Physical Characteristic and In vitro Transdermal Delivery of PCL-b-PEG Micelles Containing Quercetin and Rutin (Quercetin과 Rutin을 함유하는 PCL-b-PEG 고분자 미셀의 특성 및 피부 흡수에 관한 In vitro 연구)

  • In this study, we prepared polymer micelles containing quercetin and rutin, known as antioxidants, using poly(${\varepsilon}$-caprolactone)-b-poly(ethylene glycol), and evaluated in vitro skin permeation of the active materials. Quercetin and rutin loaded micelles were characterized by DSC (differential scanning calorimetry), HPLC (high performance liquid chromatography) and DLS (dynamic light scattering) measurements. The particle size of the polymer micelles increased in a concentration dependent manner (0.5~2.0% PCL-b-PEG). The Zeta potential of quercetin and rutin loaded micelles remained constant. To evaluate the skin penetration of PCL-b-PEG micelles, Franz diffusion cell experiment was performed. The aqueous solutions of quercetin and rutin were used as the control groups. Quercetin and rutin loaded PCL-b-PEG micelles showed more efficient skin permeation than the control groups. Safety assessment (patch test) of quercetin and rutin loaded PCL-b-PEG micelles on skin was performed to test application possibility of the polymer micelles to cosmetics. Any adverse symptoms were not observed.

Biological Potential of Enzymatic and Polyphenol Extracts from Ecklonia cava (감태 효소 추출물 및 폴리페놀 추출물의 생리활성에 관한 연구)

  • To investigate the efficacy of enzymatic extract of Ecklonia cava and its polyphenol extract (AG-DK) as cosmetic ingredients, the anti-oxidative effect, anti-glycation effect, anti-melanogenic effect, and anti-inflammatory effect of the extracts were evaluated in vitro. The enzymatic extract of E. cava ($SC_{50}$ 42.9 ppm) and AG-DK ($SC_{50}$ 6.4 ppm) showed a strong DPPH free radical scavenging activity. The anti-glycation ability of the enzymatic extract of E. cava and AG-DK was tested using bovine serum albumin (BSA), which inhibited the formation of advanced glycation end-products (AGEs) in the BSA/glucose system. The enzymatic extract of E. cava ($IC_{50}$ 97.2 ppm) and AG-DK ($IC_{50}$ 7 ppm) had inhibitory effects on tyrosinase activity. Moreover, the enzymatic extract of E. cava and AG-DK had an anti-inflammatory effect through the inhibition of nitricoxide (NO) and prostaglandin E2 ($PGE_2$). These findings suggest that the enzymatic extract of E. cava and AG-DK can be applied to skin-care products as cosmetic ingredients.

Quality Characteristics of Kimchi Fermented in Permeability-Controlled Polyethylene Containers (투과도 조절 플라스틱 용기에서 발효된 김치의 품질 특성)

  • Kimchi was fermented in permeability-controlled polyethylene containers, in glazed onggi (Korean ethnic earthenware) or glass bottles at $5^{\circ}C$ for 8 weeks. During 4 weeks of storage, kimchi fermented in the permeability-controlled container showed a stable fermentation pattern, in terms of changes in pH and acidity, compared with kimchi fermented in the other containers. With respect to changes in bacterial counts, kimchi fermented in polyethylene containers showed vigorous multiplication of lactic acid bacteria, especially Lactobacillus sp., but slow growth of total aerobic bacteria. The springiness of kimchi fermented in the polyethylene containers was optimal (about 10% more than that of glass bottle-fermented kimchi), and the overall acceptability and hardness of container-fermented kimchi were excellent upon sensory evaluation. The DPPH radical-scavenging activity of kimchi fermented in polyethylene containers was also greater (91%) than that of kimchi fermented in glazed onggi (73%) or glass bottles (63%). The $O_2$ and $CO_2$ permeabilities of the polyethylene containers were higher (458 and $357\;mmol\;h^{-1}\;m^{-2}\;atm^{-1}$, respectively) than were those of the other containers; the permeability ratio was 0.8. Glass bottles showed no permeance. The results indicate that permeability-controlled polyethylene containers may be used for kimchi fermentation.

