Characterization of Extracellular Proteolytic Enzyme of Isolated Psychrotrophic Bacteria from Cheddar Cheese (체다치즈에서 분리한 내냉성미생물의 단백질분해효소의 특성)

  • Psychrotrophs producing protease were isolated during ripening periods of Cheddar cheese and one of them containing the highest protease activity was identified as Pseudomonas fluorescens 65. The extracelluar proteolytic enzyme was partially purified from P. fluorescens 65 through the Sephadex G-100 gel filtration. The protease was eluted between 190 ml and 230 ml of elution volume of sodium phosphate buffer. The purified protease showed a single band in SDS-PAGE and its molecular weight was 47,000. The composition of amino acid for the protease was determined and the most abundant amino acids were glutamic acid (14.96%) and serine (13.86%). The optimum temperature and pH for the activity was $45{\sim}50^{\circ}C$ and 6.0, respectively.

Residues of Benomyl and Bitertanol in Apples Treated as Postharvest Fungicides under Different Storage Conditions (수확후 처리제로서 Benomyl과 Bitertanol이 처리된 사과의 저장조건에 따른 잔류량)

  • This study was conducted to clarify the degradation pattern, safety evaluation and penetration ratio of benomyl and bitertanol to apple used as postharvest fungicides during CA(controlled atmosphere) and cold storage. In CA storage, the degradation of benomyl and bitertanol in stored apple was slow at the early stage, while that in cold storage was, on the contrary, faster at the early stage. The initial concentrations of benomyl and bitertanol in apples applied at the standard application concentrations were 2.24 and 1.54mg/kg, respectively, and their residual amounts were below the maximum residue limits, 1mg/kg at 135 and 96 days in CA storage, 115 and 70 days in cold storage, respectively. The half-lives of benomyl and bitertanol in stored apples were 124 and 130 days in CA storage, 101 and 111 days in the cold storage, respectively, indicating that the degradation was faster in cold storage than in CA storage. The residual amount of benomyl and bitertanol in stored apples was retained more in peels than in pulps of apples.

  • The Korean rice of Paldal variety was milled, in March, 1969, into brown rice and 92% polished one and each 1.5kg sample was packed in the kraft paper bags. With Co-60 gamma-irradiation, the samples were treated in the various ranges of 500, 800 and 1,000 Krad, and then they were stored at the room temperature. The changes of the contents of free amino acids in the samples were determined by the amino acid autoanalyzer (Yanagimoto, Japan, LC-type). The results obtained in June and October of the year were as follows. 1) In the disinfecting irradiation of rice by the higher doses of 500, 800 and 1,000 Krad, there wasn't any particular distinction between the control lot and irradiated lot in respect to the free amino acid contents. 2) The contents of free amino acids in rice during storage had the decreasing tendencies, and especially prominent in case of glutamic acid.

  • Fate of tricyclazole in rice paddy system was studied. The effect on soil microorganism as well as the mutagenicity of the compound was also examined. The residues of tricyclazole in crops and soil with two times application before harvest were 0.37 in unpolished rice, 0.29 in polished rice, 0.14 in rice straw, and 0.15 ppm in paddy soil. With three times of application the residues were increased to 0.46, 0.39, and 0.19 ppm, respectively. Until $2{\sim}3$ weeks after treatment of pesticide the degradation of tricyclazole was progressed comparatively but very slowly afterward and the half life of that was about $140{\sim}180$ days. There was no effect for viable count of soil microorganisms and for mutagenic test by Salmonella and Saccharomyces systems.

Analysis of Wave Parametric Characteristics using WAVEWATCH-III Model and Observed Buoy Data (파랑모델과 부이 자료를 이용한 파랑인자 특성 분석)

  • The analysis of wave parametric characteristics in sea regions in the vicinity of Korean Peninsula have been carried out using the third generation wave model, WAVEWATCH-III (Tolman, 1999) and four observed buoy data of Korea Meteorological Administration (KMA). Significant wave height increases about 2-3 hours later after the increase of wind speed. Maximum correlation coefficient between two parameters appears in Donghae buoy data, which is at off-shore region. When land breeze occurs, it can be found that the correlation coefficient decreases. Time differences between wind speeds and wave heights correspond to significant tidal periods at all of the buoy locations except for Donghae buoy. After verifying the WAVEWATCH-III model results by the comparing with observed buoy data, we have carried out numerical experiments near the Kuroshio current and East Sea areas, and then reconfirmed that when there exist an opposite strong current in the propagation direction of the waves or wind direction, wave height and length get higher and shorter, respectively and vice versa. It has been shown that these modulations of wave parameters are considerable when wind speed is week or mean current is relatively strong, and corresponding values have been represented.

  • Hydrogen can extend the lean misfire limit to a large extent when it is mixed with conventional fuels for an internal combustion engine. This study is about fuel reforming to produce hydrogen enriched gas as a fuel for engine. Especially gasoline, which consists of numerous hydrocarbon fuels, considered as source of reformed gas. Various hydrocarbons, including commercial fuel were reformed and potentialities of reformed gas on vehicles were accessed. The reforming efficiency and hydrogen yield were observed. Maximum hydrogen yield were found with different gas hourly space velocity(GHSV) and O2/C ratio of reforming conditions.

