Optimization of Extraction Conditions of Antioxidant Activity and Bioactive Compounds from Rice Bran by Response Surface Methodology (반응표면분석법을 이용한 미강으로부터 항산화 활성 및 생리활성물질의 초음파 추출조건 최적화)

  • The rice's waste byproduct is known as a rice bran and produced annually about 400,000 to 600,000 tons. Most of the rice bran are used as a livestock feed or waste disposal, and needed to be used to produce high-added substances, such as bioactive materials. In this study, extraction conditions of the ultrasound-assisted extraction (UAE) of the rice bran were optimized using a statistically-based optimization. The influence of extraction variables including the extraction time ($X_1$), extraction temperature ($X_2$) and ethanol concentration ($X_3$) were investigated using the response surface methodology in order to determine optimum extraction conditions which maximize total phenolic compounds (TPC), total flavonoid compounds (TFC) and electron donating abilities (EDA). The optimal UAE from rice bran was achieved under the extraction temperature of $94.9^{\circ}C$, extraction time of 41.6 minute and ethanol concentration of 74.0% (v/v) with maximum yields of TPC 2.78 mg GAE/g DM, TFC 1.63 mg QE/g DM and EDA 42.86%. The UAE process shows its potential to the extraction of bioactive and antioxidant compounds from rice bran in a short extraction time and low temperature. Also, it is proposed that rice bran could be considered as food additives and cosmeceutical products.

Antioxidant activity of ten Lamiaceae plant seed extracts (10종 꿀풀과(Lamiaceae) 식물 종자 추출물의 항산화 활성)

  • This study explored plant-derived natural antioxidants by evaluating the antioxidant activity of Lamiaceae plant seed extracts. Plants with the percentage of filled seeds at or above 90% and seed germination at or above 50% were selected. Of the ten species studied, the total phenolic content of the seeds was high in the species Phlomis umbrosa Turcz. (6.2 mg GAEs/g of seeds) and Elsholtzia ciliata (Thunb.) Hyl. (4.5 mg GAEs/g of seeds). The total flavonoid content of the seeds was high in E. ciliata (1.0 mg QEs/g of seeds) and P. umbrosa (0.6 mg QEs/g of seeds). Based on the EC50 value of the seed extracts, 2,2-diphenyl-1-picrylhydrazyl radical scavenging activity was high in the seeds of the plants E. ciliata (27.5 ㎍/mL), Mosla dianthera (Buch.-Ham. ex Roxb.) Maxim. (29.2 ㎍/mL), and Prunella vulgaris var. lilacina Nakai (33.3 ㎍/mL). In addition, 2,2'-Azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) radical scavenging activity was high in P. vulgaris var. lilacina (25.6 ㎍/mL), E. ciliata (25.9 ㎍/mL), and M. dianthera (27.6 ㎍/mL) seeds. The ferric reducing antioxidant power of the seed extracts was high in P. vulgaris var. lilacina (2910.4 µM Fe(II)/g of extract), E. ciliata (2836.2 µM Fe(II)/g of extract), and M. dianthera (2754.4 µM Fe(II)/g of extract). According to the cluster analysis based on antioxidant activity, the seeds of the ten species were classified into three groups, from group 1 with low antioxidant activity to group 3 with high antioxidant activity; E. ciliata, M. dianthera, and P. vulgaris var. lilacina were classified as group 3.

Fermentation and quality characteristics of ALE beer with the addition of Muscat Bailey A grape (MBA 포도 첨가에 따른 ALE 맥주의 발효 및 품질 특성)

  • This study investigated the fermentation characteristics and quality attributes of ale beer brewed with MBA grapes to enhance the diversity of Korean domestic ale beers. The grapes were added to the wort in two forms: must and juice, at ratios of 10% and 20%. The results showed that while ale beer with 20% grape addition began fermentation a day later, all samples completed fermentation well on the sixth day. Increasing MBA grape content lowered the pH and increased total acidity and malic acid content without significantly affecting the sour taste. Higher grape addition also decreased the beer's IBU and bitterness scores in sensory evaluation. The addition of must addition enhanced redness due to anthocyanins, significantly increasing color intensity and EBC value of ale beer. Ale beer with the addition of must exhibited higher antioxidant capacities in total phenolic compound content, DPPH radical scavenging, and FRAP activities compared to that with the addition of juice. Sensory evaluation indicated that ale beer with 20% juice addition was preferred for its color, aroma, sweetness, body, and overall preference. The addition of MBA must improved antioxidant capacity, but ale beer with 20% juice had superior sensory qualities.

