Diagnosis of Pigs Producing PSE Meat using DNA Analysis (DNA검사기법을 이용한 PSE 돈육 생산 돼지 진단)

  • Stress-susceptible pigs have been known as the porcine stress syndrome (PSS), swine PSS, also known as malignant hyperthermia (MH), is characterized as sudden death and production of poor meat quality such as PSE (pale, soft and exudative) meat after slaughtering. PSS and PSE meat cause major economic losses in the pig industry. A point mutation in the gene coding for the ryanodine receptor (RYR1) in porcine skeletal muscle, also known calcium (Ca$^{2+}$) release channel, has been associated with swine PSS and halothane sensitivity. We used the PCR-RFLP(restriction fragment length polymorphism) and PCR-SSCP (single strand conformation polymorphism) methods to detect the PSS gene mutation (C1843T) in the RYR1 gene and to estimate genotype frequencies of PSS gene in Korean pig breed populations. In PCR-RFLP and SSCP analyses, three genotypes of homozygous normal (N/M), heterozygous carrier (N/n) and homozygous recessive mutant (n/n) were detected using agarose or polyacrylamide gel electrophoresis, respectively. The proportions of normal, carrier and PSS pigs were 57.1, 35.7 and 7.1% for Landrace, 82.5, 15.8 and 1.7% far L. Yorkshire, 95.2, 4.8 and 0.0% for Duroc and 72.0, 22.7 and 5.3% for Crossbreed. Consequently, DNA-based diagnosis for the identification of stress-susceptible pigs of PSS and pigs producing PSE meat is a powerful technique. Especially, PCR-SSCP method may be useful as a rapid, sensitive and inexpensive test for the large-scale screening of PSS genotypes and pigs with PSE meat in the pork industry.y.

Economic analysis of Frequency Regulation Battery Energy Storage System for Czech combined heat & power plant (체코 열병합발전소 주파수조정용 배터리에너지저장장치 경제성 분석)

  • According to the new climate change agreement, technology development to reduce greenhouse gases is actively conducted worldwide, and research on energy efficiency improvement in the field of power generation and transmission and distribution is underway [1,2]. Economic analysis of the operation method of storing and supplying surplus electricity using energy storage devices, and using energy storage devices as a frequency adjustment reserve power in regional cogeneration plants has been reported as the most profitable operation method [3-7]. Therefore, this study conducted an economic analysis for the installation of energy storage devices in the combined heat and power plant in the Czech Republic. The most important factor in evaluating the economics of battery energy storage devices is the lifespan, and the warranty life is generally 10 to 15 years, based on charging and discharging once a day. For the simulation, the ratio of battery and PCS was designed as 1: 1 and 1: 2. In general, the primary frequency control is designed as 1: 4, but considering the characteristics of the cogeneration plant, it is set at a ratio of up to 1: 2, and the capacity is simulated at 1MW to 10MW and 2MWh to 20MWh according to each ratio. Therefore, life was evaluated based on the number of cycles per year. In the case of installing a battery energy storage system in a combined heat and power plant in the Czech Republic, the payback period of 3MW / 3MWh is more favorable than 5MW / 5MWh, considering the local infrastructure and power market. It is estimated to be about 3 years or 5 years from the simple payback period considering the estimated purchase price without subsidies. If you lower the purchase price by 50%, the purchase cost is an important part of the cost for the entire lifetime, so the payback period is about half as short. It can be, but it is impossible to secure profitability through the economy at the scale of 3MWh and 5MWh. If the price of the electricity market falls by 50%, the payback period will be three years longer in P1 mode and two years longer in P2 and P3 modes.

Quantifying Uncertainty of Calcium Determination in Infant Formula by AAS and ICP-AES (AAS 및 ICP-AES에 의한 조제분유 중 칼슘 함량 분석의 측정불확도 산정)

  • Uncertainty was quantified to evaluate calcium determination result in infant formula with AAS (Atomic Absorption Spectrometry) and ICP-AES (Inductively Coupled Plasma-Atomic Emission Spectrometry). Uncertainty sources in measurand, such as sample weight, final volume of sample, sample dilution and the instrumental result were identified and used as parameters for combined standard uncertainty based on the GUM (Guide to the expression of uncertainty in measurement) and Draft EURACHEM/CITAC Guide. Uncertainty components of each sources in measurand were identified as resolution, reproducibility and stability of chemical balance, standard material purity, standard material molecular weight, standard solution concentration, standard solution dilution factor, sample dilution factor, calibration curve, recovery, instrumental precision, reproducibility, and stability, Each uncertainty components were evaluated by uncertainty types and included to calculate combined uncertainty. The kinds of uncertainty sources and components in the analytical method by AAS and ICP-AES were same except sample dilution factor for AAS. The analytical results and combined standard uncertainties of calcium content were estimated within the certification range $(367{\pm}20\;mg/100g)$ of CRM (Certified Reference Material) and were not significantly different between method by AAS followed by ashing and method by ICP-AES followed by acid digestion as $359.52{\pm}23.61\;mg/100g\;and\;354.75{\pm}16.16\;mg/100g$, respectively. Identifying uncertainty sources related with precision, repeatability, stability, and maintaining proper instrumental conditions as well as personal proficiency was needed to reduce analytical error.