Quality Characteristics of Yanggaeng Added with Blueberry Powder (블루베리 분말을 첨가한 양갱의 품질 특성)

  • The objective of this study was to assess the quality characteristics of yanggaeng prepared with different ratios of blueberry powder: 0%, 3%, 6%, and 9%. The moisture content of the control yanggaeng was lower than that of any other blueberry added groups. The pH decreased while the titratable acidity and the Brix increased as the amount of blueberry powder increased. The lightness (L) and yellowness (b) were the highest while the redness (a) was the lowest in the controls. Texture profile analysis showed that the hardness of the added groups were higher than that of the control which were the lowest. The total polyphenol content and DPPH (2,2-diphenyl-1-picrylhydrazyl) radical scavenging activity increased as the amount of blueberry powder increased. The results of consumer acceptance test revealed that the color preference was the highest in the 9% added group while the smell, taste, texture and the overall acceptability showed no significant differences groups. Based on these results, it is suggested that yanggaeng with up to 9% added blueberry powder can be developed as products without adverse effects on sensory characteristics.

Physicochemical Characteristics and Biological Activity of Irradiated Pectin Solution (감마선 조사 펙틴 용액의 이화학적 특성 및 생리활성 변화)

  • Pectin was dissolved in HCl, citric acid, and deionized distilled water (DW, 2%, v/v) and irradiated at different irradiation doses (2.5-50 kGy) by gamma ray to investigate its physicochemical characteristics and biological activity. Viscosity of pectin solution was significantly decreased by irradiation up to 10 kGy, then remained constant thereafter. Gamma-irradiation increased monosaccharide and polysaccharide levels up to 30-40 kDa. Electron donating ability of pectin solution was highest when DW was added and was increased by increasing irradiation dose (p<0.05). ${\beta}-Carotene$ bleaching assay revealed irradiation resulted in development of antioxidantive activity in pectin solution. Growth inhibition of cancer cell lines was observed in irradiated pectin solution in dose-dependent manner, with G36l showing the highest. Results suggested irradiation of pectin solution could be effective for preparation of functional pectin oligomer.

Antioxidants Activity of Aged Red Garlic (숙성 홍마늘의 생리활성)

  • The antioxidant activities of hot water extracts from fresh, red and black garlic processed in low temperatures were compared. The chromaticity value of browning garlic was between that of fresh and black garlic. Red garlic was similar in browning intensity to fresh garlic. Also, total phenol, flavonoids, total pyruvate and thiosulfate contents were similar between fresh and black garlic. DPPH, ABTs, NO radical scavenging activity and reducing power of red garlic were significantly higher than fresh garlic, but lower than those of black garlic. $\alpha$-Glucosidase inhibitory activity in red garlic was similar to that in black garlic. Antioxidant activities of red garlic were higher than fresh garlic but lower than black garlic, and it was confirmed that antioxidant activity by production of browning material through the thermal process was the main parameter of the biological activity in the aged red garlic.

  • Corn stover is known as a good candidate for a functional food ingredient when the main lignocellulosic material, lignin, is used as bioactive materials as form of polyphenolic compounds. The purpose of this study was to determine the microwave extraction conditions under which total phenolic compounds (TPC) and flavonoid contents of corn stover were maximized. Microwave-assisted extracts using sulfuric acid ranging from 0 to 1.0 mol with extraction time between 40 and 240 sec were conducted by using response surface methodology (RSM). Microwave power showed significant effects (p<0.05) and the concentrations of TPC and flavonoids increased with increased level of microwave power and extraction time. The optimum conditions for corn stover extraction were determined as 698.6 W, 240 sec, and 0 mol sulfuric acid, and the predicted value of TPC and flavonoid is 82.4 mg GAE/g DM and 18.1 mg/g DM, respectively. Microwave extraction was evaluated as an economic process with low energy consumption, short extraction and high extraction yield of bioactive including TPC and flavonoids compared to conventional extractions.