  • The automobile equipped with a gasoline engine uses the ignition coil, namely, a high voltage generator, to make the mixed fuel ignited and burned in the combustion chamber, which results in the power to drive the engine. The ignition coil functions to convert a low voltage of the primary into a hiか voltage of the secondary by switching method, which will be transmitted to the electrode. Here, if the ignition coil has a defect even a little, it cannot function well. In this study, it was chosen epoxy molding ignition coil in recently and epoxy resin which is insulation material as specimens, and it was measured the characteristics of the partial discharge occurring to the specimens when those were applied to a voltage, and thereby, it was researched and analyzed the distribution of phase angle, amount and count of discharge due to the changing voltage, And as the result is applying to the actual automobile ignition system, it can be expected the enhancement of the performance of the ignition coil and the reliability of the electrical equipment.

A Study on Korean Green Tea(II) -Analysis of free Amino Acid and Mineral- (한국산(韓國産) 녹다(緣茶)에 대(對)한 연구(硏究) (제 II 보) -Free amino Acid와 무기성분(無機成分)에 대(對)하여-)

  • Quantitative analysis was achieved by gas-liquid chromatographic method (GLC) with a single column system of OV-17 for 16 of free amino acids in Korean green tea and the contents of mineral in it was determined by atomic absorption flame emission. The results are summarized as follows: 1) Korean green tea contained Mn 1% or over out of the total ash content and $0.05{\sim}0.20%$ in the water extraction, as the major mineral. 2) Ba, Cr, Ni, Pb and V were analyzed also by small quantities relatively and Co, Tin and Ywere not detected in the water extraction. 3) GLC indicated the presence of 16 components in free ammo acids. 4) The quantities of free amino acids in Korean green tea were determined $2.96{\sim}6.61%$ Alanine, $1.01{\sim}l.89mg%$ Glycine, $2.07{\sim}7.81mg%$ Valine, $1.27{\sim}8.76mg%$ Leucine and Isoleucine, $94.31{\sim}316.27mg%$ Threonine, $9.10{\sim}39.91mg%$ Serine, $2.18{\sim}36.76mg%$ Hydroxyproline, $2.72{\sim}5.90mg%$ Proline, $39.64{\sim}70.02mg%$ Aspartic Acid, $25.93{\sim}101.28mg%$ Glutamic Acid and Lysine, $8.32{\sim}18.30mg%$ Phenylalanine and Tyrosine in trace amount. 5) The total free amino acid contents in Korean green tea ranged from 207.24mg% to 516.06mg% and Moo-Deoung tea contained outstandingly high, 516 mg% or over.

  • A new proposed method can separate heart sounds and lung sounds by the realization of adaptive noise canceler using adaptive lattice Wiener filter in contrast to adaptive transversal LMS filter and high pass filter as before. Lung sounds and ECG signal are detected for this purpose, and especially the second heart sounds are reduced by finding T wave location with a T wave seeking algorithm. As a result, for heart sounds reduction It was found that adaptive transversal LMS filter required 100-200's orders, 75-100's orders In adaptive transversal MLMS filter, and only 10-20's orders in adaptive lattice Wiener filter. Adaptive filtering technique has shown greater accuracy than high pass filtering without loss of low frequency component.

Effects of Treatment Process and Brushing Condition on Mechanical Properties of Brushed Fabrics (Part 1) -Mecanical Properties of Brushed Fabrics- (기모직물의 가공공정 및 기모횟수가 역학적 특성치에 미치는 영향(제1보) -역학적특성치를 중심으로-)

  • This study was accomplished to investigate the mechanical properties and handle according to treatment processes and brushing condition. For this study, grey fabrics of blended ratio of polyester 65% : cotton 35% were bleached and dyed. These fabrics were then brushed 1, 3, 5 and 7 times. The mechanical properties were measured with a KES-F system and the mechanical data were then transformed into hand values by the conversion equation, that is, KN-201-MDY. The results obtained by the analysis are as follows; 1) The variation of mechanical properties was remarkable in the initial brushing of 1∼ 3 cycles for all of grey, bleached and dyed fabrics. The values of RT, 2HB, B, G, 2HGS, MMD and SMD were decreased and MIU, WC and T were increased with the increase of number of brushing . 2) LT, B, 2HB, G, 2HG, 2HGS, MMD, SMH and W of dyed fabrics which is coincident with the end use showed lower values than those of grey or bleached fabrics, but MIU, WC and T were higher 3) From the variation of hand value, stiffness of dyed fabric showed lower value than that of grey or bleached fabric. On the other hand, smoothness, soft feeling and total hand values of dyed fabric were higher. Especially, dyed fabric showed the highest total value at the 3 cycles of brushing condition.
