본 연구팀은 태백산 지역에서 토양 시료들을 채취하여 다양한 균주들을 분리 탐색하는 과정에서 BCNU 2001 균주를 분리하였다. 분리균주의 생화학적 특징과 16S 리보좀 RNA 유전자 염기서열 분석 결과, 분리균주가 Pseudomonas aeruginosa에 속함이 확인되었다. 분리균주 BCNU 2001의 상등액은 다양한 세균과 진균에 대해 항균활성이 있었으며 특히 분리균주 BCNU 2001는 Micrococcus luteus, Proteus mirabilis, Proteus vulgaris, 그리고 Aspergillus niger의 생육을 크게 저해할 수 있었으며, Micrococcus luteus, Proteus mirabilis, Proteus vulgaris, 그리고 Aspergillus niger에 대하여 각각 18.5mm, 19.0mm, 17.0mm 그리고 13.5mm 크기의 저해환을 나타내었다. 분리균주 BCNU 2001의 핵산 분획물과 디클로로메탄 분획물은 세균에 대해 높은 항균력을 보였으며 디클로로메탄 분획물과 에틸아세테이트 분획물은 진균에 대해 높은 항균력을 보였다. 또한 Pseudomonas sp. BCNU 2001 균주는 그람양성 세균, 그람음성 세균 그리고 진균을 포함한 다양한 미생물들에 대하여 항균력이 있는 항균 펩타이드를 가지고 있음이 확인되었다. 실험을 통하여 얻은 결과는 Pseudomonas sp. BCNU 2001 균주의 유용성에 대한 예비적인 기초를 제공한다.

K/BxN serum can transfer arthritis to normal mice owing to the abundant autoantibodies it contains, which trigger innate inflammatory cascades in joints. Little is known about whether gut-residing microbes affect host susceptibility to autoantibody-mediated arthritis. To address this, we fed C57BL/6 mice with water containing a mixture of antibiotics (ampicillin, vancomycin, neomycin, and metronidazol) for 2 weeks and then injected them with K/BxN serum. Antibiotic treatment significantly reduced the amount of bacterial genomic DNA isolated from fecal samples, in particular a gene encoding 16S ribosomal RNA derived from segmented filamentous bacteria. Arthritic signs, as indicated by the arthritic index and ankle thickness, were significantly attenuated in antibiotic-treated mice compared with untreated controls. Peyer's patches and mesenteric lymph nodes from antibiotic-treated mice contained fewer IL-17-expressing cells than those from untreated mice. Antibiotic treatment reduced serum C3 deposition in vitro via the alternative complement pathway. IL-$17^{-/-}$ congenic C57BL/6 mice were less susceptible to K/BxN serum-transferred arthritis than their wild-type littermates, but were still responsive to treatment with antibiotics. These results suggest that gut-residing microbes, including segmented filamentous bacteria, induce IL-17 production in GALT and complement activation via the alternative complement pathway, which cause the host to be more susceptible to autoantibody-mediated arthritis.
Shindari is a traditional fermented drink of Jeju in Korea, which is made with boiled barley and nuruk for short fermentation periods. This study determined chemical, microbiological, and sensory characteristics of the modified Shindari with 15% carrots as an additive (carrot Shindari), and this study compared it with a traditional Shindari as a control. After fermentation at $30^{\circ}C$ for a day, the pHs of the carrot Shindari and traditional Shindari largely decreased, and the total acidities increased in both of the Shindari. The significantly higher scores of Hunter's color values were observed more in carrot Shindari than in traditional Shindari. Also, carrot Shindari (0.4954 g/100 g) had a significantly higher content of vitamin C than traditional Shindari (0.0030 g/100 g). The most abundant free sugar and organic acid were glucose and lactic acid, respectively, in both of the Shindari. The total numbers of bacteria, fungi and lactic-acid bacteria in both samples increased by log 3 CFU/mL after fermentation. Based on 16S ribosomal RNA gene analysis, the dominant lactic-acid bacteria was Pediococcus acidilactici in both samples. The DPPH (1.1-diphenyl-2-picrylhydrazyl) radical scavenging activity of carrot Shindari (60.13%) was higher than that of traditional Shindari (23.70%). In sensory evaluations (taste, flavor, color, and overall acceptance), the carrot Shindari had higher scores in all these values. In this study, the modified Shindari with carrot presenting high sensory characteristic as well as chemical and microbiologic characteristics provide an opportunity to improve the application of a traditional fermented drink of Jeju, Shindari.
