Analysis of Microbial Community Structure in Soil and Crop Root System II. Analysis of soil microbial community structure in different soil Environmental conditions by MIDI and DNA analyses (토양과 작물근계의 미생물군집 구조 해석 II. MIDI 및 DNA 분석에 의한 토양환경별 미생물 군집 해석)

  • To evaluate the correlations of microbial populations with soil healthiness and crop production and establish the criteria for microbial population of soil types. We analyzed the microbial community structure of 13 soils which were different in physical and chemical properties and cultivation methods. According to the analysis of microbial population suing the dilution plate method, the large differences of the microbial population structures among soil types were shown: aerobic bacteria $2-27{\times}10^6$, fluorescent Pseudomonas $1-1,364{\times}10^5$, Gram negative bacteria $1-126{\times}10^4$, and mesophilic Bacillus $1-110{\times}10^5$. The density of Gram negative bacteria was highest on red pepper cultivating soils (sample no. 4 and 6) of Umsung and Gesan, Chungbuk, and the density of the fluorescent Pseudomonas was highest on greenhouse soil (sample no. 7) of Jinju, Kyungnam. The crop productivity of three soils was high as compared with those of other soils. It was supposed that the density of fluorescent Pseudomonas and mesophilic Bacillus were correlated with the incresed crop production. By MIDI analysis, 579 strains isolated from 13 soils composed of a variety of microbes including 102 isolates of Agrobacterium, 112 isolates of Bacillus, 32 isolates of Pseudomonas, 44 isolates of Kocuria, and 34 isolates of Pseudomonas. Among the 624 isolates of Gram negative bacteria, Pseudomonas including P. putida and p. fluorescens occupied the highest density (51%), and Stenotrophomonas maltophilia and Burkholderia cepacia also appeared at high density. From RAPD analysis, the fluorescent Pseudomonas strains isolated from 13 soil types showed a high level of strain diversities and were grouped into 2 - 14 patterns according to soil types. Many of unknown bacteria were recovered from the paddy soil, and needed to be further characterized on the molecular basis.

  • The ultimate research goal of the current study was a development of hydrolyzed whey protein powder (7%-GNANA) manufactured with normal content of sialic acid, a marker compound, that is naturally occurring at 7% concentration in GMP obtained from the milk protein. GMP is a safe food, used worldwide in infant and baby foods, etc. The test substance was prepared using (7% sialic acid containing) GMP as a raw material, and then using alcalase, an enzyme approved as a food additive, after separation of sialic acid with 100% efficiency and 7%-GNANA (containing 7% sialic acid and protein; product name: HELICOBACTROL-7) provided by MEDINUTROL Inc. (Korea). Bacterial reverse mutation (Ames) test was conducted in accordance with GLP Guideline using the test substance specified above. To identify its mutagenic potential against microorganisms, histidine auxotrophic strains of Salmonella Typhimurium, TA98, TA100, TA1535, and TA1537, and tryptophan auxotrophic strain of Escherichia coli, WP2uvrA, were used. The bacterial reverse mutation (Ames) test was performed by dividing the test substances into five different concentration groups (0, 61.7, 185, 556, 1,670, $5,000{\mu}g/plate$). Results of this experiment did not reveal repetitive increase of colony generating values or positive criteria for reverse mutagenicity for any concentration of test substances in any of the five strains, regardless of the presence of a metabolic activation system, and no dose-dependency was identified. In conclusion, the safety of 7%-GNANA test substance was verified by bacterial reverse mutation test conducted before registration of 7%-GNANA as a food additive.

Timber Harvesting Impacts on Soil Respiration Rate and Microbial Population of Populus tremuloides Michx. Stands on Two Contrasting Soils (두 가지 서로 다른 토양에 형성된 Populus tremuloides Michx. 임분의 수확이 토양호흡률 및 토양미생물상에 미치는 영향)

  • Timber harvesting impacts on soil microbial populations and respiration rates were examined in naturally regenerating trembling aspen(Populus tremuloides Michx.) stands on two contrasting soils, an Omega loamy sand (sandy mixed, frigid Typic Udipsamment) and an Ontonagon clay loam (very fine, mixed Glossic Eutroboralf). Five timber harvesting disturbances were simulated during winter of 1990 and spring of 1991, including commercial whole-tree harvesting(CWH), winter logging trail+CWH, logging slash removal+CWH(LSR), forest floor removal+LSR(FFR), and spring compaction+FFR. Regardless of soil types, total soil respiration rates of each stand decreased slightly or remained the same after harvesting while microbial population increased progressively during the first two years following harvesting. Microbial populations increased more rapidly and constantly at the sandy site than at the clayey site, which may indicate that the soil physical and chemical conditions changed more drastically for microbial activity following timber harvesting at the sandy site than at the clayey site. However, two kinds of treatment applications-three levels of organic matter removal and two levels of compaction-did not result in significant differences in microbial population or total soil respiration rate at each site during the first two post-harvest years. Total soil respiration of the aspen stands, sum of root respiration and microbial respiration, was a poor index for the microbial activity in this study because aspen kept an active root system for the successive root-sprouts even after harvesting, which resulted in a large portion of root respiration in total respiration.

