The Study of Zn, Cu, Mn, Ni Contents of Serum, Hair, Nail and Urine for Female College Students (일부 여대생의 혈청, 소변, 두발, 손톱 중의 아연, 구리, 망간, 니켈 함량에 관한 연구)

  • The purpose of this study was to evaluat the Zn, Cu, Mn and Ni status and correlation of their concentrations in serum, urine, hair and fingernail in female college students. As the results are follows; The mean age of subjects was 22.5 years and height, weight and BMI were 160.1 cm, 51.0 kg and 20.0 respectively. The daily intake of energy 1769.5 kcal and the ratio of carbohydrate, fat and protein for energy is 60 : 20 : 15. The daily intake of energy (88.5%), vitamin B2 (86.1%), Ca (75.4%), Fe (58.3%) and Zn (63.0%) of subjects did not reach to Korean Recommended Dietary Allowance (RDA). The daily mean intake of Zn, Cu, Mn and Ni were 7.56 mg, 2.30 mg, 3.81 mg and 0.18 mg respectively. The concentrations of Zn in serum, urine, hair and nail were 85.6$\mu\textrm{g}$/dL, 391.2$\mu\textrm{g}$/day, 174.6$\mu\textrm{g}$/g and 102.4$\mu\textrm{g}$/g respectively and those Cu were 84.2$\mu\textrm{g}$/dL, 56.6 $\mu\textrm{g}$/day, 20.3$\mu\textrm{g}$/g and 4.3$\mu\textrm{g}$/g respectively. The concentrations of Mn in serum, urine, hari and nail were 0.2$\mu\textrm{g}$/dL, 1.1$\mu\textrm{g}$/day, 1.8$\mu\textrm{g}$/g and 1.6$\mu\textrm{g}$/g respectively and those of Ni were 0.6$\mu\textrm{g}$/dL, 24.5$\mu\textrm{g}$/day, 3.5$\mu\textrm{g}$/g and 3.1$\mu\textrm{g}$/g respectively. The daily intake of Mn showed the positive correlations with concentration of Mn in hair (p<0.05) and the daily intake of Ni was positively correlated with urinary exretion (p<0.05) and hair contents (p<0.001) of Ni. The more detailed studies about these trace mineral status should be required.

An Experimental Study on Mercury Compounds Poisoning (수은 중독에 관한 실험적 연구)

  • This experiment was performed to study the sequential accumulation of mercury in selected tissues of gold fish (Carassius auratus) exposed to 2, 6, 30, 120 and 300 $\mu$g Hg/1 as HgCl$_2$. In order to prepare treatment groups suitable for the present study, one control and five experimental groups, which were composed of I (2 $\mu$g/l), II (6$\mu$g/l), III (30$\mu$g/l), IV (120$\mu$g/l), V (300 $\mu$g/l), were used in 180 liter glass aquaria. The experiment was started by transfering 20 fish of average total length 140 $\pm$ 20 mm to each of the six tanks and allowing the uptake to take place for 12 weeks period. Fish were killed after time periods of 1, 2, 4, 8, and 12 weeks, and samples were disected by five parts gill, kidney liver, muscle and egg. The summarized results were as follows: 1. In control group, low concentrations of mercury(range 0.01-0.11 $\mu$g/g)were determined in the all selected tissues. 2. In experimental group, the average levels of mercury residues in the gill, kidney, liver muscle and egg were 3.61-189.54 $\mu$g/g, 13.91-182.58 $\mu$g/g, 8.56-66.49 $\mu$g/g, 0.30-20.33 $\mu$g/g, and 1.63-23.76$\mu$g/g, respectively. 3. The mercury residues in selected tissues of the experimental group were generally 230-9100 times higher than those of the control group. 4. The amounts of methylmercury per total mercury in the muscle after 12 weeks were 0.10/0.30 $\mu$g/g(33.33%) in the I group, 0.14/1.18$\mu$g/g(11.86%) in the II group, 0.25/5.76 $\mu$g/g(4.34%) in the III group, 0.39/11.48$\mu$g/g(3.40%) in the IV group and 0.40/20.33 $\mu$g/g(1.97%) in the V group.

