Expression of ssrA in non-pathogen-induced adaptation in the oral cavity through signal exchange with oral pathogens

  • Introduction: This study was conducted to evaluate ssrA expression resulting from adaptation of Escherichia coli (E. coli) to oral pathogens through signal exchange. Materials and Methods: Human cell lines Hep2 and HT29, wild-type E. coli (WT K-12), ssrA knock-out E. coli (${\Delta}K$-12), and Scleropages aureus (S. aureus) were used. A single culture consisting of Hep2, HT29, WT K-12, and ${\Delta}K$-12, and mixed cultures consisting of Hep2 and WT K-12, Hep2 and ${\Delta}K$-12, WT K-12 and S. aureus, ${\Delta}K$-12 and S. aureus, and Hep2, WT K-12, and S. aureus were prepared. For HT29, a mixed culture was prepared with WT K-12 and with WT K-12 and S. aureus. Total RNA was extracted from each culture with the resulting expression of ssrA, nuclear factor kappa-light-chain-enhancer of activated B cells (NF-${\kappa}B$), and p53 was evaluated by Reverse transcription polymerase chain reaction (RT-PCR). Results: The expression of ssrA in a single culture of WT K-12 was lower than that observed in the mixed culture of WT K-12 with S. aureus. Greater ssrA expression was observed in the mixed culture of WT K-12 with Hep2 than in the single culture of WT K-12. The expression of NF-${\kappa}B$ was higher in the mixed culture of Hep2 with ${\Delta}K$-12 than that in the mixed culture of Hep2 with WT K-12, and was lowest in the single culture of Hep2. The expression of ssrA was higher in the mixed culture of WT K-12 with Hep2 and S. aureus than in the mixed culture of WT K-12 with Hep2. Conclusion: These results suggest that ssrA plays an important role in the mechanism of E. coli adaptation to a new environment.

The effects of exercise training and acute exercise duration on plasma folate and vitamin B12

  • BACKGROUND/OBJECTIVES: Energy production and the rebuilding and repair of muscle tissue by physical activity require folate and vitamin $B_{12}$ as a cofactor. Thus, this study investigated the effects of regular moderate exercise training and durations of acute aerobic exercise on plasma folate and vitamin $B_{12}$ concentrations in moderate exercise trained rats. MATERIALS/METHODS: Fifty rats underwent non-exercise training (NT, n = 25) and regular exercise training (ET, n = 25) for 5 weeks. The ET group performed moderate exercise on a treadmill for 30 min/day, 5 days/week. At the end of week 5, each group was subdivided into 4 groups: non-exercise and 3 exercise groups. The non-exercise group (E0) was sacrificed without exercising and the 3 exercise groups were sacrificed immediately after exercising on a treadmill for 0.5 h (E0.5), 1 h (E1), and 2 h (E2). Blood samples were collected and plasma folate and vitamin $B_{12}$ were analyzed. RESULTS: After exercise training, plasma folate level was significantly lower and vitamin $B_{12}$ concentration was significantly higher in the ET group compared with the NT group (P < 0.05). No significant associations were observed between plasma folate and vitamin $B_{12}$ concentrations. In both the NT and ET groups, plasma folate and vitamin $B_{12}$ were not significantly changed by increasing duration of aerobic exercise. Plasma folate concentration of E0.5 was significantly lower in the ET group compared with that in the NT group. Significantly higher vitamin $B_{12}$ concentrations were observed in the E0 and E0.5 groups of the ET group compared to those of the NT group. CONCLUSION: Regular moderate exercise training decreased plasma folate and increased plasma vitamin $B_{12}$ levels. However, no significant changes in plasma folate and vitamin $B_{12}$ concentrations were observed by increasing duration of acute aerobic exercise.

Sox12 Is a Cancer Stem-Like Cell Marker in Hepatocellular Carcinoma

  • Recent studies on molecular carcinogenesis suggest that the chemo-resistance of some cancers is largely due to presence of cancer stem cells (CSCs), which affect the chemotherapy outcome for hepatocellular carcinoma (HCC). However, currently no consensus on a CSC phenotype in HCC has been obtained. Here, we examined Sox12 as a novel CSC marker in HCC. Sox12+ versus Sox12- cells were purified from HCC cell lines. The Sox12+ cells were compared with Sox12- HCC cells for tumor sphere formation, chemo-resistance, tumor formation after serial adoptive transplantations in nude mice, and the frequency of developing distal metastasis. We found that compared to Sox12- HCC cells, Sox12+ HCC cells generated significantly more tumor spheres in culture, were more chemo-resistant to cisplatin, were detected in circulation more frequently, and formed distal tumor more frequently. Moreover, Sox12 appeared to functionally contribute to the stemness of HCC cells. Thus, we conclude that Sox12 may be a novel marker for enriching CSCs in HCC.