Detection of Waterborne Pathogens in Public Bath Houses by PCR-Reverse Blot Hybridization Assay (PCR-REBA) (분자생물학적 방법인 PCR-REBA를 이용한 대중목욕탕 수질 중 수인성병원성미생물 검출)

  • Contamination of public bath water by waterborne pathogens can cause disease outbreaks and contribute to background rates of disease. The aim of this study is to determine the prevalence of waterborne pathogens in public baths. A total of 30 water samples were collected from 30 different public baths in seoul, Korea. Pathogens in water samples were concentrated by 0.45 ${\mu}m$ nitrocellulose membrane filter, analyzed by both cultivation and polymerase chain reaction-reverse blot hybridization (PCR-REBA) of partial 16S rRNA gene. Various microorganisms including Escherichia coli and Shigella spp. were identified by microbiological cultivation. E. coli, Shigella spp., Salmonella spp., Pseudomonas spp. and Mycobacterium spp. were identified by PCR-REBA. Our results suggest that appropriate hygiene practice and continuous monitoring is needed for reducing health risk associated with public bath houses.

Size-dependent Toxicity of Metal Oxide Particles on the Soil Microbial Community and Growth of Zea Mays (산화 금속 입자 크기가 옥수수의 성장과 토양 미생물 군집에 미치는 독성)

  • This study investigated soil microbial community and growth of Zea mays to compare the toxicity of nano and micro-sized Cu and Zn oxide particles in microcosm system. In the presence of nanoparticles, biomass of Zea mays reduced by 30% compared with micro-sized particles and inhibited growth. Dehydrogenase activity was inhibited by CuO nano although it was increased by ZnO nano particles. According to the Biolog test, the microbial diversity was decreased after exposed to CuO nanoparticles and ZnO microparticles. Therefore, though it is widely recognized that nanoparticles are more harmful than microparticles, we can conclude that the diversity of microbial community does not always influenced by the size of particles of nano and micro.

  • In this study, we determined the changes in microbial numbers in solar salts according to the manufacturing process and storage duration. The salt samples were harvested from salt farms in Shinan (area 2) and Yeonggwang (area 1). They were serially diluted ten-fold and then placed on 4 kinds of cultivable media (mannitol salt agar, eosin methylene blue, plate count agar, and trypticase soy agar). After incubation, we obtained 62 halophilic isolates from the salt samples. Coliform and general bacteria were not detected in all salt samples. By 16S rRNA sequencing analysis, we found 12 kinds of halophilic bacteria belonging to the genera Halobacillus, Halomonas, Bacillus, Idiomarina, Marinobacter, Pseudoalteromonas, Vibrio, Salinivibrio, Virgibacillus, Alteromonas, Staphylococcus and some un-known stains. In our study, we discovered two novel species that have a 16S rDNA sequence similarity below 97%.

열대식물 (파푸아 뉴기니아산)의 항암작용연구

  • 해를 거듭할수록 증가추세에 있는 악성종양은 우리나라의 3대 사망원인 질환의 하나이다. 그러나 현재 암의 화학적 치료법은 40여종의 암치료제가 주종을 이루고 있으나 그 효과면에서 한계점에 도달하고 있어 식물추출물, 해양식물, 미생물 등으로부터 얻어지는 천연물 성분에서 강력하고 부작용이 적은 항암제를 발굴하기 위한 연구가 국내외에서 많이 수행되고 있다. 본 연구는 파푸아뉴기니아산 열대식물 13과 23종의 항암작용을 검색할 목적으로 실시하였는데, 실험동물은 ICR계의 암컷생쥐를 이용하였으며 Sarcoma 180 동물복수암세포에 대한 열대 식물 엑스 투여에 의한 수명 연장 효과를 측정함으로써 항암작용의 지표로 삼았다. 항암작용의 지표는 ILS(Increased Life Span)를 사용하였으며 ILS가 20%이상인 것을 항암성이 있는 것으로 간주하였다. 실험의 결과 Apocynaceae에 속하는 Plumiera rubra의 줄기껍질 및 잎 추출물 Leguminosae과의 Pterocarpus indicus의 잎 추출물과, 같은 과의 Derris trifoliata, Moraceae과의 Artocarpus altilis의 뿌리 및 줄기 껍질, 잎 추출물과 Sterculiaceae과의 Kleinhovia hospita의 잎 추출물, Verbenaceae과의 Vitex trifolia의 5과 5종 8시료가 의의있는 항암작용이 있는 것으로 나타났다.

