Study on New Extraction Method of Microcystins from Cyanobacteria (남조류로부터 마이크로시스틴을 추출하는 새로운 추출법 연구)

  • A new analytical method of cyanobacterial toxins, i, e, microcysins was deveeloped using supercritical fluid extraction(SFE). The microcystins mcluded in the study are sparsely soluble in neat supercritical fluid CO$_2$ However, the microcystins were successfully extracted with a temary mixture(90% CO$_2$,9.0% methanol 1.0% water) at 40$^{\circ}$C and 250 atm. The SFE method developed in this study has several advantages over solid-phase extraction(SPE) sample preparation for the analysis of microcystins. Sample handling steps are minimized thus reducing possible losses of analytes and saving analysis time. No clean-up steps are employed in this SFE method. Althouhgh many methods have been described for microcystim RR and LR, the method using solid-phase extraction with ODS cartridges is the most commonly used. However, the adsorbing power of ODS caridges for microcystins is weak, so we have attempted to use a more polar CN cartridge, to increase the adsorbing power for microcystins. Lyophilized cells(100mg) were wxtracted with 5% (v/v) acetic acid. The extract was centrifuged and then the supernatant was applied to a CN cartridge. The cartridge which contained microcystins was rinsed with 5 ml of water and 5 ml of 0.5 M acetic acid. followed by 5 ml of 5% acetonitrile in water , and were determined by HPLC. Better recoveries and chromatogram were observed than with ODS cartridge.

바실러스에 의한 중금속 제거 및 EPS 추출물질 비교에 관한 연구

  • 1) 바실러스는 불리한 환경조건에서 포자를 형성하며 포자는 균체가 죽어도 살아 남고, 외부 환경이 좋아지면 다시 발아하여 영양형 균체를 형성하게 된다. 본 연구에서 바실러스를 우점화와 포자의 활성화를 위해 열처리(80$^{\circ}C$, 10min)를 한다 울산분뇨처리장 슬러지 Cake를 배양하여 포자화 전 후 상태의 미생물의 EPS 추출결과 RCF법을 적용하였을 때 Carbohydrate물질이 다른 추출법에 비해 가장 많이 추출되었고, Steaming extraction법을 적용하였을 때 Protein이 가장 많이 추출되는 것으로 나타났으며, 포자화 전 보다 포자화 후의 EPS 양이 더 많이 추출될 것이라는 가설을 가지고 실험한 결과 포자화 전보다 포자화 후의 EPS 양이 더 많이 추출되었다. 울산분뇨처리장의 슬러지 Cake를 배양하여 포자화 전 후의 Cu의 제거정도를 비교한 결과 포자화 후의 EPS물질이 포자화 전의 EPS물질보다 Cu를 더 많이 제거하는 것으로 나타났으며 EPS 물질이 중금속 제거능력이 탁월하다고 할 수 있겠다. 2) 용인분뇨처리장과 천안하수처리장의 MLSS의 중금속 제거능력을 알아보기 위해 미생물량을 고려했을 경우와 미생물량을 고려하지 않은 경우로 나누어서 실험을 하였는데 울산분뇨처리장의 슬러지 Cake를 가지고 실험한 결과와 마찬가지로 Bacillus의 EPS물질이 중금속 제거에 탁월하다는 것을 알 수 있다. 3) Baillus를 사용하는 김천 식품폐수 처리장과 활성슬러지를 사용하는 천안하수처리장과의 EPS 함량을 비교한 결과 김천 식품폐수 처리장의 EPS 물질중 Protein과 Carbohydrate가 천안하수처리장의 활성슬러지보다 더 많은 것으로 나타났다.TEX>, DIN/DIP비 표층수 $23.91\pm3.42$, 저층수 $23.43\pm3.38$이었으며, 전반적으로 해역별 수질기준 I등급 내지는 II등급을 유지하고 있었고, 공간적으로는 외해측으로 갈수록 외해수와 혼합 확산되어 양호한 수질을 나타내었다. 장기적인 변동특성은 세그룹으로 구분되어진다.기 실험결과 용출용매로 증류수와 해수를 이용했을 때, 제강 슬래그에서 용출되는 납, 구리, 카드뮴, 수은의 용출 경향의 차이를 확인할 수 있었고 이에 따라서, 납, 구리, 카드뮴의 용출 유해성은 낮기 때문에 해양구조물로의 제강슬래그 유효이용은 적합할 것으로 판단되었다.im80%$로 계산되었다. 열형광선량계로 측정된 방사선량은 각각 1.8, 1.2, 0.8, 1.2, 0.8 (70 cm 거리) cGy로 측정되었으며, 환자의 복부 표면에서의 서베이메터를 이용한 측정량은 10.9 mR/h였다. 차폐구조물의 사용 시 전체 치료 동안에 태아선량은 약 1 cGy 정도로 평가되었다. 결론 : AAPM Report No.50의 자료에 따르면, 임산부의 방사선 치료 시 태아의 방사선 피폭선량은 5 cGy 이하일 경우에 방사선 피폭에 따른 태아의 위험이 거의 없는 것으로 제시되고 있다. 본원에서 차폐 구조물을 설치하였을 경우에 측정된 태아선량은 약 1 cGy로 측정되었고, 고안된 차폐구조물은 태아에 도달하는 방사선량을 감소시키기에 적합한 설계임이 입증되었다. 아니라 일반종합병원에서도 CTX-M형 ESBL 생성 E. coli와 K. pneumoniae가 존재하며 확산 중임을 시사한다. 앞으로 CTX-M형 ESBL의 만연과 변종 CTX-M형 ESBL의 출연을 감시하기 위한 정기적인 연구와 조사가 필