Development of L-Lysine Producing Strains by Intergeneric Protoplast Fusion of Brevibacterium flavum and Corynebacterium glutamicum (Brevibacterium flavum과 Corynebacterium glutamicum의 이속간 원형질체 융합에 의한 L-라이신 생산균주 개발)

  • As a method of breeding L-lysine producing strains, the intergeneric protoplast fusion between Brevibacterium flavum and Corynebacterium glutamicum was performed. As a results, Brevibacterium flavum ATCC 21528 R showed 99% of protoplast formation and 10% of regeneration frequencies when treated with 400$\mu\textrm{g}$/$m\ell$ of lysozyme for 12hrs. In Corynebacterium glutamicum ATCC 21514 S, 99% and 12% were obtained by treatment of 300$\mu\textrm{g}$/$m\ell$ lysozyme for 12 hrs. In intergeneric protoplast fusion between Brevibacterium flavum ATCC 21528 R and Corynebacterium glutamicum ATCC 21831 S, 1.0$\times$10$^{-6}$ of recombinant frequency per regenerable cells was observed by use of PEG 6000, 30%(w/v). Among the strains obtained KR$_{43}$ strain showed 12% higher productivity of L-lysine than the parental cell. Then, the activity of aspartokinase of KR$_{43}$ was about 13% higher than the parental cell.

Phylogenetic Relationship and DNA Polymorphism of Boleophthalmus pectinirostris and Scartelaos gigas (Teleostei: Gobiidae) of Korea (한국산 짱뚱어(Boleophthalmus pectinirostris)와 남방짱뚱어(Scartelaos gigas) (Gobiidae)의 분자유전학적 계통연관과 DNA 다형화)

  • Phylogenetic relationships and DNA polymorphism among local populations of two Korean gobiidae species: Boleophthalmus pectinirostris and Scartelaos gigas were investigated based on 12S and 16S mitochondrial DNA and mitochondrial cytochrome b DNA sequences. DNA polymorphisms of B. pectinirostris between Suncheon and Gunsan populations were 100% identity from 434 bp segment of 12S rRNA gene and from 444 bp segment of mitochondrial cytochrome b genes, and 99.6% (2 bp different) identity from 484 bp segments of 16S rRNA genes. These results indicated the long period of geographic isolation between two populations of B. pectinirostris in Korea caused such high degrees of DNA polymorphisms. Based on the phylogenetic tree constructed from the two gobiid species in Korea, two genetically distinct groups of B. pectinirostris and S. gigas groups were recognized.

  • Boiler is one of the most important utilities providing steam to turbine in order to supply mechanical energy in thermal power plant. It is composed of thousands of tubes for high efficient heat transfer. The material for boiler tubes is used in such high temperature and pressure condition as $540^{\circ}C$, 22MPa. The boiler tube material is required to resist creep damage, fatigue cracking, and corrosion damages. 2.25%Cr-1Mo steel is used for conventional boiler tubes, and austenitenite stainless steel is used for higher temperature boiler tubes. But the temperature and pressure of steam in power plant became higher for high plant efficiency. So, the property of boiler tube material must be upgaded to fit the plant property. Several boiler tube material was developed to fit such conditions. X20CrMoV12.1 steel is also developed in 1980's and used for superheater and reheater tubes in supercritical boilers. The material has martensite microstructures which is difficult to evaluate the degradation. In this thesis, degrade the X20CrMoV12.1 steel at high temperatures in electric furnace, and evaluate hardness with Vickers hardness tester and strengths with Indentation tester.

