Physicochemical and Microbial Quality Characteristics of Garlic (Allium sativum L.) by Superheated Steam Treatment (과열증기 처리에 따른 마늘의 이화학적 및 미생물학적 품질 특성)

  • The objectives of this study were to investigate the effects of superheated steam (SHS) treatment on the physicochemical and microbial properties of garlic. The garlic was treated by SHS at temperatures of 100, 150, 200, 250, 300, and $350^{\circ}C$ for 60 s. The moisture content of raw garlic was lower than that of SHS-treated garlic. The total thiosulfinate and pyruvate contents were dramatically reduced by SHS treatments. The antioxidant activities of garlic measured by ferric reducing/antioxidant power, 2,2-azinobis(3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt radical scavenging assay, and total phenolics content decreased by SHS. The major volatile sulfur compounds of garlic such as diallyl disulfide, allicin, allyl sulfide, diallyl sulfide, and diallyl trisulfide were significantly reduced by SHS. The antimicrobial effects of raw garlic were stronger than those of SHS-treated garlic against three strains of bacteria, including Staphylococcus aureus, Escherichia coli, and Bacillus cereus. However, total aerobic bacteria in garlic were dramatically reduced by SHS from 8.6 to 2.9 log CFU/g. The results from the sensory evaluation show that SHS treatment of garlic above $200^{\circ}C$ provides better acceptably due to reduction of off-flavor and pungency of garlic. These results suggest that superheated steam treatment can used as an efficient process for reducing garlic off-flavor and pungency.

A Study on the Evaluation of Distribution Stability of Refrigerated Rice Cake Using Natural Antibiotics (천연항균제를 사용한 냉장떡류의 유통 안정성 평가연구)

  • Natural extract in liquid phase was adjusted to 0, 0.25, 0.5, 1, 2, and 4% concentration to check microbial changes and to measure 4, 8, $12^{\circ}C$ for refrigeration temperature. In the case of grapefruit extract, the microbial safety was maintained at all the concentrations at $4^{\circ}C$ storage, but the antimicrobial activity was maintained at $12^{\circ}C$ storage and at $8^{\circ}C$ and 21 days storage. In the case of grape seed extract, only the 4% of the culture at $8^{\circ}C$ satisfied the requirement of safety of food distribution for the last 21 days, and the safety criterion was satisfied only at 4% concentration at $12^{\circ}C$ for 18 days. Complex Scutellaria baicalensis extract showed the total number of microbial cells treated by concentration. It was confirmed that microbial flow safety was maintained at low temperature ($4^{\circ}C$). However, at $8^{\circ}C$ and $12^{\circ}C$, Exceeded the distribution limit. When polylysine was applied to brown rice cake, it showed activity in all groups except $4^{\circ}C$, but these properties were not observed at $8^{\circ}C$ and $12^{\circ}C$. At a concentration of 0.5% or more of chitosan, the growth of the microorganism is suppressed by the 21st day very stably, and a similar tendency is observed at 8 and $12^{\circ}C$, so that it may be an antimicrobial material that inhibits microorganisms. At the first day, the distribution standards for general bacterial counts were exceeded.Ethyl-pyruvate showed that microorganism safety was maintained at $4^{\circ}C$ and 1% concentration, and food safety was stable even at 2 or 4%. Glycine showed very good and stable distribution stability at $4^{\circ}C$. However, at $8^{\circ}C$ and $12^{\circ}C$, the shelf life of 14 days could not be maintained as with the addition of other antimicrobial active substances.

Growth Promotion of Tomato Plant under Drought Conditions by Treatment of Rhizobacteria Producing ACC Deaminase and Phytohormones (ACC Deaminase와 식물호르몬 생성 세균 처리에 의한 토마토 식물의 가뭄 조건에서의 생장)

  • Some rhizobacteria producing 1-aminocyclopropane-1-carboxylic acid (ACC) deaminase can make plant to continue growth under the stress conditions through lowering the level of phytohormone, ethylene which inhibits the plant growth and accelerates plant aging. In this study, some rhizobacteria producing ACC deaminase have been isolated from the rhizosphere of plants grown at sand beaches, and identified as Escherichia hermannii m-2, Enterobacter asburiae m-4, Pseudomonas thivervalensis BD2-26 and Pseudomonas brassicacearum subsp. neoaurantiaca BD3-35 through sequencing of 16S rRNA genes. Strain BD3-35 showed the highest activity of ACC deaminase among the isolates, 20.26 ${\alpha}$-ketobutyrate ${\mu}M/mg$ protein/h. Strains BD3-35 and BD2-26 secreted a phytohormone cytokinin, and strains m-4 and m-2 could produce auxin and abscisic acid, respectively. When these bacteria were applied to the 7-day old tomato plant under drought stress for 7 days, strains BD3-35, m-2, and m-4 increased the length of tomato root by 14, 15, and 35%, respectively, and strains m-2, BD2-26 and BD3-35 increased the dry weight of tomato plant by 22, 33, and 68%, respectively compared to the uninoculated control tomatoes. Therefore, these rhizobacteria may be utilized as a microbial fertilizer for the plants under drought stress.

