Antioxidant Effects of Solvent Fraction from Sanguisorbae officinalis L. with Acetone (오이풀 아세톤 추출물을 이용한 용매 분획물의 항산화 효과)

  • The solvent extracts of Sanguisorbae officinalis L. were investigated for the activities of antioxidants as a functional ingredient for cosmetic products. The electron donating effect of ethyl acetate layer and n-butyl alcohol layer was appeared similar activity with positive control butylated hydroxy anisole (BHA) at all concentrations. In addition, in 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid (ABTS) radical scavenging assay, ethyl acetate layer, n-butyl alcohol layer and water layer were over 99% effect at all concentrations and higher than that of BHA. Also in hydrogen peroxide scavenging assay, ethyl acetate layer and n-butyl alcohol layer were higher than that of positive control ascorbic acid. The measured superoxide dismutase (SOD)-like activity of n-butyl alcohol was more than 50% at concentration of 1,000 ${\mu}g/mL$ and superoxide anion radical scavenging ability showed more than 45% at 1,000 ${\mu}g/mL$ of n-butyl alcohol layer. All these findings suggested that ethyl acetate layer and n-butyl alcohol layer have a great potential as a cosmeceutical ingredient with an antioxidant effect.

Biological Activities of Solvent Fractions from Methanolic Extract of Crataegi fructus (산사(Crataegifructus) 메탄올 추출물로부터 용매분획된 분획물의 생리활성)

  • The purpose of this study is to determine the possibility of using Crataegi fructus as natural health food source. The research was conducted to determine the biofunctional activities of Crataegi fructus extract. Methanolic extract from Crataegi fructus was partitioned by using organic solvents, including n-hexane, ethyl acetate, n-butanol, and water. Ethyl acetate soluble fraction was shown to have the strongest antioxidant activity ($RC_{50}=4.39{\mu}g/m{\ell}$) among the fractions. In the antimicrobial activity assays, ethyl acetate soluble fraction was effective in bacterial inhibition, against the cases of Escherichia coli and Klebsiella pneumonia, with minimum inhibitory concentrations in $125{\mu}g/m{\ell}$. In the anticomplementary activity assays, water soluble fraction was the most effective exhibiting 18.4% inhibitory activity.

Anticancer Effects of the Extracts of Oriental Melon (Cucumis melo L. var makuwa Makino) Seeds (참외(Cucumis melo L. var makuwa Makino) 종자 추출물의 항암 활성)

  • The objective of this study was to investigate the anticancer effects of the extracts of oriental melon seeds. Various solvent extracts of oriental melon seeds were prepared and their anticancer effects were examined using in vitro MTT and CV assays. The anticancer effects of various extracts of oriental melon seeds were also examined in five human cancer cell lines including A549, AGS, HT-29, MCF-7 and HepG2. The ethanol extract of heated oriental melon seeds showed the potent cytotoxic effects especially against mouse hepatoma cell line(Hepa1c1c7), human hepatoma cell line(HepG2) and human breast cancer cell line(MCF-7). These data suggest that oriental melon seeds can be a promising anticancer agent against human liver and breast cancer.

Screening of the Immuno-stimulatory Activity of the Marine Alga Chlorella capsulate (해수 미세조류인 Chlorella capsulata의 면역활성증진)

  • A purified substance was separated from marine alga, Chlorella capsulate. Immuno-stimulatory activity was measured by observing the growth of human immune T and B cells. The growth of human T and B cells was enhanced by up to about 1.4 times by adding the fraction from C. capsulata, compared with commercially available CGF extracts from the fresh water alga, C. vulgaris. The secretion of TNF-a and IL-6 was also enhanced up to about 1.5 times by adding the separated fraction. Adding 0.4 g/L of the extract increased the production of nitric oxide from macrophages by 1.6 fold. 50% of the AGS cell growth was inhibited by adding 0.4 g/L of the water extracted sample.

The Effect of the Membrane Fluidity of Bellflower(Platycodon grandiflorum A.) Fractions on Liposomal Phospholipid Membranes (도라지 분획성분이 인지질막 Liposome의 유동성에 미치는 영향)

  • The object of this study was to investigate the effect of membrane fluidity of bellflower(Platycodon grandiflorum A. DC, ; PG) fractions in phosphatidylcholine(PC) liposomes, measured with high-sensitivity differential scanning calorimetry(DSC). We used dipalmitoylphosphatidylcholine(DPPC) bilayers which slake most stable liposomes among the other phosphatidylcholine. The sample PG was extracted and fractionated to five different types : butanol(PGMB), ethylacetate(PGMEA), ethylether(PGMEE), hexane (PGMH) and methanol(PGMM). Among five different solvent fractions, the PGMEE, PGMEA, PGMH and PGMM fractions markedly affected the thermotropic properties of DPPC liposomes, broadened and shifted the thermograms, and reduced the cooperative unit. It might be said that the incorporation of PGMEE, PGMEA and PGMH in DPPC liposomes were located in the hydrophobic core of DPPC bilayers and, PGMM and PGMB in the hydrophilic core of DPPC bilayers. These results suggest that certain substances in the PGMEE, PGMEA and PGMH fractions might have biologically significant effect on the membrane fluidity.

