Morphological and Cellular Criteria of Ovaries, Follicles and Oocytes for In Vitro Maturation in the Pig (체외배양을 위한 돼지 난소 및 난포란의 형태학적 조건)

  • 돼지난포란의 안정된 체외배양 체계를 위하여 도살장에서 재취된 난소로부터 난포 및 난자난구세포체의 혀태적인 선정기준을 설정하고 이의 이론적 배경을 확립하였다. 난소를 난포상태 및 분포나 황체 존재 여부에 따라 A,B 및 C 의 세 가지 형태로 분류하고 각 type의 난소에서 직경 3-5mm인 난포로부터 난포란을 회수하였다. 회수된 난포란을 난구세포 부착상태에 따라 Good, Fair 및 Poor 의 세 가지 형태로 분류하여 각각을 호르몬이 첨가된 M16+FCS 배양액으로 35시간 동안 체외배양하여 성숙율을 비교 검토 하였다. 난소의 형태에 따라 회수된 난포란 중 Good 또는 Fair 형태는 Type A 및 C 난소에서 85%를 차지한 반면, Type B 난소에서는 53%에 불과하였다. 또한 이들 난포란을 체외배양한 결과 Type A 및 C 에서 회수된 난포란은 90 및 85%의 높은 성숙율을 보인 반면, Type C 의 난포란은 33%의 저조한 성숙을 나타냈다. 한편 핵형 분석 및 조직학적 분석에서도 Type C 난소의 경우 난포란의 핵형이 대부분 GVBD 및 퇴화 형태를 보였으며, 폐쇄포난포의 비율도 Type A 난소의 53%에 비해 월등히 높은 85%를 나타내어 성숙율 비교실험의 결과와 일치되는 경향을 나타내었다. 따라서 본 실험의 선별기준에 의한 돼지 난소 및 난포란의 형태적 분류 작업에 의해 난포란의 체외배양 성적 향상 및 안정된 배양체계의 확립이 가능하였다.

Karyotypes of Genus Liobagrus (Pisces : Amblycipitidae) in Korea (한국산(韓國産) 퉁가리속(屬) 어류(魚類)의 핵형(核型) 분석(分析))

  • Karyological characteristics were investigated in 3 species of the genus Liobagrus from Korea. The diploid chromosome number in L. andersoni was found to be 28, with 9 pairs of metacentrics and 5 pairs of submetacentric chromosomes, and arm number (AN) was 56. L. mediadiposalis was found with 2n of 42, consisting of 13 pairs of metacentrics and 8 pairs of submetacentric chromosomes (AN=84). In the case of L. obesus 2n was 20, with 20 metacentric chromosomes (AN=40), which was the lowest among the species of the order Siluriformes. Sexual dimorphism or intraspecific polymorphism of the chromosomes was not observed in any species examined.

  • A Giemsa C-banding method was used for the identification of somatic chromosomes of Korean indigenous maize(Zea mays L.). Two Korean indigenous maize stocks and an American hybrid maize for comparison were examined. Ten deeply stained heterochromatic knobs whose position and size were different between the genotypes, two satellites and interstitial bands were observed. The length of homologous chromosomes compared by the relative lengths of chromosomes presented as a percentage of the length of chromosome 10 were different between the genotypes. The Giemsa method proved to be useful for the identification of somatic chromosomes and for the characterization of different stocks of Korean indigenous maize.

  • Morphology and karyotype of Korean Convallaria majalis plants were observed for taxonomic studies. Most plants produced two leaves (70%) and some one leaf (30%), but rarely three leaves. Plant length averaged 34.4 $\pm$ 4.6 cm. Percentage of plants bearing flowers was very low with 3.7% (n=1,346) in the field. Raceme usually grew below leaves and rarely grew over leaves. Plant beared five to ten flowers on a single raceme. Chromosomes were x=19 and diploidy with 2n=38. The chromosomes were composed of 13 pairs of median and 6 pairs of submedian chromosomes. The number of chromosome and karyotype of Korean C. majalis plant agree with those of Japanese and European plants.

