Comparison of Acute Toxicity of Ammonia in Juvenile Rockfish and Red Sea Bream (조피볼락과 참돔 유어에 대한 암모니아 독성 비교)

  • This study compared acute ammonia toxicity in juvenile rockfish and red sea bream. Oxygen concentration affected ammonia toxicity to aquatic animals. Without aeration, the 96-hr $LC_50$ values of $NH_4^+$and un-ionhized ammonia for juvenil rockfish(3.6g, 6.6cm) were 2.61 and 0.09 mg/l, respectively. The 3-hr $LC_50$ values of $NH_4^+$ for the size of 1.9g (5.2cm) and 3.6g (6.6cm) rockfish, and 1.0g (3.8cm) and 2.0g (5.0cm) red sea bream without aeration were 3.96, 3.94, 4.35 and 3.75 mg/l, respectively. When oxygen level was low, larger fish were more susceptible to ammonia toxicity than smaller one because of stress resulted from low oxygen. However, with aeration, the 96-hr $LC_50$ values of $NH_4^+$ for the size of 1.2g (3.9cm) and 2.3g (5.1cm) red sea bream with aeration were 3.84 and 3.90mg/l, respectively. The 6-hr $LC_50$values of $NH_4^+$ for the size of 3.1g (5.9cm) and 6.2g (7.0cm) rockfish with aeration were 3.83 and 3.94 mg/l, respectively. When oxygen level was high, larger rockfish and red sea bream were less susceptible to ammonia toxicity than smaller ones. The 6 hr-or 96 hr-$LC_50$ values of $NH_4^+$for rockfish with reduced ammonia toxicity. In comparing 96 hr-$LC_50$ values of $NH_4^+$ and un-ionized ammonia for juvenil rockfish with those for juvenil red sea bream, the values for rockfish were lower than for red sea bream. This indicates that juvenile rockfish is more susceptible to ammonia toxicity than juvenile red sea bream.

Isolation of an Invertebrate-type Lysozyme from the Body Wall of Spoon Worm, Urechis unicinctus (개불의 체벽으로부터 i-type 라이소자임의 정제)

  • Lysozymes are innate immune factors that play a critical role in the defense against pathogens in various invertebrate animals including spoon worms. In this study, an invertebrate-type lysozyme was isolated from the body wall of spoon worm, Urechis unicinctus. The acidified body wall extract was partially separated using a Sep-Pak C18 cartridge. Among the fractions, the materials that were eluted with 60% methanol/0.1% trifluoroacetic acid showed the most potent antimicrobial activity against Bacillus subtilis KCTC 1021. A series of high performance liquid chromatography (HPLC) steps were then utilized to isolate a single antimicrobial absorbance peak. The molecular weight of the antimicrobial peak was approximated using sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE), which was approximately 13 to 14 kDa. The partial primary structure of this antimicrobial protein that was analyzed, using LC-MS/MS, was CTGGRPPTCEDYAK (1611.69 Da). Homology search of these fourteen residues, using the National Center for Biotechnology Information Basic Local Alignment Search Tool (NCBI BLAST), revealed that the isolated protein was similar to the invertebrate-type lysozymes described in other animals. Then, the antimicrobial and lysozyme enzymatic (muramidase) activities of this protein were assessed. The isolated protein possessed antimicrobial activity and potent muramidase activity, which were comparable to those of hen egg white lysozyme. Therefore, the isolated protein was designated as Urechis unicinctus invertebrate-type lysozyme from the body wall, Uu-iLysb.

An Analysis of Seawater Effect on Groundwater Quality, in the Region of Sinan-gun area, Jeonam, Korea (전남 신안군 지역의 지하수 수질에 대한 해수의 영향 분석)

