Studies on the Biological Activity of Pleurotus ferulea (아위버섯(Pleurotus ferules) 추출물의 생리활성 탐색)

  • This study was peformed to screen the biological activities of Pleurotus ferulea (K5 and K8 strains). The cap of K5 strain is well developed than stalk, and vice versa in K8. The ethanol extract of Pleurotus ferulea exhibited significant free radical scavenging activity (35∼36%), suggesting possible effect on many degenerative diseases originated from the reaction of oxygen species. Acetylcholinesterase inhibitor is proven to be the most effective factor for Alzheimer disease induction, and ethanol extract of Pleurotus ferulea significantly inhibited acetylcholinesterase activity (25∼35%) in uitro. Moreover, ethanol extract of Pleurotus ferulea suppressed liver fibrosis by 3∼12% in uitro. However, Pleurotus ferulea feruled to inhibit glucose uptake in human intestinal cell line. Viability of gastric and colon cancer cells was also not affected by Pleurotus ferulea extract. In conclusion Pleurotus ferulea exhibited significant effect on free radical scavenging, acetylcholinesterase inhibiton and brain cell protection. However, Pleurotus ferulea failed to affect glucose uptake, and cytotoxicity of gastric and cancer cells. In general, K8 revealed more significant effects than K5.

The Effects of Antimicrobial, Antioxidant, and Anticancer Properties of Opuntia humifusa Stems (천년초 줄기의 항균, 항산화 및 항암효과에 관한 연구)

  • This study was performed to investigate antimicrobial, antioxidant, and anticancer properties of Opuntia humifusa (OH) stems. OH stems were extracted with hexane, methanol, butanol and water. The methanol and hexane fraction exhibited strong antimicrobial activities on three strains of microbes, Rhodococcus equi, Staphylococcus aureus, and Clostridium perfringens. In the peroxynitrite scavenging effect ($ONOO^-$) of OH stems, the antioxidative activity of methanol, butanol and water fraction but not hexane fraction showed significant increases in a concentration-dependent manner. The DPPH radical scavenging activities of OH stems were high in the butanol fraction compared with other fractions. Anti-proliferation effects on the B16-F10, HepG2, HT29, and MCF-7 cell lines were significantly higher in the methanol and hexane fractions than in the water and butanol fractions at $100{\mu}g/mL$ concentration of extracts. These results suggest that OH stems can be used for the development of functional foods with biological activity.

Anticancer Effects of Cultivated Orostachys japonicus on Human Prostate Cancer Cells (인체 전립선 암세포에서 재배 와송의 항암효과)

  • This study was performed to determine the anticancer effects of cultivated Orostachys japonicus (COJ) and wild Orostachys japonicus (WOJ) on primary human prostate cancer cells (RC-58T/h/SA#4 cells). The morphology of cells treated with COJ and WOJ was distorted to shrunken cell masses. In addition, cell death induced by COJ and WOJ was associated with increased population of cells in sub-G1 phase as well as the formation of apoptotic bodies and nuclear condensation. COD and WOJ markedly reduced the number of viable prostate cancer cells in a dose-dependent manner, and cell numbers were lower than control cells. COJ and WOJ also inhibited increases in cell proliferation induced by environmental hormones such as dioxin and bisphenol A in charcoal-treated FBS (cFBS) medium. COJ and WOJ methanol extracts at the tested concentrations (150, 300, and 600 ${\mu}g/mL$) also dose-dependently inhibited cell proliferation induced by environmental hormones. These results indicate that COJ and WOJ exert anticancer effects on primary human prostate cancer cells.

  • 중국(中國) 장홍화(藏紅花)는 해발 1500m~3000m에 자생하는 고원식물이다. 장홍화는 여성질환예방과 두뇌의 신경안정, 호르몬분비 촉진, 학습증진과 기억력향상에 의한 알츠하이머 치료의 효과가 있는 것으로 알려져 있으며, 함유되어 있는 크로신은 암세포를 파괴시키어 종양세포를 줄이는 작용을 하며, 카로티노이드 성분이 항산화 활성과 면역을 증진시키어 항암효과를 가져다 준다. 또한 다량의 칼륨을 함유하고 있어서 나트륨배출에 효과적이어서 혈압을 떨어뜨리는 작용을 한다. 또한 신경전달물질의 활동을 증진시키어서 스트레스 해소와 치매 및 인지력을 높여준다고도 알려져 있다. 본 실험에서는 장홍화(藏紅花)추출액의 총 폴리페놀 함량과 항산화 물질 활성을 조사했던 바, 장홍화(藏紅花) 메탄올과 에탄올추출액에서 항산화 반응이 매우 양호하였다. 또한 총 폴리페놀 함량도 에탄올 추출물과 메탄올 추출물 처리구에서도 비슷한 결과를 나타내었다. 이러한 결과들로 미루어 볼 때 장홍화(藏紅花)의 추출액은 건강음료와 화장품의 기능성원료로도 이용이 가능할 것으로 추측되었다.