Characteristics of Inclusive Playground Guidelines (통합놀이터 가이드라인의 특성)

  • The inclusive playground is a playground where disabled children and non-disabled children can play together, not a playground for the disabled. It started with the change of social awareness of the citizenship rights of disabled people in the 1960s and the resulting playgrounds. Since then, inclusive playgrounds have been developed in many countries, and these are organized in the form by guidelines. In Korea, social interest in inclusive playgrounds is increasing, but there are no systematic guidelines in Korea, and the application of overseas cases or guidelines is limited. The purpose of this study is to classify the concept of inclusive playgrounds and design guidelines, that were previously presented in inclusive playground design guideline of various countries and analyze the characteristics of, design scope, and design principles, and provide a basic framework for creating guidelines. The purpose of the design guideline was to present specific numerical values to the inclusive playground design guidelines, to link with academic research and industrial products, to present pursuit values, and to expand the value of pursuing design methods. The contents were covered by scope, conceptualization, principles of design and design process, design guidelines, and checklists. Most of the guideline covers specific autonomous governments or countries that can apply the related systems or laws, but the composition of the detailed contents is different. The guiding value of inclusive playgrounds presented in each guideline is not a playground for the disabled but a playground for all, and some guidelines refer to the difficulty in playgrounds considering non-disabled children. Based on these concepts, design guidelines are presented in each guideline. Improving the accessibility in design principles is a common theme and adds to the principles of safety, independence, convenience, and playability. None of the guidelines do not provide design guidelines. Although there is a difference in the degree and method of specificity provided by each of the guidelines, the design guidelines can be generally summarized as space, copper line, and unit facilities. As mentioned in many guidelines, an inclusive playground is not only a playground for children with disabilities. Therefore, in the design guidelines, it is also important to the support play of children with disabilities and to induce inclusive play. The design guidelines presented in the guideline can be rearranged into three stages of 'supporting the play of children with disabilities', 'securing the dimensions and materials of spaces and facilities', 'adding auxiliary devices' and 'designing new facilities'. There are three design guidelines for inducing inclusive play. First, by creating various difficulty levels and intersecting spaces, children with various abilities can play with each other, and at the same time, they can interact witheach other. Second, all children can cooperate and play without distinction between children with disabilities and non-disabled children. Finally, the guardian provides the conditions for efficient support so that the disabled child can fully enjoy the inclusive playground.

Study on the Possibility of Estimating Surface Soil Moisture Using Sentinel-1 SAR Satellite Imagery Based on Google Earth Engine (Google Earth Engine 기반 Sentinel-1 SAR 위성영상을 이용한 지표 토양수분량 산정 가능성에 관한 연구)

  • With the advancement of big data processing technology using cloud platforms, access, processing, and analysis of large-volume data such as satellite imagery have recently been significantly improved. In this study, the Change Detection Method, a relatively simple technique for retrieving soil moisture, was applied to the backscattering coefficient values of pre-processed Sentinel-1 synthetic aperture radar (SAR) satellite imagery product based on Google Earth Engine (GEE), one of those platforms, to estimate the surface soil moisture for six observatories within the Yongdam Dam watershed in South Korea for the period of 2015 to 2023, as well as the watershed average. Subsequently, a correlation analysis was conducted between the estimated values and actual measurements, along with an examination of the applicability of GEE. The results revealed that the surface soil moisture estimated for small areas within the soil moisture observatories of the watershed exhibited low correlations ranging from 0.1 to 0.3 for both VH and VV polarizations, likely due to the inherent measurement accuracy of the SAR satellite imagery and variations in data characteristics. However, the surface soil moisture average, which was derived by extracting the average SAR backscattering coefficient values for the entire watershed area and applying moving averages to mitigate data uncertainties and variability, exhibited significantly improved results at the level of 0.5. The results obtained from estimating soil moisture using GEE demonstrate its utility despite limitations in directly conducting desired analyses due to preprocessed SAR data. However, the efficient processing of extensive satellite imagery data allows for the estimation and evaluation of soil moisture over broad ranges, such as long-term watershed averages. This highlights the effectiveness of GEE in handling vast satellite imagery datasets to assess soil moisture. Based on this, it is anticipated that GEE can be effectively utilized to assess long-term variations of soil moisture average in major dam watersheds, in conjunction with soil moisture observation data from various locations across the country in the future.

