Anti-oxidative activity of the herb mixture prescribed to induce blood glucose level and effect on the differentiation of 3T3-L1 fibroblast (혈당저하를 위해 처방된 천연물 복합제의 항산화 활성 및 3T3-L1 세포의 분화에 미치는 영향)

  • We prepared five different kinds of herb mixtures prescribed for hypoglycemic effect. And the physicochemical properties of their water extracts were assessed to identify functional materials. Yields were in the range $19.52{\sim}29.79%$. Total phenolics and flavonoid contents were $349.24{\sim}1,752.21\;mg%$ and $163.06{\sim}1,118.47\;mg%$, respectively, and herb mixtures No. 2, 3 and 5 showed particularly high levels greater than 1,000 mg%. Electron-donating ability was best in herb mixtures showing high levels of total phenolics and flavonoids. Nitrite-scavenging abilities were more than 70% in herb mixtures No. 2 and 5, and decreased as pH increased. Herb mixture extracts strongly inhibited differentiation of 3T3-L1 fibroblasts, with potencies ranked in the herb mixture order 5, 1, 4, 3, and 2. The five different kinds of herb mixtures prescribed for their hypoglycemic effects may be useful as functional food materials.

Antimicrobial Activities of Zanthoxylum schinifolium Extract Against Vibrio parahaemolyticus (Vibrio parahaemoiyticus에 대한 산초 추출물의 항균활성)

  • This study was designed to investigate the possible utilization of Zanthoxylum schinifolium as a source of decontamination agents. The antimicrobial effect of Zanthoxylum schinifolium extract was investigated against Vibrio parahaemolyticus which is food-born disease organism. Ethanol extract of Zanthoxylum schinifolium was compared with water extract of Zanthoxylum schinifolium to test antimicrobial activities against Vibrio parahaemolyticus by disk method. Ethanol extract was more effective than water ,extract on the antimicrobial activities. It had remarkable antimicrobial activities against Vibrio parahaemolyticus. It was very stable on the wide range of temperature and pH. It turned out by GC-MS that estragole (4-allyl anisole) was a major antimicrobial component of Zanthoxylum schinifolium extract. These results indicated that Zanthoxylum schinifolium extract could protect against bacterial contamination and inhibit a growth of Vibrio parahaemolyticus.

Application of the Peel Extracts of Astringent Persimmon Fruits for Wastewater Treatment (폐기 떫은감 껍질 추출물의 생활하수 처리에의 적용)

  • The wastewater was quickly and fastly cleared by the peel extracts of astringent persimmon fruits. The degree of clearness increased with increasing concentrations (400-600 mg/l) of the peel extracts of astringent persimmon fruits. The removal rates of T-P and CODcr in wastewater with lime and the peel extracts of astringent persimmon fruits were lower than ferric chloride and aluminium sulfate at 100-200 mg/l concentration but higher at above 300 mg/l concentration. The removal rate of T-P increased with increasing concentration of the peel extracts of astringent persimmon fruits.

  • Antioxidative activities of ethanol extracts of fruits from three cultivars of Prunus mune with different harvest times were determined. Extracts of P. mune cutivar Okyoung showed the strongest activity at 97.5%(compared to BHT) using DPPH or thiocyanate method, although no statistically significant differences were observed among the cultivars. Antioxidative activities of the extracts measured with glutathione-S-transferase method was over 70% of that of BHT. Survival rates of cells in mouse spinal cord culture challenged by superoxide anion were 89, 79, and 62% at 40 mg/ml of Okyoung, Namgo, and Kochunme extracts, respectively, whereas 90% at 6mM glutathion.

  • Abnormal activation of the Wnt/${\beta}$-catenin pathway and subsequent up-regulation of ${\beta}$-catenin response transcription (CRT) are associated with the development of colon cancer. Thus, the Wnt/${\beta}$-catenin pathway is an attractive target for chemoprevention and treatment of this cancer. In this study, we used a cell-based screen to identify a methanol extract of Dictyota dichotoma (EDD) that suppresses the Wnt/${\beta}$-catenin pathway without altering the level of ${\beta}$-catenin protein and reduces the expression of cyclin D1, which is a known ${\beta}$-catenin/T cell factor (TCF)-dependent gene. EDD inhibited the growth of various colon cancer cells. Our findings suggest that EDD can potentially be used as a chemopreventive agent against colon cancer.

Effect of Herbal Extract on Nicotine Degradation (천연식물 추출물에 대한 니코틴의 분해효과)

  • To develop a nicotine-degrading material (NDM), the natural herbal extract was studied. For the in vitro verification, the herbal extract was mixed into the dilute nicotine solution and the ability of NDM to degrade nicotine into cotinine was measured spectrophotometically. In the in vivo study, the rats in experimental and control groups were orally fed with the herbal extract and water, respectively, for 2 weeks. And then, 3 ㎎/㎏ nicotine was administered to both groups by intraperitoneal injection. In results, the ability of NDM to degrade nicotine into cotinine was shown 2.5 fold higher after 90 min reaction in comparison with the control group. In addition, a decrease of 33% in nicotine concentration and a increase of 57% in cotinine concentration were shown in rat blood. Therefore, NDM was shown to be effective in the conversion of nicotine into cotinine.

