Molecular Cloning of nifHD from Rhizobium sp. SNU003 (Rhizobium sp. SNU003의 nifHD 클로닝)

  • Genes for dinitrogenase reductase (nifH) and dinitogenase a subunit (nifD) were found to be located on 7.9 kb of EcoRI, 6.5 kb of Sail, 7.3 kb of HindlII and 4.4 kb of Pstl fragments of the genomic blot of Rhizobium sp. SNU003. a symbiotic strain from root nodule of Canavalia lineata. Nine recombinant phage nif-clones were selected from the genomic library constructed by using EMBL-3 BamHI arms of bacteriophage lambda. Among them. Rnif-6 had insert DNA of 15.3 kb. in which 7.6 kb of BamHI!SacI fragment contained nifHD region. Therefore, the 7.6 kb fragment was subcloned into pUC19 and partial restriction map was constructed. As the results, nifH and nifD were found to be located continuously on 4.5 kb of BamHI/BglIl in the genome of Rhizobium sp. SNU003 strain.

  • The plant-homologue of Bax Inhibitor, a gene described to suppress the cell death induced by Bax gene expression in yeast, was isolated from rice (Oryza sativa L.). Nucleic acid sequence and amino acid sequence were 741 bp and 247 bp, respectively. The amino acid sequence of the predicted protein was well conserved in plant (84 % in amino acids) and contained five membrane-spanning segments.

  • Soy bean is one of the most common food material to cause food hypersensitivity reactions in Korea. In this study we have investigated the effect of heating on antigenicity and allergenicity change of soybeans by using immunoblotting and ELISA methods with serum of soybean allergic patients and polyclonal antibody against soybean proteins. Soybean proteins were extracted by one-hour heating in boiling waterbath and separated by SDS-PAGE. After heat treatment, no significant changes of soy protein patterns were observed in SDS-PAGE analysis. Furthermore, the heat treatment had no effect on the results in immunoblotting with polyclonal antibody as well as in ELISA with soybean allergic patients' serum. With these results it may be concluded that allergenicity and antigenicity of soybeans do not reduce by thermal treatment.

  • Spleen cells of mouse immunized with hCG were fused with myeloma cell (SP 2/0 Ag 14) to produce monoclonal antibody against hCG. Several clones of hybridoma secreting monoclonal antibody were established and antibodies were characterized in terms of titer, subisotyping and sensitivity in immunoassay. Several methods, for the purification of anti¬bodies, based on gel-filtration, DEAE-ion exchange chromatography and affinity chromatography. were applied and compared each other by the result of SDS-PAGE. Two-site immunochemiluminometric assay (ICMA) involving the use of an excess concentration of a specific monoclonal antibody passively adsorbed onto the walls of plastic tubes and a chemiluminescence labelled antibody conjugate were de¬veloped for the determination of hCG as a preliminary study.

  • International society has been making a lively discussion about natural disaster by climate change to mitigate natural hazard centering around intra-government association. This study dealt with a strategic mitigation and technical adaptation to support a habitual natural disaster region such as Bangladesh in terms of international cooperation for assistance to protect against the natural hazard. The land-cover on scale of one third of Bangladesh is situated lower than sea level that causes habitual flooding accident which gets increasing in the strengthen every single year. Most of people lives around exterior sea coast being faced with disaster of abnormal storm forming every three year cycle. Especially, the socio-economic status of the people in the coast is usually very low, and it need to get help from international cooperation aid. Therefor, the case study for the vulnerability of natural disaster in Bangladesh on geographical analysis is meaningful to join the international cooperation taking a part of role on technical support and education for adaptation of the natural disaster.

  • Using computational fluid dynamics (CFD) method, we investigated three-dimensional fluid flow field and particle movement with respect to the injected gas flow rate variation in typical cyclone separator system. The results of numerical investigation were deduced by coupling the analysis of fluid flow field with Wavier-stokes equation and the tracking of the particle trajectory with Langrangian approach. It was shown that the increasing of injected gas flow rate resulted in the increasing of pressure loss in the separator. This change of inner pressure had an effect on an aspect of the fluid flow in the separator. Particle movement was determined by fluid flow in the separator and was fully depended on a diameter of particles under the fixed flow rate. Increasing of injected gas flow rate was led to an increasing of the trace of particle, so the particles moved to the lower part of the separator. For this reason, the minimum diameters of the particles were decreased and increased the separation rate under the fixed particle diameter. In conclusion, the changes of injected gas flow rate have an important factor to the fluctuation of the fluid flow field and particle trajectory in the separator.