Monitoring of Indicator Microorganism Concentrations of River Sediment and Surface Water in the Geum River Basin (금강 수계 내 하천퇴적물 및 지표수의 지표미생물 농도분포)

  • Characterization of sediment quality is important for the proper management of surface water quality, yet sediment has not been monitored sufficiently. In this study, fecal indicator microorganism concentrations of sediments in the Geum River Basin were monitored. Sampling was carried out at one paddy field, one lakeshore and five monitoring stations in the lower reach of the Geum River Basin. Surface waters and sediments were sampled four times during rainy season. Total coliform concentrations of sediments were 12 times higher in average to those of surface waters while E. coli concentrations of sediments were six times higher. No correlation found between indicator microorganism concentration between surface waters and sediments.

Analysis of Indicator Microorganism Concentration in the Rice Cultural Plot after Reclaimed Water Irrigation (하수처리수 관개후 벼재배 시험구에서 지표미생물 거동 분석)

  • A study was performed to examine the effects of UV-disinfected reclaimed water on microorganism concentration during rice culture. Four treatments were used and each one was triplicated to evaluate the changes of microorganism concentrations: stream water irrigation (STR), biofilter effluent irrigation (BE), UV-disinfected water irrigation with dose of 6 mW ${\cdot}$ s $cm{-2}$ (UV-6), and UV-disinfected water irrigation with dose of 16 mW ${\cdot}$ s $cm{-2}$ (UV-16). The indicator microorganisms of interest were total coliform (TC), fecal coliform (FC), and E. coli. The biofilter effluent from 16-unit apartment sewage treatment plant was used as reclaimed water and flowthrough type UV-disinfection system was used. Concentrations of indicator microorganisms in the treatment plots ranged from $10^2$ to $10^5$ MPN/100 mL during 24 hours after irrigation in May and June, where initial irrigation water for transplanting reparation was biofilter-effluent without UV-disinfection. It implies that initial irrigation using only non-disinfected reclaimed water for puddling in paddy field can be health-concerned because of more chance of farmer's physical contact with elevated concentration of microorganisms. The concentrations of microorganisms varied widely with rainfall, and treatments using UV-disinfected water irrigation showed significantly lower concentrations than others and their levels were within the range of paddy rice field with normal surface water irrigation. The mean concentrations of STR and BE during growing season were in the range of 4 ${\times}\;10^3$ MPN/100 mL for TC, and 2${\times}\;10^3$ MPN/100 mL for FC and E, Coli, While mean concentrations of UV-S and UV-lS were less than 1${\times}\;10^3$ MPN/100 mL for all the indicator microorganisms. Overall, UV-disinfection was thought to be feasible and practical alternative for agricultural reuse of secondary level effluent in Korea.

Effects of Organic Materials on Soil Organisms in a Korean Ginseng Field (인삼재배지 유기물 시용이 토양미생물과 미소동물에 미치는 영향)

  • The aim of this study is to evaluate the effects of organic materials on soil organisms. Changes in the community structure, and population density of soil organisms (microbes, nematodes, and microarthropods) were studied in a Korean ginseng field. Phospholipid fatty acids analysis showed that the relative abundances of bacteria, fungi, and actinomycetes did not differ significantly. The aerobes/anaerobes ratio was the lowest in soils amended with leaf mold, indicating that the decomposition speed was slow. Further, the addition of leaf mold to the soil enhanced the saturated/monounsaturated fatty acid ratio and cyclopropyl fatty acid/precursor ratio, which indicated an increase in environmental stresses. Application of pig manure compost (PMC) had positive effects on the population density of nematodes, and negative effects on that of oribatid mites. The population densities of nematodes, and microarthropods remained relatively low in the plots that had been treated with leaf mold or pig manure compost. It is suggested that pre-planting soil management directed at enhancing the biological decomposition efficiency should be continued over a long period to increase the soil bioactivity in virgin soils.
