UPLC Analysis of Pinocembrin and Antimicrobial Activity of Propolis Collected from Different Regions in Korea (지역별 국산 프로폴리스의 항균활성 및 Pinocembrin의 UPLC 분석)

  • Propolis, natural antibacterial agent, which has been used traditional medicine across the globe, is resinous mixture to include abundant bioactive substances. In present study, we investigated antimicrobial activity according to quality characteristics of propolis collected from 9 different regions in Korea. Antimicrobial activity was evaluated by measuring growth inhibition zone using agar well diffusion method against Streptococcus mutans. Constituents analysis of propolis samples were measured through total phenolic contents, total flavonoid contents and quantitative analysis of major compound (pinocembrin) by ultra performance liquid chromatography. As a result, 9 different propolis (10mg/mL) and pinocembrin (1mg/mL) showed antimicrobial activity that has growth inhibition zone more than 11.9mm on S. mutans. The total flavonoid contents of 8 different propolis excluding Jeju island were in compliance with standard of health functional food in Korea and were found to affect antimicrobial activity on S. mutans when contained over 10mg/g. In addition, when content of pinocembrin in propolis was ranged from 12mg/g to 32mg/g, each propolis excluding Jeju island exhibited antimicrobial activity alike. These results indicate that pinocembrin plays a important role for antimicrobial activity of propolis collected from 8 different regions in Korea, and that it can be used as basic data for standardization of Korean propolis.

Analysis of antioxidant and anti-inflammatory effects of Mongolian wild lingonberry and blueberry, and identification of their bioactive compounds (몽골 야생 링곤베리와 블루베리의 항산화, 항염증 효과 및 생리활성 물질 분석 연구)

  • The Mongolian lingonberry and blueberry are two essential food sources found in Mongolia. This study investigated the antioxidant and anti-inflammatory effects of methanol extracts from Mongolian lingonberry (LBE) and blueberry (BBE). Compared to the LBE, the BBE showed higher total phenolic, flavonoid, and anthocyanin contents, as well as antioxidant capacities. The LBE and BBE inhibited the mRNA expression of pro-inflammatory genes, including tumor necrosis factor-α (TNF-α), interleukin-1β (IL-1β), and cyclooxygenase (COX-2) in lipopolysaccharide-stimulated RAW 264.7 macrophage cells. In addition, the LBE and BBE inhibited NADPH oxidase-2 (Nox2) mRNA expression, indicating that they have cellular antioxidant capacities. Anthocyanin derivatives of the LBE and BBE were analyzed using LC-QTOF/MS. Six anthocyanins were identified in the BBE, while one was detected in the LBE. Our findings demonstrate that the anthocyanin-rich LBE and BBE could be used as functional food sources in Mongolia.

Antioxidant and Antibacterial Activities of Extracts from Different Parts of Sophora japonica L. (회화나무 부위별 추출물의 항산화 및 항균활성)

  • In this study, the antioxidant and antibacterial activities of Sophora japonica extracts were investigated to determine the potential of S. japonica as a functional food and medicinal materials. S. japonica was divided into flowers, fruits, and branches, and ethanol extraction was used. The total polyphenol and flavonoid contents were significantly higher in the flower and fruit extracts than in the branch extracts, but the ABTS and DPPH radical scavenging activity and ORAC value were higher in the branch extracts. Among the ethanol extracts of S. japonica, branch extracts showed strong antibacterial activity against Porphyromonas gingivalis, and the MIC was 0.2 mg/ml. Branch extracts showed bacteriostatic activity against P. gingivalis at a concentration of 0.4 mg/ml or less and bactericidal activity at a concentration of 0.6 mg/ml or more. Biofilm biomass production and cell growth of P. gingivalis in the culture medium treated with the branch extract at a concentration of 0.2-2.0 mg/ml were significantly decreased in a concentration-dependent manner. In addition, the mRNA expression of fimA and mfa1 associated with fimbriae formation in these cultures was suppressed in a concentration-dependent manner. Based on these results, S. japonica branch extracts can be used as functional food and medicinal materials, as demonstrated by their antioxidant and antibacterial activities against P. gingivalis and the inhibition of biofilm formation resulting from P. gingivalis.