Effects of Dietary Fat with Various Ratios in n-6/n-3 and P/S on Immune Responses of Rats at Different Age (나이가 다른 흰쥐에서 n-6/n-3 비율과 P/S 비율이 다른 식이지방이 면역능력에 미치는 영향 연구)

  • Effect of age and dietary fatty acid composition on immune responses were investigated in Sprague-Dawley male rets. The animals weighing 88.6$\pm$2.2g were fed 10% dietary fat (W/W, 20% of calorie) with P/S ratio of 0.5, 1 and 2, and in each group, there were three different levels of n-6/n-3 fatty acid ratio; 2, 4 and 8(3$\times$3). The experimental periods were 1 month, 6 months and 12 months. The results of this study were; 1) Weights of thymus and spleen were significantly reduced with increasing age, moreover thymus weights were reduced with increasing the degree of unsaturation in dietarty fatty acid at 12 month. 2) The proportion of splenic lymphocyte in the total T-cell was increased with increased with age. The proportion of helper T-cell was not changed, while the proportion of suppressor T-cell was decreased. Thus at 12 month, the ratio of helpe $r^pressor T-cell was appeared to increase significantly, and showed the tendency to increase by consuming the low amount of dieraty n-3 fatty acid. 3) Proliferation stimulated by Con A or PWM reduced significantly at 12 month in which are high in dietary P/S ratio, representing the similar pattern with decrease in thymus weight. 4) Natural killer cell activities were shown significantly higher at 1 month than those at 6 month or 12 month.th.

Stereoselective Bioreduction of Ethyl 3-Oxo-3-(2-Thienyl) Propanoate Using the Short-Chain Dehydrogenase/Reductase ChKRED12

  • Ethyl (S)-3-hydroxy-3-(2-thienyl) propanoate ((S)-HEES) acts as a key chiral intermediate for the blockbuster antidepressant drug duloxetine, which can be achieved via the stereoselective bioreduction of ethyl 3-oxo-3-(2-thienyl) propanoate (KEES) that contains a 3-oxoacyl structure. The sequences of the short-chain dehydrogenase/reductases from Chryseobacterium sp. CA49 were analyzed, and the putative 3-oxoacyl-acyl-carrier-protein reductase, ChKRED12, was able to stereoselectively catalyze the NADPH-dependent reduction to produce (S)-HEES. The reductase activity of ChKRED12 towards other substrates with 3-oxoacyl structure were confirmed with excellent stereoselectivity (>99% enantiomeric excess) in most cases. When coupled with a cofactor recycling system using glucose dehydrogenase, the ChKRED12 was able to catalyze the complete conversion of 100 g/l KEES within 12 h, yielding the enantiopure product with >99% ee, showing a remarkable potential to produce (S)-HEES.

Effects of orthodontic force on root surface damage caused by contact with temporary anchorage devices and on the repair process

  • Objective: This study aimed to evaluate the effects of force loading on root damage caused by contact with temporary anchorage devices (TADs) during orthodontic treatment and to examine the repair process 4, 8, and 12 weeks after TAD contact by micro-computed tomography (CT). Methods: We enrolled 42 volunteers who required bilateral upper first premolar extractions. The experimental study design was as follows. For both first premolars, cantilever springs were placed, and then TADs were immediately inserted between the premolars of all volunteers. According to the removal order of the appliances, the participants were divided into the TAD group (Group T: n = 21, only TAD removal) and the spring group (Group S: n = 21, only spring removal). A splitmouth design was adopted in both groups as follows. For each volunteer, the left premolars were extracted 4, 8, or 12 weeks after TAD-root contact. The right premolars were extracted immediately after contact in both groups (Groups T-C and S-C) and used as positive controls. Resorption volumes and numbers of craters were determined by micro-CT. Results: The numbers of resorption craters were higher in Group T than in Group S at 8 and 12 weeks (p < 0.01). Crater volumes were higher in Group T than in Group S at 4 and 12 weeks (p < 0.01, both). Conclusions: Root injury was not completely repaired 12 weeks after root-TAD contact, even when the TADs were removed in cases of continuous force application.

  • We introduce (${\tau}_i$, ${\tau}_j$) fuzzy (r,s)-semiclosures and (${\tau}_i$, ${\tau}_j$)-fuzzy (r,s)-semiinteriors. Using the notions, we investigate some of characteristic properties of fuzzy pairwise (r,s)-semicontinuous, fuzzy pairwise (r,s)-semiopen and fuzzy pairwise (r,s)-semiclosed mappings.