Clonality Assay of Dysplastic Epithelial Lesions of the Stomach (위 이형성 상피 병변의 클론성에 대한 분자병리학적 연구)

  • Purpose: Dysplasia or flat adenoma of the stomach is regarded as a precancerous lesion. However, the frequency and the evolutionary process of malignant transformation of gastric dysplasia are still debated. In order to see whether the lesion was a monoclonal or a polyclonal proliferation, clonality was assayed by X-linked HUMARA polymorphism. Materials and Methods: DNA was extracted from the paraffin-embedded tissue of 16 consecutive cases of endoscopic biopsy, eight of which supplied both dysplastic and nondysplastic tissue for comparison. HUMARA was amplified by PCR with or without pretreatment with methylationsensitive restriction enzyme, HpaII. The amplification products were electrophoresed on polyacrylamide gel and silver-stained. Results: Among the 16 cases, 13 cases were informative and 3 cases noninformative. Of the 13 cases, one case showed skewed lyonization, rendering 12 cases to be analyzed further. A monoclonal band pattern was noted in 2 cases, and a polyclonal band pattern in 10 cases. A review of the histopathologies of the monoclonal and the polyclonal cases did not reveal features discriminating the two groups. Conclusion: These results suggest that gastric dysplasia is a disease entity heterogeneous in the genetic level, and many cases may be non-neoplastic.

  • Efficient production of antigen specific cytotoxic T cells is critical for appropriate adoptive immune response. In vitro culture and expansion of human T lymphocyte clones are very sophisticated and subtle procedure in immune cell therapy and hard to control. Therefore, many groups devoted their efforts to manipulate artificial antigen presenting cells (aAPCs) that can induce T cell activation and clonal expansion. To mimicking of natural antigen-presenting cells, aAPCs encompass basic signal molecules required for T cell activation: MHC:antigen complexes, co-stimulatory molecules and soluble immune modulating molecules. Orchestrated organization of these molecules is important for efficient T cell activation. Here, we discuss how those molecules have been incorporated in several aAPC models, but also how physical properties od aAPC are important for interaction with T cells.

  • The KEM1 gene is known to affect microtubule and spindle pole body function during the cell division cycle in Saccharomjyces cerevisiae. To identify new genes with functions similar or related to those of KEM1, we isolated a high copy suppressor gene (ROK1) that suppresses the kem1 mutation when cloned on a high copy number plasmid but not on a low copy number plasmid. Two clones which suppress both the benomyl hypersensitivity and the $Kar^{-}$ enhancing phenotype of kem1 null mutation were isolated and were shown to have a 9.0 kb identical insert by restriction endonuclease analysis. The restriction map constructed indicates that this suppressor gene, ROK1 is not KEM1. Subcloning experiments suggest that the functional region of ROK1 is at least 3.0kb in size.

SNU 014201. a symbiotic strain isolated from root nodule of Alnus hirsura, were found to be located in the genome on 13.5 kb of EcoRI, 18.0 kb of BamHI, 10.5 kb of BglII and 4.5 kb of KpnI fragments. Using EMBL-3 BamHI arms of bacteriophage lambda. the genomic library was constructed. from which fourteen recombinant phage nif-clones were selected. Among them, Ahnif-I2 had insert DNA of 18 kb, in which 7.9 kb of BamHl fragment contained nif-H, D, K and 3.6 kb of HindlIl/KpnI had nif-H and partial -D. Therefore, the 7.9 kb and 3.6 kb fragments were subcloned and partial restriction maps were constructed. As the results, nif-F1, D.K genes were found to be located continuously on the 6.5 kb of HindII/BamHI and 5.2 kb of SalIIBamHI fragment in the genome of Frankia sp. SNU 014201.
