Genetic Polymorphism in Corticotropin-releasing Hormone Receptor Type-1 in Preeclamptic Korean Women

  • Purpose: Placental corticotropin-releasing hormone receptor type 1 (CRHR1) expression is reduced in pregnancies with abnormal placental function such as preeclampsia (PE), and the levels and/or function of CRHR1 are genetically influenced. The aim of this study was to investigate the association between the c.33+8199C>T polymorphism in the CRHR1 gene and PE in a Korean population. Materials and Methods: Using a case-control design, the association between the CRHR1 polymorphism and the risk of PE was investigated in 203 individuals with PE and 211 normotensive controls. Genotypes were determined using a SNapShot kit and an ABI Prism 3100 Genetic analyzer. Results: Genotypes and allele frequencies for the CRHR1 polymorphism did not differ between PE and normotensive pregnancies. The variant T allele was more frequent than the ancestral C allele in both of the groups and was more frequent in the controls than in the cases. In risk analysis for PE, there was not an increased risk of preeclampsia in subjects who were concomitant homozygous rare allele genotypes (CC) (OR, 0.3; P=0.15) or heterozygous rare allele genotypes (TC) (OR, 0.8; P=0.29). There were no differences in the complications of PE such as severity or preterm delivery in patients with the CRHR1 polymorphism. Conclusion: Our findings indicate that the CRHR1 polymorphism was not associated with PE in the present Korean study group.

Comparative Evaluation on Sperm Parameter of Transgenic Pigs with General Pigs (형질전환 돼지의 정자와 일반돼지의 정자성상에 대한 비교평가)

  • Pig has been known to be one of the most feasible animals as a bioreactor to produce pharmaceuticals in milk and as a mediator in xenotransplantation research. Previously, we generated transgenic pigs for both purposes, which were expressing Factor 8, vWF, hTPA, and hEPO in milk, along with expression of MCP at GalT gene locus ($GalT^{-MCP/-MCP}$) as well as expressing MCP at GalT gene loci with CD73 expression ($GalT^{-MCP/+}/CD73$). In this study, we performed comparative analyses of sperm parameters between wild type male (WT) pig and those transgenic males to examine the effects of transgenes integrated into the pigs on motility, morphology, viability, and acrosome integrity of the spermatozoa. Our results showed that the rates of actively motile spermatozoa of WT, Factor 8, vWF, hTPA, hEPO, $GalT^{-MCP/+}/CD73$, and $GalT^{-MCP/-MCP}$ pigs were 85.0%, 83.3%, 82.5%, 83.3%, 82.5%, 77.5%, and 78.7%, respectively. Whereas, the rates of morphologically normal spermatozoa of WT, Factor 8, vWF, hTPA, hEPO, $GalT^{-MCP/+}/CD73$, and $GalT^{-MCP/-MCP}$ pigs were 90.0%, 80.0%, 80.0%, 83.3%, 85.0%, 91.8%, and 80.8%, respectively. In addition, the viability in spermatozoa of WT, Factor 8, vWF, hTPA, hEPO, $GalT^{-MCP/+}/CD73$, and $GalT^{-MCP/-MCP}$ pigs were 93.9%, 82.4%, 89.9%, 83.9%, 87.4%, 92.8%, and 83.6%, respectively. The rates of spermatozoa with normal acrosome integrity in WT, Factor 8, vWF, hTPA, hEPO, $GalT^{-MCP/+}/CD73$, and $GalT^{-MCP/-MCP}$ pigs were 98.1%, 98.6%, 98.6%, 98.7%, 98.1%, 99.5%, and 95.1%, respectively. There were no significant differences in motility, morphology, viability, and acrosome integrity of the spermatozoa among WT, Factor 8, vWF, hTPA, and hEPO, $GalT^{-MCP/+}/CD73$, and $GalT^{-MCP/-MCP}$ pigs. These mean that neither random integration nor targeted integration of the transgene into chromosome of pig effect on characteristics of spermatozoa. Ultimately, the transgenic male pigs subjected in this study could apply to propagate their progenies for production of human therapeutic proteins and advancing the xenotransplantation research.

