The effect of plant hormones on the growth of lichen-forming fungi (LFF) was evaluated. The use of 2,3,5-triiodobenzoic acid and indole-3-butyric acid resulted in a 99% and 57% increase in dry weight of the lichen-forming fungus Nephromopsis ornata. The results suggest that some plant hormones can be used as inducers or stimulators of LFF growth for large-scale culture.
Somatic embryos were formed from calli obtained from axillary shoots (raised from nodal segments of glasshouse-grown plants under aseptic conditions), internodal segments (from in vitro-raised plants), and root and coty-ledonary leaf segments (from in vitro-raised seedlings) after 8 weeks of initial culture. Embryo formation was the highest (97.33%) from cotyledonary leaf callus on Mura-shige and Skoog's (MS) medium containing kinetin (KN) (3 mg/L). Somatic embryo induction was lesser with different combinations of auxins while it increased to 100% in internodal segment and cotyledonary leaf calli with 6-benzyladenine (BA) (2mg/L) along with 2,3,5-triiodobenzoic acid (TIBA) (2mg/L). The shoots were induced from somatic embryos raised from root, coty-ledonary leaf and internodal segment calli grown on MS medium containing BA in combination with indole-3-acetic acid (IAA). Maximum of 66.67% cultures formed shoots on MS medium containing BA (1mg/L) in combination with IAA (2mg/L). The shoots raised from somatic embryos were rooted on MS medium supplemented with indole-3-butyric acid (IBA) (2mg/L). The plantlets transferred to the field showed 70% survival rate after one year.
This work was carried out to elucidate effects of endogenous and exogenous IAA transport on adventitious root formation in Phaseolus vulgaris hypocotyl cuttings. For inverted or normal incubation in distilled water, the adventitious root is always formed at the morphological base but not at the morphological apex. For inverted incubation, in both distilled water and certain chemical solution, the root formation is retarded more at the first stage (0-24 hr) than at the second stage (24-48 hr). When p-chlorophenoxyisobutyric acid (PCIB) was applied to the cuttings at the first stage, theroot formation was inhibited more than at the second stage. Treatment of 2,3,5-triiodobenzoic aicd (TIBA)markedly inhibited the adventitious root formation in Phaseolus vulgaris hypocotyl cuttings. This inhibition influenced the root according to the applied stage and period. Therefore, the root formation is more related to the stage of root primordium formation than to the stage of root elongation from the primordium. Inhibition of auxin transport oraction by TIBA or PCIB could also be reversed when hypocotyl cuttings are incubated in exogenously applied IAA solution.
Three growth regulators, TIBA (2. 3. 5-Triiodobenzoic acid). ABA(Abscisic acid) and DGLP were sprayed on soybean plants sown on April 25 and May 10 to investigate those effect on growth and yield of Hwangkeumkong cultivar. TIBA or ABA reduced stem length. and lodging. however. increased stem diamater podding rate. number of pods and seeds per plant, and seed yield. Among 3 growth regulators TIBA was most effective to healthy growth and to increase of seed yield. Optimum treatment method for healthy plant growth and higher grain yield was 2-3 times spray with 5-day interval from 6 leaf stage (V6) of soybean plants. Soybean seed yield in the plot of TIBA treatment with 3 times from 6 leaf stage was 20% higher both in early and ordinary seeding field than those of non-treatment plots.
Peach (Prunus persica L.) is a model species for stone fruit studies within the Rosaceae family. Auxin plays an important role in the development of peach fruit. To reveal the distribution of auxin in the tissues of peach fruit, immunohistochemical localization of IAA was carried out in the seed, mesocarp, and endocarp in developing peach fruit using an anti-indole-3-acetic acid (anti-IAA) monoclonal antibody. A strong IAA signal was observed throughout the outer and inner integument during peach fruit development, and the distribution was zonal. The IAA signal was mainly focused in mucilage layers in the outer integument. The outer integument may function to produce or store IAA in the seed; a strong IAA signal was detected in the cells around the vascular tissue, whereas a weak IAA signal was located in the vascular tissues. In the mesocarp, the cells around the vascular bundle tissue gave rise to an IAA signal that increased in the late phase of fruit growth, which coincided with a significant increase in fruit growth. The distribution of IAA, however, was changed when fruit was treated with auxin transport inhibitors NPA (1-N-naphthylphthalamic acid) or TIBA (2, 3, 5-triiodobenzoic acid); in mesocarp tissues, an IAA signal was detected mainly in vessels of the treated fruit. During the critical period of endocarp lignification, the vessel lignification process was negatively correlated with IAA signal. The present results confirmed that the distribution of IAA was different in various tissues of peach fruit according to the developmental stage. This research provides cytological data for further study of the regulatory mechanism of auxin in peach fruit.
It has been known that brassiolide (BL) increased the positive gravitropic response and ethylene production in maize roots. This study examined the relationship between the BL-induced gravitropic response and ethylene Production. The ethylene production was inhibited to about 90% of the control by the treatment of $10^{-4}$ M aminoethoxyvinylglycine (AVG), the ethylene synthesis inhibitor. However, the gravitropic response did not show any significant changes compared to the control at $10^{-4}$ M AVG. In the case of treatment of AVG with BL, the ethylene production decreased to 60% of the control. However, the gravitropic response increased to the level which was induced by BL. Cobalt ions, another ethylene biosynthesis inhibitor, inhibited ethylene production, but not gravitropic response. When roots were treated with BL and cobalt ions, they showed the inhibition of ethylene production and promotion of gravitropic response. To elucidate the possibility that the effect of BL is related to auxin transport, roots were treated with TIBA (2,3,5-triiodobenzoic acid), an auxin transport inhibitor. Both treatment of TIBA alone and TIBA with BL stimulated ethylene production to about 96% and 132%, respectively. However, gravitropic response was completely inhibited in both treatments. Further, roots treated with BL in the presence of TIBA and IAA showed a negative gravitropic response, which means that IAA accumulates in the upper side of horizontal roots. Root elongation was also stimulated in this treatment. Taken together, these results suggest that BL might affect the differential distribution of internal IAA on roots, causing the regulation of positive gravitropic response.
