TOSC 방법을 이용한 참당귀 뿌리 용매분획(Angelica gigas)의 항산화 활성 평가 (Evaluation of Antioxidant Activity of Solvent Fractions of Angelica gigas Root Using TOSC Assay)

  • Objectives: The aim of this study was to elucidate the antioxidant effect of solvent fractions of Angelica gigas root. Methods: The ethanol extract of Angelica gigas root was suspended in water and then partitioned with dichloromethane (MC Fr.), ethyl acetate (EA Fr.) and butanol (BuOH Fr.), sequentially. The antioxidant activities of solvent fractions of Angelica gigas root were evaluated for radical scavenging activity against stable free radicals (2,2-diphenyl-1-picrylhydrazyl) DPPH, hydrogen peroxide and superoxide anion radicals. In addition the antioxidant activities of solvent fractions of Angelica gigas root against peroxyl radicals, hydroxyl radicals and peroxynitrites were determined by the total oxy-radical scavenging capacity (TOSC) assay. Results: Among the solvent fractions of MC Fr., EA Fr., and BuOH Fr., BuOH Fr. was found to have stronger antioxidant activity with $IC_{50}$ values of 59.72, 14.36, 30.96 and $44.75\;{\mu}g/m{\ell}$ on the DPPH radical, nitrite, superoxide anion and hydrogen peroxide, respectively, than BHA used as a positive control. Moreover, specific TOSC values(564.8, 276.4 and 405.5 TOSC/mM) of BuOH fr. against peroxyl radicals, hydroxyl radical and peroxynitrite were 4 times higher than GSH (136.5, 67.4 102.6 TOSC/mM) used as a positive control. Conclusions: These results suggest that the BuOH fr. of Angelica gigas root has a high antioxidant activity and can be useful to develop functional food against oxidative stress conditions.

Antioxidant and Memory Enhancing Effects of Purple Sweet Potato Anthocyanin and Cordyceps Mushroom Extract

  • The effects of purple sweet potato anthocyanin (SPA) and Cordyceps mushroom extract (CME) on lipid peroxidation, 1,1-diphenyl-2-picrylhydrazyl (DPPH) radicals and cognitive deficits were examined. Both SPA and CME exhibited DPPH radical scavenging activities with similar potency. In contrast, only SPA was shown to effectively inhibit lipid peroxidation initiated by $Fe^{2+}$ and ascorbic acid in rat brain homogenates. Furthermore, SPA markedly enhanced cognitive performance, assessed by passive avoidance test in ethanol-treated mice. Combined treatments with SPA and CME did not significantly influence the effects of SPA alone. These results demonstrate that anthocyanin prepared from purple sweet potato exhibits memory enhancing effects, which may be associated with its antioxidant properties.

Antioxidant and anti-inflammatory activities of phenolic compounds grafted with hyaluronic aicd derived from Liparis tessellatus eggs

  • Hyaluronic acid from Liparis tessellatus eggs (HALTE) was grafted with caffeic acid (CA-g-HALTE), ferulic acid (FA-g-HALTE), gallic acid (GA-g-HALTE), and nisin (Nisin-g-HALTE) and investigated for their anti-inflammatory and antioxidant potential in lipopolysaccharides-stimulated RAW 264.7 mouse macrophages. Nitric oxide (NO) generation and prostaglandin E2 activity were measured after treatment with the grafted HALTE samples. All grafted HALTE samples exhibited more antioxidant activity against 2,2'-azinobis(3-ethylbenzothiazoline-6-sulfonic acid) radicals than 2,2-diphenyl-1-picrylhydrazyl radicals. Nisin-g-HALTE showed the least antioxidant activity. Additionally, the NO assay results showed that all grafted samples had no cytotoxic effect on RAW 264.7 macrophages and reduced macrophage activity after treatment. The most effective concentrations of CA-g-HALTE and FA-g-HALTE were found to be above 100 ㎍/mL. Increased sample concentration resulted in increased activity except with Nisin-gHALTE at 100 ㎍/mL. CA-g-HALTE, FA-g-HALTE, GA-g-HALTE, and Nisin-g-HALTE were found to have antioxidant and anti-inflammatory potential, which can be further explored for use in food, cosmetic, nutraceutical, and biomedical applications.