장내 미생물은 사람의 장에 거주하는 미생물 집단으로 자신들의 효소를 이용해 사람에게 직접적인 에너지원이 되는 대사산물을 만들기도 하고 병원성 미생물의 침입에 대응하기도 하며 소화관의 구조와 기능유지 및 면역계의 발달에 영향을 미치는 등의 중요한 기능을 수행한다. 최근 다양한 분석기술을 통해 사람의 장에 있는 다종 및 다수의 미생물의 구성을 관찰하였고 그 결과 특정 질병을 지닌 사람의 장내 미생물 구성은 건강한 사람의 일반적인 구성과 다르다는 것을 확인하였다. 이러한 발견을 바탕으로 건강한 사람의 대변에 있는 장내 미생물을 질병을 지닌 수여자에게 이식하는 fecal microbiota transplantation (FMT) 시술이 각광을 받기 시작하였다. FMT는 장내 미생물의 구성변화로 인해 나타나는 여러 질병에 치료제로 적용할 수 있는데 특히 재발률이 높은 Clostridium difficile 감염에서 활발한 연구가 이루어지고 있다. 이 논문은 지금까지 밝혀진 장내 미생물과 관련된 질병들의 종류와 실제 질병에 FMT를 적용한 사례 등을 정리하고 장내 미생물을 이용하여 현재 연구개발 중인 치료제나 진단도구의 예를 기술함으로써 장내 미생물의 사람의 건강증진과 질병치료를 위한 차세대 생물학적 치료제로서의 가능성을 고찰하고자 한다.
세균성 피부염은 항생제 치료가 필요한 흔한 질환이다. 최근 항생제 내성 세균이 전 세계적으로 증가하고 있는 추세이다. 본 연구에서는 개 피부염 병변에서 분리한 S. pseudintermedius의 항생제 내성 유전자 발현을 조사하고, 이를 디스크 감수성 시험결과와 비교하였다. 총 7개의 S. pseudintermedius 균주가 연구에 포함되었다. 세균 동정은 16S rDNA 염기 서열 분석으로 실시하였다. 조사 결과 분리한 S. pseudintermedius 모두가 1개 이상의 항생제 내성유전자(mecA, blaZ and aac[6']/aph[2"])를 보유하였다. 전체 분리균주가 blaZ 유전자에 양성을 보였으나, 두 개 균주만이 amoxicillin/clavulanate에 내성을 나타냈다. Gentamicin 내성을 나타낸 5개 균주 중, 1개는 aac(6')/aph(2") 유전자에 대한 PCR 검사에서 음성을 나타냈다. 이 결과는 내성 유전자 검출 PCR 기법과 디스크 감수성 시험 기법이 항생제 내성을 검사하는 데에 있어 상호보완적인 관계를 가짐을 제시한다.
본 연구는 2023년 4~5월 전라남도 영광군 칠산도 주변 해역에서 채집된 뱅어과(Salangidae) 자치어 총 6개체를 대상으로 분자 동정과 발달단계별 형태특징을 상세히 기재하였다. MtDNA COI 또는 16S 영역을 대상으로 분자 분석을 진행한 결과, 자치어 6개체는 도화뱅어(Neosalanx anderssoni) 성어와 유전거리 0~0.2%의 차이를 보여 도화뱅어로 동정되었다. 도화뱅어 자치어 6개체는 모두 측편형의 가늘고 긴 체형을 보였다. 전기자어~중기자어(10.24 mm NL, 13.37 mm SL) 시기에는 복강의 배쪽에 타원형의 흑색소포가, 복강 등쪽에는 옅은 점 모양의 흑색소포가 열을 이루었다. 반면 후기자어~치어 시기(22.12 mm SL, 28.43 mm SL)에는 복강 배쪽의 흑색소포가 사라지고 복강 등쪽의 흑색소포가 수적으로 증가하여 1열로 나타났다. 또한, 후기자어 시기부터는 꼬리지느러미에 크고 짙은 검은 반점 2개가 대칭적으로 나타났다. 본 연구 결과는 이전의 연구 결과(Kim and Park, 2002)와 달리 도화뱅어가 연안을 산란 또는 성육장으로 이용할 수 있음을 시사한다.
강원도를 비롯한 전국에서 발효식품 44점으로부터 유해균에 대한 생육억제 활성이 우수한 고초균주를 분리 및 동정하였으며, 생육억제 활성이 가장 우수하였던 Bacillus amyloliquefaciens MBE1283로 명명된 균주의 유해균에 대한 생육억제 활성, 전분분해 효소활성 및 생육특성을 조사하였다. Escherichia coli O157:H7 KCCM40406을 비롯한 8종류의 유해균에 대한 생육억제 활성을 평가한 결과, 막걸리에서 분리된 B. amyloliquefaciens MBE1283 균주는 다른 B. amyloliquefaciens 균주들에 비해 유의적으로 높은 유해균 생육억제 활성을 나타내었다. B. amyloliquefaciens MBE1283 균주의 ${\alpha}$-amylase 효소활성은 대조구 균주들과 비교하였을 때 현저히 낮았으나 glucoamylase 효소활성은 대조구로 사용한 균주와 유사한 수준이었다. B. amyloliquefaciens MBE1283 균주의 최적 생육온도는 $45^{\circ}C$로서 대조구 균주보다 온도에 대한 내성이 있는 것으로 판단되었으며, 균체 성장에 적합한 최적 배지는 pH 6이었다. 본 연구를 통하여 막걸리로부터 분리된 B. amyloliquefaciens MBE1283 균주는 여러 종류의 유해균에 항균활성을 지니며 높은 온도와 낮은 pH에 대하여 내성을 지닌 것을 알 수 있었다.