  • The comparative karyological analysis of the Korean treefrogs, Hyla japonica and Hyla suweonensis, were performed by C-banding method. Heteromorphic sex chromosomes, female heterogamety, has been identified in the 3rd chromosomes of H. suweonensis H. suweonensis seem to have sex chromosomes which are Zz/ZW type. The Z chromosomes contain large amount of constitutive heterochromatin, but little heterochromatin is located in the W chromosomes. This is in contrast to all previously known amphibian and most other vertebrate's W or Y chromosomes, except Gastrotheca oui'ern and G. walken (Schmid et al., 1988).

  • Cytogenetic characteristics of three Hemibarbus species (H. labeo, H. longirostris and H. mylodon) were analyzed based on erythrocyte measurement, flow cytometric estimation of cellular DNA content, and karyological analysis. Average nuclear volumes for H. labeo, H. longirostris and H. mylodon were 22.5, 21.7 and $26.0\;{\mu}m^3$, respectively. The estimated genome sizes of those three species were not significantly different from one another, being recorded as 2.51, 2.33 and 2.35 pg/cell for H. labeo, H. longirostris and H. mylodon, respectively. Modal chromosome numbers of the three species were the same as 2n = 50. However, their karyotypes and fundamental numbers (FN) were different among species; 16M+16SM+18T/A (FN = 82) for H. labeo, 18M+16SM+16T/A (FN = 84) for H. longirostris and 18M+24SM+8T/A (FN = 92) for H. mylodon.

Karyotypes of Pneumocystis carinii from Korean Rats (한국산 횐쥐 카리니주폐포자충의 핵형)

  • Molecular karyotyping was applied to Pneumocystis carinii (Pc) from two strains of experimental rats, Sprague Dawley(SD) and Fisher(F), in Korea. Field inversion gel electrophoresis and contour clamped homogeneous electric field electrophoresis resolved 15 chromosomal bands from the Pc. The size of the bands was estimated 270kb to 684kb from SD rats, and 273kb to 713 kb from F rats. The bands of 283 kb from SD rats and of 273 kb from F rats stained more brightly suggesting duplicated bands. Total number of chromosomes was at least 16, and total genomic size was estimated 7×106 bp. All of the bands from F rats hybridized to the probe of repeated DNA sequences of Pc and the band of 448 kb size was proved to contain rDNA sequences, but Pc. chromosome bands from SD rats showed no reactions to the probes. The 2 different karyotypes of p. carinii from 2 strains of rats were maintained consistently for 2 years.

  • Constitutional trisomy 8 is a relatively rare aneuploidy; most identified cases are mosaic with a normal cell line. The phenotype is highly variable from apparently normal to severe disability. The proportion of abnormal cells is dramatically different between tissues and the severity of the phenotype is not directly related to the level of mosaicism. Therefore, it is very difficult to provide a definitive prognosis. We report here a case of constitutional trisomy 8 mosaicism with agenesis of the corpus callosum, congenital heart disease and micrognathia. The trisomy 8 cell line was not detected by prenatal cytogenetic study. This is the fourth reported case of constitutional trisomy 8 mosaicism in Korea.

  • Inversion, one of the balanced rearrangements, usually does not lead to phenotypic abnormalities; all genetic information exists in the proper amount, merely in a different order or in an abnormal location. However, offspring of an inversion carrier is at risk of chromosomal imbalance because an inversion loop can be formed during crossing-over of the paternal and the maternal chromosomes in meiosis. We report a 38-year-old woman with inversion and balanced translocation and her fetus with unusual rearrangement causing chromosomal imbalance. We performed conventional cytogenetic analysis, MLPA, and subtelomeric FISH in the cells of the embryo. The results showed that the distal portion of chromosome 13q was added to the terminal portion of chromosome 9p during crossing-over. Therefore, the final karyotype of the fetus was 46,XY,rec(9)t(9;13)(p22;q32)inv(9)(p12q13)mat, confirmed using molecular-cytogenetic analyzing tools.