Effect of Removed Peel from Sweet Persimmon on Nutritional Ingredients and Antioxidant Activities (단감의 박피가 영양성분 및 항산화 활성에 미치는 영향)

  • This study was for the industrial application of functional food ingredients from whole fruits of sweet persimmon. Whole fruit and pulp of sweet persimmons were divided, and then lyophilized and powdered. Contents of crude fiber, vitamin C, and mineral were significantly higher in whole fruit than pulp of sweet persimmon. The amino acid content of whole fruit was 1.4 times higher than those of sweet persimmon pulp. In the biological activities of water and ethanol extracts from whole fruit and pulp of sweet persimmon, ethanol extract was higher than water extract, and whole fruit was higher than its pulp. The result which compared the biological activities of the water and ethanol extract from lyophilized sweet persimmon showed that total phenolic content was significantly higher in whole fruit of sweet persimmon, but flavonoid contents were not significantly different. Especially ABTS, NO radical scavenging activity, reducing power and tyrosinase inhibition activity were significantly higher in whole fruit extract than pulp extract of sweet persimmon. The relatively high content of fiber and vitamin C, and biological activity of whole fruit than pulp of sweet persimmon may be make it preferable as functional food materials for secondary processed goods.

Preparation of Nano-emulsion Containing Polygonum aviculare Extract for Enhanced Transdermal Delivery (피부 흡수 증진을 위한 마디풀 추출물 함유 나노에멀젼 제조에 관한 연구)

  • In this study, nano-emulsion containing ethyl acetate fraction of Polygonum aviculare (P. aviculare) extract was prepared and skin permeability of the nano-emulsion was evaluated. Nano-emulsion was prepared using homogenizer and microfluidizer by the high-energy method. The droplet size and loading efficiency of nano-emulsion containing ethyl acetate fraction of P. aviculare extract were determined. The mean droplet size was 238 nm and the loading efficiency was more than 98%. The size distribution of nano-emulsion was a monodispersed form and nano-emulsion was more stable than that of using typical emulsion without using the microfluidizer. The in vitro skin permeation study of nano-emulsion containing ethyl acetate fraction of P. aviculare extract was carried out using Franz diffusion cells. Compared to 1,3-butylene glycol, nano-emulsion showed greater values of cumulative permeation of ethyl acetate fraction from P. aviculare extract. These results indicate that the stability and skin permeability of nano-emulsion containing ethyl acetate fraction of P. aviculare exerting anti-oxidative and anti-aging activities were enhanced.

The Study of Physiological and Antimicrobial Activities on the Citrus Junos Extracts with its Textures and Skin (유자과즙 추출물의 생리활성 및 항균활성에 관한 연구)

  • This study analyzed the antioxidant activities and antibacterial effects of extracts from the fruit of Citrus junos. Theextracts were obtained in both 70% ethanol solution and distilled water at a distillation temperature of $50^{\circ}C$.Three experiments were carried out between November 2016 and March 2017. The averages and the standard deviations of the results were measured. The total polyphenol and tannin contents of the ethanol extracts were $7.8{\pm}0.02mgGAE/g$ and $9.9{\pm}0.01mgTAE/g$, respectively, which were higher than the concentrations in the water extracts. Furthermore, the ethanol extracts scavenged $46.1{\pm}4.76%$ of DPPH radicals and $37.1{\pm}1.23%$ of ABTS radicals. The scavenging capability of the ethanol extracts was also higher than that of the water extracts. However, the scavenging capability of both types of extracts depended on their concentrations.All the extracts showed active antibiosis effects against every bacteria tested except for C. albicans. E. coli at 25 mg/disc showed antibiosis with $18{\pm}1.73mm$ for the water extracts, while S. epidermidis and S. aureus showed antibiosis with $17{\pm}4.36mm$ and $19{\pm}2.86mm$ for the ethanol extracts, respectively. This antibiosis rate is considerably higher. The results suggest that fruit extract from Citrus junos could be useful as a primary material for beauty or skin-related products such as soaps, shampoos, and scalp enhancers.