가감온비탕합오산(加減溫脾湯合五散) 완제(完製)(HWW$^{(R)}$)의 항산화 효과에 대한 연구 (Antioxidative Activities of Wen-pi-tang-Hab-Wu-ling-san (WHW$^{(R)}$) in vitro)

  • Objectives: The objective of this study was to investigate the antioxidant effects of manufactured Wen-pi-tang-Hab-Wu-ling-san (WHW$^{(R)}$) in vitro. Methods: WHW$^{(R)}$ was prepared by the pilot manufacture of WHW water extract from a GMP system appointed company. Antioxidative activities were determined by in vitro tests as follows: the scavenging activities of oxygen free radicals including 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical, superoxide anion, hydrogen peroxide and nitric oxide radicals, as well as ferrous ion chelating capacity and Trolox equivalent antioxidant capacity (TEAC). Results: WHW$^{(R)}$ significantly scavenged oxygen free radicals such as DPPH (IC$_{50}$=115.28 $\pm$ 0.25 $\mu$g/$m\ell$), superoxide anion (IC$_{50}$=8.56 $\pm$ 0.08 $\mu$g/$m\ell$), hydrogen peroxide (IC$_{50}$=240.36 $\pm$ 3.41 $\mu$g/$m\ell$) and nitric oxide (IC$_{50}$=162.28 $\pm$ 0.21 $\mu$g/$m\ell$) radicals. WHW$^{(R)}$ also showed ferrous ion chelating activity (IC$_{50}$=543.19 $\pm$ 4.85 $\mu$g/$m\ell$) and Trolox equivalent effects (IC$_{50}$=45.311 $\mu$g/$m\ell$) in TEAC and ORAC assay, respectively. Conclusion: This study demonstrates that WHW$^{(R)}$ has strong antioxidative properties through free radical scavenging activity. These data suggest that WHW$^{(R)}$ be used as an antioxidant agent.

Phytochemical analysis of the antioxidant properties of Silybum marianum L

  • A growing body of evidence supports lipid peroxidation as having a role in the pathogenesis of liver disease. Although the probable cause of damage to human hepatocytes may be multifactorial, free radicals have been implicated in a variety of liver diseases, particularly in the presence of iron overload and toxic substances such as ethanol. Consequently, antioxidants, particularly those of plant origin such as flavonoids, may help to reduce the risk of developing these diseases. Silybum (S.) marianum, a medicinal plant widely used in traditional European medicine for the treatment of liver disorders, was evaluated for antioxidant activity. Thin layer chromatography and High Performance Liquid Chromatography analyses of crude extract of the plant confirmed the presence of a number of flavonoids reported in the literature. The antioxidant activity of these flavonoids was measured through inhibition of lipid peroxidation and 1, 1-diphenyl-2- picrylhydrazyl radical scavenging. The crude plant extract showed marked antioxidant activity in both assays. These results suggest that S. marianum contains flavonoids with antioxidant activity, capable of inhibiting or scavenging free radicals, thus supporting its traditional use as a hepatoprotective agent.

Antioxidative Activities of Kefir

  • This study aims to evaluate the antioxidative activities of cow-milk kefir and goat-milk kefir. Antioxidative mechanisms, including radical-scavenging effects, ferrous-ion chelating ability, reducing power and antioxidant activity, were investigated herein. Kefirs demonstrated significantly greater scavenging effects upon 1,1-diphenyl-2-picrylhydrazyl (DPPH) and superoxide radicals, an inhibition effect upon linoleic-acid peroxidation, and more substantial reducing power, but reduced glutathione peroxidase (GSH-Px) activity than was the case for milks. There was no significant difference between milks and kefirs as regards ferrous-ion chelating ability and superoxide dismutase (SOD) activity. These findings have demonstrated that kefirs possess antioxidant activity, thereby suggesting that kefirs are potential candidates for the role of useful natural antioxidant supplements for the human diet.

Antioxidant and Anti-inflammatory Activities of Equisetum hyemale

  • Equisetum hyemale L. has been prescribed widely as a traditional medicine for the treatment of inflammatory diseases such as rheumatoid arthritis, conjunctivitis, pyelonephritis. In order to identification the mechanism, we examined an antioxidant and anti-inflammatory activity of 85% methanol extract of E. hyemale. In this study E. hyemale exhibited strong scavenging effect on 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical, superoxide radical, and nitric oxide. To elucidate the anti-inflammatory properties of E. hyemale, we investigated the inhibition effects of nitric oxide and IL-6 by E. hyemale in IFN-gamma and LPS-stimulated mouse peritoneal macrophages. E. hyemale suppressed nitric oxide, IL-6 production and iNOS expression dose-dependently without notable cytotoxic activity. These data suggest that E. hyemale might be useful in inflammatory diseases by inhibiting the free radicals and inflammatory mediators.

