Background : In chronic airway disease, mucus secretion is increased, but extraction of mucin, which is the main component of mucus secretion, is a very complicated and limited in clinical use. Recently, monoclonal antibody for mucin was developed for possible clinical use. In this study, cellular analysis and quantification of respiratory mucin in sputum of patients with chronic airway diseases were performed. Method : Sputum was collected from patients with asthma(n=33), bronchiectasis(n=8) or chronic bronchitis (n=13) by spontaneous expectoration or by hypertonic saline induction. Collected sputums was treated by 0.1% dithiotreitol to dissociate the disulfide bond of the mucus and filtered through a nylon gauze. Total cell count, viability and differential count were measured. For detection of mucin, collected samples were treated with sodium dodecyl sulfate polyacrylamide gel electrophoresis and then with monoclonal antibody(HMO2), as the primary antibody, and PAS stain. The amount of mucin was measured with ELISA by HMO2. Correlation with clinical information, cellular analysis, and amount of measured mucin were analyzed. Results : Total cell counts of sputum were significantly increased in patients with bronchiectasis but viability remained the same. Eosinophils were significantly increased in patients with asthma, neutrophils in bronchiectasis chronic bronchitis, respectively (p<0.05). The results of Western blotting and PAS staining confirmed the presence of glycoproteins and matched? with mucin. The amounts of mucin measured by ELISA were not significantly different among the disease groups. Significant correlation was identified between the amount of mucin and viability(r=-0.482, p<0.05). Conclusion : Inflammatory cells in the sputum of those with chronic airway disease were different for each disease type. Measurement of mucin by ELISA via monoclonal antibodies may be a simple method for the evaluation of chronic airway disease.
To establish the optimum artificial light illumination method for baby leaf lettuce in closed plant factory system, the effects of red/blue light quality and short-term light quality conversion on growth and anthocyanin content were investigated. The growth of 'Hongha' lettuce was most favorable under red single wavelength LED light after 23 days of treatment, sequentially followed by the growth under red/blue mixed light, blue light, and fluorescent light. Total anthocyanin content in the mixed red/blue light (R57-B43) was 4.1-fold and 6.9-fold increased compared to the red LED and fluorescent light, respectively. With increasing the blue light ratio to 43%, the growth of lettuce was significantly decreased, while the relative chlorophyll content and Hunter's $a^*$ value was increased, indicating that the red/blue light ratio inversely affects on growth and anthocyanin pigment development. By changing light quality from red to red/blue mixed light source (R57-B43) for 9 days before harvest, the growth rate decreased compared to the continuous red light illumination, while the anthocyanin content dramatically increased compared to either red LED or fluorescent light. Whereas, when the light source was changed to red light, the growth rate was increased but anthocyanin content was reversely decreased. The result demonstrated that both growth and anthocyanin expression could be effectively regulated by shifting of light quality between red and red/blue mixed light source at a specific growth stage of lettuce in a plant factory.

Objective:In this study, we hypothesized that deficit in processing rapid linguistic stimuli is at the heart of Reading Disability(RD) and deficit in response inhibition is at the heart of Attention Deficit/Hyperactivity(ADHD). We conducted experiments to identify the core cognitive characteristics of children either with RD or with ADHD or with both, using attentional tasks and phoneme differential tests. Method:In the study 1, 28 children with ADHD, 16 children with RD+ADHD were individually administered visual/auditory performance tests. Then, the differences of performance on attentional tasks between two groups were compared while IQs of two groups were controlled. In the study 2, 13 children with RD+ADHD/RD, 13 children with ADHD, and 13 normal children were administered computerized phoneme differential tests. Result:Visual attentional tasks did not distinguish an ADHD group from a RD+ADHD group. With auditory attentional tasks, however, the comorbid group showed significantly more difficulties, causing a large variance in reaction time. RD, RD+ADHD, and ADHD groups showed more errors in phoneme differential tests than a normal control group, and each group showed distinctive performance patterns. Discussion:An ADHD group had difficulty in response inhibition and sustained attention, and children who also had RD along with ADHD magnified the auditory attentional difficulties. Even though children with RD had more trouble with responding correctly to target stimuli, their responses were not significantly different from those of children with ADHD.

