Analysis of UreB Protein Synthesis from Transgenic Lily Pollen (형질전환 백합화분을 이용한 UreB단백질의 발현분석)

  • In an attempt to produce recombinant proteins using the pollen enriched in some plant species, a 1.7 kb DNA encoding urease subunit B (UreB) amplified by PCR from Helicobacter pylori urease gene cluster in pH808 plasmid was cloned to be expressed under CaMV35S promoter in lily (Lilium longiflorum) pollen tubes elongated in vitro. Lily pollen at early germinating stage was transformed with the ureB DNA using Agrobacterium via vacuum infiltration and, incubated for a full pollen tube growth 16 - 24 h in the dark in the presence of kanamycin. DNA integration and expression in the transgenic pollen were analyzed by the standard molecular techniques and the results suggest that the pollen in vitro may be employed as a protein factory in a disposable fashion.

Chemical Composition and Antioxidant Activity of Korean Buckwheat (Fagopyrum esculentum) Pollen Grain Collected by Honey Bee, Apis mellifera (메밀화분의 성분 특성 및 항산화 활성)

  • We evaluated the nutritional composition including proximate, amino acid, vitamin, minerals, and the antioxidant activity of buckwheat (Fagopyrum esculentum) pollen grains collected by Apis mellifera bees, to be used as a species of forage plant with Quercus acutissima (acorn) and Actinidia arguta pollen grains. The content of crude protein and fat were found 14.43% and 5.67%, respectively. Eighteen amino acids from buckwheat pollen were found, including 8 essential amino acids. The predominant amino acids were glutamic acid, aspartic acid and lysine, accounting for about 42.7% of total free amino acids. The concentration of vitamin C was the highest value of 13.7 mg/100g, followed by $B_3$ (niacin) and $B_1$ among the detected vitamins. The predominant minerals were potassium (1197.95mg/100g), followed by phosphorus (962.77mg/100g) and magnesium (535.42mg/100g), whereas copper, zinc and manganese were detected as minor elements. Antioxidant activity and total phenolic content accounted for 8.1% at $500{\mu}g/ml$ extract and $2.25{\mu}g/mg$, respectively.

Chemical Components and Pulverization Conditions of the Pollens of Pinus densiflora S. et Z. and Pinus thunbergii Parl (소나무와 해송 화분의 화학적 성분과 파쇄 조건에 관한 연구)

  • This study was to analyze the chemical components of the pollens of Pinus densiflora S. et Z. and P. thunbergii Parl. and to investigate the optimal conditions for pollen pulverization. The results were as follow : (1) The contents of moisture, crude ash, crude protein, crude fat, crude filber and carbohydrate in the pollen of P. densiflora were 9.8%, 2.3%, 11.9%, 2.9%, 7.7%, and 65.4%, respectively, while those of P. thunbergii were 10.5%, 2.2%, 13.5%, 3.5%, 8.5% and 61.8%, respectively. All the contents of P. densiflora and P.thunbergii were much higher than those general crop grains. (2) The pollen of Pinus densiflora and P. thunbergii eighteen different amino acids were detected in. Among them, ten esssential amino acids were identified, which showed high nutrition values. (3) The contents of vitamins A, B1, B2, C and E in the pollen of P. densiflora were 7.9mg, 8.7mg, 19.2mg, 31.5mg and 2.0mg, respectively, while those in the pollen of P. thunbergii were 8.5mg, 9.4mg, 15.7mg, 35.3mg, and 1.8mg respectively. Vitamin C among them was abundant. (4) When the pollen grains of P. densiflora were pilverized for 40 minutes at 5,000rpm and P. thunbergii, for 50 minutes at 5,000rpm by overhead stirrer, the 100% of pollen pulverization was possible.

