Effects of Carbon, Nitrogen Sources and pH on Direct Somatic Embryogenesis in Liquid Culture of Rehmannia glutinosa Lib. (지황의 액체배양에서 탄소원.질소원 및 pH가 직접 체세포배 형성에 미치는 영향)

  • Basic informations for direct somatic embryo formation in Rehmannja glutinosa Lib. were obtained in 500ml erlenmyer flask. The ratio of ammonium to nitrate nitrogen of 825(mg/l) : 1900(mg/l) was proper condition for somatic embryo formation from stem and petiole explants and 3% sucrose was the most effective carbon source. Full strength MS medium with 2mg/l BA was better than LS medium for somatic embryogenesis. The initial pH 5.7 of medium(full strength MS with 2.0mg/l BA and 0.1mg/l NAA) was good for embryo production. Potassium ion was taken up rapidly within 2 weeks. while $Ca^{++}$ and $Mg^{++}$ ion contents were almost constant during culture period. Sucrose hydrolysis occurred throughout the culture, while glucose and fructose were absorbed simultaneously from the third week of culture.

Controlling Bacterial Regrowth Potential by the Limitation of Nutrients in Drinking Water (영양원의 제한에 의한 수돗물에서의 세균재증식능 억제)

  • In this study, the profiles of the bacterial regrowth of indigenous bacteria in tap water and Pseudomonas fluorescence P17 were investigated for cases when carbon (glucose), and/or nitrogen ($NO_3^-$-N), and/or phosphorus ($PO_4^{3-}$-P) were added below sufficient nutrient concentration (SNC) and when carbon sources (glucose and acetate) and nitrogen sources ($NH_4^+$-N and $NO_3^-$-N) were added together. The bacterial regrowth was decreased with limitation of nutrients, and were lowered relatively in the sample, which plural nutrients were limited. In addition, phosphate might be the effective nutrient to control the bacterial regrowth in drinking water because the bacterial regrowth was significantly decreased by the limitation of phosphate. In contrast, the bacterial regrowth was retarded with increasing the concentration of $NO_3^-$-N. For simultaneously adding carbon(glucose or acetate) and nitrogen sources ($NH_4^+$-N and $NO_3^-$-N), the regrowth counts appeared highly in the condition, for both glucose and acetate. And, the regrowth was increased with increasing $NH_4^+$-N concentration as a nitrogen source.

  • Using Streptomyces chibaensis, which produces a strong inulin-hydrolyzing enzyme, the optimum cultural conditions and composition of the medium for the production of inulase were studied. 1. The highest enzyme activity was obtained at pH 7.5 after 84 hours culture at $30^{\circ}C$. 2. None of carbon source better than inulin was found. 3. $(NH_4)_2HPO_4$ and corn steep liquor were favourable inorganic and organic nitrogen sources for the production of inulase. 4. $KCl,\;MgSO_4\;and\;FeSO_4$ as the metallic salts were effective for the enzyme production at their concentrations of 0.01, 0.05 and 0.0001%, respectively. 5. The highest production of inulase was obtained from the medium of inulin 1.0% and corn steep liquor 2.0% concentrations, respectively.

  • Photobiological $H_2$ production was compared using purple non-sulfur bacteria Rhodobacter sphaeroides KD131 in the medium containing various organic acids as the carbon source and electron doner under illumination of $110\;W/m^2$ using halogen lamp at $30^{\circ}C$. The organic acids used were 0~120 mM acetate, butyrate, lactate and malate. Initial pH 7.0 and cell concentration 1.0 at 660nm were increased to pH 8 and 4.4~5.1, respectively during 24hrs of photo-fermentation when lactate and malate were used. However, acetate and butyrate increased pH to 9 and cell concentration to 3.2~3.9 of malate at the same experimental conditions. Optimum ranges of organic acids concentration and carbon/nitrogen ratio were 30~60 mM and 10~20, respectively. When malate was used as the substrate, maximum $H_2$ production 1.1 ml $H_2$/ml broth, which is equivalent to 1.97 mol $H_2$/mol malate was observed.

  • 본 연구에서는 경유와 윤활유로 오염된 토양에서 유류분해능이 우수한 분해균주 5종을 분리하여 동정하였고. 분리된 미생물을 이용하여 실험실 및 현장 Pilot Test 수행으로 현장 복원에 필요한 설계인자를 도출하여 실제 현장 토양복원에 적용하였다. 미생물의 투입량은 2.0 $\times$ $10^{6}$ CFU/g 이상으로 투입하고, 투입 영양분의 조성은 오염된 탄소원의 몰비 농도와 비교하여 질소원으로는 황산암모늄, 요소, 질산암모늄 등을 질소 몰수로 첨가하구 인산원으로는 인산칼륨, 이인산칼륨 등을 인산 몰수로 공급하여 토양의 C/N/P 비율이 100:10:1~ 100:1:0.5 범위 이내로 조절되도록 오염 토양에 영양분을 공급하였으며, 경작 횟수는 3회/주 이상으로 수행하여 오염토양 TPH 5,000ppm을 40일 동안 2,000ppm 이하로 복원하였으며, 이때 생분해상수 k는 0.0229/day로 확인되었다.