  • 느타리버섯은 맛, 향기 및 영양가가 일반 채소류나 일부 버섯류에 비하여 높고, 고혈압, 당뇨병에도 효과가 있으며, 특히 항암효과 등의 약리활성이 있어서 오래 전부터 한국과 일본에서 부식으로 이용되었다. 느타리버섯의 국내 재배면적 및 생산량은 1988년 832,346평에 29,386톤에서 1997년 에는 2,100,890 평에 83,606톤으로 증가되었다. 그러나 생산량의 증가추세에 부합하는 재배기술의 축적, 적정 재배환경 관리의 모델화가 미흡하기 때문에 그 생산량은 국민적인 소비욕구를 충족시키지 못하고 있다. (중략)

Effects of Polyacetylenes in Ginseng on Activity of Enzymes Related to Post-translational Modification of Ras Protein and Effects of Petroleum Ether Extract of Ginseng on Progression of Cell Cycle (인삼의 polyacetylene 계 성분이 Ras 단백질 변형 효소의 활성에 미치는 영향과 인삼의 석유에텔 추출물이 세포주기에 미치는 영향)

  • We investigated the effects of polyacetylenes of ginseng on farnesyl protein transferase (FPTase) and carboxyl methyl transferase (CMTase) activities related to post-translational modification of Ras protein. We also investigated the effect of petroleum ether extract (PEE) of ginseng on progression of cell cycle. FPTase activity was respectively inhibited 16.2% by 10mM panaxynol and 21.3% by 10mM panaxydol, whereas CMTase activity was not inhibited by panaxynol or panaxydol. Treatment of PEE significantly reduced the numbers and size of human colon cancer cell (HT-29) and human liver cancer cell(HepG2) cultured, respectively. To investigate the mechanism of growth inhibition by PEE of ginseng, we analyzed the cell cycle progressions of PT-29 and HepG2 cells, respectively. We found that PEE significantly inhibited progression of cell cycle from G1 to S phase. These results suggest that anticancer effects of PEE were derived from the arrest of G1 phase in cell cycle progression.

  • Mountain ginseng is a perennial crop rarely found in the deep mountains of Korea. The medicinal effect of the mountain ginseng is well known as a panacea in traditional Chinese medicine for a long time. But scientific studies to elucidate the medicinal effect of the mountain ginseng have never been made on account of lack of sample. Recently an improved method of adventitious root culture system through the use of bioreactor has been developed in Panax ginseng that seems to be a reliable way of commercialization of root derived secondary metabolites. This experiment was conducted to evaluated chemotherapeutic effect against human cervical cancer cells by cisplatin (CDDP) and extract of tissue cultured mountain ginseng (ETCMG). CDDP and ETCMG-induced apoptotic cell death in human cervical cancer cell line, HeLa was confirmed by the analysis of cell growth, morphological changes, DNA fragmentation, flow cytometry showed that ETCMG is an inducer of apoptosis and synergizes with CDDP. These results suggest that ETCMG present evidence of anticancer effect and could have a possibly natural therapeutic potential in cervical cancer patients.

Anti-tumor Activity of Protein-bound Polysaccharides Extracted from Mycelia of Lentinus edodes (표고버섯 균사체로부터 추출한 단백다당체의 항암효과)

  • Protein-bound polysaccharides (PBP) were extracted from the mycellia of Lentinus edodes SR-1, and their anti-tumor activities and immunopotentiating properties were observed. The amounts of PBP needed to extend the doubling time twofold (1 unit) were found to be 1 mg for mouse leukemic cells $P_{388}\;and\;L_{1210}$; 4.4, 3.6 and 6.6 for bowel cancer cells, HCT-48, HRT-18, HT-29 respectively; and 2.6 mg for liver cancer cell, Hep G2. When $P_{388}\;and\;L_{1210}$ were treated with 4 mg of PBP, more than 90% of the cell number were reduced in 48 hours. However, 9 mg of PBP and 72 hrs of incubation time were needed to obtain the same effect for HRT-18, HT-29, and Hep G2. The significant reduction of cell size was observed as the amount of PBP and the incubation time increased. Mice spleen weight and plaque forming cell number increased when the cancer cells were treated with PBP.

  • Surgery is the only curative modality for the treatment of gastric cancer. There has been no drastic improvement in the treatment of gastric cancer with chemotherapy. Clinical trials have attempted to demonstrate the benefit of the preoperative chemotherapy for gastric cancer. The benefit of the use of preoperative chemotherapy or chemoradiotherapy has been demonstrated for other solid cancers such as breast cancer, esophageal cancer and rectal cancer. Despite the rationale of the use of preoperative chemotherapy for patients with gastric cancer, the evidence of positive results with the use of preoperative chemotherapy has not been clear. Recently the British Medical Research Council Adjuvant Gastric Cancer Infusional Chemotherapy (MAGIC) study demonstrated the survival benefit of preoperative and postoperative chemotherapy. However, this study had several problems with the use of a heterogeneous population of patients, the method of surgery and the use of perioperative chemotherapy. Further studies with new drugs are warranted to determine the role of pre-operative chemotherapy for patients with gastric cancer.

Anticancer Effects of the Extracts of Oriental Melon (Cucumis melo L. var makuwa Makino) Seeds (참외(Cucumis melo L. var makuwa Makino) 종자 추출물의 항암 활성)

  • The objective of this study was to investigate the anticancer effects of the extracts of oriental melon seeds. Various solvent extracts of oriental melon seeds were prepared and their anticancer effects were examined using in vitro MTT and CV assays. The anticancer effects of various extracts of oriental melon seeds were also examined in five human cancer cell lines including A549, AGS, HT-29, MCF-7 and HepG2. The ethanol extract of heated oriental melon seeds showed the potent cytotoxic effects especially against mouse hepatoma cell line(Hepa1c1c7), human hepatoma cell line(HepG2) and human breast cancer cell line(MCF-7). These data suggest that oriental melon seeds can be a promising anticancer agent against human liver and breast cancer.
