In order to discover physiologically active substance, we investigated a powder obtained by processing of Opuntia ficus-indica var. saboten Makino trunk. The powder was treated by sulfuric acid and then extracted by several solvents such as methanol, methylene chloride, ethanol etc. Among them, the best antimicrobial activity was showed by methylene chloride extract. To identify materials exhibiting physiological activation, the acid hydrolyzed extract was separated by 7 fractions through preparative silica gel TLC. The effective fraction exhibiting the best broad antimicrobial activity was identified, named as MBT-01108. From structural analysis of the products extracted to acid hydrolysis, a compound exhibiting the antimicrobial activities is identified to levulinic acid. Levulinic acid isolated from Opuntia ficus-indica var. saboten Makino may be applicable as a natural preservative of food or cosmetic and for prevention of bacterial diseases, an ingredient of acne, ageing and whitening cosmetics and an antimicrobial agent.
In this study, we investigated antimicrobial and cytotoxicity effects to each fraction extracted from Solanum lyratum (SL), which were extracted methanol (SLM) and then the extract was fractionated into five different types : hexane (SLMH), ethyl ether (SLMEE), ethylacetate (SLMEA), butanol (SLMB) and aqueous (SLMA). The antimicrobial activity was analyzed by the paper disc method. Among the various solvent fractions, SLMEA showed the strongest antimicrobial activies. The cytotoxicity of SL fractions on HeLa, MCF-7, HT-29 and HepG2 cells was evaluated by MTT assay. Among various partition layers, SLMEE showed the strongest cytotoxic effects to all cancer cell lines. We also observed that quinone reductase (QR) was induced by all fraction layers of SL to HepG2 cells. Since the QR-induced effects of SLMEE on HepG2 cells at $160{\mu}g/ml$ concentration showed 2.1 when compared with a control value of 1.0, inducer of QR for cancer protection may be contained in this fraction.

Purpose: The purpose of this study was to examine effects of culture conditions on the growth and antibacterial activity of Streptococcus salivarius K12. Materials and Methods: S. salivarius K12 was cultivated in medium containing animal and plant protein or in medium of neutral and acidic conditions. The growth of S. salivarius K12 was measured every 2 hours by a spectrophotometer. The antimicrobial activity of S. salivarius K12 against Streptococcus mutans and Porphyromonas gingivalis was investigated by the susceptibility assay using the spent culture medium. Results: the growth of S. salivarius K12 showed faster in medium containing plant protein and neutral pH condition. The antimicrobial and antifungal activity of S. salivarius K12 appeared stronger in medium containing plant protein than animal proteins. Conclusion: For application of S. salivarius K12 to bacterial oral disease, co-substances may be needed for S. salivarius K12 to colonize in the oral cavity and enhance the antimicrobial activity.
Broad range of fatty acids were reported to show antimicrobial activities against broad range of microorganisms. However, possible changes of the antibacterial activity of a fatty acid based on structural variations are largely unknown. This study was focused on determination and comparison of the antimicrobial activities of the medium chain fatty acids, based on the position of carboxyl groups on either terminal end, against the representative food-pathogenic bacteria. Over all, mono-carboxyl medium chain fatty acids (MC-MCFA) presented stronger antimicrobial activities against the food-pathogenic bacteria tested including methicillin-resistant Staphylococcus aureus (MRSA) than di-carboxyl medium chain fatty acid (DC-MCFA). In addition, some of MC-MCFA and DC-MCFA showed high possibility to be used as a synergistic adjuvant for both the commercial β-lactam family antibiotics and aminoglycoside family antibiotics against MRSA.
The antibacterial activities of methanol extract and systematic solvent fractions( -hexane, chloroform, ethyl acetate, and butanol) from Korean common type figs at different ripening stages were tested by the broth dilution method against 8 representative food-poisoning bacteria- : L. monocytogenes, S. aureus, S. enteritidis, E. coli O157:H7, E. coli, Y. enterocolitica, V. parahaemolyticus, and S. typhimurium. The methanol extracts of unripened I and II showed stronger activity than that of the ripened figs especially against L. monocytogenes, S. enteritidis, E. coli O157:H7, Y. parahaemolyticus and S. typhimurium in 10 mg/mL. The systematic solvent fractions showed stronger antibacterial activities than the methanol extract, even al the lower concentrations. The hexane fraction of ripened figs showed higher growth inhibition than those of unripened I and II against L. monocytogenes, E. coli O157:H7, Y. enterocolitica and V. parahaemolyticus. The chloroform fraction showed strong antibacterial activity in all ripening stages against E. coli O157:H7 and V. parahaemolyticus. The butanol fraction showed better inhibition activity in unripened I and II than in the ripened figs. The hexane and chloroform fractions showed inhibition activity of more than $75\%$ against E. coli O157:H7, V. parahaemolyticus in 0.5 mg/mL. Each fraction showed a little different antibacterial activity according to the ripening stages of the fruits and the tested strains. Especially, figs in the unripened II stage revealed superior activity relatively and the hexane and chloroform fractions revealed the strongest activity, followed by the butanol fraction, while the ethylacetate and water fractions hardly showed any activity.