Kinetic Analyses for Enzymatic Properties of Trypsins Purified from Dark-Fleshed Fish (혈합육어 Trypsin의 효소적 성질에 대한 반응속도론적 해석)

  • Kinetic properties of typsins purified from dark-fleshed fish (anchovy, mackerel, yellowfin tuna, and albacore) were examined and analyzed on $benzoyl-_{D,L}-arginine-p-nitroanilide\;(BAPNA)$. The values of Km' and $k_{cat}$ of the purified trypsins from the four dark-fleshed fish were found to be $49.3{\mu}M$ and $90.9\;min^{-1}$ for anchovy, $53.7{\mu}M$ and $61.2min-^{-1}$ for mackerel A, $96.5{\mu}M$ and $76.6min^{-1}$ for mackerel B, $62.8{\mu}M$ and $46.6min^{-1}$ for yellowfin tuna, and $98.3{\mu}M$ and $47.7min^{-1}$ for albacore, respectively. The values of $K_i$ on $tosyl-_L-lysine$ chloromethyl ketone (TLCK) were determined to be $20.90{\mu}M$ for anchovy trypsin, $2.86{\mu}M$ for mackerel trypsin A, $3.90{\mu}M$ for mackerel trypsin B, $0.96{\mu}M$ for yellowfin tuna trypsin, and $1.82{\mu}M$ for albacore trypsin. Thus yellowfin tuna trypsin was the most sensitive to TLCK among all trypsins. The activities and catalytic efficiency of the trypsins purified from the temperate zone fish, anchovy and mackerel, were higher than those of the trypsins purified from yellowfin tuna and albacore which migrate widely from the tropic zone to the temperate zone.

  • Based on the scalar wave equation of optical fibers, the dispersion characteristics of arbitrarily profiled fibers were analyzed. We used the I-D FEM employing quadratic interpolation fucntions to solve the scalar wave equation. We simulated the DC optical fibers as objects, and searched for the refractive index distribution to minimize the total dispersion. In DC fibers, we found the design parameters for which the total dispersion was almost zero at $\lambda=1.3{\mu}m and 1.55{\mu}m$ simultaneously. We also found the design parameters where the dispersion was flattened, less than 1.0 ps/km.nm for$\lambda=1.4~1.7{\mu}m$1. and the dispersion was as low as 0.65 ps/km.nm at $\lambda=1.55{\mu}m$..

Effects of Benzyl Isothiocyanate and Its N-Acetylcysteine Conjugate on Induction of Detoxification Enzymes in Hepa1c1c7 Mouse Hepatoma Cells

  • The induction of detoxification enzymes by benzyl isothiocyanate (BITC) and its synthetic N-acetyl-L-cysteine (NAC) conjugate (NAC-BITC) was examined in Hepa1c1c7 murine hepatoma cells. BITC and NAC-BITC inhibited Hepa1c1c7 cell growth in a dose-dependent manner. Cell growth was 4.5~57.2% lower in Hepa1c1c7 cells treated with $0.1{\sim}1.0{\mu}M$ BITC than in control-treated Hepa1c1c7 cells. The NAC-BITC treatment had a similar inhibitory pattern on Hepa1c1c7 cell growth; $0.5{\mu}M$ and $10{\mu}M$ NAC-BITC decreased cell growth by 13.6% and 47.4%, respectively. Treatment of Hepa1c1c7 cells with $0.1{\sim}2.0{\mu}M$ BITC also elicited a dose-response effect on the induction of quinone reductase quinone reductase (QR) activity and QR mRNA expression. Treatment with $1{\mu}M$ and $2{\mu}M$ BITC caused 1.8- and 2.8-fold inductions of QR mRNA, respectively. By comparison, treatment with $1{\mu}M$ and $2{\mu}M$ NAC-BITC caused 1.6-and 1.9-fold inductions of QR mRNA, respectively. Cytochrome P450 (CYP) 1A1 and CYP2E1 induction were lower in $0.1{\sim}2{\mu}M$ BITC-treated cells than in control-treated cells. CYP2E1 activity was 1.2-fold greater in $0.1{\mu}M$ NAC-BITC-treated cells than in control-treated cells. However, the CYP2E1 activity of cells treated with higher concentrations (i.e., $1{\sim}2{\mu}M$) of NAC-BITC was similar to the activity of control-treated cells. Considering the potential of isothiocyanatesto prevent cancer, these results provide support for the use of BITC and NAC-BITC conjugates as chemopreventive agents.

