Obesity Regulation through Gut Microbiota Modulation and Adipose Tissue Browning (장내 미생물의 조절과 지방세포의 갈색지방화를 통한 비만 조절 연구)

  • Obesity, represented by abnormal fat accumulation due to an imbalance between energy intake and expenditure, is a major public health issue worldwide, leading to multiple noncommunicable diseases, including atherosclerosis, hypertension, type 2 diabetes, and cancer. Diverse solutions have been proposed to combat obesity. Attention has focused on two types of adipose tissues as a promising therapeutic target in obesity: traditional brown and beige or brite. Unlike energy-storing white adipose (endocrine) tissue, traditional brown adipose tissue and beige adipose tissue have energy-dissipating thermogenic properties. Both types of tissue are present in adult humans and inducible through external stimuli, such as cold exposure, ${\beta}3$-adrenergic receptor agonists, and phytochemicals. Among these stimuli, microbiota present in the human intestinal tract participate in multiple metabolic activities. Modulation of gut microbiota may offer a potent and possibly curative strategy against various metabolic diseases. Numerous studies have focused on the effects of established antiobesity treatments on the gut microenvironment or brown-adipose-tissue activation. In this review, we focus mainly on stimuli known to alleviate obesity, weight gain, and metabolic diseases, in addition to known and possible inter-relations between gut microbiota modulation and similar interventions and adipose tissue browning. The findings may pave the way toward new strategies against obesity.

Oral Administration of Weissella confusa WIKIM51 Reduces Body Fat Mass by Modulating Lipid Biosynthesis and Energy Expenditure in Diet-Induced Obese Mice (생쥐 비만모델에서 Weissella confusa WIKIM51 식이에 따른 지방합성 및 에너지 대사 조절로 인한 체지방 감소 효과)

  • Obesity is closely associated with profound dyslipidemia, insulin resistance, and fatty liver disease. Recent reports have suggested that alterations in gut microbiota can be linked to diet-induced obesity. In this study, the anti-obesity effects of Weissella confusa WIKIM51 isolated from kimchi were investigated, as evidenced by: i) reduced lipid accumulation and downregulated adipogenesis-related genes in 3T3-L1 adipocytes; ii) suppressed gains in body weight and epididymal fat mass; iii) reduced serum lipid levels, for example, triglyceride and total cholesterol; iv) increased serum adiponectin levels and reduced serum leptin levels; v) downregulated lipogenesis and upregulated β-oxidation-related genes in the epididymal fat; and vi) altered microbial communities. The collective evidence indicate the potential value of W. confusa WIKIM51 as a functional food supplement for the prevention and amelioration of obesity.

Effects of High Stocking Density on the Expressions of Stress and Lipid Metabolism Associated Genes in the Liver of Chicken (닭의 고밀도 사양체계가 스트레스 및 지방대사 연관 유전자 발현에 미치는 영향)

  • The effect of high stocking density (HSD) on the expression of stress and lipid metabolism associated genes in the liver of broiler chickens was examined by chicken genome array analysis. The chickens in a control group were randomly assigned to a $495cm^2/bird$ stocking density, whereas the chickens in a HSD group were arranged in a $245cm^2/bird$ stocking density with feeding ad libitum for 35 days. The chickens assigned to the HSD group had a significantly lower body weight, weight gain, and feed intake compared with those of the control group (p<0.05). The mortality of chickens was higher in the HSD group than in the control group. The microarray analysis indicated up-regulation of stress associated genes such as HMGCR, $HSP90{\alpha}$, HSPA5 (GRP78/Bip), DNAJC3 and ATF4, and down-regulation of interferon-${\gamma}$ and PDCD4 genes. The endoplasmic reticulum stress associated genes, HSPA5 (GRP78/Bip), DNAJC3 and ATF4, were highly expressed in the HSD group. The genes, ACSL5, TMEM195 and ELOVL6, involved in fatty acid synthesis, were elevated in the HSD group. The genes, ACAA1, ACOX1, EHHADH, LOC423347 and CPT1A, related to fatty acid oxidation, were also activated in the HSD group. These results suggest that a HSD rearing system stimulates the genes associated with fatty acid synthesis as well as fatty acid oxidation in the liver of broiler chickens.