  • The purpose of the study was to evaluate the groundwater quality of Sinan-gun, Jeollanam-do, an island located in the southern part of the Korean peninsula where the effect of seawater on the groundwater quality had not been investigated in the past. In order to evaluate its effect, the hydrogeological parameters including groundwater quality and major dissolved components were investigated. The water quality was measured four times in the field, and 74 of 163 samples that showed the high conductivity value of more than $500{\mu}S/cm$ and the influence of seawater on the groundwater were analyzed by $Cl^-/HCO_3{^-}$ molar ratio. The results showed that, 40 samples out of 74 were found to have a value of 2.8 or more, indicating severe and very severe effects. According to the type of groundwater quality, the ratio of samples belonging to Na-Cl type, which is considered to be influenced by the direct seawater, is 35.3% for bedrock groundwater and 52.5% for weathered zone and alluvial groundwater. In the evolution stage of groundwater due to seawater infiltration, the type of Ca-Cl prior to the Na-Cl type is 44.1% in bedrock groundwater and 45% in weathered zone and alluvial groundwater. The effect of sea water on the aquifer is likely to be influenced by distance from the shore, pumped water, and tide.

Structural and Electrical Characterization of $LiMn_2O_4$ Synthesized by Solid State Reaction (고상 반응법에 의하여 합성된 $LiMn_2O_4$의 구조와 전기적 특성)

  • We have studied structural characterization, electrical charge and discharge, and impedence properties for the $LiMn_2O_4$ synthesized by solid state reaction. From the X-ray diffraction results, diffraction patterns for compounds of the four kinds of $LiMn_2O_4$ compounds were observed to peaks of (111), (311), (004) and (400). Also, Small similar peaks (110), (222) and (313) were observed for sample C1 and C2. Concerning to new peaks of (113) and (331) for the sample C4, we can found that structural transitions were accompanied from C1 and C2 to C4 via C3 structure. A electrical charge and discharge data showed that efficiency of C1 was very poor state and efficiencies of C2 and C3 were about 60-70% of theoretical capacity. But, the C4 showed good charge and discharge properties of more than 70% of theoretical capacity. From the impedence properties for finding the effect of ions and electrons transfer of battery, C2 and C3 of high frequency of real number showed good electrical impedence properties.

Identification and Characterization of an Agarase- and Xylanse-producing Catenovulum jejuensis A28-5 from Coastal Seawater of Jeju Island, Korea (제주 연안해수로부터 한천 분해 효소 및 자일란 분해 효소를 생산하는 Catenovulum jejuensis A28-5의 동정 및 특성 규명)

  • Strain A28-5, which can degrade xylan and agar in solid medium, was isolated from a coastal seawater sample collected from Jeju Island, South Korea. This strain was found to be a gram-negative, $Na^+$-requiring bacterial strain with a polar flagellum for motility. Additionally, the strain was tolerant to antibiotics such as ampicillin and thiostrepton. The G+C content of the genome was 43.96% and menaquinone-7 was found to be the predominant quinone. Major fatty acids constituting the cell wall of the strain were $C_{16:1}$ ${\omega}7c/iso-C_{15:0}$ 2-OH (23.32%), $C_{16:0}$ (21.83%), and $C_{18:1}$ ${\omega}7c$ (17.98%). The 16S rRNA gene sequence of the strain showed the highest similarity (98.94%) to that of Catenovulum agarivorans YM01, which was demonstrated by constructing a neighbor-joining phylogenetic tree. A28-5 was identified as a novel species of the genus Catenovulum via DNA-DNA hybridization with Catenovulum agarivorans YM01, and thus was named as Catenovulum jejuensis A28-5. The formation of tetramers and hexamers of xylooligosaccharides and (neo)agarooligosaccharides, respectively, were confirmed by thin-layer chromatography analysis using an enzyme reaction solution containing xylan or agarose with two crude enzymes prepared from the liquid culture of the strain.

Relation of the Physico-chemical Properties of Forest Soil to Site Indices of Larix leptolepis Stands (산림 토양의 이화학적 성질과 낙엽송 임분의 지위지수와의 관계)

  • This study was carried out in order to estimate site indices in un-stocked land by the physico-chemical properties of forest soil and to investigate the distribution of the physico-chemical properties by soil horizons. In Larix leptolepis Gordon stand, 80 sites were selected respectively to A and B horizon in soil. About the analysis method of data there was used stepwise regression analysis. Soil pH and T.N. of the physico-chemical properties turned out to be positively related to the site index. However, O.M. and Avail. $P_2O_5$ in soil were found out negatively related. In the degree of contribution of the variables to site index by each one of the stands, the highly effective variables were Base Sat., C.E.C., Sand, Exch. $Ca^{{+}{+}}$, and Exch. $Mg^{{+}{+}}$ in A horizon, Exch. $Ca^{{+}{+}}$, Base Sat., T.N., Avail. $P_2O_5$, and Clay in B horizon, and Exch. $Ca^{{+}{+}}$ and Base Sat. in both of A and B horizon. In conclusion, these results will provide not only the important criteria for establishment of management forest plan in un-stocked land but also the useful guidance for selecting the suitable sites and trees.