Optimal Extract Condition for the Enhancement of Anticancer Activities of Artemisia princeps Pampanini (강화 사자발쑥의 항암활성 증진을 위한 추출조건의 최적화)

  • Extractions of Artemisia princeps Pampanini were obtained by using water and ethanol at different temperatures ($60^{\circ}C,\;100^{\circ}C$) with or without ultrasonification process (40 kHz). Yield of ultrasonificated extracts were about 20% higher than that of control group. Cytotoxicity of all conditions through adding 1.0 mg/$m{\ell}$ was below 37%, and treated with ultrasonification group was lower than the other group, about $5{\sim}8%$. $100^{\circ}C$ water extract with ultrasonification was higher anticancer activities as maximum 73% and higher selectivities at concentrations over 0.8 mg/$m{\ell}$. The extracts treated with ultrasonification were higher anticancer activities than the control. Densitometric analysis of bcl-2 revealed that extracts of high anticancer activity had low density. This results suggest that expression of bcl-2 protein by adding of Artemisia princeps Pampanini extracts relative to taking cancer. To conclude, optimum condition for efficient extraction of Artemisia princeps Pampanini is using water with ultrasonification at over $60^{\circ}C$ below $100^{\circ}C$.

Changes of the Prefrontal EEG(Electroencephalogram) Activities according to the Repetition of Audio-Visual Learning (시청각 학습의 반복 수행에 따른 전두부의 뇌파 활성도 변화)

  • In the educational study, the measure of EEG(brain waves) can be useful method to study the functioning state of brain during learning behaviour. This study investigated the changes of neuronal response according to four times repetition of audio-visual learning. EEG data at the prefrontal$(Fp_{1},Fp_{2})$ were obtained from twenty subjects at the 8th grade, and analysed quantitatively using FFT(fast Fourier transform) program. The results were as follows: 1) In the first audio-visual learning, the activities of $\beta_{2}(20-30Hz)$ and $\beta_{1}(14-19Hz)$ waves increased highly, but the activities of $\theta(4-7Hz)$ and $\alpha$ (8-13Hz) waves decreased compared with the base lines. 2). According to the repetitive audio-visual learning, the activities of $\beta_{2}$ and $\beta_{1}$ waves decreased gradually after the 1st repetitive learning. And, the activity of $\beta_{2}$ wave had the higher change than that of $\beta_{1}$ wave. 3). The activity of $\alpha$ wave decreased smoothly according to the repetitive audio-visual learning, and the activity of $\theta$ wave decreased radically after twice repetitive learning. 4). $\beta$ and $\theta$ waves together showed high activities in the 2nd audio-visual learning(once repetition), and the learning achievement increased highly after the 2nd learning. 5). The right prefrontal$(Fp_{2})$ showed higher activation than the left$(Fp_{1})$ in the first audio-visual learning. However, there were not significant differences between the right and the left prefrontal EEG activities in the repetitive audio-visual learning. Based on these findings, we can conclude that the habituation of neuronal response shows up in the repetitive audio-visual learning and brain hemisphericity can be changed by learning experiences. In addition, it is suggested once repetition of audio-visual learning be effective on the improvement of the learning achievement and on the activation of the brain function.

  • For the purpose of obtaining basic data which can be applied to processing of retort pouched shellfishes, retort pouched seasoned ark shell, Anadara broughtonii, was prepared. The frozen ark shell was thawed and seasoned with a mixed seasoning powder prepared with $10.0\%$ of sorbitol, $2.0\%$ of table salt and $0.5\%$ of monosodium glutamate at $5^{\circ}C$ for 10 hours, and then dried at $45^{\circ}C$ for 4 hours. The dried seasoned ark shell was coated with $1.0\%$ sodium alginate solution, dried with cola air blast for 2 hours and then vacuum-packed in the laminated plastic film bag (polyester/casted polypropylene= $12{\mu}m/70{\mu}m,\;15{\times}16cm$), and finally sterilized up to Fo=6.0 in hot water circulating retort at $121^{\circ}C$ for 10 minutes. The major fatty acids of raw ark shell and retort pouched seasoned ark shell products were 16:0, 20:5, 22:6, 18:0 and 18:3, and predominant free amino acids of those were lysine, arginine, glycine, alanine, glutamic acid and leucine. In nucleotides and its related compounds of raw ark shell and retort pouched seasoned ark shell products, the most abundant one was AMP, and total extract-N of those was chiefly consisted of free amino acids, betaine and nucleotide and its related compounds. During the processing procedure such as drying and sterilization, unsaturated fatty acids slightly decreased while saturated fatty acids increased, and total extract-N content decreased about a half. From the results of chemical and microbial experiments during storage, it was concluded that the products could be preserved in a good condition for 100 days at room temperature, and their duality could be improved by the coating treatment of sodium alginate solution.

