Effects of Different Nitrogen Levels and Planting Densities on the Quality and Yield of the Black Rice Cultivar 'Shinnongheugchal' (재식밀도 및 시비량 차이가 신농흑찰 품질 및 수량에 미치는 영향)

  • The late-maturing black rice cultivar Shinnongheugchal from Jeollabuk-do Agricultural Research and Extension Service was used as the plant material for estimating growth characters, quality and yield from the vegetation period to harvest age. This study was performed to select an optimum combination of nitrogen level and planting density for the maximum yield of Shinnongheugchal. The plant height, number of tillers, and SPAD index were higher when the combination of 70 hills per $3.3m^2$ and 13 kg/10 a nitrogen level was used at 30 days after transplanting. The heading date for the combination of 70 hills per $3.3m^2$ and 15 kg/10 a nitrogen level, and 80 hills per $3.3m^2$ and 15 kg/10 a nitrogen level was August 22. The heading date for the other combinations was August 21. The combination of 70 hills per $3.3m^2$ and 13 kg/10 a nitrogen level yielded the highest number of tillers at 40 days after flowering. Even though the lodging index was increased with increasing nitrogen levels, field lodging did not occur until harvest time. Seed nitrogen concentration in the combination of 70 hills per $3.3m^2$ and 13 kg/10 a nitrogen level showed a significant difference when compared with the other combinations. The black rice yield varied significantly, and the highest yield was observed in the combination of 70 hills per $3.3m^2$ and 13 kg/10 a nitrogen level. The yield was significantly correlated with seed nitrogen concentration. The maximum yield was estimated to be 14.67 kg/10 a nitrogen level by using the regression equation. On average, the coloring degree of the black rice was higher at planting density of 70 hills per $3.3m^2$ than at 80 hills per $3.3m^2$. The highest yield of perfect black rice was obtained using the combination of 70 hills per $3.3m^2$ and 13 kg/10 a nitrogen level. Our findings demonstrate that a nitrogen level of 13-14 kg/10 a can be used to obtain the maximum yield from Shinnongheugchal with yield, cyanidin 3-glucoside content, and perfect black rice yield as the standard.

  • A 21-residue peptide corresponding to amino acids 84-104 of $p16^{INK4A}$, the tumor suppressor, has been synthesized and its structure was studied by Circular Dichroism, $^1H$ NMR spectroscopy and molecular modeling. A p16-derived peptide (84-104 amino acids) forming stable complex with CDK4 and CDK6 inhibits the ability of CDK4/6 to phosphorylate pRb in vitro, and blocks cell-cycle progression through G1/S phase as shown in the function of the full-length p16. Its NMR spectral data including NOEs, $^3J_{NH-H{\alpha}}$ coupling constants, $C_{\alpha}H$ chemical shift, the average amplitude of amide chemical shift oscillation and temperature coefficients indicate that the secondary structure of a p16-derived peptide is similar to that of the same region of full-length p16, which consists of helix-turn-helix structure. The 3-D distance geometry structure based on NOE-hased distance and torsion angle restraints is characterized by ${\gamma}$-turn conformation between residues $Gly^{89}-Leu^{91}$(${\varphi}_{i+1}=-79.8^{\circ}$, ${\varphi}_{i+1}=60.2^{\circ}$) as evidenced in a single crystal structure for the corresponding region of p18 or p19, but is undefined at both the N and C termini. This compact and rigid ${\gamma}$-turn region is considered to stabilize the structure of p16-derived peptide and serve as a site recognizing cyelin dependent kinase, and this well-defined ${\gamma}$-turn structure could be utilized for the design of anti-cancer drug candidates.

The Effect of Lattice Topology on Benzyl Alcohol Adsorption on Kaolinite Surfaces: Quantum Chemical Calculations of Mulliken Charges and Magnetic Shielding Tensor (캐올리나이트 규산염 층과 벤질알코올의 반응에 대한 양자화학계산에서 결정학적 위상이 멀리켄 전하와 자기 차폐 텐서에 미치는 영향)