Flavor Composition of Garlic from Different Area (국내산 마늘의 향미성분)

  • Garlic from 4 main growing area was analyzed for the composition related to garlic flavor. Twenty-eight volatile compounds in garlic were separated by GC and twenty five were identified by GC/MS. Total peak area was high in garlic from $S\u{o}san$, and low from $Hampy\u{o}ng$. However, no significant difference was found in total peak area of sulfur compounds for garlic from Namhae, $S\u{o}san$, and $Uis\u{o}ng$. The results of free sugars analysis showed that relatively large amount of 1-kestose, 1-nystose, and 1-F-fructosyl nystose known as fructooligosaccharide were found in garlic in addition to glucose, fructose, and sucrose. Garlic from $S\u{o}san$ contained high amount of 1-nystose and 1-F- fructosyl nystose. Total free amino acid content in garlic was in a range of $2036.1{\sim}2704.0mg%$, and it was higher in garlic from $S\u{o}san$ and $Uis\u{o}ng$. Lactic acid, pyruvic acid, oxalic acid, malonic acid, fumaric acid, levunic acid, succinic acid, malic acid, citric acid and pyroglutamic acid were found in garlic. Total organic acid content in garlic was $1905.7{\sim}2359.2mg$, and garlic from $Hampy\u{o}ng$ had relatively low organic acid contents.

The Production and Geochemistry of Evaporite from the Acid Mine Drainage (산성 광산배수로부터 형성되는 증발잔류광물의 생성량과 지구화학)

  • This study has focused on the amount of evaporites and geochemical characteritics of evaporites from the acid mine drainage and on the variation of constituents in acid mine drainage during evaporation. The various colors of evaporites are frequently observed at the rock surfaces contacting acid mine drainage. In order to produce evaporites in the laboratory, acid mine drainages were sampled from the abandoned mine areas (GTa, GTb, GH and GB) and air-dried at room temperature. During the evaporation of acid mine drainages, TDS, EC values and the concentrations of major and minor ions increased, whereas ER and DO values decreased with time. The concentration of Fe increased gradually with evaporation time in the GTb and GB, whereas GH founded in one day but rapidly not detected in the other day after due to removal of Fe by formation-precipitation of amorphous Fe hydroxide. The amounts of the evaporites were produced in amounts of 4 g (GTa), 5 g (GB), 15 g (GH), and 24 g (GTb) from 4 liter of acid mine drainage after 80 days of the evaporation, respectively. In linear analysis from the products with the parameters which are the EC, TDS, salinity, ER, DO and pH contents in field, the determination coefficients were 0.98, 0.99, 0.98, 0.88, 0.89, and 0.25 respectively. If we measure the parameters in field, it would be easy to estimate the amount of evaporites in acid mine drainage. Gypsum and epsomite were identified in all of the evaporites by x-ray powder diffraction studies. Evaporite (GTb) was heated at 52, 65, 70, 95, 150, 250, and 350oC for one hour in electrical furnaces. Gypsum, $CaSO_4\cdot1/2H_2O$ and kieserite were identified in the heated evaporite by XRD. With increased heating temperature, the intensity of the peak at $7.66/AA$ (diagnostic peak of gypsum), the peak at 5.59A ($CaSO_4{\cdot}1/2H_2O)$ and the peak at $4.83{\AA}$ (kieserite) decreased in x-ray diffraction due to dehydration. In the SEM and EDS analysis for the evaporite, gypsum of well-crystallized, radiating cluster of fibrous, acicular, and columnar shapes were observed in all samples. Ca was not detected in the EDS analysis of the flower structures of GTb. Because of that, the evaporite with flower structures is thought to be eposmite.

