Toxic Effects of Ethylene Glycol on Mammalian Embryo Survivability (Ethylene Glycol이 포유류 초기배자의 생존성에 미치는 독성 효과 분석)

  • This study was carried out to evaluate the effects of embryonic stage and toxicities of cryoprotectant on the rates of survival and development of the cryopreserved mouse early embryo and finally to establish the cryopreservation method of surplus embryos obtained during assisted reproductive technology. Toxicities of two cryoprotectants, dimethyl sulfoxide (DMSO) and ethylene glycol (EG) were investigated using a murine embryo model. Female F-1 mice were stimulated with gonadotropin, induced ovulation with hCG and mated. Two cell embryos were collected and cultured after exposure to either DMSO or EG. Embryo development was evaluated up to the blastocyst stage. The total cell count of blastocysts that were treated with DMSO ($68.1{\pm}24.1$) at the 2-cell stage was significantly lower than that were treated with EG ($81.2{\pm}27.0$) or the control ($99.0{\pm}18.3$) (p<0.001). On comparison of two cryoprotectant treated groups, the DMSO treated group showed a decreased cell count compared with the EG treated group (p<0.05). Both DMSO ($15.4{\pm}1.5$) and EG ($10.2{\pm}1.4$) treated groups showed higher apoptosis rates of cells in the blastocyst compared with the control ($6.1{\pm}0.9$, p<0.0001). In addition, the DMSO treated group showed more apoptotic cells than the EG treated group (p<0.001). The potential toxicity of cryoprotectants was uncovered by prolonged exposure of murine embryos to either DMSO or EG at room temperature. When comparing two cryoprotective agents, EG appeared to be less toxic than DMSO at least in a murine embryo model.

Preparation of MWCNTs/Poly(methyl methacrylate) Composite Particles via the Emulsion Polymerization of Methyl Methacrylate Using MWCNTs Modified by Silanization Reaction and Their Morphological Characteristics (실란화 반응으로 표면 개질된 다중벽 탄소나노튜브(MWCNTs)와 Methyl Methacrylate의 유화중합을 통한 MWCNTs/Poly(methyl methacrylate) 복합 입자 제조 및 그 형태학적 특성)

  • In this study, multi-walled carbon nanotubes (MWCNTs) were oxidized with a mixture of nitric acid and sulfuric acid. After oxidation, oxidized MWCNTs were treated with thionyl chloride ($SOCl_2$) and 1,4-butanediol (BD) in sequence at room temperature to introduce hydroxyl groups on the surface of MWCNTs. The prepared MWCNT-OH was silanized with 3-methacryloxypropyltrimethoxylsilane (MPTMS) to make MWCNT-MPTMS. The MWCNT-MPTMS was used as fillers in emulsion polymerization to make MWCNT-MPTMS/PMMA composite particles with 3 kinds of emulsifiers, hexadecyltrimethylammoniumbromide (CTAB) as a cationic, sodium dodecylbenzene sulfonate (SDBS) as an anionic and polyethylene glycol tert-octylphenyl ether (Triton X-114) as a nonionic emulsifier. Morphologies of composite emulsions were confirmed by a particle size analyzer (PSA) and a scanning electron microscope (SEM). Morphologies of emulsion polymerized MWCNT-MPTMS/PMMA with SDBS showed more uniform particle size distribution compared to those of other two emulsifiers used emulsions. MWCNT-MPTMS/PMMA showed $3.4^{\circ}C$ higher $T_g$ compared to pristine MWCNT/PMMA due to covalent bond formation at interface of MWCNT-MPTMS and PMMA.

Effect of Seeding Date on the Growth and Yield of Cynanchum wilfordii Hemsley (백하수오(白何首烏) 파종기(播種期)가 생육(生育) 및 수량(收量)에 미치는 영향(影響))

  • This experiment was carried out to investigate the effect of seeding date on the growth and yield of Cynanchum wilfordii. Hemsley emergence date was May 16 with seeding on March 20 and was June 4 with seeding on April 30, and required days to emergence was shortened as seeding date was delayed. Vine length was longer about 5cm with seeding on March 30 than that of seeding on march 20 and no. of leaves was small about 5 to 16 as seeding date was delayed. Root length was longer about 0. 9cm to 1. 8cm as seeding date was delayed than that of seeding on March 20, but no. of roots had no differences among seeding dates. Yield of root was more increased 18% with seeding on March 30, 10% with seeding on April 10 and April 20 than 207kg/10a with seeding on March 20. As the results of regression analysis, the optimum seeding date of Cynanchum wilfordii Hemsley was considered April 6 in plain area of central inland.