Characteristics of Glycyrrhiza uralensis extract-loaded chitosan nanocapsules and their antioxidant activity (감초 추출물 함유 키토산 나노캡슐의 특성 및 항산화 활성)

  • To improve the oxidative stability of Glycyrrhiza uralensis extract (GU), GU extraction conditions were optimized for maximal antioxidant activity, and GU-loaded nanocapsules were prepared by chitosan ionic gelation. The optimized ethanol concentration and extraction time were 83.0% and 32.6 min, respectively, using response surface methodology. The particle size of the GU-loaded nanocapsules ranged from 280 to 370 nm. A GU extract of 0.8 mg/mL and chitosan concentration of 2.0 mg/mL were selected as the optimal conditions for entrapment and loading efficiency. Both free GU and GU-loaded chitosan nanocapsules exhibited concentration-dependent antioxidant activity. However, the antioxidant protection factor of GU was effectively maintained when it was entrapped within the chitosan nanocapsules. In conclusion, chitosan nanoencapsulation is a potentially valuable technique for improving the oxidative stability of GU.

Antioxidative Activities and Protective Effects on Alcohol-Induced Oxidative Stress in the Human Hepatic HepG2 Cells of Undaria pinnatifida and Costaria costata Extracts (미역과 쇠미역 추출물의 항산화 및 간 보호 효과)

  • We investigated the antioxidant and hepatoprotective effects of extracts from the Undaria pinnatifida and Costaria costata against ethanol-induced oxidative damage. The total polyphenol and flavonoid contents were highest in the 70% ethanol extract from Undaria pinnatifida and Costaria costata. Also, the radical scavenging activity of DPPH (IC50 0.33± 0.21, 0.48±0.47 mg/ml) and ABTS (IC50 0.34±0.30, 0.47±0.17 mg/ml) in the 70% ethanol extract was higher than that of the hot water and 10% ethanol extracts. To determine the hepatoprotective effects of extracts in ethanol-induced oxidative damage, cell viability was measured using an MTT assay. In the pre-treatment of Undaria pinnatifida and Costaria costata hot water extracts, the concentration-dependent increased the cell viability compared with the ethanol treated cells (73.95%) by 89.91~97.63% and 84.99~90.54%, respectively. The data suggests that 70% ethanol extracts have antioxidant activity and hot water extracts exhibit hepatoprotective effects. Therefore, Undaria pinnatifida and Costaria costata may be considered potential agents for control ethanol-induced liver damage.

Antioxidant and Elastase Inhibitory Effects of Equisetum hyemale Extract (속새(Equisetum hyemale) 추출물의 항산화 및 Elastase 저해 효과)

  • Background: The presence of antioxidants was not confirmed in the medicinal plant Equisetum hyamale grass. Purposes: This study was to determine the antioxidant and elastase inhibition effects of extracts of E. hyemale. Methods: Antioxidant functions of E. hyemale stems and roots were measured and extracted with hot water (HW) and ethyl alcohol (70EOH, 100EOH). Results: The extraction yield of stems was higher in HW extraction than in ethyl alcohol extraction. The polyphenol content was significantly higher in the root extract than in the stem. Total flavonoid content of 70EOH extract was significantly higher in root extract than in stem. The elastase inhibitory function of the extract was 46% in the root and 49% in the stem at 100 ppm of the extract. The ABTS free radical scavenging function was in the order of HW<70EOH<100EOH

Antioxidant, α-amylase and α-glucosidase inhibitory ability effects of sesame meal ethanol extract (참깨박 에탄올 추출물의 항산화 및 α-amylase및 α-glucosidase 저해 활성)

  • In this study, a sesame meal was used in order to analyze the proximate composition and mineral contents. The sesame seed meal, pressed from roasted Sesame seed, contains various polyphenols. The defatted sesame meal was extracted using 70% ethanol, and its antioxidant activity and antidiabetic effects were evaluated. Proximate composition of sesame meal was showed that moisture 6.51%, carbohydrate 16.22%, crud protein 46.30%, ash 9.88%, crude fat 21.09%. Mineral contents were K 1128.08 mg/100 g, Ca 1356.27 mg/100 g, Fe 12.29 mg/100 g, P 2022.14 mg/100 g, Cu 2.08 mg/100 g, Mg 643.40 mg/100 g, Na 7.29 mg/100 g. The results showed the sesame meal of 70% ethanol extract had higher polyphenol content (184.98 mg GAE/g) and flavonoid content (27.63 mg QE/g). The 2,2-diphenyl-1-picrylhydra-zyl and 2,2'-aziono-bis(3-ethylbenzthiazoline-6-sulfonic acid radical scavenging activities of defatted sesame meal (IC50 ) were 891.84 and 340.09 ㎍/mL. According to the test results, the defatted sesame meal extracted using 70% ethanol had significant antioxidant activity and inhibitory ability to diabetes-related enzymes, indicating that it has good potential as a functional food or nutritional food for prevention and treatment of oxidation.