Environmental Changes after Timber Harvesting in (Mt.) Paekunsan (백운산(白雲山) 성숙활엽수림(成熟闊葉樹林) 개벌수확지(皆伐收穫地)에서 벌출직후(伐出直後)의 환경변화(環境變化))

  • The objective of this study was to investigate the impacts of large-scale timber harvesting on the environment of a mature hardwood forest. To achieve the objective, the effects of harvesting on forest environmental factors were analyzed quantitatively using the field data measured in the study sites of Seoul National University Research Forests [(Mt.) Paekunsan] for two years(1993-1994) following timber harvesting. The field data include information on vegetation, soil mesofauna, physicochemical characteristics of soil, surface water runoff, water quality in the stream, and hillslope erosion. For comparison, field data for each environmental factor were collected in forest areas disturbed by logging and undisturbed, separately. The results of this study were as follows : The diversity of vegetational species increased in the harvested sites. However, the similarity index value of species between harvested and non-harvested sites was close to each other. Soil bulk density and soil hardness were increased after timber harvesting, respectively. The level of organic matter, total-N, avail $P_2O_5$, CEC($K^+$, $Na^+$, $Ca^{{+}{+}}$, $Mg^{{+}{+}}$) in the harvested area were found decreased. While the population of Colembola spp., and Acari spp. among soil mesofauna in harvested sites increased by two to seven times compared to those of non-harvested sites during the first year, the rates of increment decreased in the second year. However, those members of soil mesofauna in harvested sites were still higher than those of non-harvested sites in the second year. The results of statistical analysis using the stepwise regression method indicated that the diversity of soil mesofauna were significantly affected by soil moisture, soil bulk density, $Mg^{{+}{+}}$, CEC, and soil temperature at soil depth of 5(0~10)cm in the order of importance. The amount of surface water runoff on harvested sites was larger than that of non-harvested sites by 28% in the first year and 24.5% in the second year after timber harvesting. The level of BOD, COD, and pH in the stream water on the harvested sites reached at the level of the domestic use for drinking in the first and second year after timber harvesting. Such heavy metals as Cd, Pb, Cu, and organic P were not found. Moreover, the level of eight factors of domestic use for drinking water designated by the Ministry of Health and Welfare of Korea were within the level of the first class in the quality of drinking water standard. The study also showed that the amount of hillslope erosion in harvested sites was 4.77 ton/ha/yr in the first year after timber harvesting. In the second year, the amount decreased rapidly to 1.0 ton/ha/yr. The impact of logging on hillslope erosion in the harvested sites was larger than that in non-harvested sites by seven times in the first year and two times in the second year. The above results indicate that the large-scale timber harvesting cause significant changes in the environmental factors. However, the results are based on only two-year field observation. We should take more field observation and analyses to increase understandings on the impacts of timber harvesting on environmental changes. With the understandings, we might be able to improve the technology of timber harvesting operations to reduce the environmental impacts of large-scale timber harvesting.

The Impact of monsoon Rainfall (Changma) on the Changes of Water Quality in the Lower Nakdong River (Mulgeum) (장마기의 강우가 낙동강 하류 (물금) 수질에 미치는 영향)

  • The impact of summer monsoon on water quality of the lower Nakdong River was evaluated during the summer (June-August) in 1997. Several limnological variables were measured in the interval of $1{\sim}3$ day using an automatic monitoring system (Hydrolab $Recorder^{TM}$) to detect water quality changes caused by rainfall on onehour basis. During the monsoon period (from late June to mid July), 5 times of major rainfall events of >50 mm were recorded in the river basin. Dynamic changes of water quality were observed during the monsoon, and the first rainfall event (June$25{\sim}27$) had a significant influence on the water quality at the lower part of the river. All Parameters were largely changed due to the first rain event, and the changed level was maintained until the end of monsoon period. Nutrient concentrations and turbidity increased and values of the other parameters were declined as a result of water dilution. This rainfall event, Changma, is a meteorological phenomenon caused by the East-Asian monsoon climate. The magnitude and frequency of the rainfall during the early monsoon play an important role in change of water quality and ecosystem characteristics of large river systems.
