Screening was performed to isolate biopolymer-producing microorganisms from natural sources. The bacteriological characteristics of this strain and physicochemical properties of the biopolymer produced were investigated. The bacterial strain was identified as a Bacillus coagulans. Crude biopolymer treated with ethanol and actone was purified to fraction I and II by ion exchange chromatography and gel chromatography (Shephadex G-100 and Shepharose CL-2B). Analysis of chemical composition and various color reaction revealed that the polymer is composed of amino acids. It was confirmed that fraction II is a homopolymer of glutamic acid and fraction I is a homopolymer of lysine by analysis data of amino acid analyzer, GC and IR.
To investigate the distribution of nutritive salts and organic compounds in six stations at the coastal area of East Sea, environmental factors, nutritive salts, organic matters and chlorophyll were analysed four times from July 1994 to April 1995. The value of BOD in Ulsan Chuyongam was the highest among 6 stations. The concentration of nutritive salts was generally high through the investigation period near Pohang and Ulsan Chuyongam. From the value of dissolved nutrient salts, the Chuyongam area seemed to be under hyper-eutrophication and the Changsengpo area under eutrophication. Suspended particulate matter (SPM) of 3~10${\mu}{\textrm}{m}$ in size was distributed up to 70% of total volume. The amounts of SPM and particulate organic matter (POM) were considerably high in Pohang and Ulsan, which seemed to be resulted from the input of pollutants of cities and factories nearby.
This study was conducted to investigate immunofunctional activity of the polyphenol fractions isolated from Korean pear. In the experimental of Rosette forming cell, the results showed that all the polyphenol fractions enhance the cell count compared with the control group. Especially polyphenol fraction II and III showed highly significant effect on Rosette forming cell, and allergy inhibition. After antigen challenge, histamine content of blood of the polyphenol groups was lowered to near the normal group.
Brewing method and quality of 10 sample of traditional Dungge-Jang in Kyungsang-Do area were investigated. In order to improve the taste of Dungge-Jang, some amount of boiled bean was added in the Dungge-Jang at early stage of fermentation. The level of amino nitrogen turned out to be low while that of water soluble protein and salt soluble protein was high. Glutamic acid, aspartic acid and proline were the major amino acid in water and salt soluble protein in traditional Dungge-Jang in Kyungsang-Do area. The content of total sugar and free reducing sugar were found to be considerably high, and among the free sugar, glucose was the highest$(2.16{\sim}4.02\;mg/ml)$, followed by maltose and maltotriose. Activities of acid protease and liquefying amylase were $0.13{\sim}1.36$ unit per milliliter and $10.18{\sim}15.19D^{40o}_{30}$ respectively. Result of sensory evaluation showed that the good Dungge-Jang turned out to have well harmonized taste of flavor, sweetness and sourness while the color looked slightly dark and yellow.
The quality characteristics of traditional Kyungsangdo Myungtae (Alaska pollack) sikhae including optimal composition of the product were investigated. In the sensory evaluation, the Myungtae sikhae made from Gampo accepted to be best for traditional Kyungsangdo Myungtae sikhae and it is called standard sikhae. The standard sikhae was prepared as follows: dried-Myungtae 200g, cooked rice 230g, red pepper 92g, garlic 112g, ginger 100g, radish 1.6kg, salt 20g, and malt meal 65g. The Myungtae sikhae made from Gampo showed higher values in total sugar, free reducing sugar, and nitrogen compounds than the Myungtae sikhae made from another locations. The sugar of standard sikhae consisted of four kinds including glucose. And, the contents of free amino acid was increased in the order of glutamic acid, alanine, and methionine. The composition of free amino acid in Myungtae sikhae contained 17 kinds, and the contents was increased in order of glutamic acid, aspartic acid, and proline. The organic acid of standard Myungtae sikhae consisted of 7 kinds including lactic acid during fermentation. The contents of citric acid, succinic acid, and lactic acid gradually increased during fermentation, and especially, increasing of lactic acid remarkably increased than the different organic acid.
Effect of enzymatic modification with pepsin, papain and trypsin was studied on functional properties of isolated sesame meal protein hydrolysates. Solubility of protein hydrolysates distinctively increased from 2% to $53{\sim}94%$ at pH 4. Emulsifying properties showed marked increase 6 fold and 4.5 fold at degree of 10%, 20% hydrolysis by trypsin and degree of 10% hydrolysis by papain. The emulsion stability of the protein was unstable by heat treatment for 30 min. at $80^{\circ}C$. Foaming properties were also enhanced by enzymatic hydrolysis except at degree of 30% hydrolysis. Bulk density and water absorption of protein with trypsin and papain decreased about 0.1 g/ml and $0.3{\sim}0.7\;ml/g$, but oil absorption was increased about 1 ml/g.
The polyphenol compounds of Korean pears were extracted with 60% acetone for 4 days at room temperature and purified using Sephadex LH-20 column chromatography, MCI gel column chromatography, Bondapak $C_{18}$ column chromatography, TLC, and HPLC. As a result, three compounds were isolated. The chemical structures of each compound were determined and identified using NMR, FAM-mass, and FT-IR. The compounds were confirmed as (+)-catechin (compound A), (+)-gallocatechin (compound B), (-)-epigallocatechin (compound C), and procyanidin B-3-3-o-gallate (compound D).
Changes in polyphenol, free sugar, and organic acid concentrations and their antioxidative activities in Korean pear during storage were investigated. The antioxidant activity of polyphenols was evaluated. Polyphenol concentration in different parts of pear, peel, pulp and core, reached 25.7%. pH decreased while concentrations of free sugars, including fructose, glucose, and sucrose, increased during storage. Four organic acids, citric malic, succinic, and fumaric acids, were determined. Succinic acid was the major organic acid in the pulp and decreased after 5 month of storage. The antioxidative of polyphenol fractions I, II, and III on cottonseed, linseed, and fish oils were higher than those of the control group.
As a series of study for an anti-gout agent, two flavan-3-ols have been isolated from Oolong tea. They showed positive reaction in $anisaldehyde-H_2SO_4$ solution and $FeCl_3$, which were able to be confirmed in TLC with dimeric flavan-3-ols. The components were identified as procyanidin B-2 and procyanidin B-2-O-3, 3'-di-gallate by spectroscopic analysis. Their inhibitory effect on xanthine oxidase was also investigated and procyanidin B-2-O-3, 3'-digallate showed 60.6% inhibition at $50\;{\mu}mole$. The compound competitively inhibited the xantine oxidase and dimeric flavan-3-ols contanining gallate had higher inhibition activity.
To examine utilization of Korean black soybean as a natural pigment, the effects of temperature, carbohydrate, organic acid, vitamin C and metal ion were investigated. Korean black soybean pigment was more stable than other anthocyanin pigments when temperature was abused. There was no darkness effect in carbohydrate. In organic acid, the color intensity was increased. The vitamin C addition negatively affected on color of anthocyanin. Among the metal ions tested, $Cu^{2+}$ and $Zn^{2+}$ was not affected but $Mn^{2+}$ makes it unstable. Results indicate that Korean black soybean pigment was more stable than other anthocyanin pigment in various environment at pH 3.