  • Prunus mume has been used as a Korean medicine. It is effective in treating diarrhea and an abdominal pain. This experiment was carried out to optimize extraction conditions of prunus mume flower tea and to analyze volatile compounds. Three kinds of samples treated with fresh, freeze dry, and shade dry, were used, and prunus mum flower tea was manufactured by the mixed ratio of green tea and prunus mume flower. The result was valued by the Hunter's value, flavor and taste. The optimum conditions of extraction time and temperature were 3 min and $80^{\circ}C$ respectively. Sensory evaluation shows that optimum ratio was adaptive 90% green tea with 10% prunus mume flower. The major volatile compound in prunus mume flower was benzaldehyde.

The Anti-Oxidant and Whitening Activities of Seaweeds Mixture Fermentation Extracts (복합해조류 발효추출물의 항산화, 미백 활성)

  • Studies on seaweed-based materials have been progressing steadily day by day. In this experiment, we checked the anti-oxidant, whitening, and moisturizing activities of fermented extract from a mixture of Undariapinnatifida, Saccharina japonica, and Gloiopeltis furcate. Also, Lactobacillus sakei strains of kimchi were used as the lactic acid bacteria. The physiological status of the combined seaweed extracts was also investigated. The 1,1-diphenyl-2-picrylhydrazyl (DPPH) and 2,2'-azino-bis-3-ethylbenzothiazoline-6-sulphonic acid (ABTS) radical scavenging results showed that the inhibitory effects of the combined seaweed extracts were higher than the positive control. Furthermore, 3,4-dihydroxy-L-phenylalanine (L-DOPA) and Mushroom tyrosinase tests were conducted during the whitening efficacy experiment. Hence, it was confirmed that the whitening activity of fermented extracts was greater than the extracts without fermentation. HPLC analysis of fucose (an active ingredient of seaweed) was also performed and a standard method for solvent conditions was newly established. This study suggests that the composite of algae extract has potentials to be used as anti-oxidant and whitening agents in cosmetics.

Damaged hair improvement effect of natural complex extract (천연 복합추출물의 손상모발 개선효과)

  • In this study, NF Complex of 0%, 0.5%, 2.5%, 12.5%, and 100% was prepared by complexing and extracting Indian gooseberry, Rosa multiflora Thunberg roots, and saw palmetto fruit in a ratio of 5:1:1. The manufactured NF Complex was applied to bleached sample hair and then compared and analyzed with damaged hair. To confirm the improvement effect, tensile strength, gloss, absorbance, and brightness were measured. As a result of the measurement, the tensile strength increased. The gloss content decreased by 100%, but the remaining content increased. The change in absorbance was minimal. There was also a change in brightness, but it was minimal. It was confirmed that there is a significant difference in the average value of NF Complex, and it is judged necessary to study various ratios in the future.

