A Study on the Stability of Langmuir-Blodgett Films Mixed with Myristic Acid and Stearic Acid (미리스트산과 스테아르산 혼합 LB막의 안정성에 관한 연구)

  • We were investigated by cyclic voltammetry to the stability through the electrochemical characteristics of Langmuir-Blodgett films mixed with myristic acid and stearic acid. Fatty acid mixture monolayer LB films was deposited by the LB method on the indium tin oxide(ITO) glass. The electrochemical properties was measured by cyclic voltammetry with a three-electrode system in 0.01 N $NaClO_4$ solution. The measuring range is continuously oxidized to 1650 mV, with an initial potential of -1350 mV was reduced. Scanning rates of 50, 100, 150, 200, and 250 mV/s was set. As a result, LB monolayer films of fatty acid mixture was appeared on irreversible processes by the oxidation current from the cyclic voltammogram. Diffusion coefficient (D) of fatty acid mixture was calculated $7.9{\times}10^{-2}cm^2s^{-1}$ at 0.01 N $NaClO_4$ solution.

The Effect of Acyl Chain Structure on the Hydrolysis of Fatty Acids from Fish Oil by Lipase-OF 360,000 (아실체인 구조가 효소 Lipase-OF 360,000에 의한 지방산의 가수분해 특성에 미치는 영향)

  • The hydrolysis characteristics of each fatty acid composing the fish oil by means of the lipase from Candida cylindracea was investigated. The saturated fatty acids, C14:0, C16:0 and C18:0, and the unsaturated fatty acids with one double bond, C16:1, C18:1(n-7), C18:(n-9), C20:1 and C22:1 were more easily hydrolyzed than the $\omega$-3 polyunsaturated fatty acids. when the number of carbon of the $\omega$-3 fatty acids was same but that of double bond was different, the hydrolysis of the $\omega$-3 fatty acids having lower number of double bond was more rapidly carried out. When the degree of polyunsaturation was same but the number of carbon was different, the lipase acted more rapidly upon the $\omega$-3 fatty acids with lower number of carbon. Docosahexaenoic acid(DHA) was most highly concentrated in the glyceride mixture after 120hr hydrolysis among the various $\omega$-3 polyunsaturated fatty acids. The weight percentage of DHA to the $\omega$-3 fatty acids in the fish oil was 31.87% but that in the glyceride mixture after 120hr hydrolysis was 51.89%.

  • Sugar esters are produced by transesterification of fatty acids and sugars in organic solvent media. Chromatographic separation of sugar esters and fatty acids was investigated. $C_{18}$ column($3.9{\times}300mm$) was used in the chromatography experiment. The effect of flow rate, column temperature, and sample loading on the charateristics of chromatogram was studied. Sugar esters and fatty acids tested in this study was successfully separated with a high resolution.

  • The hydrolysis characteristics of various fatty acids composing the fish oil was investigated for function of acyl chain specificities using Lipase-OF 360,000 from Candida cylindracea. The hydrolysis of fatty acids decreased with the increase of the number of carbon and double bond in the fatty acids, in case that the number of double bond and the position of the first double bond from the methyl group of fatty acids were the same. The position of the first double bond was found to be an acyl chain specificity of Lipase-OF 360,000 for the hydrolysis of fish oil. Lipase-OF 360,000 also showed the another acyl chain specificity that the increase of double bond of fatty acids, having the same number of carbon and the position of double bond, brought about the decrease of hydrolysis.

사과가 콜레스테롤을 급여한 흰 쥐의 콜레스테롤 대사 및 항산화 보호인자에 미치는 효과

  • 사과급여가 지질대사에 미치는 효과를 40마리의 Wistar rats에 0.3% 콜레스테롤을 첨가한 반정제식이를 급여하여 조사하였다. 사과첨가군는 일반식이중 전분함량(15%)을 동결건조시킨 사과(Gala 품종)로 대체하였으며, 대조군은 사과에서 유래되는 당함량을 대신하여 전분중 13%를 과당/포도당/ 전분의 혼합물로 대체하였다. 실험식이에서 지방원은 옥배유 사용하였으며, 식이중 비타민E는 요구량의 1/3정도 첨가하였다. 실험쥐는21일간의 적응기간을 걸쳐 선별하였다. 사과급여군에서, 사과에서 유래되는 식이섬유소량이 소량(약 2%)임에도 불구하고 혈장내 콜레스테롤이 유의적으로 낮아지는 경향을 보였으나, 콜레스테롤 첨가군에서는 간에 콜레스테롤 에스테르가 축적되는 결과를 보였다. 사과급여군에서 지단백질 형상이 크게 다르게 나타났는데, 이는 중성지방이 많은 지단백질(TGRLP) 콜레스테롤이 감소되고 HDL 콜레스테롤은 증가되는 결과를 보여 심장관련질환 예방에 효과가 있을 것으로 추정되었다. 또한, 사과가 콜레스테롤 흡수를 크게 저하시키는 것으로 추정되나 담즙산의 분해에는 영향을 주지 않는 것으로 나타났다. 이와 같은 결과로 사과식이군이 대조군에 비해 혈장 FRAP가 높고 뇨중의 MDA 추출물이 감소하는 경향을 보여 산화억제효과도 있는 것으로 나타났다. 따라서 후식으로 적당량의 사과를 섭식하는 것이 지질과 과산화성 인자에 바람직한 영향을 주는 것으로 나타났다.