Effects of Ginseng Radix on the Cell Cycle Regulation in Human Fetal Osteoblast (인삼이 사람태아골모세포의 세포주기조절에 미치는 영향)

  • Ginseng Radix(GR) had been used widely from oriental medicine and the effects of it have been investigated by many researchers. The purpose of present study was to investigate the effects of GR on the cell cycle progression and its molecular mechanism in human fetal osteoblast. The results were as follows. Increased cell proliferation was observed in cells exposed to 100 ng/ml, 10 ng/ml of GR-1 at 12 hours and 24 hours, 1 ${\mu}g$/ml of GR-1 at 48 hours, and 100 ${\mu}g$/ml, 10 ${\mu}g$/ml of GK-2 at 12 hours, all treatment groups of GR-2 at 24 hours(p<0.05). S phase and G1 phase was increased in the group of treated with 100 ng/ml of GR-1, with 10 ${\mu}g$/ml and 1 ${\mu}g$/ml of GR-2, with 100 ${\mu}g$/ ml and 10 ${\mu}g$/ml of GR-3 in the cell cycle analysis. The cell cycle regulation protein levels of Cyclin D1, Cyclin E, CDK 2. CDK 4 and CDK 6 were increased in the group of treated with 1 ${\mu}g$/ml and 100 ng/ml of GR-1, with 10 ${\mu}g$/ml and 1 ${\mu}g$/ml of GR-2, with 100 ${\mu}g$/ ml and 10 ${\mu}g$/ml of GR-3. On the other hand, p21 was decreased in the treatment group with 1 ${\mu}g$/ml and 100 ng/ml of GR-1, with 10 ${\mu}g$/ml and 1 ${\mu}g$/ml of GR-2, 10 ${\mu}g$/ml of GR-3, and p53 and p16 was decreased in the treatment group with 100 ng/ml of GR-1, 100 ${\mu}g$/ml and GR-3 and pRb was decreased in the all treatment groups except 1 ${\mu}g$/ml of GR-1. These results suggested that GR increases the cell proliferation and the cell cycle progression in human fetal osteoblast, which is linked to increased cell cycle regulation protein levels of Cyclin D1 , Cyclin E, CDK 2, CDK 4, CDK 6 and decreased cell cycle regulation protein levels of p21, pRb.

A Study on Several Mineral Elements in Korea Tobacco Leaves (한국산 잎담배 중의 몇가지 무기성분에 관한 조사 연구)

  • The contents of Sodium, Magnesium, Iron, nickel , Copper, Maganese and Zinc in Korean tobacco leaves Produced in 1978, were investigated. The contents of each mineral element in tobacco leaves of various varieties were as follows ; Mg was 5370$\mu{g}/g-7640\mu{g}/g$, Burley> Flue -cured >Native. Mn was 210$\mu{g}/g-1290\mu{g}/g$, Burley>Flue -cured>Native. Fe was 330$\mu{g}/g-590\mu{g}/g$, Burley>Flue -cured>Native. Zn was 500$\mu{g}/g$ -100, Native > Burley>Flue-cured. Na, Ni and Cu showed no difference between the types. The contents of Ha, Mg, Mn, Ni and Cu were reduced in the order of Heavy 5, 3, 1 and increased in the order of Light 1, 3, 5 grades. The correlation coefficient (r) between Mg and grade in Hicks, SC 72 and HC 2326 were 0.770-0.981, and significance level was 0.1 % .