Clinical Laboratory Aspect of Carbapenem-Resistant Enterobacteriaceae (카바페넴내성장내세균속균종의 임상검사 측면)

  • The correct distinction of carbapenem-resistant Enterobacteriaceae (CRE) and ccarbapenemase producing Enterobacteriaceae (CPE) and the rapid detection of CPE are important for instituting the correct treatment and management of clinical infections. Screening protocols are mainly based on cultures of rectal swab specimens on selective media followed by phenotypic tests to confirm a carbapenem-hydrolyzing activity, the rapid carbapenem inactivation method, lateral flow immunoassay, the matrix-assisted laser desorption ionization-time-of-flight test and molecular methods. The CPE is accurate for detection, and is essential for the clinical treatment and prevention of infections. A variety of phenotypic methods and gene-based methods are available for the rapid detection of carbapenemases, and these are expected to be routinely used in clinical microbiology laboratories. Therefore, to control the spread of carbapenemase, many laboratories around the world will need to use reliable, fast, high efficiency, simple and low cost methods. Optimal effects in patient applications would require rapid testing of CRE to provide reproducible support for antimicrobial management interventions or the treatment by various types of clinicians. For the optimal test method, it is necessary to combine complementary test methods to discriminate between various resistant bacterial species and to discover the genetic diversity of various types of carbapenemase for arriving at the best infection control strategy.

Enhancement of Tomato Tolerance to Biotic and Abiotic Stresses by Variovorax sp. PMC12 (Variovorax sp. PMC12 균주에 의한 토마토의 생물학 및 비생물학적 스트레스 저항성 증진)

  • Rhizobacteria play important roles in plant growth and health enhancement and render them resistant to not only biotic stresses but also abiotic stresses, such as low/high temperature, drought, and salinity. This study aimed to select plant growth promoting rhizobacteria (PGPR) with the capability to mitigate biotic and abiotic stress effects on tomato plants. We isolated a novel PGPR strain, Variovorax sp. PMC12 from tomato rhizosphere. An in vitro assay indicated that strain PMC12 produced ammonia, indole-3-acetic acid (IAA), siderophore, and 1-aminocyclopropane-1-carboxylic acid (ACC) deaminase, which are well-known traits of PGPR. The aboveground fresh weight was significantly higher in tomato plants treated with strain PMC12 than in non-treated tomato plants under various abiotic stress conditions including salinity, low temperature, and drought. Furthermore, strain PMC12 also enhanced the resistance to bacterial wilt disease caused by Ralstonia solanacearum. Taken together, these results indicated that strain PMC12 is a promising biocontrol agent and a biostimulant to reduce the susceptibility of plants to both abiotic and biotic stresses.

Effect of Low Temperature on the Qualities of Long-term Fermented Kimchi (Korean Pickled Cabbage) (저온이 묵은지의 발효에 미치는 영향)

  • Recently, many Koreans have started to consume aged kimchi fermented long-term at low temperatures. In the present study, the effect of low temperature ($5^{\circ}C$) on pH, titratable acidity (TA), organic acid level, viable microbial cell count, amino acid concentration, and polygalacturonase activity (PG) during long-term fermentation (46 weeks) of kimchi, were evaluated. After 10 weeks of fermentation, kimchi had a pH of 4.1 and a TA of 1.0%, respectively after 46 weeks fermentation, these values were 3.9 and 1.3%, respectively. Lactic acid, the ratio of lactic acid to acetic acid, and the ratio of Lactobacillus species/Leuconostoc species in kimchi increased as fermentation progressed from 10 weeks to 46 weeks. However, total viable cell counts of aerobic bacteria, yeasts, Lactobacillus species, and Leuconostoc species, free amino acid levels, and PG decreased as the fermentation period was extended from 10 weeks to 46 weeks.