Study on Fatty Liver Duagnosed by Abdominal Ultrasonography and Clinical Laboratory Findings (초음파로 진단된 지방간과 임상검사수치의 관련성에 관한 비교 연구)

  • This study obtained the following conclusions by making a comparative study on fatty liver diagnosed by abdominal ultrasonography and clinical laboratory findings. I surveyed the value of abdominal ultrasound in 400 patients without clinical symptoms at C Health Clinic Center, Seoul. 1. Compare with blood pressure was high(systolic/diastolic) in 7.5%/4.5% on persons who were diagnosed fatty liver. 2. At the time of the diagnosis, Total cholesterol level was increased in fatty liver patients, HDL-cholesterol level was high in fatty liver patients. And Trigryceride level was increased in fatty liver persons, LDL-cholesterol was high in fatty liver persons. 3. SGOT level was increased in 5.5% on patients who were diagnosed fatty liver, 0% on persons who were normal and SGPT level was high in 29.5% on people who were diagnosed fatty liver, 0% on patients who were diagnosed normal.

  • 노년기에 들어서더라도 신체가 요구하는 기본적인 영양소는 변하지 않지만 열량 요구는 감소된다. 이는 젊었을 때보다 활동량이 적어지는 노년기의 환경적 원인과 함께 체질량 감소 및 상대적으로 증가하는 지방세포 때문이다. 지방세포는 비지방세포와 비교해 늦게 대사되며 칼로리도 신속히 소모되지 않는 특징이 있다. 노인일수록 모든 영양소를 골고루 섭취하되 열량은 줄이는 식습관을 형성하는 것이 중요한 이유가 여기에 있다.

Mountain-cultivated Ginseng Ripened into Persimmon Vinegar Ingestion on Fat Storage and Metabolic Protein Expression in Diet-controlled Rats (산양삼 혼입 숙성 감식초 섭취에 의한 식이 제한 흰쥐의 지방 저장 및 에너지 대사 단백질 발현)

  • This research is to investigate the four years growth mountain-cultivated ginseng ripened twenty-two weeks into four years fermented persimmon vinegar (tentatively: Sansamcho) ingestion on obese-related factors during dietary control. The Sansamcho was ingested orally, two times a day, after every meal for six weeks to the male rats. Groups were divided into the control (CON), the restricted diet (RD), and the weight cycling (WC). And, each groups has its own sub-groups as the -control (-CON), 2.5 times diluted Sansamcho ingestion (-MPV2.5), and 5.0 times diluted Sansamcho ingestion (-MPV5.0) groups, respectively. The number of rat was consisted of seven in each group. After six weeks rearing periods was done, abdominal fats (retroperitoneal fat, mesentery fat, and epididymal fat) and energy metabolic-related protein (AMPK: AMP-activated protein kinase; PPAR-${\alpha}$: peroxisome proliferator-activated receptor-${\alpha}$; and CPT-1: carnitine palmitoyltransferase-1) were weighed and analyzed. Amount of stored fat was significantly or tended to decrease by Sansamcho ingestion. In addition, sum of fats increasing were suppressed by the material. On the contrary, energy metabolism-related protein expression was significantly increased or tended to increase by Sansamcho ingestion. This results suggested that increased energy metabolism using Sansamcho was restrained effectively visceral fat store by high-fat diet and/or dietary control. In other words, it has a good function to suppress weight cycling which is the most insoluble problem. Therefore, the fusion material, Sansamcho, may expect to utilize as the obese-suppression-food.