국내 잡곡산업의 현황과 발전방안

  • 잡곡은 혈압강하, 당뇨병 질환, 고혈압 예방, 면역성 증가, 혈당강하, 항산화, 항염증 효과 및 항암효과 등 기능적 특성을 가지는 작물로서 최근 사람들의 건강에 대한 인식이 증대되면서 다양한 생리활성을 가지는 잡곡에 대한 관심이 증대되고 있다. 그러나 잡곡은 현재 재배면적 및 생산량의 감소, 수입산에 대한 경쟁력 약화 및 농가 생산기피 등의 이유로 국내산 잡곡 생산량 및 재배면적은 감소하는 추세이다. 따라서 잡곡의 부가가치 상승 및 경쟁력 강화를 위한 다양한 노력이 요구된다. 최근 국내 식품의 트렌드는 바쁜 사회생활 및 불균형한 식습관으로 인하여 편이식이 또는 식사대용 형태의 편이식이 제품의 수요가 높아지고 있다. 따라서 다양한 생리활성을 가지는 잡곡을 활용한 indoor용 맞춤형 가공식품 개발은 바쁜 일상생활으로 인한 영양 불균형으로 건강에 대한 우려가 있는 현대인들 특히 영양요구성이 가장 높은 청소년 및 고령층의 영양보충뿐만 아니라 수입산 잡곡에 대한 국내 잡곡시장의 경쟁력 강화 및 농가소득 상승에 도움이 될 것으로 여겨진다.

  • The objective of this study was carried out in order to evaluate the antioxidative activities of n-hexane fractions from medicinal herbs. Certain parts from 6 species of edible and medicinal herbs were extracted with methanol. Radical scavenging activity of the n-hexane fractions against DPPH and ABTS radicals were observed to be high in Psoralea corylifolia, and Scutellsria barbata; it also significantly increased (p<0.05). For a comparison of the effect of various concentration of fractions, as the concentration of treatment increased, the antioxidative effects on radical scavenging activity also oncreased; yet it was not significant. The antioxidative effect of medicinal plants was dependent on the concentration but was not significant. Therefore, these results indicate that edible wild plants, such as Psoralea corylifolia, and Scutellsria barbata may be useful as potential antioxidant sources for improving the human antioxidant defense system against disease.

The Effect of Dissolved Cement Powder on Carassius auratus (용해된 시멘트 분말이 붕어에 미치는 영향)

  • In this study, we studied the effect of dissolved cement powder on Carassius auratus by analysis of the morphophysiological changes. The gill exposed to dissolved cement powder showed the thickened primary lamellae and the activity of chloride cells and mucous cells was also significantly increased and the proliferation, separation and clubbing of gill filament was observed in the secondary lamellae. In the kidney tissue, the space in Bowman's capsule was widen and the arrangement of dermis was irregular due to the thinned epidermis in the integument tissue. The activities of antioxidant enzymes and LDH tended to increase with the duration of cement exposure. It was confirmed that the up-regulated proteins were identified as involved in glycolysis and energy metabolism and down-regulated proteins were myofibrillar proteins which were involved in muscle contraction by the cement exposure to the integument. With these results, dissolved cement powder was thought to be a big threat to the survival of the fish because it causes the morphological changes and weakens the physiological activity in C. auratus tissues.

Stress Resistance of Zacco koreanus due to Exposure of Cement Powder (시멘트 분말 노출에 따른 참갈겨니의 스트레스 저항성)

  • This study investigated the effects of dissolved cement powder on Zacco koreanus by analyzing morphophysiological changes. The gills exposed to dissolved cement powder developed abnormal shapes in their secondary lamellae and increased numbers of mucous cells after long-term exposure. Additionally, clubbing, edema, and exfoliation of the epithelial cells were observed in the secondary lamellae. In the kidney tissue, the space in Bowman’s capsule was widened, and in the integument tissue the arrangement of the dermis was irregular due to the thinned epidermis. These results suggest that long-term exposure to cement powder can significantly affect morphological change, resulting death. The activities of antioxidant enzymes and LDH tended to increase commensurately with the duration of cement exposure. It was concluded that up-regulated proteins were the stress proteins involved in myofibrillar-protein production and down-regulated proteins were involved in glycolysis and energy metabolism after the integument’s exposure to cement. According to these results dissolved cement powder is a serious threat to the survival of fish because it causes morphological changes and weakens physiological activity in Z. koreanus tissues.

  • The multi-lamellar and liquid crystal structures have drawn great public attention in the functional cosmetic and skin-related medicinal areas recently. The structure of an emulsion containing aqueous phase as a binding water and fixed oil phase components forming an association compound of the multi-lamellar structure can reconstruct the intercellular lipid lamellar structure in the stratum corneum and restore barrier function of the skin. In this study, we investigated the beneficial effect of bio-mimic liquid crystal emulsions (BLCE) containing higher fatty alcohol, lecithin, and cholesterol on the skin barrier function, and evaluated its cytotoxicity.
