  • Ultrastructures of the cutaneous xanthophores and formation of pigment granules in the goldfish, Carassius auratus L., are studied with electron microscope. The cutaneous xanthophores are observed only in dermis and pigment granules of these pigment cells are pterinosomes and carotenoid vesicles. By the differentiated level, pterinosomes are subdivided into 3 types; while type I pterinosomes have clear limiting membranes and contain some amorphous fine fibrous structures, type II pterinosomes have thick and densely aggregated fibrous materials. Type III pterinosomes have concentric lamellar structures in the granules. Pigment granules of the xanthophores are originated from the Golgi complexes and pinocytotic vesicles of plasma membrane as well as rER-rich cells among the chromatophores are presumed to be associated with the accumulation of pigment materials.

Dyeabilities with Various Fabrics and Chemical Composition of Brown Colorants from Pine Bark (적송수피 색소 성분의 화학적 조성과 섬유 염색성)

  • Chemical compositions and biological functions of brown colorants extracted from pine bark(Pinus densiflora) have been studied. Dyeing test using multifiber fabrics with extracted colorants were preliminary carried out. Dyeing conditions and fastness tests of selected fabrics have been also studied. The brown colorants were produced 1.5% concentrations by solvent extraction from milled pine bark using methanol. The colorants were extracted with 80% methanol as best choice by a criteria of solid quantity and dyeability on fabrics. The chemical compositions were identified as mixtures of taxifolin epicatechin and procyanidin by LC/MS analysis. The brown colorants could be dyed not only natural fibers such as cotton, silk and wool but also synthetic fiber as nylon and semi-synthetic fiber as viscose rayon. Maximum K/S values was shown at 400 nm according to different fiber with color appearance of redish brown. Optimum pH and temperature of dyeing conditions was 4 and above $80^{\circ}C$, respectively. The brown colorants had a strong antioxidant activity compared to Butylated hydroxyanisole as standard and weak antimicrobial activity against E. coli. compared to kanamycin. Washing, rubbing, perspiration, dry cleaning and light fastness for cotton, nylon and silk dyed with the brown colorants were carried out by KS K method. Most of color fastness such as washing, rubbing, perspiration, and dry cleaning were represented as 4-5 grade. However, light fastness was reported as 2-3 grade. From this studies, brown colorants produced pine bark have a high potentials for natural dyeing on fabrics with antioxidant activity.

Immunological Characterization and Localization of the Alcohol-dehydrogenase in Streptococcus pneumoniae (폐렴구균 알코올탈수소효소의 세포 특이성 및 세포내 분포)

  • Heat shock proteins serve as chaperone by preventing the aggregation of denatured proteins and promote survival of pathogens in harsh environments. In bacteria, ethanol shock induced the major chaperone GroEL and DnaK, but in Streptococcus pneumoniae, it induced neither GroEL nor DnaK but alcohol dehydrogenase (ADH). In this study, ADH gene encoding a 104-kDa (p104) protein was identified and characterized. The deduced amino acid sequence of pneumococcal ADH shows homology with other members of the ADH family, and particularly with Entamoeba histolytica ADH2 and E. coli ADH. S. pneumoniae adh is composed of 883 amino acids and its estimated isoelectric point is 6.09. Although ADH is conserved between S. pneumoniae and E. coli, immunoblot analysis employing antisera raised against pneumococcus ADH demonstrated no cross-reactivity with ADH analog in Eschericha coli, Staphylococcus aureus and human HeLa cells. Also secretion of ADH was demonstrated by subcellular fractionation and immunoblot analysis of proteins. These results suggest that S. pneumoniae ADH could be a highly feasible candidate for both diagnostic marker and vaccine.

Isolation and Identification of Antimicrobial Active Substance from Sophora flavescens Ait. (고삼으로부터 항균활성 물질의 분리 및 구조 동정)

  • The ethanol extract and its chloroform fraction of Sophora flavescens Ait. exhibited growth inhibition on some food poisoning bacteria. The minimum inhibitory concentration of the above extracts were $50{\sim}500\;ppm$ and below 50 ppm Listeria monocytogenes (ATCC 19111, 19112, 19113, 19114, 15313). By silica gel column chromatography twice, antimicrobial active compound S-10-6 was isolated from chloroform fraction of Sophora flavescens Ait. The fraction S-10-6 showed strong growth inhibition at 10 ppm on 5 strains of L. monocytogenes, Bacillus subtilis ATCC 14593 and Staphylococcus aureus KFCC 11764 but Esherichia coli ATCC 25922 was not inhibited at 100 ppm and also confirmed bactericidal effect at 30 and 50 ppm on 5 strains of L. monocytogenes. The antimicrobial compound S-10-6 was identified as kushenol F, a kind of flavanone compound, by EI/MS, $^1H-NMR\;and\;^{13}C-NMR$.

  • The ethyl acetate fraction from ethanol extract of Glycyrrhiza uralensis FISCH showed growth inhibition on 5 strains of Listeria monocytogenes (ATCC 19111, 19112, 19113, 19114 and 15313) at $50{\sim}100\;ppm$. By silica gel column chromatography twice, antimicrobial active compound G-4-4 was isolated from ethyl acetate fraction of Glycyrrhiza uralensis FISCH. The G-4-4 fraction showed growth inhibition at $10{\sim}50\;ppm$ and confirmed bactericidal effect at 50 and 100 ppm on 5 strains of Listeria monocytogenes. Staphylococcus aureus KFCC 11764 was inhibited at $30{\sim}50\;ppm$ of G-4-4 but Escherichia coli ATCC 25922 was not inhibited at 100 ppm. The antimicrobial active substance G-4-4 was identified as liquiritigenin, a kind of flavanone compound, by IR, MS, $^1H-NMR\;and\;^{13}C-NMR$.