Convenient Evaluation of Stored Apple Pollen Viability by Fluorochromatic Reaction (형광염색반응에 의한 장기 저장 사과 화분의 활력 측정)

  • In order to evaluate stored apple pollen viability, in vitro germination test was performed on a microscope slide coated with the culture medium containing fluorescein diacetate (FDA). However, the inclusion of FDA to the culture medium declined pollen germination. Alternatively, the fluorochromatic reaction procedure was tested. The procedure involved dusting pollen grains onto drops of 10% sucrose solution containing 0.002% FDA and allowing them to accumulate fluorescein. Within 30 min after the fluorochromatic reaction, viable pollen grains clearly fluoresced under ultraviolet light. Both the in vitro germination test and the fluorochromatic reaction procedure revealed that stored apple pollen viability was not considerably decreased over storage up to at least 39 months. Of the cultivars examined by both methods, 'Fuji' and 'Senshu' pollen viability was highest, 'Tsugaru' was intermediate, and 'Jonagold' was lowest. The fluorescing percentages appeared approximately comparable to the germination percentages except for the 'Senshu' pollens stored for 3 months, although the fluorescing percentages was slightly higher than the germination percentages. Strong and highly significant correlations were found between the two methods. It can thus be concluded that the fluorochromatic reaction procedure provides a convenient and reliable evaluation of stored apple pollen viability.

Comparative Morphology of Pollen Grains from Some Korean Gymnosperms (한국산 나자식물 화분의 비교형태)

  • Pollen grains of 19 taxa belonging to 12 genera in 5.families of the Korean gymnosperms were investigated by LM, SEM and TEM, and their morphological characters were described and palynological keys were made for each family. From the taxonomic viewpoint, palynological characters are very available to intergeneric and intrageneric classifications since their distinctive differences in pollen shape, size, condition of surface, sculpture pattern and presence of air bladders and projection except for the intrageneric classification of the genus Pinus since the overlapped characters such as pollen size and sculpture pattern. Pollen grains of Picea koraiensis in the family Pinaceae are distinguished from thc genera Pinus and Abies for the reasons that shape of air bladders are irregular, air bladders are bigger than pollen body, and pollen wall is thin. Compared with the genera Abies and Pinus that have air bladders, Picea koraiensis is closely related to Abies in that pollen body wall become gradually thin from pole of proximal face to air bladder, surface of pollen body is not rough, and pollen size is similar to that of the genus Abies. The presence of ektexine and enexine in proximal face, and the connections of the former to air bladder wall and the latter to distal face were elucidated by the investigation of pollen grains with air bladders through TEM. Endexine of the families of Taxaceae, Taxodiaceae and Cupressaceae was also obse-rved as a pile of laminae. From the observation by SEM, sculpture pattern of air bladder was foveolate with fine pits on surface. Sculpture pattern of air bladder is reticulate under LM, but observed by TEM, it resulted from the transmission of netted structure in air bladder.

Beneficial Effects of Nano-sized Bee Pollen on Testosterone-induced Benign Prostatic Hyperplasia in Rodents (테스토스테론-유도 양성전립선비대증에서 나노화 벌 화분의 효능 연구)

  • Bee pollen is one of many types of alternative remedies, and it has been used for a long time throughout the world. It has numerous health effects, including antifungal, antibacterial, and antioxidant properties, immune modulation, enhanced cell proliferation, and even anti-carcinogenic effects. This study was designed to elucidate the effects of bee pollen on benign prostatic hyperplasia in rodents. For this experiment, we used nano-sized bee pollen produced through wet-grinding technology, thereby the extraction efficiency of the active ingredients in the bee pollen was significantly enhanced. First, We found that nano-sized bee pollen significantly reduced the size of prostates enlarged by chronic testosterone administration. In addition, nano-sized bee pollen significantly reduced the plasma concentration of the prostate-specific antigen (PSA). Interestingly, nano-sized bee pollen did not reduce the testosterone-induced increase in the plasma concentration of prostaglandin $E_2$ ($PGE_2$). The beneficial effects of nano-sized bee pollen in reducing both the size of the prostate and the plasma concentration of PSA was comparable to that of dutasteride. Finally, nano-sized bee pollen did not cause damage in LNCaP cells which are androgen-sensitive human prostate adenocarcinoma cells. Together, these data indicate that nano-sized bee pollen may be able to be used as a good alternative remedy for the treatment of benign prostatic hyperplasia.