Dietary Characteristics of Macronutrient Intake and the Status of Metabolic Syndrome among Koreans (한국인의 대사증후군과 다량 영양소의 섭취패턴)

  • Metabolic syndrome (MetS), which is heavily dependent on dietary and lifestyle practices, is prevalent in Korean adults. Because dietary practices are unique for each race, nationality, and culture, it is important to identify Korean adult dietary practices that are associated with MetS. Macronutrient intake patterns were analyzed and compared across health status using the 2007-2008 Korean National Health and Nutrition Examination Survey data (8,143 adults who had dietary data and biomarkers related to MetS). Although intake levels were different by age, gender, and health status, carbohydrate intake was high and fat intake was low among Korean adults. The carbohydrate energy ratio was higher and the fat energy ratio was lower for subjects > 60 years old who were diagnosed with diabetes, hypertension, or MetS than those in other age groups. The main source of fat from food differed depending on the group. These results demonstrated that macronutrient intake patterns were different between age, gender, and health status subgroups. Hence, it is recommended that nutritional policy and practices aimed at controlling MetS should be based on the dietary characteristics of the target group.

Effects of Hatha Yoga Exercise on Serum Leptin and Metabolic Syndrome Factors in Menopause Obese Middle-Aged Women (하타요가 운동이 폐경기 비만중년여성의 렙틴농도와 대사증후군 인자에 미치는 영향)

  • The purpose of this study was to analyze the effects of Hatha Yoga exercise on serum leptin and metabolic syndrome factors in obese and menopausal middle-aged women. The subjects were 26 obese women divided into the Hatha Yoga exercise group (n=13), which trained for 16 weeks, and the Control group (n=13). Variables of body composition, serum leptin and metabolic syndrome factors were measured in all the subjects before and after the 16-week Hatha Yoga training. The results of the study in the Hatha Yoga group were as follows: body weight, % fat, BMI, WC, WHR and VFA had significantly decreased, but SMM had increased. HDL-C had significantly increased, but leptin, TC, TG, LDL-C, insulin, glucose and HOMA-IR had decreased. The main variables affecting changes in VFA were % body fat, BMI, WHR, TC, LDL-C, glucose, and HOMA-IR. Therefore, regular and continuous Hatha Yoga exercise was effective in improving body composition, visceral fat and serum lipids. Consequently, Hatha Yoga exercise will be effective in preventing cardiovascular disease caused by obesity in obesew and menopausal middle-age women.

Effects of an aqueous extract of purple sweet potato on nonalcoholic fatty liver in high fat/cholesterol-fed mice (고지방/고콜레스테롤 식이를 섭취한 마우스에서 자색고구마 열수추출물 보충이 지방간 저항성에 미치는 영향)

  • Purpose: Anthocyanins from purple sweet potato (PSP) have been investigated in vitro and in animals and found to have a protective effect against oxidative hepatic damage. In this study, we investigated that aqueous extract of PSP can ameliorate the dysfunction of lipid metabolism in mice fed a high fat/cholesterol diet. Methods: Forty C57BL/6J mice were randomly divided into 5 groups (n = 8) and fed one of the following diets for 8 weeks; normal fat (NF) diet; high fat/cholesterol (HFC) diet; HFC with 1.25% PSP (HFPL) diet; HFC with 2.5% PSP (HFPM) diet; HFC with 5% PSP (HFPH) diet. Results: Non-alcoholic fatty liver was manifested in the HFC group by showing increased levels in plasma alanine aminotransferase (ALT) activity, total cholesterol (TC) and low density lipoprotein cholesterol (LDL-C), increased level of TC and presence of many large lipid droplets in the liver, and increased fat cell size in the HFC group compared with the NF group. However, administration of HFC induced a significant decrease in food intake, resulting in decrease in fat mass. Co-administration of PSP did not lead to reversal of body weight changes, ALT activity, and lipid levels in plasma and the liver, but suppressed excess enlargement of the fat cell size through increasing carnitine palmitoyltransferase-1 (CPT-1) gene expression in the liver. Accordingly, the number of fat droplets in the liver was reduced in PSP administered groups. Conclusion: Taken together, these results suggest that PSP may have a protective effect on the dysfunction of lipid metabolism. Conduct of further studies on the coordinated regulation of PSP for lipid metabolic homeostasis at the liver-adipose tissue axis is needed.
