Occurrence of Barley Virus Diseases in Southern Part of Korea (국내 남부지역의 맥류 바이러스병 발생 현황)

  • Virus disease surveys on barley cultivation areas in Jeonnam Jeonbuk Gyungnam Gyungbuk-do were conducted during 2014-2015. In this study, we detected Barley yellow mosaic virus (BaYMV), Barley mild mosaic virus (BaMMV) and Soil-borne wheat mosaic virus (SBWMV) by RT-PCR from barley leaves. These viruses are of great economic importance for wheat and barley, causing significant quantitative and qualitative losses in yield. The result of investigation showed that the field incidence of BaYMV in Buan, Gimje was more than 90% in 2014. The infection field rate of barley virus including Boseong, Gangjin, Haenam, Jangheung in Jeonnam was ${\approx}$ 30%. In 2014, double infections by BaYMV and BaMMV was detected in Boseong, Gangjin, Haenam and Jangheung. Only as a low rate BaYMV occurred in various fields of Jeonbuk in 2015. At the same time high infection field ratio of 70% was observed in Gunsan. Also in Yeonggwang was double infection of BaYMV and BaMMV. BaYMV occurred single infection has been confirmed in all of the study area of Gyungbuk and Gyungnam except for Goseong during the investigation period.

Phylogenetic Relationship and DNA Polymorphism of Boleophthalmus pectinirostris and Scartelaos gigas (Teleostei: Gobiidae) of Korea (한국산 짱뚱어(Boleophthalmus pectinirostris)와 남방짱뚱어(Scartelaos gigas) (Gobiidae)의 분자유전학적 계통연관과 DNA 다형화)

  • Phylogenetic relationships and DNA polymorphism among local populations of two Korean gobiidae species: Boleophthalmus pectinirostris and Scartelaos gigas were investigated based on 12S and 16S mitochondrial DNA and mitochondrial cytochrome b DNA sequences. DNA polymorphisms of B. pectinirostris between Suncheon and Gunsan populations were 100% identity from 434 bp segment of 12S rRNA gene and from 444 bp segment of mitochondrial cytochrome b genes, and 99.6% (2 bp different) identity from 484 bp segments of 16S rRNA genes. These results indicated the long period of geographic isolation between two populations of B. pectinirostris in Korea caused such high degrees of DNA polymorphisms. Based on the phylogenetic tree constructed from the two gobiid species in Korea, two genetically distinct groups of B. pectinirostris and S. gigas groups were recognized.

  • The National Center for Intensive Observation of Severe Weathers (NCIO) as a part of METRI's principal project "Korea Enhanced Observing Period; KEOP" was established at Haenam Weather Observatory in order to effectively monitor and observe heavy rainfall in summer, which is essential for the identification of the structure and evolution mechanism of mesoscale severe weather system. The intensive field-based experiments in 2002 within southwestern Korea toward various meteorological phenomena ranging from heavy rainfall to snowfall were conducted in collaboration with KMA(Korea Meteorological Administration) and universities. In this study, preliminary analysis results using intensive observation data obtained from these experiments are presented together with the introduction of NCIO and its operational structure.

Suppressive Effects of Bio-Sulfur on Citrus Scab (바이오 황을 이용한 감귤 더뎅이병 발병 억제 효과)

  • The aim of the present study was to investigate the suppressive effects of the bio-sulfur used by eco-friendly farms on the outbreak of citrus scab. To evaluate the inhibiting effect of bio-sulfur on citrus scab germ tube growth, the citrus scab pathogen Elsinoe fawcettiiwas cultured in PDB and agar media, and germ tube growth was observed after bio-sulfur treatment. At both 40 and 88 h after inoculation, germ tube formation was inhibited by 500-, 1000-, and 2000-fold diluted bio-sulfur, and at dilutions above 4000-fold, germ tube formation was observed, although growth was still inhibited, when compared to untreated cultures. Meanwhile, the occurrence of citrus scab on spring-flush leaves in the field was 40.3% in the untreated control and 5.3, 10.3, 12.3, 15.3, and 24.0% when treated with imibenconazole, 2-4 and 6-6 lime-Bordeaux mixtures, which are also used by eco-friendly farms, 500-fold diluted bio-sulfur, lime sulfur, and 1000-fold diluted bio-sulfur, respectively. The occurrence of citrus scab on citrus fruit was 79.3% in the untreated control and 4.0, 33.8, 42.0, 43.3, 44.8, and 78.0% when treated with imibenconazole, 2-4 lime-Bordeaux mixture, 6-6 lime-Bordeaux mixture, 500-fold diluted bio-sulfur, lime sulfur, and 1000-fold diluted bio-sulfur, respectively. Because citrus scab can infect citrus leaves as early as May, as the spring flush begins, preventative control should be implemented by mid- to late-April, thereby increase disease control and reducing both labor and farming costs.