  • In order to have better insights into adsorption of organic molecules on kaolinite surfaces, we performed quantum chemical calculations of interaction between three different model clusters of kaolinite siloxane surfaces and benzyl alcohol, with emphasis on the effect of size and lattice topology of the cluster on the variation of electron density and magnetic shielding tensor. Model cluster 1 is an ideal silicate tetrahedral surface that consists of 7 hexagonal rings, and model cluster 2 is composed of 7 ditrigonal siloxane rings with crystallographically distinct basal oxygen atoms in the cluster, and finally model cluster 3 has both tetrahedral and octahedral layers. The Mulliken charge analysis shows that siloxane surface of model cluster 3 undergoes the largest electron density transfer after the benzyl alcohol adsorption and that of model cluster 1 is apparently larger than that of model cluster 2. The difference of Mulliken charges of basal oxygen atoms before and after the adsorption is positively correlated with hydrogen bond strength. NMR chemical shielding tensor calculation of clusters without benryl alcohol shows that three different basal oxygen atoms (O3, O4, and O5) in model cluster 2 have the isotropic magnetic shielding tensor as $228.2{\pm}3.9,\;228.9{\pm}3.4,\;and\;222.3{\pm}3.0ppm$, respectively. After the adsorption, the difference of isotropic chemical shift varies from 1 to 5.5 ppm fer model cluster 1 and 2 while model cluster 2 apparently shows larger changes in isotropic chemical shift. The chemical shift of oxygen atoms is also positively correlated with electron density transfer. The current results show that the adsorption of benzyl alcohol on the kaolinite siloxane surfaces can largely be dominated by a weak hydrogen bonding and electrostatic force (charge-charge interaction) and demonstrate the importance of the cluster site and the lattice topology of surfaces on the adsorption behavior of the organic molecules on clay surfaces.

Dust Spatial Distribution and Emission of Tunnel Ventilated Windowless Broiler Building (터널환기 무창육계사의 분진크기별 분포와 배출농도에 관한 연구)

  • This study investigated the spatial distribution of dust originating from tunnel-ventilated windowless broiler building measuring 12 m wide, 61 m long, with a side wall height of 3 m and a capacity of 16,982 birds. Dust concentrations in terms of total suspended particles (TSP), and particulate matter of sizes $10\;{\mu}m$(PM10), $2.5\;{\mu}m$(PM2.5), and $1\;{\mu}m(PM1)$ were measured at 30 minutes interval by using GRIMM Aerosol Monitor (GRIMM AEROSOL). The spatial distribution of dust showed the lower dust concentration in the inlet than in the outlet of the tunnel ventilation, and dust concentration decreasing as the dust size decreased, as follows: $317.9\;{\mu}g/m^3$ TSP; $74.7{\mu}m/m^3$ PM10; $9.7\;{\mu}g/m^3$ PM2.5; and $6.2\;{\mu}g/m^3$ PM1 in the inlet; and $2,678.5\;{\mu}g/m^3$ TSP; $555.5\;{\mu}g/m^3$ PM10; $33.3\;{\mu}g/m^3$ PM2.5; and $10.2\;{\mu}g/m^3$ PM1 in the outlet. The dust concentration emitted from the tunnel ventilated fan was $446.6\;{\mu}g/m^3$ TSP; $129.1\;{\mu}g/m^3$ PM10; $15.8\;{\mu}g/m^3$ PM2.5; and $6.1\;{\mu}g/m^3$ PM1 in the 3 meters from the fan and $25.1\;{\mu}g/m^3$ TSP; $8.8\;{\mu}g/m^3$ PM10; $5.6\;{\mu}g/m^3$ PM2.5; and $4.9\;{\mu}g/m^3$ PM1 in the 50 meters from the fan.

  • The purpose of this study was to compare the shaping ability of the three $ProTaper^{(R)}$ instrumentation techniques in simulated canals. Thirty resin blocks were divided into 3 groups with 10 canals each. Each group was instrumented with manual $ProTaper^{(R)}$ (Group M), rotary $ProTaper^{(R)}$ (Group R), and hybrid technique (Group H). Canal preparation time was recorded. The images of pre- and post-instrumented root canals were scanned and superimposed. The amounts of canal deviation, total canal width, inner canal width, outer canal width and centering ratio were measured at apical 1, 2, 3, 4, 5 and 6 mm levels 1. Canal preparation time was the shortest in R group (p < 0.05). 2. The amounts of total canal width in R group was generally larger than the other groups, but no significant differences were observed except at the 1, 3 mm levels (p > 0.05) .3. The amounts of inner canal width in R group was larger than M group at the 1 mm level and H group was larger than R group at the 6 mm level (p < 0.05). The amounts of outer canal width in R group was larger than H group only at the 1 mm level (p < 0.05). 4. The direction of canal deviation in H, R group at the 1, 2, 3 mm levels was outward and that in M group at the 1, 2 mm levels was inward. The amounts of canal deviation in H group was larger than R group at the 6 mm level (p < 0.05). 5. The amounts of centering ratio in H group was larger than R group at the 6 mm level (p < 0.05).

