The Application of Single Nucleotide Polymorphism Markers for Discrimination of Sweet Persimmon Cultivars (단감 품종 판별을 위한 single nucleotide polymorphism 마커 적용 검정)

  • The recent development of next-generation sequencing technology has enabled increased genomic analysis, but very few single nucleotide polymorphism (SNP) markers applicable to sweet persimmon (Diospyros kaki Thunb.) cultivars have been identified. In this study, SNP primers developed from five pollination-constant astringent (PCA) persimmons native to Korea were applied to discriminate between cultivars and verify their usability. The polymerase chain reactions of 19 SNP primers developed by Jung et al. were checked, with 11 primers finally selected. The other eight were very difficult to analyze in the agarose gel electrophoresis and QIAxcel Advanced System used in this experiment and were therefore excluded. The 11 SNP primers were applied through first and second verification to 76 cultivars and collection lines including 20 pollination-variant non-astringent (PVNA), 30 pollination-constant non-astringent (PCNA), 20 PCA, and six pollination-variant astringent (PVA). Of these, 38 were indistinguishable (eight PVNA, 18 PCNA, nine PCA, and three PVA). However, the results of applying the 11 SNP primers to new sweet persimmon cultivars, namely Gamnuri, Dannuri, Hongchoo, Jamisi, and Migamjosaeng, showed that they have the potential to be used as a unique marker for simultaneously determining between them.

  • This study was carried out to compare isozyme band pattern, growth characteristics and tuber yield of Dioscorea batatas to get basic information for varietal classification. It could be identify clearly by protein and esterase band pattern of leaf, petiole and stem tissue in three cultivar but with peroxidase band it was difficult to identify because of similar pattern in leaf and petiole tissue. Three cultivars has different leaf shape as Dan-ma and Jang-ma were lanceolate, Sukunea was long-heart shape in upper part of plants. In phyllotaxis, Dan-ma and Jang-ma shows alternate and opposite but Suwon 2 shows opposite in upper parts of stem. Root length shows significant difference from 9.7cm to 51cm respectively as 30cm, 51cm and 9.7cm in Dan-ma, Jang-ma and Sukunea. Tuber yield of three cultivars were 27.5M/T in Dan-ma, 22.8M/T in Jang-ma and 22.8M/T/ha in Sukunea.

Use of Simple Sequence Repeat (SSR) Markers for Variety Identification of Tomato (Lycopersicon esculentum) (Simple Sequence Repeat (SSR) Marker를 이용한 토마토 품종 식별)

  • This study was carried out to evaluate the suitability of simple sequence repeat (SSR) markers for varietal identification and genetic diversity in 28 commercial tomato varieties. The relationship between marker genotypes and 28 varieties was analyzed. Of the 219 pairs of SSR primers screened against ten tomato varieties, 18 pairs were highly polymorphic with polymorphism information content (PIC) ranging from 0.467 to 0.800. Among the polymorphic loci, two to nine SSR alleles were detected for each locus with an average of 3.3 alleles per locus. Genetic distances were estimated according to Jaccard's methods based on the probability that the amplified fragment from one genotype would be present in another genotype. These varieties were categorized into cherry and classic fruit groups corresponding to varietal types and genetic distance of cluster ranging from 0.35 to 0.97. The phonogram discriminated all varieties by marker genotypes. The SSR markers proved to be useful variety identification and genetic resource analysis of tomato.

Genetic Diversity of Korean Barley (Hordeum vulgare L.) Varieties Using Microsatellite Markers (Microsatellite 마커를 이용한 한국 보리 품종의 유전적 다양성)

  • Microsatellite markers were utilized to investigate genetic diversity among 70 Korean barley varieties (Hordeum vulgare). Ninety nine microsatellite primer pairs were screened for 9 varieties. Twenty primer pairs showed highly polymorphic. The relationship between markers genotypes and 70 varieties was analyzed. A total of 124 polymorphic amplified fragments were obtained by using 20 microsatellite markers. Two to nine SSR alleles were detected for each locus with an average of 6.2 alleles per locus. Average polymorphism information content (PIC) was 0.734, ranging from 0.498 to 0.882. A total of 124 marker loci were used to calculate Jaccard's distance coefficients for cluster analysis using UPGMA. Clustering group was divided 2 groups corresponding to 2-rowed and 6-rowed barley varieties. The phenogram was discriminated all varieties by markers genotypes. These markers may be used wide range of practical application in variety identification and genetic purity assessment of barley.

