Objectives : We examined the effect of Hwangryunhaedoktang(HH) on the experimental ulcerative colitis induced by dextran sulfate sodium (DSS). Methods : Experimental colitis was induced in rats by daily treatment with 5% DSS in the drinking water for 5 days. Afterward, the mts were divided into two groups: the control group was administered water and the sample group was administered HH for 7 days. Results : The sample group provided HH for 7 days demonstrated fast recovery of body weight compared with the control group. Histologic change showed fast regeneration of crypt and surface epithelial cells and decreased edema of the submucosa and decreased lymphatic follicle of mucosa compared with the control group. Immunohistochemical stain usingCOX-2 gene was decreased and there was localized Ki-67 positive reaction. Regeneration of surface epithelial cell and goblet cell in mucosa was observed by transmission electron microscope. These results indicate therapeutic effect of HH on DSS-induced colitis in rats.
Objective : We analyzed 3 clinical cases to estimate the possibility of shanghanlun as a completed clinical text which has its own diagnostic system. Methods : We collected 3 cases treated by huang lian e jiao tang conducted by a practiced shanghanlun doctor and defined the conceptional meaning of each provisions by analyzing the application of them in each case. Results : Shanghanlun provisions dualized with Je Gang(提綱) and Jo Mun(條文) is the record of preceding factors which describe the phenomenon initiating the diseases. Conclusions : Rudimentarily a factor can bring about many outcomes, so if shanghanlun is structured record of the factors which can make a human sick from many diseases, it can be used as the completed clinical text only with those 113 prescriptions. Factors that shanghanlun recorded seem somewhat different from western conventional medicine because the field of each factors are very varied from biological phenomenon to psychological phenomenon. However it is not that weird because psychosomatic medicine in western conventional medicine also point out the factors come from mental, society and behavior.
This study investigate the patent trend of natural feed additives for avian influenza prevention and immunity enhancement. The search scope is Korean and Chinese patents published before December 31, 2017. As a result, I found 53 Korean patents and 105 Chinese patents. Korea and China has increased the number of patent applications since late 2000. In particular, China filed the most patent applications in 2016. Both Korea and China, the oriental medicine materials were mainly used in patent claims. Korea has many patent applications that are submitted by universities, research institutes and bio companies, but China has a high proportion of individual patent applicants.
Strawberry and cucumber were dipped with Korean medical herb extracts such as Rheum palmatum L. and Coptis chinensis Franch. to maintain the keeping quality during storage at 5$^{\circ}$C and 10$^{\circ}$C, respectively. Treatment with 500 ppm solutions gor strawberry retarded weight loss, microbial load increase, and decay during storage. Decay ratio was shown 68.8% for control, 45.0% for Rheum palmatum L. and 42.5% for Coptis chinensis Franch. at 10 days. Strawberry treated with Coptis chinensis Franch. showed a higher hardness and soluble solid content than others. Cucumber treated with 1,000 ppm solutions revealed a different O$_2$ and CO$_2$ concentrations after 16 days. Korean medical herb extracts showed an antimicrobial activity for the quality retention of cucumber during storage. Ascorbic acid content was generally increased till 8 days, but decreased thereafter.
Objectives : Coptidis Rhizoma has been known traditional medicine with antimicrobial activities. We investigated inhibitory effects of Coptidis Rhizoma extract on lipopolysaccharide(LPS)-induced nitric oxide production from mouse macrophages. Methods : After Coptidis Rhizoma extract was pretreated in BV2, mouse brain macrophages and RAW264.7 mouse macrophages, cells were activated with LPS. To investigate cytotoxicity Coptidis Rhizoma extract, cell viability was measured by MTT assay. The production of nitric oxide(NO) and inducible nitric oxide synthase(iNOS) was determined in each culture supernatant and mRNA by Griess reaction and RT-PCR. The production of $TNF-{\alpha}$ from cells was measured by ELISA. Results : Coptidis Rhizoma extract significantly inhibited LPS-induced NO production in BV2 and RAW264.7 cells. Coptidis Rhizoma extract also greatly suppressed mRNA expression of iNOS in BV2 and RAW264.7 cells activated by LPS. Conclusion : These data suggests that Coptidis Rhizoma extract may have an anti-inflammatory effect through the inhibition of NO production.