Quality Characteristics of Brown Rice Dasik Prepared with Pollen Powder (화분 분말을 첨가한 현미다식의 품질 특성)

  • To enhance the quality of traditional Dasik, pollen Dasik containing brown rice flour fortified with 5, 10, 15 and 20% pollen powder were prepared, and their physicochemical properties were evaluated. Pollen Dasik possessed higher amount of mineral such as K and Ca than brown rice Dasik. The L value of pollen Dasik decreased with increasing pollen powder compared to brown rice Dasik, while the b value of pollen Dasik was increased with increasing pollen powder compared to brown rice Dasik(p<0.05). Hardness of pollen Dasik decreased with increasing amounts of pollen compared to the brown rice Dasik(p<0.05). Sensory evaluation on pollen Dasik preparewd with over 10% pollen showed the highest sensery evaluation value in overall preference.

Effects of Cattail Pollen Powders on the Rheology of Dough and Processing Adaptability of White Pan Bread (부들화분을 첨가한 밀가루 반죽의 물성과 제빵 적성)

  • To explore cattail pollen powder as a functional food ingredient, we analyzed the general components of pollen powder, tested changes in the physical properties of dough containing the powder, and investigated the process ability of powder-containing dough in bread making by adding 3%, 6%, or 9% by weight of pollen powder to wheat flour. Cattail pollen powder consisted of (all w/w) 12.7-13.2% water, 15.7-17.8% crude protein, 1.3% crude fat, 7.5-7.7% free sugar, 14.7-18.6% crude fiber, 3.4-4.9% pollen, and 49.7-55.9% soluble nitrogen-free extract (NFE). Analysis of the physical properties of dough mixed with pollen powder showed that as more pollen powder was added, the absorption rate increased, but dough stability decreased. With increasing levels of cattail pollen powder, the falling number decreased, and amylase activity increased. Fermentability was highest in dough made with 3% by weight of pollen powder, and the bread product made from such dough had the greatest volume. As more cattail pollen powder was added, the moisture activity in dough tended to decrease to a greater extent than seen in control dough, and this tendency increased with time. We found that longer storage periods were associated with greater hardness and springiness, which indicated degradation in product quality. Therefore, it is suggested that bread products containing cattail pollen powder should be consumed within 3 days of preparation. In a taste survey, bread baked with 3% (w/w) cattail pollen powder scored highest in all questionnaire items.

Investigation of Various Pesticide Residues in Commercial Bee Pollen Products Sold in South Korea (한국에서 유통되는 화분식품의 잔류농약 함량 분석)

  • To analyze the pesticide residues in commercial bee pollen products in South Korea, 61 samples were collected and screened for 339 pesticides. Results revealed that approximately 34% (>LOQ) of samples were contaminated with at least one pesticide. The pesticide residue detection rates of domestic and imported samples were 31% and 44%, respectively. Furthermore, the pesticide residue detection rate of online distribution (60%) was higher than that of offline distribution (27%). Fifteen pesticides were discovered in bee pollen, and pendimethalin, chlorfenvinphos, chlorpyrifos, and fluazinam were detected in 7, 6, 3, and 2 order of frequency, respectively. Even though its concentration was low, chlorfenvinphos which is banned in food crops in the United States, European Union, and Korea, was detected in bee pollen samples commonly. Therefore, continuous investigation of pesticide residues in bee pollen products and their acceptance criteria is required for safety.

A Study on the Pollen Morphology of the Genus Acer L. in Korea (한국(韓國)에서 생육(生育)하는 단풍나무속(屬) 화분(花粉)의 형태학적(形態學的) 연구(硏究))

  • The pollen morphology of 19 species and 1 variety of the genus Acer L. in Korea was investigated by light and scanning electron microscopy. The results are as follows ; 1. A. pollen key of the genus Acer L. in Korea was provided in four types. 2. Based on the aperture types and sculpture patterns, the four major pollen types of the genus Acer L. in Korea were recognized which included palmatum, barbinerve, negundo and saccharum types. 3. Pollen grains are tricolpate or tricolporate on the aperture, and striate, rugulate or reticulate on the sculpture pattern. 4. As the result of the correlation analysis between the five measured pollen parameters in pairs, a highly significant positive correlation was found between polar axis length(PL) and colpus length (CL).
