Mass Mortality of Doctor Fish(Garra rufa obtusa) Caused by Citrobacter freundii Infection (Citrobacter freundii 감염에 의한 Doctor fish(Garra rufa obtusa)의 집단 폐사)

  • In this paper, we described a case of mass mortality of doctor fish from a private fish hatchery farm in Korea with a history of abnormal swimming behavior, diffuse bleeding on the skin and fins and sudden death caused by fish pathogenic bacteria, Citrobacter freundii. Twelve moribund fish fingerling samples were submitted to College of Veterinary Medicine, Seoul National University in October 2008 for diagnostic examination. Diagnostic results showed that the morphological and biochemical properties of the bacteria isolated from the moribund fish were C. freundii. The remaining diseased fish from the hatchery farm were given treatment based on our recommendation and successfully recovered.

Characteristics of the Ekman Layer Flow over a Rough Bottom (거친 바닥 위의 에크만 경계층 내의 흐름의 특성)

  • Ekman layer equation with rough-bottom boundary condition has been solved to determine the effect of roughness on the magnitude of Ekman veering. The bottom boundary condition and the flow field were expanded in a power of roughness (h) which is always smaller than the Ekman layer thickness (${\delta}_E$). By changing the magnitude of roughness parameter (h/${\delta}_E$), the magnitude of the veering, which rotates counterclockwise from the interior geostrophic flow, has been computed. At a fixed depth within the Ekman layer, the magnitude of veering increases as the roughness parameter increases. However, the cross-isobar flux turns out to decrease with increasing roughness. To verify the analytic solution, laboratory experiments were carried out. Rough-bottom cylinderical container filled with homogeneous fluid was sit on a rotating table. The flow pattern during the period of steady spin-up shows that the degree of veering coincides well with the analytic results for various roughness parameters.

Control of Powdery Mildews of Cucumber by Using Mayonnaise (마요네즈 살포에 의한 오이흰가루병 방제)

  • This study was conducted to develop an organic control of powdery mildews of cucumber by using mayonnaise in green house. The treatment of 0.1~2% mayonnaise resulted in 8.3%~99.2% control efficiencies against powdery mildew of cucumber. 0.5% mayonnaise treatment resulted control values over 97% in disease. It did not adversely affected the photosynthesis of foliages. Although one application of mayonnaise to the foliage was not practically effective enough, two or three application of mayonnaise to the foliage at the 0.5% concentration resulted in excellent control against powdery mildews. This treatment could provide protection for 10~14 days after application. Among the type of mayonnaise, general mayonnaise revealed 97.5% control value, but mayonnaise containing low oil content revealed 39.3%~97.5% control values on powdery mildews at the 0.5% concentration. Therefore, oil content in mayonnaise played a essential material to control powdery mildew. Results indicated that mayonnaise could be used as organic control of powdery mildews of cucumber. This control might be environmentfriendly as well as cost-effective.

Development of the Empirical Model for Estimating the Delivered Pollutant Loads Considering Geomorphic and Hydraulic Characteristics (유역형상과 수리특성을 고려한 경험론적 유달부하식 개발)

  • This study was conducted to develop a model equation to estimate the delivered point and nonpoint pollutant loads, which are critical factor to determine the water quality of watersheds. The model equation was developed by considering various factors such as biological removal and delivered distance of pollutants, basin shape and geomorphic runoff condition. The parameters for the model equation were estimated in 3 periods, which are October to March, April to June, and July to September. As a parameter, ${\alpha}_p$, ${\alpha}_n$, ${\beta}$, a and b for $BOD_5$-delivered pollutant loads were estimated to be 0.010~0.0155, 0.051, -0.033, 0.018~0.050 and 0.93, respectively. For T-N, ${\alpha}_p$, ${\alpha}_n$, ${\beta}$ a and b were estimated to be 0.0060~0.0140, 0.014, -0.02, 0.044~0.079 and 0.93, respectively. The same parameters for T-P were estimated to be 0.0160, 0.014, -0.0250, 0.015 and 1.21, respectively. The relationship, $E^2$ (Model efficiency), between observed and calculated delivered pollutant loads showed 0.65 for $BOD_5$, 0.81 for T-N, and 0.66 for T-P, respectively. Consequently, the model equation is effective to estimate delivered pollutant loads for TMDL.

Investigation on Water Quality Variation Characteristics during Dry Season in Namhan River Drainage Basin (남한강수계 저수기 수질변동 특성에 관한 연구)

  • From the direct outflow of Chungju Dam to the junction of water body and watershed in Paldang lake is the scope of this research. This study performed to investigate the main cause of water quality deterioration and the influenced region in the middle field range of the Namhan river Basin with the onsite measurement of water quality and flow rate simultaneously during spring dry season. The purpose of this study is to find out the time-spatial variation characteristics of water quality and flow rate. Following the flow direction $BOD_5$ and $COD_{Mn}$ concentration increased to the highest value of 3.7 mg/L, 5.9 mg/L at Wolgesa respectively. Chl.a concentration increased to $50mg/m^3$ or so at Kangsang, after that it decreased to $37mg/m^3$ at the junction of Paldang lake. Organic matter concentration variation trend showed similar than that of Chl.a. Also $BOD_5$ concentration tendency was similar to Chl.a in every measuring sites except Paldang lake mixing zone. The major factors of water quality deterioration in Namhan river and Paldang lake during dry season were algal bloom and followed internal production. High phosphorus load from Dalcheon and Seom river caused dry season algal bloom and internal production in transitional zone which was stagnant area in downstream of Namhan river.