The human skin is constantly exposed to environmental irritants such as ultraviolet, smoke, chemicals. Free radicals and reactive oxygen species (ROS) caused by these environmental facts play critical roles in cellular damage. These irritants are in themselves damaging to the skin structure but they also participate the immensely complex inflammatory reaction. The purpose of this study was to investigate the skin cell protective effect of Juniperus chinensis xylem extract on the UV and SLS-induced skin cell damages. We tested free radical and superoxide scavenging effect in vitro. We found that Juniperus chinensis xylem extracts had potent radical scavenging effect by 98% at 100 $\mu\textrm{g}$/mL. Fluorometric assays of the proteolytic activities of matrix metalloproteinase-l(MMP-1, collagenase) were performed using fluorescent collagen substrates. UV A induced MMP-1 synthesis and activity were analyzed by enzyme-linked immunosorbent assay (ELISA) and gelatin-based zymography in skin fibroblasts. The extract of Juniperus chinensis showed strong inhibitory effect on MMP-1 activities by 97% at 100 $\mu\textrm{g}$/mL and suppressed the UVA induced expression of MMP-1 by 79% at 25 $\mu\textrm{g}$/mL. This extract also showed strong inhibition on MMP-2 activity in UVA irradiated fibroblast by zymography. We also examined anti-inflammatory effects by the determination test of proinflammatory cytokine, interleukin 6 in HaCaT keratinocytes. In this test Juniperus chinensis decreased expression of interleukin 6 about 30%. Expression of prostaglandin E$_2$, (PGE$_2$) after UVB irradiation was measured by competitive enzyme immunoassay (EIA) using PGE$_2$ monoclonal antibody. At the concentrations of 5-50 $\mu\textrm{g}$/mL of the extracts, the production of PGE$_2$ by HaCaT keratinocytes (24 hours after 10 mJ/$\textrm{cm}^2$ UVB irradiation) was significantly inhibited in culture supernatants (p〈0.05). The viability of cultured HaCaT keratinocytes was significantly reduced at the doses of above 10 mJ/$\textrm{cm}^2$ of UVB irradiation, but the presence of these extracts improved cell viability comparing to control after UVB irradiation. We also investigated the protective effect of this extract in sodium lauryl sulfate (SLS)-induced irritant skin reactions from 24 hour exposure. Twice a day application of the extract for reducing local inflammation in human skin was done. Irritant reactions were assessed by various aspects of skin condition, that is, erythema (skin color reflectance) and transepidermal water loss (TEWL). After 5 days the extract was found to reduce SLS-induced skin erythema and improve barrier regeneration when compared to untreated symmetrical test site. In conclusion, our results suggest that Juniperus chinensis can be effectively used for the prevention of UV and SLS-induced adverse skin reactions such as radical production, inflammation and skin cell damage.

In this study, the extracts of Inula britannica var. chinensis (I. britannica) flower were extracted at three different temperatures (room temperature, $45^{\circ}C$, and $65^{\circ}C$) and their anti-aging effects were studied. Before investigating anti-aging effects of the extracts, their cytotoxicity was tested on B16F10, Hs683, and HaCaT cells. All extracts showed no cytotoxicity at the concentration less than 0.1% (v/v). Melanin synthesis inhibitory activities in B16F10 cells and reverse transcriptase polymerase chain reaction (RT-PCR) in Hs683 and HaCaT cells were used to see their anti-aging effects. The room temperature extract at 0.1% showed 24.5% melanin synthesis inhibition, which was better than the $45^{\circ}C$ and $65^{\circ}C$ extracts. In addition, expression rates of the room temperature extract at 0.1% on HAS-1, HAS-2, and HAS-3 related to hyaluronan synthase genes were 123.3%, 137.8%, 133.2%, respectively. which were higher than reference material of L-ascorbic acid. Expression rates of the $45^{\circ}C$ extract at 0.1% on TNF-${\alpha}$, COX-2, and IL-$1{\alpha}$, which are inflammatory related genes, was suppressed to 30.3%, 12.8%, 25.7%, respectively. It was better in anti-in flammatory effect than the room temperature and $65^{\circ}C$ extracts. As results, we showed that I. britannica var. chinensis flower extarcts decreased melanin production and expression of inflammatory related genes and increased the expression rate of hyaluronan synthase genes. Thus, it is believed that the extracts affect anti-aging effects of skin through whitening, moisturizing, and anti-inflammatory processes and could be applicable to cosmetics as a functional cosmetic ingredient.