Evaluation of Antibacterial Effects of a Combination of Coptidis Rhizoma, Lonicerae Flos, Paeonia Japonica Extracts, and Dioctahedral Smectite Against Salmonella Typhimurium in Murine Salmonellosis

  • The present study was undertaken to estimate the antibacterial effect of a combination of C. rhizoma, L. Flos, and P japonica (1:1:1) extracts (CLP1000) and a combination of the herbal extract mixture and dioctahedral smectite (CLPS1000) against murine salmonellosis. At the concentration of CLP1000 and CLPS1000 0.5 mg/ml, the antibacterial effect was not showed on Salmonella typhimurium (S. typhimurium). On the other hand, the antibacterial effect against S. typhimurium was observed at the concentration of CLP1000 and CLPS1000 1.0 mg/ml. Oral administration of Smectite, CLPl000, and CLPS1000 at the dose of 10 mg/ml showed a therapeutic effect for S. typhimurium infected BALB/c mice. The mortality of Smectite, CLP1000 and CLPS1000-treated mice was 90%, 90%, and 70% at 12 days, respectively, while that of untreated mice was 100% at 9 days after a lethal dose of S. typhimurium infection. The results of our study strongly indicate that CLPS1000 has potential as an effective of salmonellosis.

  • Manuka oil named New Zealand’s tea tree oil is oil-soluble and comes firom nature. Manuka oil and its extract $\alpha$-pinene, Oxy’less clear, R-limonene which is one of the component of Citron extracted from Grapefruit seed and Citrex were used to estimate the antimicrobial activity and to improve the capability of antiseptic. Disk diffusion method was used to measure the antimicrobial activity. Escherichia coli which is gram-negative bacteria and Staphylococcus aureus which is gram-positive bacteria were used as strain. The antimicrobial activity of Manuka oil and $\alpha$-pinene for Escherichia coli, Staphylococcus aureus was similar when the concentration of Manuka oil and $\alpha$-pinene are 10u/paper disk. However, antimi-crobial activity of Manuka oil fDr Escherichia coli, Staphylococcus aurem was better than that of $\alpha$-pinene when the concentration of Manuka oil and $\alpha$-pinene was low. Antimicrobial activity of Oxy’less clear is better than that of propyl para hydroxybenzoate(PPHB), R-limonene at all the concentration and is similar to that of $\alpha$-pinene.   • An organic acid material, which can be manufactured by plants extraction, encourages microbe proliferation over time. Microbial activity, which is affected by organic acid, encourages accelerating consolidation with biochemical penetration; soil particles are compacted by microbes and pore water is dissipated quickly. Additionally, $CaCO_3$ for cementation was made by proliferating microbes. Accordingly, tests were conducted to investigate the unconfined compressive strength and permeability of soil samples aged with and without an organic acid. In the 96 days of aging, the strength was generally 1.5~2.5 times greater than those without an organic acid material and permeability was definitely decreased to 74.2~93.1%. SEM analysis showed the change of pore structure and the change of the total bacteria counts revealed the activity of microbes reflecting the engineering characteristics and this material would be an environment-friendly for soil improvement.

  • In order to improve the usability of glue, a traditional adhesive material, and the gelation process of seaweed extract carrageenan, a liquid adhesive for wood preservation was prepared by mixing the two materials. The prepared wood adhesive was mixed with a 15 wt% aqueous solution of glue, λ-carrageenan, an antibacterial agent, an antifoaming agent, and the maximum adhesive strength of 1.80 Mpa was confirmed. As a result of comparison, the adhesive strength was superior to that of 5 different types of traditional natural adhesive ingredients (aqueous solution) and 1 type of polyvinyl acetate-based adhesive.

Antioxidant Effects of Crotalaria sessiliflora L. Leaf Extracts Fractionated from Each Solvent (활나물 잎 추출물의 분획별 항산화 활성)

  • In this study, Crotalaria sessiliflora L., leaf was extracted with a solvent mixture of $CH_2Cl_2$ and $CH_2OH$ (3:1). In order to draw or separate effective components from the leaf extract with high activity, a 1:1 solvent mixture of water and butanol was used after removal of the sugar and fat from the extract. From the decompressed and concentrated butanol layer, 10 small fractions (Fr1, Fr2, Fr3, Fr4, Fr5, Fr6, Fr7, Fr8, Fr9, and Fr10) were obtained using a chromatography column filled with sephadex LH-20. The activity of each fraction was measured by the same method as that used to measure the antioxidant effect of each part(p<0.05). The Fr1, Fr4, and Fr6 fractions showed the highest activity levels(p<0.05). The vital unknown material that vitality exists in the strong butanol fractions of F1, F4 and F6 will be isolated and identified. Currently, we are performing for a single compound from an unknown peak by instrumental analysis.