This study was undertaken to determine inhibitory compounds from extracts of the softwood (larix leptolepis, Pseudotsuga menziesii) sawdust against Trichoderma spp. The sawdust of L. leptolepis and P. menziesii were hot water extracted, which were with fraction extracted organic solvents. The organic solvent extractions were carried out by n-hexane, methylene chloride, ethyl acetate. The antifungal activity of hot water extracts of L. leptolepis sawdust was determined to be 20.6% inhibition at a concentration of 1,000 ppm against Trichoderma spp. The antifungal activity of P. menziesii sawdust was outstanding about 60.3% against Trichoderma spp. The yields of the fractions of n-hexane soluble, methylene chloride soluble and ethyl acetate soluble from the hot water extract of L. leptolepis sawdust were 4.0%, 6.0% and 8.0%, repectively. However, the yields of the fractions of three solvents of P. menziesii sawdust were 8.0%, 13.0 and 14.0% correspondingly. The antifungal activity of n-hexane soluble fraction from hot water extracts of L. leptolepis sawdust was highest to about 68.5% to 79.9% against Trichoderma spp. compared to others. The antifungal activities of n-hexane soluble fraction from hot water extracts of P. menziesii sawdust showed 68.5%, 71.4%. 71.9%, 75.7% and 82.3% against T. aggressivum, T. atroviride, T. harzianum, T. koningii and T.viride, respectively. The n-hexane soluble fraction revealed much higher antifungal activity than the other fractions did. This study demonstrated that the n-hexane fraction of the hot water extracts of L. leptolepis and P. menziesii exhibited the greatest antifungal activity against Trichoderma spp.