Effect of Apple Leaf Hair Density on the Occurrence and Infection of Alternaria Leaf Spot Caused by Alternaria mali Roberts (모용(毛茸)이 사과나무 반점낙엽병(斑點落葉病) 발생(發生)과 병원균(病原菌)의 감염(感染)에 미치는 영향(影響))

  • In the 82 apple varieties tested, the distribution of varieties grouped on the damage degree to Alternaria leaf spot were 15 varieties(l8.3%), which damaged above 50%, and 51 varieties(62.2%) damaged below 5%. Susceptible varieties to Alternaria leaf spot were Raritan, Indo, Winesap and Starkrimson, moderately resistant varieties Sekaiichi and Starking, and resistant Jonathan, Tsugaru, Molliies, Jonagold, Gala etc. The leaf hair density on the backside of leaf were rarer inresistant varieties than susceptible one. In inoculation test, penetration and infection of Alternaria mali were easier on the backside than surface of apple leaf at the same varity, and infection rate with Alternaria mali were higher at removal treatment than non-removal one of leaf hairs, so we can guess that leaf hair are related to resistance of variety to Alternaria mali.

  • This study was carried out to discriminate potato cultivars and breeding lines by specific molecular markers using random amplified polymorphic DNA (RAPD) analysis. The genotypes of potatoes used for analysis were eight cultivars and five breeding lines. Some of those show much phenotypic resemblances among them because 'Jopung', 'Daekwan70', 'Gawon', and 'Daekwan72' have immediate parental relationship with 'Superior', 'Irish Cobbler', 'Namsuh', and 'Atlantic', respectively. So, there are many difficulties to distinguish the varieties by the morphological characteristics. Three URP primers, URP2, URP4, and URP8 were selected for promising primers to discriminate potato genotypes or cultivars. The three URP primers were shown very high reproducibility because of the relatively high annealing temperature and long primer size. Although the results of similarity analyses did not always reflect the genetic relationship between potato varieties, the reproducible pattern of amplified DNA bands by URP primers showed possibility for molecular markers for discrimination of potato genotype or cultivar.

Identification of Domesticated Silkworm Varieties Using a Whole Genome Single Nucleotide Polymorphisms-based Decision Tree (전장유전체 SNP 기반 decision tree를 이용한 누에 품종 판별)

  • Silkworms, which have recently shown promise as functional health foods, show functional differences between varieties; therefore, the need for variety identification is emerging. In this study, we analyzed the whole silkworm genome to identify 10 unique silkworm varieties (Baekhwang, Baekok, Daebaek, Daebak, Daehwang, Goldensilk, Hansaeng, Joohwang, Kumkang, and Kumok) using single nucleotide polymorphisms (SNP) present in the genome as biomarkers. In addition, nine SNPs were selected to discriminate between varieties by selecting SNPs specific to each variety. We subsequently created a decision tree capable of cross-verifying each variety and classifying the varieties through sequential analysis. Restriction fragment length polymorphism (RFLP) was used for SNP867 and SNP9183 to differentiate between the varieties of Daehwang and Goldensilk and between Kumkang and Daebak, respectively. A tetra-primer amplification refractory (T-ARMS) mutation was used to analyze the remaining SNPs. As a result, we could isolate the same group or select an individual variety using the nine unique SNPs from SNP780 to SNP9183. Furthermore, nucleotide sequence analysis for the region confirmed that the alleles were identical. In conclusion, our results show that combining SNP analysis of the whole silkworm genome with the decision tree is of high value as a discriminative marker for classifying silkworm varieties.

Molecular Phylogenetic and Dendrological Study of Paper-mulberry (B. kazinoki) in Gyeongsang-do Region (경상도지역 닥나무의 수목학 및 분자계통학적 연구)