백렴 추출물의 Matrix Metalloproteinase-1 발현 저해 효과 (Effect of Melothria heterophylla Extract on Expression of Matrix Metalloproteinase-1 in Human Skin Fibroblasts)

  • Matrix metalloproteinases (MMPs) are known to play an important role in photoaging by mediating the degradation of extracellular matrix proteins. In this study, to develop a n ew anti-aging agent, we investigated the antioxidant activity and the inhibitory effect of Melothria heterophylla extract on expression of MMP-1 in UVA-irradiated human dermal fibroblasts and MMP-1 activity. The M.heterophylla extract was found to scavenge radicals and reactive oxygen species (ROS) with the $SC_{50}$ values of $13{\mu}g/ml$ against 1,1-diphenyl-2-picrylhydrazyl (DPPH) radicals and $20{\mu}g/ml$ against superoxide radicals in the xanthine/xanthine oxidase system, respectively. UVA-induced MMP-1 expression was reduced about 80% by $100{\mu}g/ml$ of the M.heterophylla extract but MMP-1 Mrna expression was not inhibited. Therefore, we conclude that the M.heterophylla extract significantly inhibits MMP-1 expression at the protein level. Also, the M.heterophylla extract inhibited MMP-1 activity in a dose dependent manner. From these results, we suggest that the M.heterophylla extract can be used as a new anti-aging agent by antioxidant activity, regulation of UVA-induced MMP-1 production, and inhibition of MMP-1 activity.

The Isolation and Antioxidative Effects of Vitexin from Acer palmatum

  • Free radicals and reactive oxygen species (ROS) caused by UV exposure or other environmental factors are critical players in cellular damage and aging. In order to develop a new antiphotoaging agent, this work focused on the antioxidant effects of the extract of tinged autumnal leaves of Acer palmatum. One compound was isolated from an ethyl acetate soluble fraction of the A. palmatum extract using silica gel column chromatography. The chemical structure was identified as apigenin-8-C-beta-D-glucopyranoside, more commonly known as vitexin, by spectral analysis including LC-MS, FT-IR, UV, $^{1}H-$, and $^{13}C-NMR$. The biological activities of vitexin were investigated for the potential application of its anti-aging effects in the cosmetic field. Vitexin inhibited superoxide radicals by about $70\%$ at a concentration of $100\;{\mu}g/mL$ and 1,1-diphenyl-2-picrylhydrazyl (DPPH) radicals by about $60\%$ at a concentration of $100\;{\mu}g/mL$. Intracellular ROS scavenging activity was indicated by increases in dichlorofluorescein (DCF) fluorescence upon exposure to UVB $20\;mJ/cm^2$ in cultured human dermal fibroblasts (HDFs) after the treatment of vitexin. The results show that oxidation of 5-(6-)chloromethyl-2',7'-dichlo-rodihydrofluorescein diacetate ($CM-H_{2}DCFDA$) is inhibited by vitexin effectively and that vitexin has a potent free radical scavenging activity in UVB-irradiated HDFs. In ROS imaging using a confocal microscope we visualized DCF fluorescence in HDFs directly. In conclusion, our findings suggest that vitexin can be effectively used for the prevention of UV-induced adverse skin reactions such as free radical production and skin cell damage.

고분자물질과 접촉한 자기조립단분자막 전극 물질의 기계화학 현상 분광학적 연구 (Mechanochemistry on Self-Assembled Monolayer(SAM) /Electrodes after Contacting with Polymeric Stamp)

  • We investigated mechanochemical radical, which is concomitant with chemical lift-off lithography(CLL), on the self-assembled monolayer(SAM)/electrodes and a polydimethylsiloxane(PDMS) using a colorimetric and a spectroscopic method. The 11-mercaptoundecanol(MUO)/Au or the 11-hydroxyundecylphosphonic acid (HUPA)/ITO were contacted with bare or activated PDMS. After contact, the each of SAM/substrates and the PDMS were immersed in a 2,2 Diphenyl-1-picrylhydrazyl(DPPH) radical scavenger. The color of the DPPH exposed to the PDMS was changed from purple to yellow and the absorbance decreased definitely at 515 nm wavelength. The SAM/substrates, however, have caused small changes in spectroscopic property, indicating no existence of radical species. We concluded that mechanochemical radicals were formed by homolytic cleavage of PDMS molecules upon external force and hardly transferred on the SAM/substrates.