Change in Potential Productivity of Rice around Lake Juam Due to Construction of Dam by SIMRIW (벼 생장모형 SIMRIW를 이용한 주암호 건설에 따른 주변지역의 벼 잠재생산성 변이 추정)

  • To estimate the change in rice productivity around lake Juam due to construction of artificial lake, growth, yield components and yield of rice were measured at different locations around lake Juam for three years from 1994 to 1996. Automated weather stations(AWS) were installed nearby the experimental paddy fields, and daily maximum, average and minimum temperature, solar radiation, relative humidity, and precipitation were measured for the whole growing period of rice. Plant height, number of tillers, leaf area and shoot dry weight per hill were observed from 8 to 10 times in the interval of 7 days after transplanting. Yield and yield components of rice were observed at the harvest time. Simulation model of rice productivity used in the study was SIMRIW developed by Horie. The observed data of rice at 5 locations in 1994, 3 locations in 1995 and 4 locations in 1996 were inputted in the model to estimate the unknown parameters. Comparisons between observed and predicted values of shoot dry weights, leaf area indices, and rough rice yield were fairly well, so that SIMRIW appeared to predict relatively well the variations in productivity due to variations of climatic factors in the habitat. Climatic elements prior to as well as posterior to dam construction were generated at six locatons around lake Juam for thirty years by the method of Pickering et al. Climatic elements simulated in the study were daily maximum and minimum temperature, and amount of daily solar radiation. The change in rice productivity around lake Juam due to dam construction were estimated by inputting the generated climatic elements into SIMRIW. Average daily maximum temperature after dam construction appeared to be more or less lower than that before dam construction, while average daily minimum temperature became higher after dam construction. Average amount of daily solar radiation became lower with 0.9 MJ $d^{-1}$ after dam construction. As a result of simulation, the average productivity of habitats around lake Juam decreased about 5.6% by the construction of dam.

  • A study was conducted to find out the varietal difference in some important agronomic characters in response to different seeding time in barely from 1975 to 1976 at Chikugo Agric. Expt. Station in Japan. Thirteen varieties which are diverse in growth period were used. Nine seeding time were tried. Seedling emergence, primary tillering, and formation of flower primodia were delayed in proportion to the seeding time. And also heading and maturing time showed the same tendency. This tendency appeared more conspicuous in the late maturing varieties. Therefore, days to heading and maturing duration were inversely shortened by the delay of seeding time and by earliness of variety. Number of spikes as well as culm length and spike length was reduced in the late seeding. Grain yield was reduced in all varieties by late seeding, easpecially with late maturing varieties. In this test, widely adaptable variety to late seeding time was not found, but earliness and high tillering in growth habit appeared more favorable to yield increase.

Effects of Planting Density and Tiller Removal Growth and Yield of Sweet Corn Hybrids (재식밀도와 얼자제거가 단 옥수수의 생육 및 수량에 미치는 영향)

  • Two sweet corn hybrids, 'Tanok l' and 'Golden Cross Bantam 70 (GCB 70)' were grown at five plant densities, of 4, 167. 5,556, 6,667. 8, 333, and 11, 111 plants per 10 ares, with or without tiller removal, to determine effects of tiller removal on growth and yield of sweet corn hybrids at various plant densities. Tillers were pulled when less than 15 cm tall. The number of tillers per plant linearly decreased as plant density increased. The two hybrids had similar plant height, ear length and diameter, ear weight and the number of ears per plant and 10 ares. Tanok 1 lodged approximately 20% at above 8,333 plants per 10 ares, while GCB 70 did not lodge at all, at any plant density. Tanok 1 had higher leaf area index (LAI), ear and stover yields than GCB 70. Except for root lodging and LAI, hybrid x plant density interaction was not significant at 5% probability level. Plant density did not affect silking data. Increasing plant density linearly increased plant height, LAI, and stover yield, but linearly decreased ear length, ear weight, and the number of ears per plant. Increase in LAI was greater in Tanok 1 than in GCB 70, with increasing plant density. The relationships between the number of ears and ear yield per 10 ares and plant density were Quadratic. The optimum plant density was estimated to be approximately 6500 plants per 10 ares, using the equation based on ear yield. Except for ear height and LA I, hybrid x tiller removal and plant density x tiller removal interactions were not significant. Hybrid x plant density x tiller removal interaction was not significant for any characters. When averaged over hybrids and plant densities, tiller removal reduced plant height and ear and stover yields by about 3, 10, and 16%, respectively, but did not significantly affect silking date, root lodging, ear length and diameter and the number of ears per plant and per 10 ares. The results indicate that the optimum plant density is approximately 6500 plants per 10 ares, regardless of tiller removal and tillers are not to be removed at any plant density.