The purpose of this study was to evaluate the microleakage of composite resin($Z250^{(R)}$) and compomer(Dyract AP) cured with various curing lights(Elipar $TriLight^{(R)}$, $Flipo^{(R)}$, and Elipar FreeLight $2^{(R)}$). Box-shaped class V cavities were prepared on the buccal and lingual surfaces of extracted human third molars. The sectioned specimens were evaluated for dye penetration and following results were obtained. 1. Regarding microleakage in the enamel margin with different curing light sources, there was no significant difference in Dyract AP whereas $Z250^{(R)}$ with $Flipo^{(R)}$ showed higher microleakage score than those of FreeLight $2^{(R)}$ or $TriLight^{(R)}$(p<0.05). 2. Regarding microleakage in the dentin margin with curing light sources, there were no significant differences in both Dyract AP and $Z250^{(R)}$(p>0.05). 3. Regarding microleakage in the enamel margin with filling materials. there were no significant differences in both filling materials with $Flipo^{(R)}$ whereas $Z250^{(R)}$ showed less microleakage than Dyract AP with FreeLight $2^{(R)}$ or $TriLight^{(R)}$ (p<0.05). 4. Regarding microleakage in the dentin margin with filling materials, there wer no significant differences in both filling materials with $TriLight^{(R)}$ whereas $Z250^{(R)}$ showed less microleakage than Dyract AP with FreeLight $2^{(R)}$ or $Flipo^{(R)}$ (p<0.05). 5. Enamel margin showed less microleakage than dentin margin when filled with Dyract AP and cured with $Flipo^{(R)}$ or FreeLight $2^{(R)}$ (p<0.05), but there were no statistically significant differences between the enamel and the dentin in the rest groups. This study suggested that $Z250^{(R)}$ showed lower microleakage score than Dyract AP. and that $Flipo^{(R)}$ showed higher microleakage score than FreeLight $2^{(R)}$ and $TriLight^{(R)}$ in the enamel margin filled with $Z250^{(R)}$.
This study investigates the physicochemical quality characteristics of tomato sauce added with different kinds of pimpinella brachycarpa. The results are as follows. The pH of tomato sauce added with freeze-dried pimpinella brachycarpa, in contrast to the tomato sauce with raw pimpinella brachycarpa, decreased as the addition quantity of the sample increased. As for sugar content, PBP0 with Pimpinella brachycarpa contained was shown to be the highest with 10.83. For chromaticity, all values (e.g, L, A and B) of tomato sauce added with pimpinella brachycarpa and with freeze-dried pimpinella brachycarpa decreased showing significant differences among the samples as the addition quantity of the samples increased. With increased addition quantity of the sample with freeze-dried Pimpinella brachycarpa, its water content decreased. PBP4, addition group with 4% of freeze-dried Pimpinella brachycarpa, was shown to be the highest DPPH free radical scavenging activity as 29.58. When adding Pimpinella brachycarpa to tomato sauce, a total number of micro-organisms decreased. In case of adding raw Pimpinella brachycarpa rather than freeze-dried Pimpinella brachycarpa, it created a further effect on an inhibitive action of growing and developing micro-organisms. In the preference of tomato sauce with Pimpinella brachycarpa, PBF2 with 2% of Pimpinella brachycarpa had the highest score as 5.1 for the color. For taste, 3.7, the lowest taste score, was shown in PBF1with 1% of Pimpinella brachycarpa. The overall preference revealed that PBF2 was the highest scoring 5.8.. PBF3 was scored highest with 5.9 for overall preference. From the comprehensive findings, 2~3% of the addition ratio between raw Pimpinella brachycarpa and freeze-dried Pimpinella brachycarpa in the production of tomato sauce is considered the most preferable.