The aim of this study was to investigate the tyrosinase inhibitory activity and antimicrobial activity by mixtures of ultrasonicated chitosan and Maillard reaction products. Analysis of tyrosinase was purified from potato and confirmed by active staining after SDS-PAGE. Maillard reaction products (MRPs) were formed from various sugars (glucose, fructose, galactose, xylose, arabinose or ribose) and cystein. MRPs inhibited the tyrosinase purified from potato. The highest tyrosinase inhibitory activity was shown by MRP from glucose and cystein. Ultrasonicated chitosan (over 1 hr) showed antimicrobial activity at the concentration of 1% against E. coli and S. aureus. For the development of antibrowning agent with antimicrobial activity, tyrosinase inhibitory and antimicrobial activity by the mixtures of ultrasonicated chitosan and MRP were tested. 1:1 mixture of ultrasonicated chitosan and MRP from glucose and cystein was the best antibrowning agent having antimicrobial activity.
Antimicrobial activities of plant extractives were investigated to develop a natural fungicide. Two stilbenoids and five flavonoids were isolated from wood extractives of Hovenia dulcis (Rhamnaceae) which had been selected due to its high antifungal activity among the tested species. The chemical structures of isolated compounds were determinded as : 5-hydroxy-7-methoxyflavone, 5,7-dihydroxyflavone (chrysin), 5,7-dihydroxyflavanone (pinocembrin), 3,5,7-trihydroxyflavanone (pinobanksin), 3,4',5,7-tetrahydroxyflavanone (aromadendrin), 3-hydroxy-5-methoxystilbene and 3,5-dihydroxystilbene (pinosylvin) on the basis of Mass and NMR spectroscopic data. According to the results of antifungal test, 3-hydroxy-5-methoxystilbene was evaluated as the strongest antifungal compound among the tested compounds and next were pinocembrin and pinosylvin, but those also had high hyphal growth inhibition activities against C. parasitica, T. versicolor, T. palustris and T. viride. However, pinobanksin, 5-hydroxy-7-methoxyflavone, chrysin and aromadendrin showed very low antifungal activity. In this regard, it could inferred that high antifungal activity of wood extractives of H. dulcis were derived from 3-hydroxy-5-methoxystilbene, pinocembrin and pinosylvin, respectively.

Antimicrobial effect of chitosan and its oligosaccharides was examined on Vibrios sp., Edwardsiella tarda and Streptococcus sp., which are major pathogenic bacteria inducing bacterial diseases of acquacultured flounder. Chitosan oligosaccharides (COS ) were produced by enzymatic hydrolysis of chitosan in an ultrafiltration mombrane bioreactor system which was established with three membranes with different molecular weight cut-off (MWCO) 1,000, 5,000 and 10,000, and fractionated into three kinds of COS, based on their molecular weight sizes. The three kinds of COS were as follows : relatively high molecular weight COS [HMW-COS, molecular weight distribution of 7,000 to 24,000 Da〕, medium molecular weight COS 〔MMW-COS, 1,500 to 6,000 Da〕, and low molecular weight COS 〔LMW-COS, 1,000 to 1,500 Da). Chitosan and HMW-COS effectively inhibited the growths of Vibrio sp. and Streptococcus sp. and their antimicrobial activities were superior to the others with smaller molecular weights. This result suggested that antimicrobial effect of chitosan preparations extremely depend on their molecular weight sizes. Antimicrobial effect of chitosan and HMW-COS on E. tarda was improved by longer inoculation times. Scanning electron microscopy in morphological change of E. tarda treated with chitosan preparations showed that chitosan and HMW-COS bound to the cells and suppressed the growth of the cells. This observation appears to prove the fact that positive charged amines of chitosan electrostatically bind to negative charged compounds of cell walls.

A study was carried out to produce antimicrobial peptides from zein treated with pretenses of six kinds. Among the pretenses of six kinds, zein hydrolysate treated with pepsin showed the highest antimicrobial activity. The zein hydrolysate with pepsin was fractionated with membrane filter (30,000 10,000 and 3,000 molecular weight cut-off) and antimicrobial activity was measured for each fractions. Antimicrobial activity appeared greatly in the fraction below 3,000 (molecular weight cut-off) . The fraction was re-fractionated by HPLC and substances of two peaks collected as a sample to measure antimicrobial activity. All of both peaks showed the antimicrobial activity but 1st peak exhibited a consistently higher antimicrobial activity than 2nd peak. Minimum inhibitory concentrations (MIC) were between 2.5 and 3.0 mg/mL. The peptide was heat-stable since antimicrobial activity was maintained after treated with heat for 20 min at $121^{\circ}C$. N-terminal amino acid sequence of peptide fractionated by HPLC was leucine, glutamic acid, proline, phenylalanine, aspartic acid and argenine. These results indicated that peptide isolated from zein hydrosate with pepsin can use as a natural preservative ingredient in food industry.