Evaluation of Agronomic Characteristics and Fatty Acid Composition of Flax Germplasms (아마 유전자원의 농업적 특성 및 지방산 조성 평가)

  • In this study, agronomic characteristics and fatty acid composition of 121 flax (Linum usitatissimum L.) germplasms were analyzed. The flax germplasms had a fairly short growth period of 75-95 days. The plant height at maturity varied from 52 to 124 cm. The flower color varied from white to lilac and even purple. Early flowering flax germplasms had white flowers and were resistant to lodging. In early flowering flax germplasms, all agronomic characteristics, including growth period, plant height, branch number per plant, stem diameter, number of capsules per raceme, seed number per capsule, 1000-seed weight, and seed yield, were decreased. The average fatty acid composition of 121 flax germplasms comprised palmitic acid (5.3%), stearic acid (4.3%), oleic acid (29.7%), linoleic acid (13.1%) and linolenic acid (46.4%). The total content of unsaturated fatty acids ranged from 84~94% with an average of 88%. Among the fatty acids, linolenic acid (omega-3) content was the highest with a range of 36~54%. In five selected flax germplasms (Hollandia 1803, Red son, C & F Res Br 1767, Wiera, and Ireland 1657) with high linolenic acid (${\geq}51%$), total lipid and protein contents were 30~36% and 5.4~6.9%, respectively. Overall, flax can practically be adoptable into cropping systems in South Korea and utilized as a scenery crop, since flax has a short cultivation period with diverse flower colors.

Responses of Photosynthetic Characters to Waterlogging in Soybean [Glycine max (L.) Merrill] (과습에 따른 콩 광합성 관련 형질 반응)

  • Stress due to excess water is one of the most limiting factor for soybeans to high yield under wet climates. This study aimed to identify the photosynthetic responses of soybeans to waterlogged growing condition with 5 soybean varieties by waterlogging for 10 days at V5 and R2 stage, respectively. Chlorophyll fluorescence decreased more rapidly at R2 stage waterlogging than at V5 stage waterlogging in all soybean tested varieties. The degree of recovery was much more in Pungsannamulkong and Muhankong( 95~97% of control) than in Jangyeobkong and Myungjunamulkong at 5 days after waterlogging. Photosynthetic rate, transpiration and stomatal conductance were also increased more rapidly in Pungsannamulkong and Muhankong than in Jangyeobkong and Myungjunamulkong after waterlogging irrespective of waterlogging stages. As the result of multiple regression analysis in order to identify the effects of stomatal conductance and transpiration to the photosynthetic rate, the R2 value of stomatal conductance in control and waterlogging treatment was 0.7293 and 0.7582, respectively. If the transpiration, another dependent variable, was added to the regression formula, there was not so big difference in the variation of photosynthetic rate. This result means that if just one factor of them(the stomatal conductance and transpiration) be measured in the case of waterlogged condition, the changes of photosynthetic rate can be estimated.

Genetic Variations in Geographic Venus Clam(Gomphina aequilatera, Sowerby) Populations from Samcheok and Wonsan (삼척과 원산의 지리적 민들조개(Gomphina aequilatera, Sowerby) 집단의 유전적 변이)

  • Genomic DNAs(gDNAs) were isolated from the venus clam(Gomphina aequilatera) from Samcheok(venus clam from Samcheok; VCS) and Wonsan(venus clam from Wonsan; VCW) located in the East Sea of the Korean Peninsula. The amplified products were generated by agarose gel electrophoresis(AGE) with oligonucleotides primer, detected by staining with ethidium bromide and viewed by ultraviolet ray. The seven arbitrarily selected primers BION-21, BION-23, BION-25, BION-27, BION-29, BION-31 and BION-33 generated the shared loci, polymorphic, and specific loci, with the molecular sizes ranging from 150 bp to 2,400 bp. In this study, 147 polymorphic loci(147/954 loci, 15.41%) in VCS population and 274(274/996 loci, 27.51%) in VCW population were generated with seven primers. These results suggest the genetic variation in VCW population is higher than in VCS population. Especially, the 700 bp bands generated by the primer BION-21 were identified commonly in two Gomphina populations, which identified populations and/or species. This specific primer was found to be useful in the identification of individuals and/or population, resulting from the different DNA polymorphism among individuals/species/population. Two Gomphina populations between the individual SAMCHEOK no. 03 and WONSAN no. 22 showed the longest genetic distance(0.696) in comparison with other individuals used. The complete linkage cluster analysis indicating three genetic groupings and dendrogram revealed close relationships among individual identities within two geographical populations of venus clam(G. aequilatera) from the Samcheok and Wonsan. The intra-species classification and clustering analyses inferred from molecular markers supported the traditional taxonomy of the species based on morphological characters such as shell size, shape and color. Accordingly, as mentioned above, RAPD analysis showed that VCS population was more or less separated from VCW population.

