Expression of Anthrax Lethal Factor, a Major Virulence Factor of Anthrax, in Saccharomyces cerevisiae (Yeast내에서 탄저병 원인균인 Bacillus anthracis의 치사독소인 Lethal Factor 단백질 발현)

  • Anthrax is an infectious disease caused by the gram-positive bacterium, Bacillus anthracis. Anthrax toxin is a tripartite toxin comprising of protective antigen (PA), lethal factor (LF) and edema factor (EF). PA is the receptor-binding component, which facilitates the entry of LF or EF onto the cytosol. LF is a zinc-dependent metalloprotease, which is a critical virulence factor in cytotoxicity of infected animals. Therefore, it is of interest to develop its potent inhibitors for the neutralization of anthrax toxin. The first step to identify the inhibitors is the development of a rapid, sensitive, and simple assay method with a high-throughput ability. Much efforts have been concentrated on the preparation of powerful assays and on the screening of inhibitors using these system. In the present study, we have tried to construct anthrax lethal factor in yeast expression system to prepare cell-based high-throughput assay system. Here, we have shown the results covering the construction of a new vector system, subcloning of LF gene, and the expression of target gene. Our results are first trial to express LF gene in eukaryote and provide the basic steps in design of cell-based assay system.

Derepression of a Methionine Biosynthetic Gene by Utilizing a Promoter Isolated from Corynebacterium glutamicum (Corynebacterium glutamicum에서 분리된 프로모터를 이용한 메치오닌 생합성 유전자의 조절해제)

  • A transcriptionally active fragment $(P_{19})$ isolated by utilizing the promoter-probe shuttle vector pSK1Cat was analyzed. By subcloning analysis, the 180 bp region $(P_{180})$ responsible for the activity was determined. Transcriptional fusion of the C. glutamicum metX gene to $P_{180}\;(P_{180}-metX)$ resulted in a 24-fold increase in MetX activity in a complex medium, while a 13-fold increase was observed with the $P_{tac}$ promoter. Additionally, the expression conferred by $P_{180}$ was not affected by methionine added to the growth medium, suggesting that the $P_{180}$ clone is useful for the deregulated expression of biosynthetic genes in C. glutamicum during amino acid fermentation. Introduction of $P_{180}-metX$ into a lysine-producing C. glutamicum resulted in the production of methionine to 0.8 g/l.

  • Some factors of wounding methods, solidifying agents, origin of leaf explants, cone. of acetosyring-one, and MES affecting regeneration and transformation by Agrobacterium tumefaciens were investigated to establish an efficient transformation system of apple. Wounding by cutting the leaves merely showed the tendency of regeneration and transformation with higher efficiency compared with that of wounding by non-traumatic forcep when carrying out co-cultivation for three days after bacterial inoculation. While examining the solidifying agents of medium with the combination of agar (A)+Gelrit $e^{ }$ (G) in g. $L^{-1}$ , the higher concentration of Gelrit $e^{ }$ increased the efficiency of regeneration. However, there was no difference in the efficiency of transformation from the treatments of 2.5 G, 3.5 A+1.2 G, and 7.0 G. The origin of leaf explants showed no difference statistically in the efficiency of regeneration and transformation, but that from the shoots of proliferation medium showed the tendency with higher efficiency. The concentration of above 0.1 mM acetosyringon had an increase in the efficiency of regeneration and transformation and the concentration of 0.15 mM had the highest efficiency of transformation in the treatment of acetosyringone with different concentration. There was no effect of MES on regeneration and transformation.ion.