Screening of Traditional Medicines for Antioxidative and Anti-proliferative Effects on Rat Mesangial Cells (한약재 추출물의 항산화 및 사구체혈관간세포 증식 억제활성 탐색)

  • In the present study, anti-oxidative and the RMC proliferation inhibitory propeties of 80% ethanol extracts from 63 kinds of traditional medicines were investigated. Inhibitory effects of RMC proliferation were showed that Dalbergia odorifera T. Chen., Melia azedarach Linn$\acute{e}$ and Hydnocarpus anthelmintica Pierre. Among them Hydnocarpus anthelmintica Pierre had the highest anti-oxidative activity ($ORAC_{PE\;value}=1.6$, DPPH = 81.1), but Dalbergia odorifera T. Chen. and Melia azedarach Linn$\acute{e}$ had no effects. These results suggest that the Hydnocarpus anthelmintica Pierre could prevent or protect from kidney disease as antioxidant and anti-proliferative agent for RMC.

Screening of Korean Native Plant Extracts for Herbicidal Activities (국내 자생식물 추출물의 살초활성 탐색)

  • 본 연구는 국내에 자생하는 자원식물들의 살초활성을 조사하여 식물생장억제물질을 활용한 환경친화형 제초제 개발에 요구되는 기초 자료를 확보하기 위해 수행되었다. 국내 자생 식물 101종을 식물체 부위별로 나누어 MeOH로 추출하여 획득된 시료 129점에 대해 돌피(Echinocholoa crus-galli P.B. var. formosensis Ohwi)를 이용한 살초활성 검정을 실시하였다. 시료를 소량의 methanol로 녹여 직경 5.5cm petri dish 상의 Whatman No. 2 여지에 균일하게 흡착시킨 후, fume hood 내에서 용매를 제거하고 1mL의 증류수를 첨가하였다. 돌피 종자를 15립씩 3반복으로 파종하여 $28^{\circ}C$, 5,000lux의 growth chamber에 치상하고, 7일 후 시험구 당 균일하게 자란 10개의 유묘를 선발해 초장 및 근장을 조사하였으며, 살초활성 평가를 위해 대조구 개체의 초장과 근장에 대한 생장억제율을 산출하였다. $1,000{\mu}g/mL$ 농도에서 101종 129점 MeOH 추출물들의 돌피 유묘 뿌리 생장에 대한 억제율을 조사한 결과, 80% 이상을 나타낸 식물 시료는 가죽나무 줄기, 누린내풀 지상부, 단풍취 뿌리, 두릅나무 지상부, 백양꽃 전초, 백양꽃 지하부, 병조희풀 경엽부, 산사나무 잎 가지, 삽주 뿌리, 상사화 전초, 상사화 지하부, 석산 뿌리, 애기송이풀 전초 등 17점이었다. 이들 중 90% 이상 고활성을 나타낸 단풍취 뿌리, 두릅나무 지상부, 백양꽃 전초, 상사화 지하부, 석산 뿌리 유래의 추출물에는 새로운 환경친화형 제초제 개발소재로써 활용할 수 있는 살초활성물질들이 함유되어 있는 것으로 생각된다.

Processing of Powdered Jujube Juice by Spray Drying (분무건조에 의한 분말대추음료의 가공)

  • Spray dried powdered juice was processed from concentrated extract of jujube(Zizyphus jujuba MILLER). Spray drying of the extract solution could not be operated to have powder product by itself over whole concentration range and required addition of some carrier or support material. The concentrated extract of 26$^{\circ}$Bx was combined with carrier material solution to have a final concentration of 30$^{\circ}$Bx, and then spray dried. Proper addition level of carrier solid for physical and flavor quality of the powder product was determined to be 1 : 1 ratio to jujube solid. Combined use of maltose and gum arabic produced the best quality product among the studied carrier materials, which were maltose, dextrin, condensed milk and gum arabic. Enzymatic treatment in extraction process could increase the yield by 13~39%, but hurt the sensory quality of powdered juice. Treatment by 0.5% pectinase(0.05 unit/ml) may be used with lesser quality change for improved yield.

Antioxidative Effect of Pimpinella brachycarpa Ethanol Extract. (참나물 에탄올 추출물의 항산화 효과)

  • This study was performed to investigate the antioxidant effect of 80% ethanol extracts from Pimpinella brachycarpa in vitro. The extraction yields of 80% ethanol extract was 9.23 g/100 g. The extract was further fractionated subsequently by n-hexane, chloroform, ethylacetate, n-butanol and water. Water fraction showed the highest extraction yield among fractions. Antioxidative activities of different fractions were examined by 1,1-diphenyl-2-picryl hydrazyl (DPPH) radical generation, Rancimat test, thiobarbituric acid (TBA) value, nitrite scavenging activity, inhibition of lipid peroxidation and peroxide value in linoleic acid in comparison with the commercial antioxidant butylated hydroxytoluene (BHT). Antioxidant activities of n-hexane fraction of Pimpinella brachycarpa ethanol extract were the highest among fractions. Furthermore, the antioxidative capacity of the n-hexane fraction was similar to that of BHT.