Studies on the Estimation of the Genetic Parameters on All Traits in Korean Native Ogol Fowl III Estimations of the Heritabilities and Genetic Correlations on the Egg Shape Index and Egg Qualities (한국재래오골계의 제형질에 대한 유전모수추정에 관한 연구 III, 난형지수 및 난질에 대한 유전력 및 유전상관추구)

  • This study was conducted to estimate heritabilities and genetic correlations on egg shape index and egg qualities in Korean Native Ogol fowl. The date analysis were a total of 58,320 eggs in 450 pullets bred from 150 dams and 20 sires of korean Native Ogol fowl raised at Chungnam National University from June 18, 1987 to April 6, 1989. The results obtained are summarized as follows:1. On the egg shape index and egg qualities, the egg shape index at first egg, 300 and 500 days of age were 75.044, 74.169 and 72.601 ; the shell thickness were 0.342, 0.320 and 0.326 mm: the albumen height were 6.014, 5.161 and 4.807mm:the Haugh units were 83.903, 71.348 and 71.136, respectively. 2. The heritabilities estimates of egg shape index and egg qualifies based on the varience of sires, dams and combined components were 0.120-0.827, 0.485-0.503 and 0.232-0.872 for egg shape index at first egg, 300 days and 500 days of age: 0.197-0.819. 0.184-0.756 and 0.279-0.557 for shell thickness at first egg, 300days and 500days of age:0.202-0.678, 0.119-0.394 and 0.225-0.527 for albumen height at first egg, 300 and 500 days of age 0.108-0.669, 0.237-0.251 and 0.354-0.443 for Haugh units at first egg 300days and 500days of age. 3. The genetic correlation coefficients of egg shape index and egg qualifies were as follows; between egg shape index and shell thickness, albumen height and Haugh units were 0.596-0.909, 0.384-0.943 and 0.121-0.619:between shell thickness and albumen height. Haugh units were 0.082-0.596, -0.076-0.167:between albumen height and Haugh units were 0.374-0.964.

A Study on Effects of Breeding Combination for Feeding and Economic Analysis in Broiler Stock (육용종계의 교배조합이 실용계의 사양과 경제성에 미치는 영향)

  • In order to identify the best superior cross combination of breeder stocks for broiler production, combining ability test and analyses of phenotypic performances for parent stocks were examined on records of 1,440 broiler chicken which were produced from 4 parental strains and 3 maternal strains at Hanhyup Poultry Breeding Farm from September 28, 1978 to January 5, 1979. The results obtained were as follows; 1. There was not found heterosis effect in viability but it seems to be desirable to select Hubbard strain in paternal line to improve viability. 2. As the paternal and maternal lines, selection of Ross strain showed the best paternal and maternal performance and the best general combining ability in body weight at 8 weeks of age is expected to be able to improve body weight of it s crossbred And the most superior cross combinations based on the specific combining ability and performance of rack crossbred were identified as Hubbard x Ross ana Ross x Hypeco crossbreds. 3. The best paternal and maternal lines on the smallest feed consumption for 8 weeks were Hubbard and Ross strains, and Hypeco strain, respectively. Especially Hubbard x Hypeco cross combination was proved as the smallest feed consumption compared with other cross combinations. 4. In feed requirement per Kg body weight increase, Hubbard strain for paternal line, Hypeco strain for naternal line, and cross combinations of Hubbard x Hypeco, Hubbard x Ross and Ross x Hypeco were certified as the most superiors. 5. Also superior cross combinations of Hubbard x Hypeco and Hubbard x Ross earned the most profit per bird through economic analysis. According to results as shown above, this experiment seems to be able to reach a such conclusion that production of superior cross combinations Hubbard x Ross, Hubbard x Hypeco and Ross x Hypeco through selection of Ross and Hubbard strains to paternal line and Hypeco and Ross strains for maternal line may become to considerable improvement for important economic characters of broiler; viability, body weight, feed consumption and feed requirement.