Comparison of Solid Fat Index and Triacylglycerol Composition of the Blends from Natural Vegetable Fats and Palm Stearin Fraction (천연 식물고체지와 팜스테아린 분별유 혼합물의 Solid Fat Index 및 Triacylglycerol 조성 비교)

  • Palmitoyl-oleoyl-oleoyl (POO) and palmitoyl-oleoyl-palmitoyl triacylglycerol rich fraction (PSL) was obtained from the acetone fractionation of palm stearin. The fatty acid composition (total and positional), tri-acylglycerol species, and solid fat index (SFI) were compared among the blends of natural vegetable fats (sal fat, illipe fat, kokum fat, shea stearin fat, and shea butter) and PSL with different ratios (50:50, 60:40, 65:35, 70:30). In total fatty acid composition of PSL, palmitic, oleic, and linoleic acids were the major fatty acids, whereas in natural vegetable fats stearic and palmitic acids were the major fatty acids. Moreover, oleic acid was a major fatty acid at sn-2 position in sal fat, illipe fat, and kokum fat. The TAG species was analyzed by reversed-phase HPLC, from which the PN value ranged from 46 to 54. When natural vegetable fats and PSL were blended with different ratios, decreasing the amount of PSL resulted in increasing SFI in most cases. Among blends, the SFI of sal fat and PSL were most similar to commercial cocoa butter equivalent (CBE).

Proximate Composition and Microbial Content Change of Broiler Waste Silage by Mixing with Wheat Bran and Oven-drying (닭폐기 부산물 Silage와 소맥피 혼합 및 오븐건조에 따른 일반성분과 미생물 총균수 변화)

  • Broiler processing waste(offal) was homogenized and treated with the combination of acids. The offal was autolyzed(ensiled) at $25^{\circ}C$ for 72 hrs and analyzed for pH and fatty acid profile. The proximate composition and microbial content change of the autolyzed offal by mixing with wheat bran and ovendrying were evaluated. The initial pH value of the homogenized offal, 6.52 came down to 2.75 within 5 min after acidification and increased silightly to $3.06{\sim}2.92$ during autolysis. The proximate composition and fatty acid profile of the autolyzed offal were not substantially different from the unautoylzed offal. However, the log CFU(colony forming units)/g of total plate counts and fungal counts decreased from 7.45 and 7.11 to 3.39 and 2.03 after autolysis, respectively.

  • Fish oil was hydrolyzed with Aspergillus oryzae lipase, Lipolase-100T. The hydrolysis characteristics of Lipolsae-100T were investigated. Lipolase-100T showed 1,3-positional specificity which hydrolyzed acyl chains combined on the 1 or 3 position of triglyceride into free fatty acids. Lipolase-100T represented another property that the saturated fatty acids composing the triglyceride were hydrolyzed more easily that the polyunsaturated fatty acids(PUFAs). n-3 PUFAs, such as C16:4, C20:5 and C22:6, were hardly hydrolyzed, so that the concentrations of those in the mixture of glycerides were increased according to hydrolysis time. Especially docosahexaenoic acid(DHA), C22:6 showed the highest increase in the concentration. This result explained that n-3 PUFAs were combined on 2-position of triglyceride. When the hydrolysis of fish oil with Lipolase-100T 0.4 wt% was performed for 120 hr, n-3 PUFAs wt% was increased to 50 wt% in the mixture of glycerides. This result was obtained due to the 1,3-positional specificity of Lipolase-100T and positional specificity of n-3 PUFAs.

  • Antioxidative characteristics of freeze dried soybean sauce powder (SSP) on the oxidation reaction of linoleic acid mixture(LA) were evaluated by the determinations of peroxide formation, synergistic property, hydrogen donation and lipoxygenase activity. SSP was found to possess a considerable potentiality of antioxidant activity on the formation of hydroperoxides in the LA oxidation reaction system at $50^{\circ}C$ for 144hrs. This antioxidative effect was increased by the concentration from 0.02% to 0.5% of SSP in the reaction system. Under the condition of presence of ferric chloride (10 ppm) in the reaction system. appreciable effect of SSP on the synergistic antioxidation were observed. On the other hand, hydrogen donation property of SSP onto ${\alpha},\;{\alpha}'-diphenyl-{\beta}-picrylhydrazyl$ was found and inhibitory ability of SSP on LA oxidation was also shown in the reaction system of lipoxygenase-catalized oxidation.

A Study on the Photoisomerization and Physical Properties for Mixtures of Phospholipid and Fatty Acid containing Azobenzene (아조벤젠기를 가지는 지방산과 인지질의 혼합물에 대한 광이성화 현상 및 물성에 관한 연구)

  • We carried out this subject to observe photoisomerization using 8A5H and phospholipid(DLPC) containing azobenzene group which has reversible to cis-trans by light irradiation. We investigated the photoisomerization and property of the monolayer film on quartz substrate prepared by the mixture of 8A5H and phospholipid deposited using Langmuir-Blodgett(LB) method. It was found that the absorption spectrum of the 8A5H and phospholipid LB monolayer was induced to photoisomerization by alternate light irradiation. And the absorbance of both the solution of mixtures of 8A5H-phospholipid and LB film was reversibly changed by the acid-base exposure and alternate temperatures.