Phosphorus Phases in the Surface Sediment of the South Sea (남해 표층 퇴적물에서의 인의 존재상)

  • To understand the role of shelf sediment in phosphorus biogeochemical cycle, we carried out sequential sediment extraction (SEDEX) of P and porewater analysis on 14 core samples collected in the South Sea of Korea, SEDEX classified P-pools into 5 phases and results are grouped into two categories: reactive P (loosely sorbed-P and Fe bound-P) and refractory P (detrital inorganic-p, authigenic mineral-P and organic-P). Total P concentrations are decreased with sediment depth in all samples as a result of dissolution to porewater. Reactive P comprises about $20\~50\%$ of total P, and iron bound-P is the major form consisting $70\~80\%$ of reactive P-pool. Iron bound-P decreases sharply with depth. Depth profiles of dissolved P concentration in porewater show mirror image of iron bound-P, revealing the role of FeOOH as a regulator of reactive P supply to overlying water column. Authigenic mineral-P consists less than $5\%$ of total P, thus removal of reactive P by converting into refractory P seems inefficient in shelf sediment. This implies that continental shelf sediment sequesters P temporarily rather than permanently. Results show local variation. Nakdong estuary receiving large amount of terrigenous input shows the highest concentration of total P and reactive P. Here iron oxyhydroxides at the surface sediment control the water column flux of P from sediment. Although total P content at the surface is comparable (500$\~$600 ${\mu}g{\cdot}g^{-1}$) between the South Sea and East China Sea, the former contains more iron bound-P and less derital inorganic-P than the latter. Reasons for the difference seem due in part to particle texture, and to biological productivity which depends roughly on the distance from land.

  • Relationship between distribution of tree species and salt concentration in soil was studied in order to understand the salt tolerance of tree species in the middle part of Korean east coast and its results were as follows; 1) The tree species in the area mostly consist of Pinus thunbergii, Pinus densiflora, Rosa rugosa, Lespedeza bicolor, Amorpha fruticosa, Quercus dentata, Rhododendron mucronulatum, Rohinia pseudoacacia and others. 2) Pinus thunbergii was dominant species and Rosa rugosa gradually disappeared at the distance from the beach line to 200 meters toward inland. Pinus thunbergii tended to disappear gradually and Pinus densiflora was dominant at the distance from 200 meters to 300 meters inland. 3) Pinus thunbergii was dominant below 50 meters in altitude while Pinus densiflora was dominant above 50 meters. 4) NaCl content tended to decrease as proceeding to inland, higher altitude and to shallow depth of soil. 5) Change in pH in terms of distance toward inland and altitude was not significant, but change in pH along soil depth was conspicuously decreasing from pH 7.0 of top soil to pH 6.5 50cm below. 6) Pinus thunbergii was densely distributed to the area where soil NaCl content was higher than 100 vpm, while Pinus densiflora dominated the area of less than 100 vpm. Rosa rugosa was shown to dominate the area of over 100 vpm soil NaCl content. 7) NaCl content in tree tended to decrease, as proceeding to inland, rapidly within 150 meters distance from the beach line and gradually at further distances. NaCl content in leaf was about 600 ppm, branch 350ppm and root 250 ppm.

Changes in Genetic Diversity of a Test Plantation of Liriodendron tulipifera L. by simulated Practices for Seed Trees (백합나무 시험림(試驗林)의 모의간벌(模擬間伐)에 따른 유전다양성(遺傳多樣性) 변화(變化))

  • Changes in genetic diversity after seed-tree practice were simulated in test plantation of tulip tree at Sokdal-ri, Banwol-myon, Hwasung-gun, Kyungki Provence. I-SSR markers were analyzed in a total of 305 tulip trees. A total of 89 amplicon variants were observed by PCR with 9 I-SSR primers. Genetic diversity for 305 trees was relatively high (S. I. =0.4532). Individuals originated from the seed orchards in U.S.A. showed the highest level of genetic diversity (0.4530), those from Anyang showed the medium level (0.4152), and those from Cheonbuk showed the lowest (0.3929). Simulation of seed-tree practice accompanied by 2 consecutive thinnings was performed on the basis of morphological characteristics and planted distances of the individual trees, which left 37 trees as candidates for seed-trees. Decreasing rate of genetic diversity within seed sources was greatest for individuals from Cheonbuk (28.3%), moderate for those from Anyang (16.3%), and smallest for those from U.S.A. (8.0%). In spite of little difference in decreasing rate of individuals for the 3 seed sources (87.5~88.2%), large difference in decreasing rate of genetic diversity within seed sources might be due to difference in number of mother trees for the 3 seed sources. For example, whereas individuals originated from the seed orchard in U.S.A., which showed the smallest decreasing rate of genetic diversity, might be originated from relatively large number of mother trees, those from Anyang and Cheonbuk might be originated from relatively small number of mother trees. Although mean of 17.5% of the genetic diversity within seed sources was decreased through 2 consecutive thinnings, a decrease in genetic diversity for the whole individuals (37 trees) was only 6.1%. This observation suggests that the seed-tree practice on the basis of the criteria established in the present study may not result in great reduction in overall genetic diversity of the progenies.