Modeling and Intelligent Control for Activated Sludge Process (활성슬러지 공정을 위한 모델링과 지능제어의 적용)

  • The main motivation of this research is to develop an intelligent control strategy for Activated Sludge Process (ASP). ASP is a complex and nonlinear dynamic system because of the characteristic of wastewater, the change in influent flow rate, weather conditions, and etc. The mathematical model of ASP also includes uncertainties which are ignored or not considered by process engineer or controller designer. The ASP is generally controlled by a PID controller that consists of fixed proportional, integral, and derivative gain values. The PID gains are adjusted by the expert who has much experience in the ASP. The ASP model based on $Matlab^{(R)}5.3/Simulink^{(R)}3.0$ is developed in this paper. The performance of the model is tested by IWA(International Water Association) and COST(European Cooperation in the field of Scientific and Technical Research) data that include steady-state results during 14 days. The advantage of the developed model is that the user can easily modify or change the controller by the help of the graphical user interface. The ASP model as a typical nonlinear system can be used to simulate and test the proposed controller for an educational purpose. Various control methods are applied to the ASP model and the control results are compared to apply the proposed intelligent control strategy to a real ASP. Three control methods are designed and tested: conventional PID controller, fuzzy logic control approach to modify setpoints, and fuzzy-PID control method. The proposed setpoints changer based on the fuzzy logic shows a better performance and robustness under disturbances. The objective function can be defined and included in the proposed control strategy to improve the effluent water quality and to reduce the operating cost in a real ASP.

Effects of Feeding Levels and Particle Size of Germanium Biotite on Pig Performance (돼지 생산성에 있어 게르마늄흑운모의 첨가수준 및 입자도의 효과)

  • Two experiments were conducted to investigate the effect of germanium biotite (GB) on growth performance, nutrient digestibility and fecal gas emission in pigs. In Exp 1., a total of one hundred nursery pigs (initial body weight 13.12${\pm}$0.15kg) were used in a 21 d growth assay. The five treatments were control (CON, basal diet), GB0.5-200 (basal diet + 0.5% GB, 200mesh), GB1.0-200 (basal diet + 1.0% GB, 200mesh), GB0.5-325 (basal diet + 0.5% GB, 325mesh), GB1.0-325 (basal diet + 1.0% GB, 325mesh). Each treatment had four replicates with five pigs per replicate. ADG, ADFI and gain/feed were not significantly different among the treatments. Fecal NH3-N concentration of pigs fed the GB325 diet was lower than that of pigs fed the GB200 diet (P=0.01). The GB treatments reduced fecal volatile fatty acids significantly compared to the CON (propionic acid, P=0.01; butyric acid, P=0.01; acetic acid, P=0.02). Especially, fecal propionic acid concentration of pigs fed the GB325 diets was lower than that of pigs fed the GB200 diets (P=0.02). In Exp 2., a total of seventy five pigs (initial body weight 21.18${\pm}$0.15kg) were used in a 28 d growth assay. The treatments were same as described for Exp. 1. Each treatment had five replicates with three pigs per replicate. The GB1.0 treatments significantly increased the ADG compared to the GB0.5 treatments (P=0.03). The DM and N digestibility of pigs fed the GB1.0 diets were higher than that for pigs fed the GB0.5 diets (P=0.01). Also, the Ca digestibility of pigs fed the GB diets was higher than that for pigs fed the CON diets (P=0.01). The fecal NH3-N concentrations for the GB treatments were lower than that for the CON (P=0.01). The GB325 treatments significantly decreased the fecal NH3-N concentration compared to the GB200 treatments (P=0.03). The fecal butyric acid concentration for the GB325 treatments was lower than that for the GB200 treatment (P=0.04). In conclusion, the results obtained from these feeding trials suggest that the dietary GB for nursery pigs affects fecal noxious gas emission. In growing pigs, dietary GB was effective to improve ADG and decrease fecal noxious gas emission.