Effect of Monascus Fermentation on Content of Monacolin K and Antioxidant Activities of Germinated Brown Rice (홍국균 발효가 발아현미의 Monacolin K 함량과 항산화 활성에 미치는 영향)

  • This study was performed to investigate the changes in monacolin K content and antioxidant activities of Monascus-fermented brown rice with different germination temperatures and periods. Brown rice was germinated at 32, 35 and $37^{\circ}C$ for 1~4 days, after white rice (WB), brown rice (BR), and germinated brown rice (GBR) were fermented with M. pilosus 305-9 at $30^{\circ}C$ for 20 days. The redness, yellowness and Monascus pigments increased after germination. Total monacolin K content increased from 215.85 mg/kg of BR to 1,263.04 mg/kg of GBR ($32^{\circ}C$/1 day), whereas monacolin K content decreased with increase in germination period. Citrinin was not detected in any of the samples. Total polyphenol (TPC) and flavonoid contents (TFC) increased with increase in germination temperature and period, whereas electron donating ability (EDA) and total antioxidant activities (TAA) decreased due to reduction of Monascus pigment content. The TPC and TFC showed the highest values (13.80 mg/g and 1.30 mg/g, respectively) in GBR ($37^{\circ}C$/4 day), whereas EDA and TAA showed the highest values (22.16 mg Trolox equivalent/g and 62.27 mg ascorbic acid equivalent/g, respectively) in GBR ($32^{\circ}C$/1 day). These results indicated that the optimal germination temperature and period for increasing monacolin K content and antioxidant activities was found to be at $32^{\circ}C$ for 1 day. In addition, it was found that M. pilosus 305-9 was a useful strain for increasing monacolin K content without producing citrinin in functional foods and pharmaceutical industrial regions.

Antioxidant Activities of Prunus salicina Lind cv. Soldam (Plum) at Different Growth Stages (생육 시기에 따른 피자두(Prunus salicina Lindl. cv. Soldam)의 항산화 효과)

  • This study was performed to evaluate the antioxidant effect of Prunus salicina Lindl. cv. Soldam at different growth stages (sample 1-8). Previous studies shows that this fruits possess hematopoiesis effect, osteoporosis prevention, and antimutagenic effects. Prunus salicina Lindl. cv. soldam was picked in every 5 days from the 40th day before harvesting date for marketing in Gimcheon, Gyeongbuk. The fruits at different growth stages (sample 1-8) were extracted with 60$\%$ acetone and chlorophyll in the extracts was removed. In proximate compositions, the contents of moisture of sample 1, 5, 8 were 88.52, 87.01, 83.56$\%$ ; crude ash were 7.12, 3.35, 3.57$\%$ ; crude protein were 7.52, 5.55, 3.85$\%$ ; crude fat contents were 3.20, 0.99, 5.15$\%$, respectively. The contents of total polyphenols and condensed tannin in the acetone extracts from sample 1, 5, 8 were 10.67, 4.05, 2.57$\%$, and 8.36, 3.11, 1.88$\%$, respectively. The antioxidantive effect of acetone extracts from immature fruits showed strong scavenging effect on DPPH free radicals. The RC$_{50}$ values of the extracts from sample 1, 2 were 2.23, 9.70 $\mu$g/mL, respectively while those of butylated hydroxyanisole (BRA) was 5.25 $\mu$g/mL. The extracts from immature fruits showed over 85$\%$ inhibition on peroxidation of linoleic acid at 100 $\mu$g/mL as determined by both the ferric thiocyanate (FTC) and the thiobarbituric acid (TBA) method.

Chemical Components and Biological Activity of Stauntonia hexaphylla (멀꿀의 화학성분과 생리활성)