Studies on the Tissue Culture of Korean-Ginseng III. Effects of NAA on the Callus Induction and Organ Differentiation trom Korean-ginseng Explants (고려인삼의 조직배양에 관한 연구 제3보 NAA가 인삼 Callus의 유기 및 기관의 변화에 미치는 영향)

  • These experiments were carried out to define the effects of NAA, 2, 4-D and Benzyladenine on the callus induction and the organ differentiation from the explants and to find out the vegetative propagation method of Korean ginseng. The results obtained are summarized as follows; 1. NAA was significantly effective in forming roots from the ginseng stem segment and the number of roots was increased by increasing NAA concentration in the medium. The roots were formed from both distal and proximal ends of the ginseng stem segments grown on the medium containing more than 2mg/L of NAA. 2. The amount of callus growth increased proportionatly with NAA concentration in the range of 4.0mg per liter in the medium. The callus was easly induced from stem segment than leaf segment and 2, 4-D was more effective in callus induction and growth than NAA. 3. The benzyladenine showed the significant inhibition effect in forming roots from ginseng explant. The callus was not induced with BA alone, but in BA and 2, 4-D or BA and NAA added medium, the callus was easily induced and its growth was also accelerated. The interaction effects between 2, 4-D and BA on the callus induction and growth were significantly higher than those between NAA and BA. 4. As the ginseng embryos were cultured on the M.S. medium supplemented with 2mg per liter NAA, number of shoots was significantly increased and the percentage of embryo which had shown more than 4 shoots later was 22.2%. On the medium containing 8mg per liter NAA, the ginseng embryo showed the normal growth of shoots and leaves, but increased roots and callus induction on the basal part of shoots. 5. When the shoots with 3 leaflets were cut in 1.5cm long and grown on the Blayde's medium containing NAA 1.0mg per liter, roots were formed at the proximal end of shoot, and a new ginseng seedling was successfully obtained.

  • This study was conducted to evaluate the effect of herbIcides on weed control, growth characteristics and yield in Angelica gigas NAKAI, after transplanting it to the field. All herbicids treated had no effect on the emergency period, bolting rate, and growth characteristics of A. gigos. Simazin wp, methabenzthiazuron wp were slightly harmful, but linuron wp pendimethalin ec, triflurain ec had no injury on the A. gigas, even with double dosage level. Under the treatment of pendimethalin ec, linulon wp, effect of weed control valuae at 55 days after transplanting was 89.6, and 88.3 respectively with 5 or 7 species of weeds appearing. Broadleaf weeds f?ere abundant than grasses, and the major weeds were C. albam, C. flexuos, D.sanguinalis, and E. arvense. Dry yield were increased somewhat more with herbicides than by hand weeding, even with double dosage level.

  • A new species of the rajid genus Okamejei is described from a single specimen (295 mm TL) from off Shantou, Gwangdong in the South China Sea. The new species differs from all other congeners in the following combination of characters: snout pointed, dorsal head length 6.7 times interorbital width, tail moderately wide and long, its length 48.5% TL, interdorsal distance less than length of first dorsal fin base, postdorsal tail short as 5.8% TL, small evenly distributed dark brownish spots, without ocelli on dorsal surface of disc, pores of ampullae of Lorenzini on ventral surface distributed from snout tip to distal end of metapterygium, scapulocoracoid high, its height about 1.4 times rear corner height, trunk vertebrae 23, predorsal tail vertebrae 50 and pectoral fin radials 96.

Optimal Density of Initial Inoculation of Rotifer and, Optimal Density of Chlorella for Sustainable Dai]y Harvest of Rotifer (윤충의 연속반복 수확을 위한 적정윤충접종밀도와 적정 Chlorella 농도에 대하여)

  • For daily supply of rotifers to feed juvenile fish and/or larval crustaceans such as shrimp in their seedling culture procedures, the maximum daily increase of cultured rotifers in relations with the inoculation densities of rotifers and the concentrations of food organism, Chlorella sp. were initially investigated in an experimental culture unit. The best results were obtained from the 10 inds./ml of the rotifer inoculation density with $3\times10^6\;cells/ml$ of Chlorella showing the maximum daily increase of 73.0 inds./ml/day on the 7th day. Based on the best results of the initial experiments a trial was made for continuous daily harvests of rotifers. As in the initial experiments, the maximum daily increase of rotifer was observed on the 7th day, when the daily harvest was initiated and continued once a day for 6days. For the 6 days, the amount of the mean daily harvest was $1.42\times10^5$ individuals per culture flask ($3{\iota}\;filled\;with\;2{\iota}$culture water) shelving a possibility of continuous daily harvests.

