Optimum Culture Conditions for the Growth of Spirulina platensis NIES 39 (Spirulina platensis NIES 39의 성장을 위한 최적배양조건)

  • Recently, as the interest in the accelerated global warming and the food shortage problem is increased, the concerns for microalgae as photosynthetic microorganisms are also increased. Specially, photosynthetic microalgae, Spriulina platensis have been an attractive source for $CO_2$ gas fixations and for a vast array of valuable nutritious compounds. In this paper, to culture the microalgal Spirulina platensis NIES 39 in a batch culture with high mass, optimal conditions for the culture temperature, initial pH, light intensity and concentration of carbon and nitrogen, were tested. At the most favorable culture condition, $35^{\circ}C$, initial pH 9.5, 4500 lux and carbon and nitrogen concentration of 16.8 g/L $NaHCO_3$ and 2.5 g/L $NaNO_3$, the excellent yields of 2.10 g/L biomass and 29.53 mg/L chlorophyll were obtained.

Production of Hydantoinase from Streptomyces sp. (방선균으로부터 Hydantoinase의 생산)

  • In order to investigate hydantoinase-producing strain of the genus Streptomyces, 523 strains of Streptomycee sp. isolated from soils were cultivated in various media and conversion activity of the enzyme was measured to DL-5-phenylhydantoin. A number of strains producing hydantoinase were detected and among them, the strain of Streptomyces sp. Y-183 was selected as a most powerful strain to producing the enzyme. The optimal culture conditions for the production of hydantoinase of the strain were studied, and it was found that almost all hydantoinase activity was produced in the cell fraction. The maximum activity of the enzyme, 17.8 unitstg of dried cells weight, was obtained when the strain was cultured at $30^{\circ}C$ for 72 hr in a medium containing 1.0% of glycerol. 0.5% of yeast extract. 0.5% of soytone, 0.5% of beef extract, 0.6% of KCI, 0.002% of $K_2HP0_4, 0.25% \;of \;CaC0_3, \;0.0002% \; of \; ZnSO_4, \; 0.0002%\; of\; FeS0_4$, and 0.4% of uracil as an inducer, and the pH of culture broth was adjusted ranging from 7.0 to 7.5.

  • To produce ${\gamma}$-linolenic acid (GLA) by cell cultures of Lithospermum erythrorhizon, we optimized medium compositions including carbon sources, nitrogen sources and growth regulators. MS basal medium supplemented with 1.0 mg/L 2, 4-D was effective for callus induction from mesophyll tissue. Addition of sucrose at 88mM concentration induced active proliferation of suspension cells and increased GLA content. Increased supplement of potassium nitrate as nitrogen source resulted in proliferous cell growth and increased total fatty acid content Abscisic acid increased cell growth and fatty acid content in callus culture, whereas as it had an inhibitory effect in suspension cell culture.

Production of Single Cell Lipid and Treatment of Wastewater Alcohol Manufactory (알코올 발효공장의 폐수처리를 겸한 단세포지질의 생산)

  • In order to reduce BOD of wastewater from alcohol distillery manufactory, the production of single cell lipid was attempted. Among five yeast strains tested, Rhodotorula glutinis was most desirable for lipid production. Wastewater was treated with 2N NaOH and used as a medium. The optimum pH and temperature for lipid production were found to be 5.0 and $30^{\circ}C$, respectively. The addition of monobasic phosphate was good for cultivation of Rhodotorula glutinis. The C/N ratio was an important factor for lipid production and composition. The best C/N ratio was 50 for the production of single cell lipid. By cultivation Rhodctorula glutinis for 4 days, 4g/L of single cell lipid was harvested and BOD of wastewater reduced by 88.7%.

  • Agrobacterium tumefaciens KU12 isolated from Korea is able to induce tumors on various plants and catabolize octopine as a sole carbon and nitrogen source. A, tumefaciens KU12 contains three plasmids. Their sizes are 45.5 kb. 240 kb. and > 240 kb. respectively. For the purpose of identification of octopine type Ti plasmid, avirulent A, tumefacients A136 is transformed with plasmids isolated from KU12 by direct transformation. Transformants containing Ti plasmid were grown on AB medium containing octopine as a sole nitrogen source. The isolated strain, named KU911, contains only 240 kb plasmid. As a result of induction of crown gall and Southern hybridization with other octopine Ti plasmid pTiAch5, 240 kb plasmid named pTiKU12 was Ti plasmid.

  • Agrocybe aegerita is Hymenomycetes fungus belonging to the order Agaricales and family of Bolbitiaceae. Much less is known about liquid culture of Agrocybe aegerita. Thus, the present study was to investigate the liquid cultural characteristics of Agrocybe aegerita my-celium. The optimal medium for the mycelial growth and density was ME medium, optimal tem-perature and initial pH were 25$\pm$1$^{\circ}C$ and 5.5, respectively. And optimal culture time for mycelial growth was 12 days. The modified optimal medium compositions were dextrin 3% (w/v), yeast extract 2% (w/w), MgSO$_4$0.05% (w/v), and KH$_2$PO$_4$0.15% (w/v). Under optimal culture conditions, the mycelial growth of modified optimal medium was higher than that of ME medium.

  • Productivity of bacterial cellulose by Acetobacter xylinum GS11 was investigated in the several culture conditions. In various carbon sources, others with the exception of glucose were not found to be effective for cellulose production, and 2% was better in yield than other concentration of glucose. Yeast extract and soytone among several organic nitrogens were effective, but inorganic nitrogen sources tested were not efficient for cellulose production by A. xylinum GS11. The effects of various inorganic salts, amino acids and vitamins were also investigated: $MgSO_4$, phenylalanine and $\alpha$-tocopherol gave the cellulose yield of 1.5, 1.4 and 1.4 fold, respectively, compared with basal medium. In our experiment, cellulose production by A. xylinum GS11 added with 10% coconut milk and 0.5% lignosulfonate in basal medium, was the most efficient among the several material sources employed here, and these were 2.2 and 2.1 fold, respectively.

  • This experiment was conducted to study about nutrient absorption of flag leaf and chaff of rice plant different transplanting date with elevations. Heading stage was delayed by destructive cold temperature or late transplanting. Plant analyses revealed that above poor plants also had higher total nitrogen content, but ower silicate in the flag leaf and sterile chaff. Total nitrogen and silica contents to accumulated in flag leaf between yield was significant high correlation. The chaff of late transplanting and sterility was high total nitrogen. Relationship between silicate absorption and total nitrogen of chaff was significant high correlation. Ripening temperature after heading stage was influenced total nitrogen of chaff. Phosphate, potassium, calssium and silicate contents of chaff increased hight ripening temperature but total nitrogen decreased. Therefore, inorganic element contents of chaff was closely connected with grain sterility.

  • This study was conducted to investigate optimum conditions for mass production of antagonistic microbes Pseudomonas azotoformans HC5. P. azotoformans HC5 is a potent biological control agent to control brown blotch disease caused by Pseudomonas tolaasii. This markedly showed the antagonistic activity against P. tolaasii, the most destructive pathogen of cultivated mushrooms. To define the optimum conditions for the mass production of the P. azotoformans HC5, we have investigated optimum culture conditions and effects of various nutrient source on the bacterial growth. The optimum initial pH and temperature were determined as pH 6.0 and $15^{\circ}C$, respectively. The optimal concentration of medium elements for the growth of pathogen inhibitor bacterium was determined as follows: 0.6% adonitol, 1.5% yeast extract, 0.8% $NH_4H_2PO_4$, 5mM $MgSO_4$, and 0.2% asparagine.
