E2, weight of a femur, ash content of the tibia, and area and thickness of trabecular bone were measured. Results: Regarding the density of ALT, ALP, and estradiol from serum, the Lycii Fructus group was significantly decreased compared to the control group, and the density of phosphorous and calcium also decreased slightly. Moreover, albumin, triglyceride, and T4 showed an increasing propensity, but AST and total-cholesterol showed a decreasing propensity, but not significant. Regarding the variation of bone, femur weight and ash content of tibia, the Lycii Fructus group was increased compared to the control group but significance wasn't shown. The Lycii Fructus group increased significantly over the control group in area and thickness of trabecular bone and osteoclast number. Conclusions: From the results of the above study, Lycii Fructus should be effective for osteoporosis treatment and prevention.

Gene Expression Profiling of Acetaminophen Induced Hepatotoxicity in Mice

  • Microarray analysis of gene expression has become a powerful approach for exploring the biological effects of drugs, particularly at the stage of toxicology and safety assessment. Acetaminophen (APAP) has been known to induce necrosis in liver, but the molecular mechanism involved has not been fully understood. In this study, we investigated gene expression changes of APAP using microarray technology. APAP was orally administered with a single dose of 50 mg/kg or 500 mg/kg into ICR mice and the animals were sacrificed at 6, 24 and 72 h of APAP administration. Serum biochemical markers for liver toxicity were measured to estimate the maximal toxic time and hepatic gene expression was assessed using high-density oligonucleotide microarrays capable of determining the expression profile of >30,000 well-substantiated mouse genes. Significant alterations in gene expression were noted in the liver of APAP-administered mice. The most notable changes in APAP-administered mice were the expression of genes involved in apoptosis, cell cycle, and calcium signaling pathway, cystein metabolism, glutatione metabolism, and MAPK pathway. The majority of the genes upregulated included insulin-like growth factor binding protein 1, heme oxygenase 1, metallothionein 1, S100 calcium binding protein, caspase 4, and P21. The upregulation of apoptosis and cell cycle-related genes were paralleled to response to APAP. Most of the affected gene expressions were returned to control levels after 72 hr. In conclusion, we identified potential hepatotoxicity makers, and these expressions profiling lead to a better understanding of the molecular basis of APAP-induced hapatotoxicity.

The Effect of Ear Reflexotherapy on Back Pain of Working Women in Middle Age (귀 자극요법 적용이 중년 여성 근로자의 요통에 미치는 효과)

  • Purpose: The purpose of this study was to investigate the effect of ear reflexotherapy on back pain of working women in middle age. Methods: The quasi-experimental, nonequivalent control group pretest-posttest design was used for the study. Total of 56 participants composed of experimental group (n=26) and control group (n=30), who were working in four hospitals in urban area. The degree of back pain after ear reflexotherapy was measured by VAS. The data were analyzed by frequency, percent, t-test and $X^2$-test. Results: The degree of back pain after ear reflexotherapy in the experimental group was significantly reduced, while there was no difference in control group. There was a significant difference between the experimental group and control group in the degree of back pain after ear reflexotherapy (t=3.702, p=.001). Conclusion: Application of ear reflexotherapy was effective for employed women in middle age and it needs to be further evaluated among the other population.

DNA Ploidy and S-phase Fraction Analysis in Paediatric B-cell Acute Lymphoblastic Leukemia Cases: a Tertiary Care Centre Experience

  • DNA ploidy is an important prognostic parameter in paediatric B-ALL, but the significance of the S-phase fraction is unclear. In present study, DNA ploidy was assessed in 40 pediatric B-ALL cases by flow cytometry. The DI (DNA index) and percentage of cells in S-phase were calculated using Modfit software. Aneuploidy was noted in 26/40 (65%) cases. A DI of 1.10-1.6 (hyperdiploidy B) was noted in 20/40 (50%) and 6/40 (15%) had a DI>1.60 (triploid and tetraploid range). Some 14/40 (35%) cases had a diploid DI between 0.90-1.05. None of the cases had a DI <0.90 (hypodiploid) or in the 1.06-1.09 (hyperdiploid A) range. The mean S-phase fraction was 2.6%, with 24/40 (60%) having low and 16/40 (40%) high S-phase fractions. No correlation was noted with standard ALL risk and treatment response factors with DI values or S-phase data, except for a positive correlation of low S-phase with high NCI risk category (p=0.032). Overall frequency of hyperdiploidy in our cohort of B-ALL patients was very high (65%). No correlation between hyperdiploidy B and low TLC or common B-phenotype was observed in our study as 42% cases with DI 1.10-1.6 had TLC> $50{\times}10^9$ and 57.1% CD 10 negativity. The study also highlighted that S-phase fraction analysis does not add any prognostic information and is not a useful parameter for assessment in ALL cases. However, larger studies with long term outcome analysis are needed to derive definitive conclusions.