Optimization of a Process for Extraction of Petasin from Petasites japonicus Leaves by Response Surface Methodology (반응표면분석법에 의한 머위 잎의 petasin 추출공정 최적화)

  • Petasin extracted from Petasites japonicus leaves has been well known to be effective in the treatment of allergic asthma. This study was carried out to optimize the extraction process of petasin from P. japonicus leaves by response surface methodology (RSM). The dried powder of P. japonicus leaves was extracted at ethanol concentrations ranging from 40% to 80%, extraction rpm ranging from 125 rpm to 225 rpm, and extraction time ranging from 1 to 3 hours. The effects of the extraction conditions on the dry yield and petasin content of the extracts were investigated using a second-order Box-Behnken design. The petasin content was significantly affected by ethanol concentration, extraction rpm, and extraction time, tending to increase more with increasing ethanol concentration. The optimum condition for petasin extraction from Petasites japonicus leaves was 79.92% in ethanol concentration, 178.10 rpm in extraction rpm, and 2.06 hours in extraction time, respectively.

  • In an aero gas-turbine engine, a surface air-oil heat exchanger (SAOHE) is used to cool the oil system for the gearboxes and electric generators. The SAOHE is installed inside the fan casing of the engine in order to dissipate the heat from the oil system into the bypass duct stream. The purpose of this study was to develop an effective numerical method for designing an SAOHE for an aero gas-turbine engine. A two-dimensional model using a porous medium was developed to evaluate the aero-thermal performance of the fins of the heat exchanger, and a one-dimensional flow and thermal network program was developed to save time and cost in the evaluation of the heat exchanger performance. Using this network program, the pressure drop and heat transfer performance of the heat exchanger were predicted, and the results were compared with two-dimensional computational fluid dynamics results and experiment data for validation.

  • In the future network such as Internet of Things (IoT), the number of computing devices are expected to grow exponentially, and each of the things communicates with the others and acquires information by itself. Due to the growing interest in IoT applications, the broadcasting in Opportunistic ad-hoc networks such as Machine-to-Machine (M2M) is very important transmission strategy which allows fast data dissemination. In distributed networks for IoT, the energy efficiency of the nodes is a key factor in the network performance. In this paper, we propose a fuzzy logic based probabilistic multi-hop broadcast (FPMCAST) algorithm which statistically disseminates data accordingly to the remaining energy rate, the replication density rate of sending node, and the distance rate between sending and receiving nodes. In proposed FPMCAST, the inference engine is based the fuzzy rule base which is consists of 27 if-then rules. It maps input and output parameters to membership functions of input and output. The output of fuzzy system defines the fuzzy sets for rebroadcasting probability, and defuzzification is used to extract a numeric result from the fuzzy set. Here Center of Gravity (COG) method is used to defuzzify the fuzzy set. Then, the performance of FPMCAST is evaluated through a simulation study. From the simulation, we demonstrate that the proposed FPMCAST algorithm significantly outperforms flooding and gossiping algorithms. Specially, the FPMCAST algorithm has longer network lifetime because the residual energy of each node consumes evenly.

An Efficient Filtering Technique of GPS Traffic Data using Historical Data (이력 자료를 활용한 GPS 교통정보의 효율적인 필터링 방법)

  • For obtaining telematics traffic information(travel time or speed in an individual link), there are many kinds of devices to collect traffic data. Since the GPS satellite signals have been released to civil society, thank to the development of GPS technology, the GPS has become a very useful instrument for collecting traffic data. GPS can reduce the cost of installation and maintenance in contrast with existing traffic detectors which must be stationed on the ground. But. there are Problems when GPS data is applied to the existing filtering techniques used for analyzing the data collected by other detectors. This paper proposes a method to provide users with correct traffic information through filtering abnormal data caused by the unusual driving in collected data based on GPS. We have developed an algorithm that can be applied to real-time GPS data and create more reliable traffic information, by building patterns of past data and filtering abnormal data through selection of filtering areas using Quartile values. in order to verify the proposed algorithm, we experimented with actual traffic data that include probe cars equipped with a built-in GPS receiver which ran through Gangnam Street in Seoul. As a result of these experiments, it is shown that link travel speed data obtained from this algorithm is more accurate than those obtained by existing systems.