  • 투어멀린(Tourmaline)은 비대칭 쌍극자를 가진 천연 유극성 결정체로 광물 중에서 영구전기분극을 띄고 있는 유일한 물질로써 일명 "전기석"이라고 알려져 있다. 분극에 의한 자체 미약전류(약 $60{\mu}A$)와 함께 음이온 및 원적외선의 발생으로 최근 우리주변에서 건강과 환경을 위한 관심고조의 대상인 육방정계의 압전성 및 초전성을 띄는 붕규산염(주요성분: Mg, Fe, B, Si, Ca)으로, 투어멀린 원석이나 그 복합세라믹체의 면역지수가 ~1000에서 ~400,000 이상인 점을 비롯하여 그 유용성을 이용하려는 새로운 아이디어 신상품들이 계속 개발되고 있다. 뿐만 아니라 물분자를 만나면 수소$(H^+)$와 수산기 $(OH^-)$로 전기분해 하여 친수기와 소수기 부분으로 구분하며, $H^+$$OH^-$는 각각 $H_2O$와 결합하여 활성이 강한 hydronium ion$(H_3O^+)$을 생성하고, 물 속에서 계속 불안정한 상태로 존재하는 수산기는 계면활성 작용이 있는 hydroxyl ion$(H_3O_2^-)$을 형성하여 물을 약 알카리성 (pH~7.4)으로 바꿔주며, 물분자 클러스터 세분화와 미네랄을 용출, 공급하여 이 물이 체내에 흡수되면 살균, 항균성의 면역기능을 갖게 되며, 혈액정화 기능과 자율신경 자극으로 교감신경의 흥분을 억제해 준다. 이와 함께 전기분극을 띈 투어멀린 입자는 다른 물질에 비하여 경이적 수치의 원적외선을 발생하여 강력한 열 효과와 높은 침투력으로 인체의 혈액순환 촉진과 혈전용해 작용을 주어 건강과 활력에 도움을 주는 것으로 확인되어 많은 관심을 끌고 있다. 따라서 유익한 이용가치를 인정받고 있는 천연 투어멀린을 단독 형태나 또는 서로 상승효과를 줄 수 있는 유용한 타 물질과 혼합물을 구성하여 성형 또는 EPD 방법으로 전착한 후 소결 처리하여, 소결체의 특성 분석과 함께 물에 대한 전기 화학적 작용 및 항균 효과를 조사, 확인하여 수질개선 효과를 분석하고 그 응용성을 개발, 확보하였다.

Cloning and Characterization of Zebrafish Microsomal Epoxide Hydrolase Based on Bioinformatics (생물정보학을 이용한 Zebrafish Microsomal Epoxide Hydrolase 클로닝 및 특성연구)

  • A gene encoding for a putative microsomal epoxide hydrolase (mEH) of a zebrafish, Danio rerio, was cloned and characterized. The putative mEH protein of D. rerio exhibited sequence similarity with mammalian mEH and some other bacterial EHs. A structural model for the putative mEH was constructed using homology modeling based on the crystallographic templates, 1 qo7 and 1 ehy. The catalytic triad consisting of $Asp^{233}$, $Glu^{413}$, and $His^{440}$ was identified, and the characteristic features such as two tyrosine residues and oxyanion hole were found to be highly conserved. Based on bioinformatic analysis together with EH activity assay, the putative protein was annotated as mEH of D. rerio. Enantiopure styrene oxide with enantiopurity of 99%ee and yield of 33.5% was obtained from racemic styrene oxide by the enantioselective hydrolysis activity of recombinant mEH of D. rerio for 45 min.

Characteristics of Antioxidative and Antimicrobial Activities of Various Cultivars of Sweet Potato (고구마의 품종별 항산화성과 항미생물 특성)

  • Antioxidative and antimicrobial activities of ethanol extracts from 2 colored sweet potatoes such as purple and yellow, 5 pale yellow sweet potatoes, and 8 general cultivars of sweet potato were investigated. Colored sweet potatoes showed the highest antioxidative activity, followed by Saengmi and Yeonmi, which were pale yellow sweet potato cultivars, and Hwangmi, which was a general cultivar of sweet potato. Colored sweet potatoes also showed distinctively lower mold and surface microbial number (cfu) than the pale yellow sweet potatoes and the general cultivars of sweet potato. Although all sweet potatoes tested did not represent any antimicrobial activity against yeast and mold, they exhibited strong antimicrobial activity against Streptococcus faecalis. Especially, purple and yellow sweet potatoes showed antimicrobial activity against wide range of bacteria.

  • 넙치 (Paralichthys olivaceus)로부터 elastase-like serine protease (PoElSp)를 암호화하는 cDNA를 클로닝하여 그 서열을 분석한 결과, PoElSp 유전자는 269 아미노산을 암호화하는 978염기쌍으로 구성되었다. PoElSp 유전자의 조직 특이적 발현 양상을 RT-PCR법으로 조사한 결과, 간, 비장 및 소장에서 그 발현이 크게 나타났다. lipopolysaccharide (LPS)로 인위적 세균감염을 유도한 후, 1시간째에 콩팥에서, 3시간째에는 근육에서, PoElSp 유전자의 발현이 크게 증가하였다. 또한, 이 유전자의 발현은 비장에서 LPS 주입 후 1-24시간동안 점차로 증가하였다. pro-mature PoElSp (proPoElSp)에 해당하는 cDNA를 pET32a 벡터 시스템을 이용하여 대장균에서 발현시켰다. 이 재조합 proPoElSp 단백질의 활성은 gelatin zymography 방법과 합성형광 Z-Phe-Arg-AMC의 분해법을 이용하여 측정하였다. 단백질 분해효소 활성을 위한 최적 pH는 7.5였다. 실험결과들을 종합하면, PoElSp 단백질은 면역 반응에서 중추적 역할을 하리라 판단된다.