A Study on the Concentration of Aflatoxin B1 in Granule and Globular Types of Herbal Medicines (엑스과립과 환으로 만들어진 한방생약제제의 aflatoxin B1 연구)

  • This study is an endeavor to evaluate the risk assessment of hazardous(aflatoxin $B_1$) in medicines from oriental medical prescription which are circulated much recently. For that, twelve globular and granule types, seven liquid types of herbal medicine were bought to compare and analyze the content of aflatoxin aflatoxin B_1), which are harmful to human body. Woo Hwang Cheong Sim Hwan of Aflatoxin $B_1$ concentration lower than the standard accepted by all the products have been detected, B company(tradition) is the concentration of $1.24\;{\mu}g/kg$, C company $1.04\;{\mu}g/kg$, A company(tradition) and B company did not detect. And the general pill of aflatoxin B1 concentration lower than the standard accepted by all the products have been detected, S-1 is the concentration of $1.8\;{\mu}g/kg$, S-2 of $1.04\;{\mu}g/kg$, S-3 of $0.88\;{\mu}g/kg$, S-4 of $9.32\l\;{\mu}g/kg$, S-6 of $7.8\;{\mu}g/kg$, S-5 did not detect. All the products eundan allowed in the concentration of aflatoxin $B_1$ levels were lower than detection, D company of $0.96\;{\mu}g/kg$, E company concentration was not detected. The liquid product of aflatoxin $B_1$ concentration was found liwer than the standard accepted by all the product, L-3 concentration of $0.8\;{\mu}g/kg$, K-4 was detected in the $1.16\;{\mu}g/kg$, L-1 and L-2 is not detected, L-5 concentration of $15\;{\mu}g/kg$, L-7 is detected as $1.08\;{\mu}g/kg$ and, L-6 was not detected.

Comparative Morphology of Eggs of Heterophyids and Clonorchis sinensis Causing Human Infections in Korea (한국의 인체기대 이형흡충류 및 간흡충 충란의 비교형태학적 검토)

  • In order to provide some clues for differential diagnosis of trematode infections in fecal examination, the comparative morphology of eggs of 5 kinds of heterophyid flukes (Metagonimus yokogawai, Heterophyes heterophyes nocens, Heterophyopsis continua, Stellantchasmus falcatus and Pygidiopsis summa) and Clonorchis sinensis was studied. The eggs were obtained from distal portion of uteri of worms which were recovered from men after treatment. The characteristic shape and appearance of each kind of eggs were observed in detail under light microscope, and their length and width measured and compared one another. The results are as follows: 1. Eggs of C. sinensis are elongated ovoidal in shape with attenuated anterior end, 25.3~33. 2 (28. 3 in average) ${\mu}m$ long and 14.2~17.4(5.9) ${\mu}m$ wide with length/width ratio of 1.60~2.00 (1.78). They differ from all heterophyid eggs in that they have prominent wrinkling (muskmelon pattern) at their shell surface. 2. P. summa eggs are ovoid to pyriform in shape and characterized by the smallest size of all kinds examined, 19.8~22.9 (21.6) ${\mu}m$ long and 11.1~13.4 (12.1) ${\mu}m$ wide and the ratio 1.63~1.99 (1.78). 3. Eggs of S. falcatus are elongated ovoidal and most slender form, 25.3~29.2 (27.2) ${\mu}m$ long and 11.1~13.4 (12.5) ${\mu}m$ wide with the ratio of 2.00~2.57 (2.17). 4. Eggs of M. yokogawai are ellipsoid to elliptical in shape with round both ends, 26.9~31.6 (28.5) ${\mu}m$ long and 14.2~18.2 (16.8)${\mu}m$ wide with the ratio of 1.48~2.11 (1.70). 5. H. continua eggs are oval in shape, sometimes similar to M. yokogawai or H. h. nocen$ eggs, however, the relative breadth is broadest among all kinds, with maximum width at posterior half portion. They are 23.7~27.7 (25.0) ${\mu}m$ long, 15.8~18.9 (16.4) ${\mu}m$ wide with the ratio of 1. 33~1.75 (1.53). 6. Eggs of H. h. nocens are ellipsoid to ovoid in shape but sometimes more slender than M. yokogawai and have slightly pointed both ends. They are 23.7~29.2 (25.7) p.m long, 14.2~15.8 (15.4) ${\mu}m$ wide, and the ratio 1.50~2.06 (1.67). From the results, it is concluded that eggs, of 5 kinds of heterophyids and C. sinensis can be morphologically differentiated one another, however, careful observation and measurement on sufficient number of eggs are needed.