To elucidate the biodiversity of plant growth-promoting rhizobacteria (PGPR) in Korea, 7,638 bacteria isolated from the rhizosphere of plant species growing in many different regions were screened. A large number of PGPR were identified by testing the ability of each isolate to promote the growth of cucumber seedlings. After redundant rhizobacteria were removed via amplified rDNA restriction analysis, 90 strains were finally selected as PGPR. On the basis of 16S ribosomal RNA sequences, 68 Gram-positive (76%) and 22 Gram-negative (24%) isolates were assigned to 21 genera and 47 species. Of these genera, Bacillus (32 species) made up the largest complement, followed by Paenibacillus (19) and Pseudomonas (11). Phylogenetic analysis showed that most of the Grampositive PGPR fell into two categories: low- and high- G+C (Actinobacteria) strains. The Gram-negative PGPR were distributed in three categories: ${\alpha}$-proteobacteria, ${\beta}$- proteobacteria, and ${\gamma}$-proteobacteria. To our knowledge, this is the largest screening study designed to isolate diverse PGPR. The enlarged understanding of PGPR genetic diversity provided herein will expand the knowledge base regarding beneficial plant-microbe interactions. The outcome of this research may have a practical effect on crop production methodologies.
We cultured a tubular marine green alga, originally identified as Capsosiphon groenlandicus (J. Agardh) K.L. Vinogradova, from Amaknak Island, Alaska. The alga had an alternation of heteromorphic generations in which tubular monoecious fronds produced quadriflagellate zoospores and/or biflagellate isogametes. The gametes fused to produce cysts or Codiolum-like zygotes with long, tortuous stalks. Cysts and codiola produced 8-16 aplanospores, which germinated in situ to yield upright fronds. Fronds arising from both aplanospores and zoospores displayed a distinctive development in which non-septate colorless rhizoids from the base of the initially uniseriate, Ulothrix-like filament were transformed into septate uniseriate Ulothrix-like photosynthetic filaments. These transformed filaments then developed new basal non-septate rhizoids. This pattern of rhizoids becoming filaments, which then produced new rhizoids, was repeated to yield a tuft of up to 50 fronds. Periclinal and longitudinal divisions occurred in each filament, starting basally, until the mature tubular thallus was achieved. Pyrenoid ultrastructure revealed several short inward extensions of chloroplast lamellae, each of which was surrounded by pyrenoglobuli. Analysis of ribosomal SSU and ITS sequences placed this alga in the family Ulotrichaceae, order Ulotrichales, together with but as a distinct species from North Atlantic Capsosiphon groenlandicus. Analysis of a partial ITS sequence from authentic Capsosiphon fulvescens, the current name of the type of the genus Capsosiphon, indicated that neither our material nor C. groenlandicus belongs in that genus, and we propose a new genus, Pseudothrix, to accommodate both species. We propose P. borealis for the North Pacific entity formerly called C. groenlandicus and make the new combination P. groenlandica for the Atlantic species.
Background : Infection caused by rapidly growing mycobacteria (RGM) is not uncommon, and the prevalence of RGM infection has been increasing. Clinical diagnosis is difficult because there are no characteristic clinical features. There is also no standard antibiotic regimen for treating RGM infection. A small series of patients with RGM infections was studied to examine their treatments and outcomes. Methods : A total of 5 patients who had developed postoperative infections from January 2009 to December 2010 were retrospectively reviewed. Patients were initially screened using a mycobacteria rapid screening test (polymerase chain reaction [PCR]-reverse blot hybridization assay). To confirm mycobacterial infection, specimens were cultured for nontuberculous mycobacteria and analyzed by 16 S ribosomal RNA and rpoB gene PCR. Results : The patients were treated with intravenous antibiotics during hospitalization, and oral antibiotics were administered after discharge. The mean duration of follow-up was 9 months, and all patients were completely cured of infection with a regimen of a combination of antibiotics plus surgical treatment. Although none of the patients developed recurrence, there were complications at the site of infection, including hypertrophic scarring, pigmentation, and disfigurement. Conclusions : Combination antibiotic therapy plus drainage of surgical abscesses appeared to be effective for the RGM infections seen in our patients. Although neither the exact dosage nor a standardized regimen has been firmly established, we propose that our treatment can provide an option for the management of rapidly growing mycobacterial infection.