  • 닥나무(Paper mulberry)는 뽕나무과(Moraceae) 닥나무속(Broussonetia)에 속하는 낙엽 활엽 관목으로 중국, 일본, 한국 등에 자생하며 Hutchinson(1967)에 의하면 닥나무 속은 열대, 아열대, 난대지방에서 자라는 낙엽성 관목으로 세계적으로 약 6종이 있다고 보고되었다. 일반적으로 닥나무의 품종을 구분하는 것은 수목학적 관점으로 잎의 성상, 줄기의 색과 무늬 유무로 구분한다. 그러나 상기의 수목학적 특징은 닥나무(Broussonetia kazinoki Siebold)와 꾸지나무[Broussonetia papyrifera (L.) L'Her. ex Vent.]가 유사하여 오동정의 사례가 발생하기도 한다. 경상도지역에서는 닥나무를 참닥나무, 머구닥나무, 개닥나무 3가지의 향명으로 구분하고 있다. 본 연구에서는 경상도지역 9개체 닥나무를 대상으로 수목학적 특징을 확인하였다. 나아가 식물종의 기준을 명확히 규명하기 위하여 엽록체 속의 matK, trnL-F, ndhF, 3개 마커와 핵에 존재하는 ITS, 총 4개 마커의 염기서열을 생산하였고 상기 구간에서 얻어진 염기서열 비교분석 및 계통학적 분류를 통해 유연관계를 파악하였다. 수목학적 관점으로는 품종을 명확하게 구분하기가 어려웠으며 분자계통학적 연구로 모든 시료는 닥나무와 꾸지나무의 교잡종으로 확인되었다. 본 연구 결과는 우리나라 전통한지의 원재료로 사용되는 닥나무류 식물자원의 분류체계의 확립을 위한 기초자료로 활용 될 것이다.

Mechanisms of Tolerance to Diphenyl Ether Herbicide Oxyfluorfen in Rice Cultivars (Diphenyl Ether계 제초제 Oxyfluorfen에 대한 벼 품종간 저항성기구)

  • In this study, the relationships between sensitivity to oxyfluorfen, absorption of the herbicide, protoporphyrin IX(Proto IX) accumulation and activities of antioxidative enzymes were examined to identify the tolerance mechanism against oxyfluorfen in various rice cultivars having different level of tolerance to this herbicide. Absorption of oxyfluorfen in tolerant rice cultivars was slower than in susceptible cultivars. Proto IX accumulation in various rice cultivars treated with oxyfluorfen was higher in susceptible cultivars than in tolerant ones. In susceptible cultivars especially, Proto IX accumlated rapidly during the herbicide treatment in the dark. Large amounts of Proto IX accumulation were considered to cause membrane lipid peroxidation in the light. However, among the tested rice cultivars, there was little relationship between their tolerance to oxyfluorfen and the activities of antioxidative enzymes. Therefore, it is assumed that differential susceptibility of rice cultivars to oxyfluorfen was due to difference in their capability to absorb the herbicide and to subsequently accumulate Proto IX.

Evaluation of Quality Characteristics and Definition of Utilization Category in Korean Potato (Solanum tuberosum L.) Cultivars (국내산 감자(Solanum tuberosum L.)의 품종별 품질특성 평가 및 용도구분)

  • Total twenty cultivars of potato (Solanum tuberosum L.) were grown at Gangneung where is located in low land of Korea. Their tubers were harvested from 100 to 120 days after planting, and over 150 g of tubers were selected for quality evaluation. Dry matter, starch, amylose in starch, ascorbic acid, protein, individual sugars and mineral were analyzed and mealiness of cooked potatoes were also evaluated by panel test. Dry mater content was the highest in 'Haryeong', 'Jayoung' and 'Shepody', and the biggest starch granules were observed in 'Jayoung' and 'Sinnamjak'. In addition, the content of amylose was highest in 'Haryeong', 'Chudong' and 'Goun'. Mineral content showed the slight difference between cultivars, but it's tendency was not clear. Vitamin C content was highest in 'Jayoung' as 62.5 $mg{\cdot}100g^{-1}$ FW, and 'Chugang' exhibited the highest content of protein. Sugar content was lower in cultivars for single one than double cropping. The lowest sugar content was observed in 'Atlantic' and 'Namseo' among cultivars for single cropping, and in 'Goun' for double cropping. As a result of analysis for quality factors, we could select nine cultivars ('Atlantic', 'Gahwang', 'Gawon', 'Goun', 'Hareong', 'Irish Cobbler', 'Jasim', 'Jayoung' and 'Shepody') with high dry matter content and low reducing sugar as a cultivar group for processing. Additionally, seven cultivars ('Chugang', 'Gawon', 'Goun', 'Hareong', 'Irish Cobbler', 'Jasim' and 'Seohong') with high mealiness and amylose content and five cultivars ('Atlantic', 'Chudong', 'Gahwang', 'Jopung' and 'Jowon') with low mealiness and amylose content were classified as groups for boiled or steam cooking and for soup or pot dishes, respectively.