Geochemistry of the hydrocarbon gases in the Pohang Area (포항 지역 지하수에 분포하는 탄화수소 가스의 지화학)

  • Chemical components of water, chemical and isotopic compositions of extractable gases were analyzed to characterize the properties of the natural gases which are dissolved in ground water in the Pohang area. Amount of total extracted gases ranges from 27 ml/l to 50.1 ml/l. Hydrocarbon gases are composed of methane ($27{\~}376,420 ppm$) and ethane ($19{\~}127 ppm$). Amount of total hydrocarbon gases is related to the lithology and geological factors around the reservoir. Quantity of hydrocarbon gases tends to increase in the Tertiary reservoirs and in the reservoirs where the Tertiary formations are thick enough. According to the relationship between hydrocarbon gases and total solids in the ground water, it is believed that the hydrocarbon gases were dissolved in the Tertiary formation water. Based on the methane content ($>99.9\%$) and isotopic composition (${\delta}C^{13};-73.1\%_{\circ}{\~}-43.22\%_{\circ}$), we interpret the gases to be of predominantly biogenic origin which were generated by the methanogenic bacterial processes under the low temperature and anoxic conditions.

Studies on the Water and Na Metabolism of Roosters Fed Various Sources of Dietary Fiber (여러 가지 종류의 사료 섬유질을 섭취하는 수탉의 수분 및 Na대사에 관한 연구)

  • Fifteen 1-yr old roosters were employed in order to investigate the influence of various dietary fibers on the water and Na metabolism of chicken. Wheat bran, pure cellulose and ground rice straw replaced corn in the control diet at 10% level; Pectin replaced corn at 8% level. Three roosters were allotted per dietary treatment and one rooster per replicate. The limit of probability accepted as being significant was P$\leq$.05. The dry matter metabolizabilities of cellulose and rice straw diets were significantly lower, and those of control and pectin diets were significantly higher than that of the wheat bran diet. Pectin fed roosters seemed to drink more water than did the other birds. Also, excreta from roosters fed the pectin diet were significantly wetter than those from other treatments. However, no difference in water intake and fecal moisture were found when !he roosters were colostomized. It was also found that the colostomized birds drank more water than did intact ones. Data of water holding capacity and bulk density of dried excreta showed that cellulose, rice straw and wheat bran in decreasing order were strong in fibrous property. There were no significant differences in C$_{Na+}$ , Cosm and C$_{{H_2}O}$ among all treatments. When the values of urine volume, urine osmolality and C$_{{H_2}O}$ were put together, it can be concluded that 11 out of 15 roosters developed diuresis. More severe diuretic symptoms were developed in roosters when they were fed more fibrous diets. In conclusion, when the four dietary fibers were graded in terms of fibrous property, it turned out to be pectin, cellulose, rice straw and wheat bran in decreasing order.

The Operational Status of the 7th Elective-Centered Curriculum (제7차 선택중심 교육과정의 운영 실태)

  • The purpose of this study is to survey the status of the 7th elective-centered curriculum and find problems in operating the system. 485 students in the tenth and eleventh grade were randomly selected as subjects. The results are as follows. (1) Among the students, 29% of them selected their science subject without considering their future jobs, 48% did it without enough overview of the system, and 25% did without clearly understanding the details the choice centered curriculum. (2) Among the students who participated in the survey, 65% of them showed positive responses concerning the needs of individual choice centered curriculum and 96% said it increased their participation in class. However, only 39% believed that operating this type of curriculum is realistically possible. (3) 89% of students selected their science subjects accordingly with the college preparatory courses and 14% selected based on their hopes. (4) The percentage of science subjects chosen are 40% for chemistry, 25% for biology, 22% for Physics, and 13% for earth science, but 74% of students wanted to change their choices. These results showed that students recognized the object of the 7th curriculum, but selected science subjects as a means of getting into college entrances, rather than carefully considering their future and aptitude.