  • This study was conducted to gather the basic data on the increase of utilization for the Japanese staunton vine (Stauntonia hexaphylla), native plants which were grown in the southern districts in Korea. We have also determined their partial physical and chemical compositions and their physiological activities. Vitamin C contents in fruit skin was 85.23 mg/100 g, and that in flesh was 61.67 mg/100 g. Total amino acids contents in fruit skin increased much more by 762.72 mg/100 g DW compared to that in flesh by 434.05 mg/100 g DW. Inorganic matter contents were more increased in the fruit skin (108.48 mg/$\ell$) and its main components were K (76.53 mg/$\ell$), Ca (20.20 mg/$\ell$) and Mg (6.22 mg/$\ell$). Total phenol compound and flavonoid contents in 1,000 mg/$\ell$ methanol extracts were 7.3-9.6 mg/$\ell$ and 5.1-6.7 mg/$\ell$. Nitrite radical scavenging activity in 4,000 mg/$\ell$ methanol extracts of fruit skin and flesh for Stauntonia hexaphylla were 79.5% and 77.8%, however, that in seeds was 17.1%. Overall mushroom tyrosinase inhibition activity (% of control) was less than 10.8%. Anti-microbial activities of methanol extracts from the fruit skin against the gram negative and positive microbial strains were not significant in the lower concentration of extracting solution, however, that from flesh and seeds in terms of the inhibition diameter were $8.91{\sim}12.25\;mm$.

Nutritional Analyses and Antioxidant Activity of Apple Pomace (사과 부산물의 영양성분 분석 및 항산화 효과)

  • To enhance the applications of apple pomace, which is a by-product of apple, this study analyzed the nutritional components, ursolic acid content, and antioxidant activity of different solvent (distilled water, fermented alcohol, and methanol) extracts. The samples included hot air-dried and freeze-dried apple pomace. The moisture, protein, fat, ash, and total dietary fiber contents of hot air-dried apple pomace were 3.2%, 3.9%, 2.4%, 2.0%, and 28.5%, respectively, and those of freeze-dried apple pomace were 8.2%, 3.4%, 2.4%, 1.8%, and 33.0%, respectively. Ursolic acid was not detected in the distilled water extract of either sample. However, in hot air-dried apple pomace, the methanol extract was 1,753.32 ㎍/ml, and the fermented alcohol extract was 1,532.94 ㎍/ml. In freeze-dried apple pomace, the methanol extract was 1,407.04 ㎍/ml, and the fermented alcohol extract was 1,221.81 ㎍/ml. The total polyphenol and flavonoid contents were 306.7 ㎍/ml and 950.1 ㎍/ml, respectively in methanol extracts of hot air-dried apple pomace and 277.6 ㎍/ml and 925.0 ㎍/ml, respectively in methanol extracts of freeze-dried apple pomace. 2, 2'-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activities of hot air-dried apple pomace were 73.3% in methanol extract and 59.4% in fermented alcohol extract, and those of freeze-dried apple pomace were 76.1% in methanol extract and 66.0% in fermented alcohol extract. Both samples had the lowest antioxidant activity in distilled water extracts. Similar to DPPH radical scavenging activity, both samples showed increasing 2,2'-azino-bis(3-ethylbenzothiazoline-6- sulfonic acid) (ABTS) radical scavenging activity in the order of methanol, fermented alcohol, and distilled water. All samples had stronger reducing power than ascorbic acid (311.5 ㎍/ ml) as a positive control.

In vitro antioxidant, anti-diabetic, anti-cholinesterase, tyrosinase and nitric oxide inhibitory potential of fruiting bodies of Coprinellus micaceus (갈색먹물버섯 자실체의 메탄올과 열수추출물의 항산화, 항당뇨, 항콜린에스테라아제, 항티로시나아제 및 Nitric oxide의 저해 효과)

  • Coprinellus micaceus, belongs to family Psathyrellaceae of Agaricales, Basidiomycota, has been used for edible purposes in the world. This study was initiated to evaluate the antioxidant, anti-diabetic, anti-cholinesterase, anti-tyrosinase, and nitric oxide inhibitory activities of fruiting bodies from C. micaceus extracted with methanol and hot water. The HPLC analysis of phenolic compounds from the mushroom extracts identified 4 phenolic compounds including procatechuic acid, chlorogenic acid, (-)-epicatechin, and naringin. In 1,1-diphenyl-2-picrylhydrazyl(DPPH) free radical scavenging assay, the scavenging activities of methanol and hot water extracts were lower than that of positive control, BHT. The chelating effects of methanol and hot water extracts were significantly higher than that of BHT, the positive control at the all concentrations tested. In the reducing power assay, methanol and hot water extracts exhibited the lower activities compared with positive control at the 0.125-0.2 mg/ml. The methanol and hot water extracts of the mushroom inhibited the ${\alpha}$-glucosidase activity by 62.26% and 67.59%, respectively at the 2.0 mg/ml, while acarbose, the positive control, inhibited the ${\alpha}$-glucosidase activity by 81.81% at the same concentration. In the acetylcholinesterase(AChE) inhibitory activity assay, methanol and hot water extracts of the mushroom inhibited the AChE by 94.64% and 74.19%, respectively at 1.0 mg/ml, whereas the galanthamine, standard drug, inhibited the AChE activity by 97.80% at the same concentration. The tyrosinase inhibitory activities of methanol and hot water extracts were 91.33% and 91.99% at 2.0 mg/ml, while the inhibitory activity of kojic acid, the positive control, was 99.61% at the same concentration. Nitric oxide(NO) production in lipopolysaccahride (LPS) activated RAW 264.7 cells were inhibited by the methanol and hot water extracts in a concentration dependent manner. Therefore, it is concluded that fruiting bodies of C. micaceus contained natural antioxidant, anti-acetylcholinesterase and ${\alpha}$-glucosidase inhibitory, anti-inflammatory, anti-tyrosinase substances which might be used for promoting human health.