Evaluation of the Knowledge and Perceptions with Regards to Pictorial Health Warnings on Tobacco Products among Tobacco Users Diagnosed with Head and Neck Carcinoma: a Study from the Kumaon Hills of India

  • Background: Tobacco products continue to be used in large quantities in India despite the mandatory inclusion of pictorial health warnings (PHWs) on all tobacco packaging. The circumstances as to how people could continue the use of tobacco to the point of developing head and neck cancer despite enhanced awareness about the ill effects of tobacco is the main focus of this study. Materials and Methods: This study concerned patients with least 5-years history of tobacco use, having been diagnosed with histopathologically proven malignancies of the hypopharynx, larynx, oropharynx and oral cavity presenting at the Government Medical College-Haldwani, Nainital, India. A total of 183 patients were eligible for inclusion during July 1 2013 - June 30 2014. Of these, 59 patients used smoked tobacco exclusively, 22 patients used smokeless tobacco exclusively, and 102 patients used both forms of tobacco. Among users of smoked forms, 75.2% (n=121) were beedi users, and 24.8% (n=40) were cigarette users. Patients were asked direct questions as to whether they had noticed the presence of PHWs upon tobacco products. The reasons as to why PHWs were not effective in stopping the patients from tobacco use were investigated. Results: Of the 183 patients, 146 reported being aware about the presence of PHWs, and when they were asked reasons as to why they continued tobacco despite being aware of ill-effects, the commonest reason chosen (by 53.4%) was that patients had not regarded themselves as using tobacco heavy enough to cause cancer. Among the 36 patients who reported as being oblivious to the presence of PHWs on tobacco products, 63.9% reported that the products they used never displayed any PHWs, and 36.1% reported never having paid attention to the packaging. The awareness about PHWs was higher among cigarette smokers in comparison to beedi smokers (100% vs 76.1%, p=0.0002). Conclusions: Locally produced and marketed tobacco products such as beedis and oral tobacco often fail to display PHWs. The presence of PHWs without doubt enhances awareness about the carcinogenic risks of tobacco. However, enhanced awareness alone may not be enough, and as elucidated by this study, some persons continue to use tobacco to the point of developing malignancies. The need of the hour is the implementation of legal and economic sanctions discouraging the use of tobacco products.

Evaluation of the Health Status of the Aged in rural Area by Anthropometry and Clinical Laboratory Tests (농촌지역(農村地域) 노인(老人)들의 신체계측(身體計測) 및 임상검사(臨床檢査)에 의한 건강상태(健康狀態) 평가(評價))

  • To evaluate the health status of the aged in rural area, 476 men and women of 65 years old and older among the residents of Gunwee County in Kyungpook were examined at the county health center between 4th and 22nd of June, 1984. Physical examination included measuring height, weight, chest circumference and blood pressure and chest x-ray. Laboratory tests were done for serum cholesterol, glucose and GOT, and urine glucose and protein. The mean heights of male and female were 160${\pm}$7.0cm and 147.7${\pm}$6.9cm, respectively. The height decreased as the age increased. The mean body weight of male was 53.5${\pm}$7.8kg and 46.7${\pm}$7.5kg for female and the older, the eighter. The mean chest circumference of male was 85.4${\pm}$7.5cm and 79.5${\pm}$79cm for female and it had no remarkable differences by age group. The average height, body weight, chest circumference, and serum cholesterol level of the age 65 years and older in rural area were lower than the standard this may be due to undernutrition of the aged. The mean diastolic and systolic blood pressure of male were 75.9${\pm}$13.5 mmHg/126.3${\pm}$24.1 mmHg and those for female were 73.1${\pm}$13.9 mmHg/125.8${\pm}$32.4 mmHg. Both male an d female in the age group of 65-69 years showed the lowest mean blood pressure. Among the study population 18.3% the borderlin hypertension and 14.3% had hypertension. Average hemoglobin level of the male(12.5${\pm}$0.8 g/dl) was higher than that of the female(11.6${\pm}$0.8 g/dl) (p < 0.01). Mean serum G.O.T. level was 25.2${\pm}$8.7 IU for male and 23.6${\pm}$7.5 IU for female and all of the study population were within the normal range. Mean serum glucose level was 110.4${\pm}$16.0 mg/dl for the male and 110.5${\pm}$15.7 mg /dl for the female and 41.4% of the study population were above the normal limit of serum glucose level. However, there was only one person she had glycosuria. Hyperglycemia could be due to non-fasting before the test. Two of the study population had proteinuria and five persons (1.05%) were diagnosed as pulmonary tuberculosis by the chest x-ray.