  • Inhibitors of acetylcholinesterase(AchE), such as organophosphates and carbamates, interfere the action of AchE in nerve and may lead to a severe impairment of nerve functions or even death. Therefore, insect AchE is the biological target of predominant insecticides used in agriculture. Biosensors are sensitive and can be used as dispoisable sensors for environmental control. In recent years, the use of AchEs in biosensor technology has gained enormous attention, in particular with respect to insecticide detection. The principle of biosensors using AchE as a biological recognition element is based on the inhibition the catalytic activity by the agents to be detected. We here present a strip-type biosensor based on AchE inhibition. In this study, acetylcholinesterase and PVA-SbQ(polyvinyl alcohol functionalized with methyl pyridinium methyl sulfate) were co-immobilized on immobilone-P membranes. Immobilization of the enzymes showed a stability in 6 months without activity loss in $4^{\circ}C$ storage. Enzymes immobilized on surfaces of membrane responded to organophosphates and carbamate more sensitivitive than enzyme in solution. Organophosphates and carbamates concentrations could be detected by entrapped and surface immobilized enzymes, in 5 min. For chlorpyrifos, carbofuran, cabaryl, and methidathion, the detection limits of AChE-strip were similar to that of HPLC/GC method.

Distribution of Arbuscular Mycorrhizal Fungi(AMF) at Ginseng Cultivated Fields in Jeonnam Province (전남지역 인삼재배 토양의 Arbuscular 균근균 분포 특성)

  • This study was carried out to investigate the distribution characteristics, colonization patterns of arbuscular mycorrhizal fungi (AMF) at Ginseng- cultivated fields in Jeonnam province including Gokseong, Yeonggwang, Yeongam and Haenam area. Average density of AMF spores in all of analyzed areas was in the range of $3.4{\sim}5.9spores\;g^{-1}$ air-dried soil and spore density in the soil was increased gradually up to Ginseng cultivated fields grown for 3 years. However, AMF spore densities in the marginal field soil were $6.3{\sim}14.7spores\;g^{-1}$ air-dried soil and it was higher than those in Ginseng-cultivated field soils. A $45{\sim}106{\mu}m$ in diameter of AMF spores by size was dominant (74.3%) in Ginseng cultivated fields. AMF colonization association type in Ginseng roots was a Paris-type. Average rates of AMF infection ranged from 40% to 50%, while 64.4% of AMF infection occurred in Yeongam area. External hyphae length (EHL) in the rhizosphere soil of Ginseng cultivated field was in the range of $0.5{\sim}4.4m\;g^{-1}$ air-dried soil and the hyphae quantity was gradually increased proportional to increment of Ginseng cultivation years.

Study of Population Dynamics of Birds Using Unmanned Monitoring System in Rice Paddy (무인모니터링 시스템을 활용한 논습지에 도래하는 조류 개체군 동태 연구)

  • This study was conducted to identify the characteristics of bird population dynamics using unmanned monitoring system in rice paddy. We compared the similarity of population dynamics of birds between unmanned monitoring system and field survey, and evaluated the spatial and temporal patterns of population dynamics of birds using the unmanned monitoring system. The monitoring using the system was conducted from January 1, 2014 to December 31, 2016 in Cheolwon, Dangjin, Buan, and Heanam. The images from the system were obtained at 10-min intervals from 6:00 to 20:00. The field survey was conducted once a month in Dangjin from January to December 2016. Total 91,980 images were obtained from the unmanned monitoring system. We extracted the number of individuals for herons, shorebirds, and waterfowl from the images. The population dynamics of waterbirds using the unmanned monitoring system were similar to that in field survey. Especially, population dynamics of herons was more similar than other waterbirds. It was identified that the population dynamics of herons using the unmanned monitoring system was different among the Cheolwon, Dangjin, Buan, and Heanam. Furthermore, the unmanned monitoring system was available on various time scale such as month, day, and minute. It is expected that long-term data storage using the unmanned monitoring system can be used to identify in detail and forecast the population dynamics of birds in rice paddy.