Influence of cell-wall degrading enzyme treatment and Saccharomyces cerevisiae fermentation on the antioxidant and antibacterial activities of green tea leaf (세포벽 분해 효소 처리 및 Saccharomyces cerevisiae 발효가 녹차 잎의 항산화 및 항균 활성에 미치는 영향)

  • This study was conducted to suggest an extraction method for preparing the extract from green tea leaves that possess enhanced antioxidant and antibacterial activities. Different ethanol concentrations were tested to recover phenolics and flavonoids, and 50% ethanol was the best under heat treatment (121℃, 15 min). The ethanol extract exhibited excellent DPPH (2,2-diphenyl-1-picrylhydrazyl) radical scavenging activity and growth inhibition against B. cereus, B. licheniformis, S. aureus subsp. aureus, and A. hydrophila subsp. hydrophila. To enhance the antioxidant and antibacterial activities, cell-wall degrading enzymes (2.5% cellulose+2.5% pectinase, v/w dry sample) treatment and Saccharomyces cerevisiae fermentation were applied singly or in combination. The enzymatic treatment of green tea leaves notably increased extraction yield. However, the antioxidant and antibacterial activities of the extract were lower than those of the control (heat-treated 50% ethanol extract). In contrast, the yeast fermentation alone did not affect the yield, but enhanced antioxidant and antibacterial activities, contributing to the increase in the extract's total phenolic and flavonoid contents.

Anti-oxidant activities of ethanol extract and fractions from defatted Camellia japonica L. seeds (동백 유박 에탄올추출물 및 분획물의 항산화 활성)

  • The aim of this study was to investigate in vitro antioxidant activities of defatted Camellia japonica L. seeds (DCJS). The DCJS were extracted using ethanol and then fractionated with butanol (BuOH), ethyl acetate (EtOAc), chloroform, and hexane. To evaluate antioxidant activity of extract and fractions from DCJS, we investigated free radical scavenging activities such as 2,2-diphenyl-1-picrylhydrazyl (DPPH), 2,2'-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) (ABTS+), hydroxyl radical (OH), and superoxide anion (O2-) radicals. The five extract and fractions of DCJS dose-dependently increased DPPH, ABTS+ and O2- radical scavenging activities. The BuOH fraction of DCJS showed the highest free radical scavenging activities among other extract and fractions. The contents of total polyphenol and flavonoid in BuOH fraction of DCJS were 23.26 mg GAE/g and 32.39 mg QE/g, respectively. The polyphenol and flavonoids contents of BuOH fraction has highest than other extract and fractions. In addition, BuOH and EtOAc fraction of DCJS contained 102.37 and 165.05 ㎍/g of camelliaside B, respectively. Therefore, DCJS has higher antioxidant activity and may be useful as a natural antioxidant material.

The Antioxidant and Nitrite Scavenging Activity of Wild Grape(Vitis coignetiea) Wine (머루와인의 항산화성 및 아질산염 소거능)

  • Total anthocyanin and color intensity contents of wild grape wine were $4.3{\pm}0.3%$ and $10.2{\pm}0.8%$, respectively. The contents of total phenols and flavonoid in wild grape wine were $18.8{\pm}3.9$ mg/100 g, $0.5{\pm}0.2$ mg/100 g, respectively. Total mineral content in wild grape wine was $22.6{\pm}0.2$ mg/100 g and the 10 minerals of the potassium content ($5.3{\pm}0.2$ mg/100 g) was the highest. Electron donating abilities of wild grape juice at concentration of 1,000 ${\mu}L/mL$ were $90.4{\pm}1.8%$. Reducing power of wild grape wine at concentration of 1,000 ${\mu}L/mL$ were 0.932. The electron donating abilities and reducing power were increased significantly by increased the sample concentration in the reaction mixture. The nitrite scavenging ability was dependent on pH of reaction mixture and sample concentration. The nitrite scavenging ability of wild grape win was $76.3{\pm}1.3%$ at concentration of 1,000 ${\mu}L/mL$ under pH 1.2.