Recently, there have been various lifeline installations constructed in the underground space of urban area due to the effective use of land. For newly installed lifelines or the management of the installed lifelines, many construction activities of excavation and backfilling are observed. Around these area, there are possibilities of collapse or excessive settlement due to the leaking of the pipe or unsatisfactory compaction of backfill material. Besides, construction costs can be saved since the on-site soils are used. The application of this liquidity filling material is not only to the lifeline installation but also to underpin the foundation under the vibrating machinery. On the evaluation of the applicability of this method to this circumstance, the strength should be investigated against the static load from the machine load as well as the vibration load from the activation of the machine. In this study, the applicability of the liquidity fill material on the foundation under the vibrating machinery is assessed via uniaxial compression and resonant column tests. The liquidity filling material consisting of the on-site soils with loess and kaolinite are tested to investigate the static and dynamic characteristics. Furthermore, the applicability of the reclaimed ash categorized as an industrial waste is evaluated for the recycle of the waste to the construction materials. The experimental results show that the shear modulus and 7 day uniaxial strength of the liquidity filling material mixed with reclaimed ash show higher than those with the on-site soils. However, the damping ratio does not show any tendency on the mixed materials.

The severity of carotid Intima-media thickness (IMT) is an Independent predictor of atherosclerosis which causes transient cerebral ischemia, stroke, and coronary events such as myocardial Infarction. The IMT consists of Intima thickness (IT) and media thickness (MT). However, the Individual clinical significance of IT and MT has not been well studied. We devised a method of measuring IT, MT, and IMT using B-mode ultrasound Image processing technique for the diagnosis of atherosclerosis. To inspect the clinical significance of IT, MT, and IMT, one hundred forty-four consecutive patients (mean age; 57 years old, 72 males) were underwent common carotid artery scanning using high-resolution ultrasound. Results showed that, the IT (p<0.05), MT (p<0.05) as well as IMT (p<0.01) of patients with atherosclerotic disease were significantly thicker than that of the patients without atherosclerotic disease. Patients with hyperiension showed significantly thicker IT (p<0.01), MT (p<0.001), and IMT (p<0.001). However, only IT was thicker in patients with smoking (p<0.01). The IT (r=0.374, p=0.001), MT (r=0.433, p=0.000), and IMT (r=0.479, p=0.000) showed positive correlation with age. The coefficients of determination ($r^2$) were estimated to be $92.4\%$ for IMT and MT, $49.1\%$ for IMT and IT, and $27.4\%$ for IT and MT. This result suggests that the Intima layer of the carotid artery has a different physiology with the media layer.
The job burnout of the private security guards who provide services through contact with customers can affect the overall the private security industry. So the managers need to make efforts to diagnose and evaluate the job burnout and provide continuous provision for the repatriation effect. However, up to the present, the job burnout measure for the private security guards has not been provided, so the objectivity of the information is questionable. For this reason, in order to establish the goodness of fit of the job burnout scale for private security guards based on the contents of the job burnout scale that was being provided to other service employees, this research aims to provide the scale to provide objective information compared with that of the past by applying the Rasch model of the item response theory. To achieve this purpose, the data on 379 private security guards working for the private security guard companies in the Gyeonggi region were collected, and the data were processed to suit the research purpose using SPSS 18.0 and AMOS 18.0, Winsteps programs. Through these research procedures, the following conclusion was obtained. With regard to the private security guards' measure of job burnout, as a result of factor analysis through the collected data, 4 questions were deleted, and as a result of the analysis of the goodness of the fit based on 18 questions, 4 questions were found to be inappropriate questions that did not satisfy the standard value, and the scope of the difficulty level of the questions from - 98 to 1.30 and the response reaction of a 5-point scale were found to suit the final private security guards' job burnout scale. In other words, the conclusion that it is appropriate to measure the private security guards job burnout scale with 3 factors and 13 questions was obtained. So if private security guards companies collect systematic data using the scale established in this research, and practice refresher training accordingly, they can reduce the private security guards' job burnout.