Differences in swine gut microbiota in southern region of Republic of Korea (한국 남부 지역별 돼지 장내 미생물생태 비교분석)

  • Since the banning of antibiotic growth promoters (AGPs), the death of livestock has been increased, thus there is a strong demand for AGP-alternatives. Modulation of gut microbiota has been reported to affect host physiological functions and suggested to be a novel approach for developing AGP-alternatives. However, little has been understood about livestock gut microbiota compared to that of humans. We conducted preliminary study provide fundamental information regarding to regional differences in swine gut microbiota. Swine fecal samples were obtained from farms in Jeju (n=40), Gwangju (n=28), and Haenam (n=30). MiSeq was used to sequence 16S rRNA V4 region, and Mothur pipeline (Schloss et al., 2009) was used for data processing. A total of 5,642,125 reads were obtained and 3,868,143 reads were remained after removing erroneous reads. Analysis of taxonomic composition at the phylum level indicated greater abundance of Firmicutes among Jeju swine, and cluster analysis of distribution of operational taxonomic units also showed regional differences among swine gut microbiota. In addition, correlation analysis between non-metric multidimensional scaling and abundance of phyla suggested that the phyla Actinobacter, Verrucomicrobia, Firmicutes, and Fibrobacteres were driving factors for the regional differences. Livestock gut microbiota may be affected by diet and practices in farms. Our results indicated significant regional differences in swine gut microbiota, suggesting that future livestock gut microbiota studies should be designed with the regional differences in mind.

Chemical Characteristics and Water Dispersible Colloid Content of Jeju Citrus Orchard Soils (제주도 감귤원 토양의 화학적 특성과 물 분산성 콜로이드 함량)

  • Water-dispersible colloids are suspected to facilitate transport of contaminants to groundwater. This study evaluated some soil chemical properties in relation to the stability of colloids in soils of Jeju citrus orchards. Thirty surface soil samples were collected, and pH, organic matter content oxalate-extractable Al and Fe contents, and water-dispersible colloid content were measured. In soils of higher pH, water-dispersible colloid contents were higher. The stability of colloids was found to be significantly promoted at pH above 5$\sim$6. Since organic matter can act as a flocculant organic matter content significantly enhanced the colloid stability. In soils of less than 5% organic C, water-dispersible colloid content was expected to be significantly higher. In soils of higher oxalate-extractable Al and Fe contents, colloids remaining in suspension were lower. This indicated that amorphous oxides and hydroxides play important stabilizing roles in soil structure and can stabilize soil clay against dispersion. Therefore in soils of higher pH, lower organic matter, and lower amorphous clay minerals, the stability of water-dispersible colloids and the potential of colloid-mediated transport of organic chemicals to groundwater could be higher.

Inferring Regional Scale Surface Heat Flux around FK KoFlux Site: From One Point Tower Measurement to MM5 Mesoscale Model (FK KoFlux 관측지에서의 지역 규모 열 플럭스의 추정 : 타워 관측에서 MM5 중규모 모형까지)

  • Korean regional network of tower flux sites, KoFlux, has been initiated to better understand $CO_2$, water and energy exchange between ecosystems and the atmosphere, and to contribute to regional, continental, and global observation networks such as FLUXNET and CEOP. Due to heterogeneous surface characteristics, most of KoFlux towers are located in non-ideal sites. In order to quantify carbon and energy exchange and to scale them up from plot scales to a region scale, applications of various methods combining measurement and modeling are needed. In an attempt to infer regional-scale flux, four methods (i.e., tower flux, convective boundary layer (CBL) budget method, MM5 mesoscale model, and NCAR/NCEP reanalysis data) were employed to estimate sensible heat flux representing different surface areas. Our preliminary results showed that (1) sensible heat flux from the tower in Haenam farmland revealed heterogeneous surface characteristics of the site; (2) sensible heat flux from CBL method was sensitive to the estimation of advection; and (3) MM5 mesoscale model produced regional fluxes that were comparable to tower fluxes. In view of the spatial heterogeneity of the site and inherent differences in spatial scale between the methods, however, the spatial representativeness of tower flux need to be quantified based on footprint climatology, geographic information system, and the patch scale analysis of satellite images of the study site.