In order to evaluate acute toxicity of Coptidis rhizoma, 6 week- and 13 week-old male ICR mice received Coptidis rhizoma extract (600~4,800 mg/kg body weight) orally, and toxicological responses were observed for consecutive 7 days. In the mice received relatively high concentration of Coptidis rhizoma($\geq$1,200mg/kg), death occurred within 3 hrs after oral administration, and its ratio in 13 week-old mice was conspicuously higher than that in 6 week-old mice. $LD_{50}$ of Coptidis rhizoma were estimated to bi 2,575 mg/kg and 1,490 mg/kg body weight in 6 week and 13 week-old mice, respectively. Coptidis rhizoma-treated animals manifested a variety of abnormal clinical findings such as ptosis, crouching, lethargy, convulsion, bizarre behavior and truning sideway. These abnormalities also ranked highly in the 13 week-old mice compared to those in the 6 week-old mice. In addition to abnormal behaviors, Coptidis rhizoma($\geq$1,200 mg/Kg) significantly elevated the urinary contents of bilirubin, urobilirubin, protein and glucose, and values in 13 week-old mice was higher than those in 6 week-old animals. No toxicological response was observed at concentration less than 600 mg/kg. Our results clearly demonstrate that susceptibility of mice to Coptidis rhizoma may be related with age, indicating that younger age mice is more resistant to the Coptidis rhizoma than the older, and toxicological mechanism of Coptidis rhizoma may be closely associated with its pharmacological mechanism.
The Coptidis rhizoma is known for containing protoberberine alkaloids. Berberine, coptisine and palmatine are the major constituents of protoberberine alkaloids. The alkaloids were isolated and determined by forming complex compounds from Coptidis rhizoma in preparation I(Sam-Hwang-Sa-Sim-Tang) and II(Hwang-Ryen-Tang). For the determination of these alkaloids, a new spectrophotometric method was developed with a simple and selective sample clean-up using thiocyanatocobaltate[II] complex compound ion. The absorbance of alkaloidal complex compounds in l.2-dichloroethane solution was measured at 625 nm. Calibration curve for the alkaloids isolated from Coptidis rhizoma was linear over the concentration range of 0.2-0.3 mg/ml. The method was proved to be rapid, simple and reliable for the isolation and the determination of the alkaloids in Coptidis rhizoma preparation I and II.
A simple and reliable reverse phase HPLC method of Coptidis Rhizoma and Salviae Miltiorrhizae Radix were developed to determine pharmacologically active marker compounds. The water-extract of two natural medicines were evaluated by the HPLC methods. The stability test for two extract were examined for five months. However, no significant change in the content of the marker compounds of each extract observed during the time of investigation.
Our previous data showed the protein isolated from Coptidis Rhizoma (CRP) had antifungal activity. In present study, we examined mode of action of the CRP and its activity against subcutaneous candidiasis due to C. albicans yeast cells. Results showed that the CRP blocked hyphal production from yeast form of C. albicans. The CRP also activated RAW 264.7 monocyte/macrophage cell line, which resulted in nitiric oxide (NO) production from the cells. This activation seemed to increase macrophage phagocytosis to destroy the invaders. Like other antimicrobial peptides, CRP was influenced by ionic strength, thus resulting in a decrease of antifungal activity. In murine model of a subcutaneous candidiasis, the sizes of infected areas of the nude mice given the CRP after subcutaneous injection of C. albicans yeast cells to the dorsal skin were $90\%$ less than those of the nude mice groups that received DPBS instead of the CRP. All data indicate that the CRP, which appeared to act like an antimicrobial peptide and to inhibit the morphological transition from blastoconidia, was effective against the subcutaneous disease.

Objectives : This study was planned to examine the enhancing effect of Coptidis Rhizoma mask pack on water, oil contents and skin texture, and to check the possibility of commercialization. Methods : We measured the change of water, oil contents and skin texture before and after the use of Coptidis Rhizoma mask pack. Measuring was conducted twice at an interval of ten days using accurate SK-03 skin analyzer. Measuring site was right and left SI18 and CV24 acupoints. Results : We can examine the improvement of water, oil contents and skin texture on every acupoints measured, except for CV24. On CV24, no difference was observed from water contents of both 1st, 2nd visit, and skin texture of 1st visit. Conclusions : We could confirm the boosting effect of Coptidis Rhizoma mask pack on water, oil contents and skin texture, especially around the SI18 acupoints. We should take follow-up measures to improve the absorption of a liquid herbal medicine around the CV24 acupoint, and heighten the potential for commercialization.