Antioxidative and Antigenotoxic Effect of Omija (Schizandra chinensis B.) Extracted with Various Solvents (다양한 용매에 의해 추출된 오미자의 항산화능과 항유전독성 효과)

  • The purpose of this study was to evaluate antioxidant and antigenotoxic effects of Omija (Schizandra chinensis B.) extracted with various solvents (acetone, ethanol, and methanol). The total polyphenol content (TPC) of methanol extract (ME), ethanol extract (EE) and acetone extract (AE) from Omija were 1183.3, 1009.4, and 747.3 mg/100 g (garlic acid equivalents: GAE), respectively. Antioxidant effects of the Omija extracts was measured by DPPH radical-scavenging activity (RSA) and superoxide dismutase (SOD)-like activity. The $IC_{50}$ for DPPH RSA was in the order of EE $(1411.1\;{\mu}g/mL)$=AE $(1462.0\;{\mu}g/mL)$>ME $(1585.0\;{\mu}g/mL)$. The $IC_{50}$ for SOD-like activities was the highest in ME $(905.7\;{\mu}g/mL)$=EE $(970.3\;{\mu}g/mL)$>AE $(1579.4\;{\mu}g/mL)$. The antigenotoxic effect of Omija on DNA damage induced by $H_2O_2$ in human leukocytes was evaluated by comet assay. $H_2O_2$ induced DNA damage was effectively protected by all of the Omija extracts. Aectone extract of Omija showed the highest antigenotoxic effect ($IC_{50}$ value of AE is $14.6\;{\mu}g/mL$) followed by EE, and ME (21.4 and $34.4\;{\mu}g/mL$), respectively. As a result, we propose that Omija (Schizandra chinensis B.) can serve as a new natural source enriched with potent antioxidant and antigenotoxic agents.

A study on Physiochemical Property of Salicornia herbaciea & Suaeda japonica (함초와 칠면초의 이화학적 특성 조사연구)

  • This study was conducted to investigate for mineral contents, total polyphenol compounds, betaine, choline and DPPH free radical scavenging activity of halophyte. The mineral concentrations of Salicornia herbacea (top part) were Na 100,006 mg/kg, K 1,385 mg/kg, Mg 6,263 mg/kg, Ca 2,750 mg/kg, Fe 90.4 mg/kg, Mn 98.9 mg/kg, Zn 33.3 mg/kg, Cu 3.4 mg/kg respectively. And Suaeda Japonica (top part) were Na 85,332 mg/kg, K 710 mg/kg, Mg 7,005 mg/kg, Ca 4,344 mg/kg, Fe 1,434.9 mg/kg, Mn 119.1 mg/kg, Zn 19.2 mg/kg, Cu 2.7 mg/kg respectively. The betaine contents of Salicornia herbacea (top part) were 15.09 mg/g and Suaeda Japonica (top part) were 14.64 mg/g. The choline contents estimated by the DBAP-choline derivatives of Salicornia herbacea (top part) were 20.9 mg/100 g, Salicornia herbacea (root) were 23.4 mg/100 g, Suaeda Japonica (top part) were 23.1 mg/100g and Suaeda Japonica (root) were 23.8 mg/100 g. Total polyphenol compounds of Salicornia herbacea (top part) were high 36.0 mg/g in growth phase. The DPPH radical scavenging activities of methanol extract Salicornia herbacea (top part) were high 90.1% in growth phase. The frozen dried powder of Salicornia herbacea (top part) 1 g was equal to Quercetin 30.26 mg, Rutin 42.65 mg, TBHQ 20.32 mg, BHA 25.86 mg, BHT 40.75 mg, Ascorbic acid 22.86 mg in DPPH radical scavenging activities.