Study on Estimation and Application of Discharge Coefficient about Nonpoint Source Pollutants using Watershed Model (유역모형을 이용한 유량조건별 배출계수 산정 및 활용방안 연구)

  • TPLMS (Total water pollutant load management system) that is the most powerful water-quality protection program have been implemented since 2004. In the implementation of TPLMS, target water-quality and permissible discharged load from each unit watershed can be decided by water-quality modeling. And NPS (Non-point sources) discharge coefficients associated with certain (standard) flow are used on estimation of input data for model. National Institute of Environmental Research (NIER) recommend NPS discharge coefficients as 0.15 (Q275) and 0.50 (Q185) in common for whole watershed in Korea. But, uniform coefficient is difficult to reflect various NPS characteristics of individual watershed. Monthly NPS discharge coefficients were predicted and estimated using surface flow and water-quality from HSPF watershed model in this study. Those coefficients were plotted in flow duration curve of study area (Palger stream and Geumho C watershed) with monthly average flow. Linear regression analysis was performed about NPS discharge coefficients of BOD, T-N and T-P associated with flow, and R2 of regression were distributed in 0.893~0.930 (Palger stream) and 0.939~0.959 (Geumho C). NPS Discharge coefficient through regression can be estimated flexibly according to flow, and be considered characteristics of watershed with watershed model.

Enzymatic Characteristics of Two Forms of the Purified Glucoamylase from Rhizopus oryzae (Rhizopus oryzae로부터 정제한 두가지형의 Glucoamylase의 효소적(酵素的) 특성(特性))

  • These experiments were conducted to investigate general enzymatic characteristics of two forms(glucoamylase I and glucoamylase II) of the purified glucoamylase produced by Rhizopus oryzae. Molecular weights of glucoamylase I and glucoamylase II estimated by Sephadex G-100gel filtration, were approximately 101,000 and 115,000, respectively, and those estimated by SDS-polyacrylamide gel electrophoresis being 120,000 and 127,000, respectively. Isoelectric points of the above enzyme were pH 7.25 and pH 7.75. The optimum temperature was $50^{\circ}C$ and the enzyme was stable below $45^{\circ}C$. Optimum pH of both glucoamylase I and glucoamylase II was about pH 5.0. The stable pH range of them were pH 3.5-8.0 and 4.5-8.0, respectively. Michaelis constants of glucoamylase I and glucoamylase II toward souluble starch were 4.545 mg/ml and 5.560 mg/ml, respectively. $Hg^{++}$, $Pb^{++}$, p-CMB and IAA were inhibitors of glucoamylase I and $Hg^{++}$, $Mn^{++}$, p-CMB and IAA were inhibitors of glucoamylase II.

Hydrolysis of Various Substrates by Two Forms of the Purified Glucoamylase from Rhizopus oryzae (Rhizopus oryzae로 부터 정제(精製)한 두가지형의 Glucoamylase의 각종기질(各種基質)의 가수분해(加水分解))

  • These experiments were conducted to investigate the substrate specificity, the hydrolysis products on the various carbohydrates and the hydrolysis rate on the various raw starches of the two purified glucoamylase produced by Rhizopus oryzae. Both of the glucoamylases hydrolyzed amylose, amylopectin, glycogen, soluble starch, pullulan, maltose, maltotriose, maltotetraose, maltopentaose, maltohexaose, maltoheptaose and maltooctaose, but did not act on ${\alpha}-cyclodextrin$, ${\beta}-cyclodextrin$, raffinose, sucrose and lactose. When the reaction mixture of glucoamylase and polysaccharides were incubated $37^{\circ}C$for 32 hours, glucoamylase I hydrolyzed amylopectin, soluble starch and amyloses completely, but hydrolyzing glycogen up to only about 88%. Glucoamylase II hydrolyzed the previous four polysaccharides up to about 100%. Both of the glucoamylases produced only glucose for various substrates and did not have any ${\alpha}-glucosyl$ transferase activity. Both of the glucoamylases hydrolyzed raw glutinous rice starch almost complety, wheras they acted on raw potato starch, raw green banana starch, raw arrow root starch, raw corn starch, raw yam starch and raw high amylose corn starch weakly. Glucoamylase II hydrolyzed raw starches at the higher rate than glucoamylase I.

  • Recently, structure and type of GIS data is various and complicated. Therefore, there is a research on efficient data collecting and processing method. Digital maps and orthophotos are needed for efficient usage in the field of GIS. Digital maps which are one of the part of NGIS and digital cadastral maps are different, and data construction isn't performed. Furthermore, there are no research on the improvement of difference between digital maps and cadastral maps. In this paper, manufactured digital orthophotos using aerial photos are used for change detection in a central area of a town. Selecting a town which has various changes, and digital maps and cadastral maps overlap each other, and analyze them for correction, renovation and possible application.

Accuracy of Mid Point Computation for Boundary Delimitation on Ellipsoid (타원체상에서 경계획선을 위한 중간점계산의 정확도)

  • The general rule of boundary delimitation is a the principle of equidistant. The principle of equidistant is a method that determine boundary delimitation from fixed distant of baseline or basepoint. In this paper, study Two-Point Algorithm and Three-Point Algorithm that are widely used. and developed the Boundary Delimitation Program to verify the result and error. This program is specially useful for maritime boundary delimitation problem because there is no artificial and natural object in sea to determine boundary. As a result The mid-points computed on Ellipsoid have small error rather than mid-points on plane or sphere without any distortion by map projection. Through developing boundary delimitation program, can eliminate the various manipulation error using paper map, and quickly cope with maritime boundary delimitation negotiation. Also, verify that the error of basepoint in baseline is propagate the mid-point in mid-line, and determine suitable reference plane.