Effect of Nitrogen Compounds and Organic Carbon Concentrations on $N_2O$ Emission during Denitrification (탈질에서 질소성분 및 유기탄소 농도가 $N_2O$ 배출에 미치는 영향)

  • The effects of the compounds and concentrations of nitrogenous electron acceptor, the ratio of electron donor/electron acceptor (C/N), and the complexity of electron donor on the emission of $N_2O$ during wastewater denitrification were quantitatively investigated in this study. The higher ${NO_3}^-$ and ${NO_2}^-$ concentrations, the more $N_2O$ emission was observed. ${NO_2}^-$ has strong effect on $N_2O$ emission as it emitted morc $N_2O$ than ${NO_3}^-$, 50 mg/L of ${NO_2}^-$-N gave the highest conversion (9.3%) and yield (9.8%) of $N_2O$ while ${NO_3}^-$-N (50 mg/L) gave 5.6% conversion and 11.0% yield. Lower C/N ratio decreases nitrogen removal efficiency, but it increases the conversion of $N_2O$ because of the incomplete denitrification by the limited organic carbon. When real domestic wastewater is used as the electron donor of the denitrification, $N_2O$ emission is reduced to 1/10 of the emission when single carbon (acetate) is used. It is thought that multiple carbon source utilizes many denitrification pathways and it seems to be helpful for the reduction of $N_2O$ emission.

Optimization for the Production Factors of Cellulolytic Enzymes of a Fungus, Strain FJ1 by Response Surface Methodology (반응표면 분석에 의한 사상균 Strain FJ1의 Cellulolytic Enzymes 생산조건의 최적화)

  • The production conditions of cellulolytic enzymes by a fungus, strain FJ1, were optimized using response surface analysis. The culture factors which largely affected the production of enzymes such as cultivation time, carbon source concentration, nitrogen source concentration, and composition ratio of carbon sources were employed. Optimizedconditions of the factors above corresponding to each cellulolytic enzyme production were as fellowing: CMCase production was obtained in the conditions of cultivation time of 5.4 days, carbon source concentration of 3.5%, nitrogen source concentration of 0.6%, and composition ratio of carbon sources of 52:48 (avicel:CMC), xylanase appeared in the conditions of 5.3 days, 3.5%, 0.8%, and 54:46, respectively, and $\beta$-glucosidase were 7.0 days, 5.0%, 1.0%, and 83:17, respectively, and avicelase were 6.5 days, 4.0%, 0.9%, and 64:36, respectively. The activities of CMCase, xylanase, p-glucosidase, and avicelase predicted by the response surface methodology were 33.5, 52.6, 2.88, and 1.84 U/mL, respectively, and $\beta$-glucosidase activity was enhanced up to 74% when compared to that obtained in the experimental conditions.

Optimization of Heteropolysaccharide-7 Production by Beijerinckia Indica (Beijerinckia Indica 배양을 통한 Heteropolysaccharide-7 생산 최적화)

  • Beijerinckia indica was cultured in mineral salts medium (MSM) medium with various carbon and nitrogen sources to improve the production yield of heteropolysaccharide-7 (PS-7). At high C/N ratio, the high concentration of PS-7 was produced until 40 h of the culture, whereas most of the glucose as a carbon source was used for the cell growth at low C/N ratio. However, at the high C/N ratio, PS-7 accumulation stopped at 48 h of the culture due to the increasing viscosity of the culture broth would inhibit the cell growth. Therefore, the optimized value of C/N ratio was 33.3 (20 g/L glucose, 7.5 mM $NH_{4}NO_3$) for the high production of PS-7. In the culture with various carbon sources, B. indica effectively used the hexoses or glucose-generating sugars for PS-7 formation. Especially, sucrose was the best carbon source for the high production of PS-7 (6.96 g/L) with a high viscosity (40772 cp). In the culture of B. indica with MSM medium containing 20 g/L glucose and 7.5 mM $NH_{4}NO_3$ in a 51 fermentor, the highest cell concentration was 2.5 g/L and the highest concentration of PS-7 was 7.5 g/L (35174 cp). The additional nitrogen sources of 7.5 mM $NH_{4}NO_3$, glutamine and glutamate at 12 h of the culture after exhaustion of a nitrogen source regulated the metabolism of carbon sources, therefore the nitrogen sources could control PS-7 synthesis.

  • In this study, transglucosidase (TG), an enzyme produced by Aspergillus niger, synthesized isomaltooligosaccharide from ${\alpha}-(1{\rightarrow}4)$ linked substrates. The highest TG-producing A. niger KCTC6913 was selected from six kinds of species, and optimized TG producing conditions were established. Five different carbon sources (potato starch, sweet potato starch, corn starch, wheat starch, and dextrin) and three different nitrogen sources (yeast extract, malt extract, and beef extract) were tested to establish the carbon and nitrogen sources favorable for TG production. Measurements of TG activity after an initial culture at pH 5.0 for 15 days revealed that potato starch and yeast extract, which are basic culture media, resulted in the highest TG activity. In addition, A. niger KCTC6913 increased TG production under aerobic conditions and a controlled carbon/nitrogen ratio. In conclusion, to evaluate TG activity in the established optimal medium, it is confirmed that the basal and potato dextrose broth medium were used as a control, and the highest TG production was measured, which was highlighted in the established optimal medium.