Genomic analysis of Mycobacterium fortuitum by pulsed-field gel electrophoresis (Pulsed-field Gel Electrophoresis를 이용한 Mycobacterium fortuitum의 유전형 분석)

  • Epidemiological studies are important in both the prevention and treatment of mycobacterial infections. This study was initiated to establish the pulsed-field gel electrophoresis (PFGE) method, which are not yet extensively studied. The most apprpriate restriction endonucleases included DraI, AsnI, and XbaI. The optimal PFGE condition was different according to the enzymes used. Two stage PFGE was performed, in case of DraI first stage was performed with 10 seconds of initial pulse and 15 seconds of final pulse, while the second stage was performed with 60 seconds of initial pulse and 70 seconds of final pulse. The electrophoresis time for DraI-PFGE was 14 hours for each stage. Electrophoresis was performed for 22 hours, in case of XbaI, with 3 seconds of initial pulse and 12 seconds of final pulse. Electrophoresis was performed for 22 hours, in case of AsnI, with 5 seconds of initial pulse and 25 seconds of final pulse. In all cases the voltage of the electrophoresis was maintained constantly at 200 voltage. Standard mycobacterial strains, which included Mycobacterium bovis BCG, M. tuberculosis, and M. fortuitum, could not be differentiated by PFGE analysis. PFGE analysis was performed to differentiate 9 clinically isolated M. fortuitum strains using AsnI. All M. fortuitum strains showed different genotypes except 2 strains. Cluster analysis divided M. fortuitum strains into 2 large groups. PFGE analysis was performed to further differentiate M. fortuitum isolates using XbaI. The undifferentiated 2 M. fortuitum strains showed different PFGE patterns with Xba I. Cluster analysis of the XbaI-PFGE patterns showed more complex grouping than AsnI-PFGE patterns, which showed that XbaI-PFGE analysis was better than AsnI-PFGE in M. fortuitum genotyping. The top dissimilarity values of AsnI-PFGE and XbaI-PFGE were 0.74 and 0.75, respectively. This value was higher than that of arbitrarily primed polymerase chain reaction (AP-PCR) analysis and lower than that of restriction fragment length polymorphism (RFLP) analysis. This suggested that PFGE can be used as a supportive or alternative genotyping method to RFLP analysis.

Fructification, Fruit Characteristics, and Yield According to Tree Age of Jujube Tree (Zizyphus jujuba var. inermis) 'Hwangsil' (대추나무 '황실'의 수령에 따른 결실 및 과실특성과 수확량)

  • The fructification characteristics, fruit quality, and yield of the 'Hwangsil' jujube tree were analyzed at various stages of growth age (3-8 years old) to obtain basic data for developing high-quality jujube production technology. The average height, crown area, stem diameter near the root, stem clear length, the number of the main branches, and the distance between any two main branches were 230.8 cm (224.4~247.2 cm), 3.0 m2 (2.1~3.8m2), 4.8 cm (2.4~6.2 cm), 69.1 cm (46.6~78.0 cm), 12.9 (8.6~19.6), and 8.1 cm (7.4~9.0 cm), respectively. Tree age was positively correlated with the crown area, stem diameter near the root, but stem clear length negatively correlated with the number of main branches. The average number of fruits per mother bearing shoot and tree was 20.0 (14.3~26.3) and 302.8 (257.3~373.5), respectively. There was no correlation between tree age and fructification characteristics, such as the number of fruit-bearing mother shoots per the main branch, the number of fruit-bearing shoots per fruit-bearing mother shoot, and the number of fruits per fruit-bearing shoot. Since the shape of the jujube tree is constantly managed based onthe growing area and type of greenhouse where the tree grown. The average fruit weight, fruit hardness, and soluble solid content were 24.2 g (22.4~26.8 g), 28.4 N (27.3~30.0 N), and 19.0% (17.1~19.8%), respectively, with no correlation between the tree age and fruit quality. The average yield was 7.4 kg per tree (5.7~9.1 kg), with significantly high quantities were produced in six and seven years old trees.