Preparation and Evaluation of Physiological Activity of White Bread Containing Korean Blue Mussel (국산 양식 홍합 함유 식빵의 제조 및 생리활성 평가)

  • White breads containing Korean blue mussel (Mytilus edulis) powder were prepared and characterized. WB (white bread without blue mussel) and four different MBs (white breads containing blue mussel; number in front of MB means added % of blue mussel powder per wheat flour) were prepared by the straight dough method. With addition of blue mussel to bread, lightness decreased, whereas redness and yellowness increased. Addition of blue mussel did not significantly affect specific volumes of breads. DPPH and ABTS radical scavenging activities significantly increased with increasing blue mussel content. Addition of blue mussel to breads also increased alcohol dehydrogenase and acetaldehyde dehydrogenase activities. In the sensory test, 1MB acquired relatively high points for taste, flavor, texture, and preference. The results indicate that blue mussel can be applied to white bread to improve physiological functions without reduction of physicochemical characteristics.

Reproductive Ecology of the Freshwater Melania Snail, Semisulcospira coreana (v. Martens) in Bukhan River (북한강 참다슬기, Semisulcospira coreana (v. Martens) 의 번식생태)

  • To clarify reproductive ecology of the melania snail Semisulcospira coreana (v. Martens, 1886) in Bukhan River, gonad development, fatness, gonad index, sex ratio, first sexual maturity of population, monthly change of larvae number and developmental stages in brood pouches were investigated by six identification methods. As maturation progrsses, the sex of the snali can be distinguishable easily by color:: the ovary being blue-green and testis light yellow. The sex ratio of female to male individuals over 13.95 mm shell height was significantly different from 1:1 (${\chi}^2$ = 38.45, p < 0.05). The sex ratio of female to male individuals changed drastically according to the season, Based on the monthly variations of fatness, gonad index and histological analysis, spawning occurred twice a year (spring and autumn) and the mean size of matured eggs was $450{\mu}m$ in diameter. The monthly change of larval number in brood pouch showed also two distinct peaks in March and September during the year. The average number of larvae in brood pouches was 286 - 862 individuals. In this study, the number of larvae in the brood pouches were a minimum in December and a maximum in March (975 larvae). The biological minimum size (the size at 50% of group sexual maturity) of the melania snail was 13.95 mm in shell height in females and males. All females over 15 mm in shell height possesed brood pouches.

  • In order to develop the method for mass production of natural food colorant from Monascus anka, optimum cultivation conditions for producing red and yellow pigments by cultiva- ting the mold in a jar fermenter and their color characteristics were investigated. The mold produced red and yellow pigments both intracellularly and extracellularly. These pigments showed unique light absorption characteristics with maximum absorption of 494, 380, 506, and 388 nm for extracellular red pigment (ERP), extracellular yellow pigment (EYP), intracellular red pigment (IRP), and intracellular yellow pigment (IYP), respectively. Optimum conditions for producing red pigments were found to be temperature 30$\circ$C, initial pH 6.0, rice powder 3-5%, peptone 0.05%, magnesium sulfate 0.25%, aeration rate 0.1 vvm. Optimum temperature for producing yellow pigments was around 35$\circ$C which is higher than that of producing red pigments. The initial pH and rice powder concentration for producing yellow pigments were the same as those of producing red pigments. The higher concentration of nitrogen source and inorganic salt, aeration rate, the more the yellow pigments were produced. The optimum agitation speed was 100 - 300 rpm for pigment production.

Mucormycosis in a group of chickens (닭에 집단 발병한 뮤코마이코시스)

  • The etiologic agents of mucormycosis are several nonpathogenic fungi of the order Mucorales and are ubiquitous. A total of 82 chickens were infected with the disease and developed anorexia, diarrhea, malnutrition, dyspnea, and paralysis in a chicken farm, Sokcho, Kwangwon. At necropsy there were multiple nodular lesions and hemorrhages in livers, spleens, kidneys, gastrointestinal track, and respiratory system. On histopathological examination it was found that the nodular lesions were consisted of granoulomatous inflammation accompanying characteristic hyphae ($4{\sim}24{\mu}$ wide) of the fungi.

  • The cutaneous xanthophore differentiation from larvae to adult in Bombina orientalis-the Korean fire bellied toad-is studied with light and electron microscopes. General structure of adult xanthophore which is composed of many pterinosomes and a small quantity of carotenoid vesicles is forming chromatophore complex with other dermal pigment cells. And the cytoplasmic process of xanthophore is distributed just beneath the basement membrane. The first differentiated xanthophore is originated from both rER rich cells and Golgi complex rich cells before and after the feeding larval stage. Formation of the pigment granule is proceeded gradually along the sequental metamorphic stages. After the metamorphosis, rapid multiplication of pterinosome is observed and enlargement of carotenoid vesicle is appeared after hybernation. These pigment granules are seen several structures by the differentiated level.