Effects of Types and Application Levels of Swine Manure on Herbage Productivity, Improvement of Soil Fertility and Environmental Pollution in Mixed Grassland (가축분뇨의 처리형태와 시용수준이 영년초지의 생산성, 지력증진 및 환경에 미치는 영향)

  • This study was conducted to investigate the effects of the type and application level of swine manure on herbage productivity, efficiency of nitrogen utilization and environmental pollution by the leaching of nitrogen compounds in mixed grassland. The field experiment was carried out on established grassland sward growing on silt clay loam soil. Main plots were the types of swine manure, such as swine manure fermented with sawdust(SMFWS), swine manure fermented without sawdust(SMF) and swine slurry(SS), and mineral fertilizer(Urea). Subplots were the application levels of swine manure, such as 100, 200 and 400 kgN/ha, The results obtained were summarized as follows; 1. Herbage productivity, nutritive value and nitrogen yields were the highest with mineral fertilizer and followed by swine slurry(SS), and the lowest with swine manure fermented with sawdust(SMFS). As compared with dry matter(DM) yield by application of mineral fertilizer(100%), the DM percents of SMFWS, SMFS and SS were 90.6, 80,9 and 76.8%, respectively. 2. Organic matter(OM) contents of the soil were increased by the applications of swine manure. OM contents were the high set with SMFS and the lowest with mineral fertilizer. 3. The amounts of nitrate leaching by types and application levels of swine manure were the highest in the end of Aug. and early of Sep. during the seasons and ranged from 10 to 25ppm.

Cost Analysis of Wrap Silage Production in the Paddy Field for Forage Crop Cropping System (답리작 사료작물의 랩사일리지 생산비 분석)

  • Mechanized operation model and mechanical cultivation technology for winter barley, rye, Italian ryegrass and sudan grass wrap silage production system at the paddy field for cropping system was developed. Also, a series of experiment were performed and lots of data were collected and analyzed to develope mechanical technology, coverage area, and optimum size of the farm (break-even point) for wrap silage production system. The coverage area for winter barley or rye wrap-silage production system is determined around 61.9, 73.4, 77.5, 88.2 ha in the case of drill seeding and different ripening species by tractor power 50, 75, 100, 130 ps, respectably. The break-even point of the farming size is analyzed as 20 ha and its production cost is estimated around 367, 383, 430, 443 won/TDN-kg in the case of winter barley wrap-silage by tractor power 50, 75, 100, 130 ps, respectably. The break-even point of the farming size is analyzed as coverage area and its production cost is estimated around 237, 215 won/TDN-kg in the case of winter barley wrap silage and sudan grass by the tractor power 50, 100 ps, respectably.

Pretreatment Characteristics of Potatoes by Soft Steam Treatment (저온스팀 열처리 방식에 따른 감자의 전처리 특성)

  • This study aimed to investigate the effects of various soft steam treatments, namely, forced convection-boiler, forced convection-fan, and natural convection, on the pretreatment characteristics of potatoes. In this study, potatoes were exposed to various cooking conditions, including steaming method, treatment time (0-60 min), and temperature (60, 70, $80^{\circ}C$). Then, changes in temperature, cook value, ascorbic acid content, moisture content, and weight loss in the fresh and steam-treated samples were measured and evaluated. The results clearly showed that natural convective steaming was superior to other treatments in terms of heating characteristics, cook value (FC-b: $46.4{\pm}1.7$, FC-f: $21.8{\pm}1.1$, NC: $52.1{\pm}1.9min$ at $80^{\circ}C$), ascorbic acid content (FC-b: $36.5{\pm}2.7$, FC-f: $28.5{\pm}2.9$, NC: $48.2%{\pm}2.5%$ at $80^{\circ}C$), moisture retention (FCb: $74.6{\pm}0.8$, FC-f: $71.5{\pm}0.5$, NC: $77.6%{\pm}0.4%$ for 60 min at $80^{\circ}C$), and weight loss (FC-b: $13.9{\pm}0.8$, FC-f: $15.6{\pm}0.6$, NC: $10.6%{\pm}0.7%$ for 60 min at $80^{\circ}C$) for thermally processed potatoes.
