Effects of Growth Regulators, Temperature, and Light on the Germination in Wild Vegetable (Chwinamul) Seeds (生長調節劑 , 溫度 및 光이 취나물류의 種子發芽에 미치는 影響)

  • This study was conducted to determine the effects of growth regulators, temperature, and light on the germination of Chwinamul(Aster scaber, Aster tataricus, Synurus deltoides, Solidago virga-aurea var. asiatica, Ligularia fisheri) seeds. Compared to GA, IAA, and kinetin, BA (150ppm) stimulated best seed germination of all five species under the both light and dark condition at $25^{\circ}C$. The effect of growth regulators was not observed significantly under the light condition except of Ligularia fisheri and Synurus deltoides while it was shown markedly under the dark condition. Germination rates of Ligularia fisheri and Synurus deltoides in different concentration of BA were relatively higher under the light at $15^{\circ}C$ while those of other species varied in different concentration and temperature.

Effects of Temperature, Light Condition, and Priming Treatment on Seed Germination of Bidens bipinnata L. (온도, 광 및 Priming 처리가 도깨비바늘 종자의 발아에 미치는 영향)

  • This study was carried out to investigate the effects of light, temperature, and priming treatment on the germination of Bidens bipinnata seeds for the purpose of cultivation of new vegetable crop. The results showed that the treatment of $25^{\circ}C$ yielded the highest germination percentages and the highest germination speed in both light and dark conditions. Light condition had no effect on the germination of B. bipinnata. Priming treatments demon-strated the following improved germination percentages: $Ca(NO_3)_2$ 0.1 M (68.3%), $K_3PO_4$ 0.1 M (63.3%), and $KNO_3$ 0.1 M (53.3%), with $Ca(NO_3)_2$ 0.1 M treatment giving the greatest improvement. Consequentially, the results suggested that $25^{\circ}C$ light condition, and $K_3PO_4$ 0.1 M or $Ca(NO_3)_2$ 0.1 M priming treatments would be effective method to improve the germination of Bidens bipinnata seeds.

Sodium Hypochlorite Solution As a Chemical Wounding Agent for Improving Agrobacterium-mediated Chinese Cabbage Seed Transformation (Sodium hypochlorite처리에 따른 배추종자의 Agrobacterium이용 형질전환 증대)

  • Chinese cabbage (Brassica campestris ssp. napus var. pekinensis Makino) seeds/seedlings were transformed via vacuum-infiltration with recombinant Agrobacterium tumefaciens LBA4404 cells. The agroinfiltration method was determined to be unsuccessful for Chinese cabbage transformation during the analysis of hepatitis B surface antigen expression by ELISA. However, treatment of sodium hypochlorite solution, prior to agroinfiltration, to pregerminated or germinating 1 day- or 2 days-old seeds was proven effectively to enhance transformation efficiency, suggesting that chemical wounding caused by sodium hypochlorite reaction might facilitate Agrobacterium infection and, therefore, transient gene expression in Chinese cabbage sprouts.

Comparison of Germination Power, Germination Speed, and Germination Peak Time among Five Festuca Species (훼스큐 속 잔디 5종의 발아력, 발아세 및 발아 피크 기간 비교)

  • Research was conducted to investigate germination and establishment characteristics of five fescue species. Alternative conditions of 8-hr light at $25^{\circ}C$ and 16-hr dark at $15^{\circ}C$ were applied. Significant differences were observed in seed germination power, germination speed and germination peak time. Germination power was 62.25% in CRF 'Audubon' to 96.75% in TF 'Olympic Gold'. It was greater with TF over FF, being TF > HF > SF > CF > CRF. Germination speed, measured as days to seed germination of 50% through 80%, was fastest with TF, medium with CF and HF, and slowest with CRF and SF, being TF > CF, HF > CRF, SF. Germination peak time was much faster with TF over FF. It ranged from 0.94 to 1.47 days with TF. But it varied with 2.80 to 12.36 days among FF species. The peak time was longer in order of HF, SF < CF < CRF. Considering germination power, germination speed and germination peak time, TF 'Arid III' was the best cultivar under alternative conditions, while CRF 'Audubon' the poorest. Overall establishment characteristics among Festuca species were even better with TF over FF in order of TF > CF, HF > SF > CRF.

Effects of Temperature, Light and Plant Growth Regulators on the Seed Germination of Lavandula angustifolia Mill (잉글리쉬 라벤다의 종자발아(種子發芽)에 대한 온도(溫度), 광(光) 및 생육조절물질(生長調節物質)의 영향(影響))

  • This study was conducted to establish mass propagation system from seeds of Lavandula angustiolia Mill. Only 4% of dry seeds were germinated when they were incubated in $4^{\circ}C$. Germination rate of stratified seeds for 9 weeks was 15% higher in light than darkness. Soaking with 1000mg/l of $GA_3$ was helpful to overcome the effect of darkness. The optimal temperature for germination was $25^{\circ}C$ for the seeds that were treated with$GA_3$, solution. For the improvement of germination rates, pretreatment of $GA_3$ at 500-2000mg/l showed about 75% of germination, and in the combination treatment of $GA_3$, and BA, germination rate increased by about 10% in the treatment of 1000mg/l $GA_3+10mg/l$ BA compared with the 1000mg/l $GA_3$ treatment. Cold stratification treatment was very effective for seed germination, and over 70% of seeds were germinated when they were incubated in $4^{\circ}C$ for 9-12 weeks. In addition, there was synergic effect on the seed germination subject to stratification and 1000mg/l $GA_3$, treatment for 6 months. In the mixture of vermiculite: peatmoss (1 : 1, v/v), emergence rate was 6.7% in control and 65% in 1000mg/l $GA_3$ treatment, respectively.

  • Theses studies were conducted to clarify the germinating characteristics of seed treated with different temperature, light, and ($Gibberellin(GA_3)$) to produce the homogeneous seedlings Angelicogigas NAKAI. The results obtained are summarized as follows: Rate and speed of germinating at $25^{\circ}C$ were higher than $15^{\circ}C$and $20^{\circ}C$ by 87.3% and 68.9% and also the required day for germination at 25°C was shortest by 14.1 days. Germination rate at the light condition was 90.7% and that at the dark was 65.7%. Germinating ability of seed stored at $4^{\circ}C$and $-10^{\circ}C$ for 165 days as dried states was 92.0% and 91.3%, while that at room temperature was 50.3%, but the longer storage of seed than 165 days decreased the ability. Gibberellin treatment by 10ppm to seed stored at $4^{\circ}C$ and by 6ppm to seed stored at $-10^{\circ}C$ increased the germination rate more by 20.9% and 31.7% than nontreated seed.

Seed Dispersion and Seedling Spatial Distribution of the Tree of Heaven in Urban Environments (도시환경에서 가중나무 종자의 확산 및 발아 분포)

  • The tree of heaven naturalized plant grows in the various habitats of the city. The principal reason is that it is reproduced not only through sexual reproduction but through asexual reproduction. By the spreading of sucker and a bud appearance of the seed it forms a group easily inside the city habitat. The distribution of seed and spreading of the seedlings of the tree of heaven were investigated like the followings. As the result of the sexual reproduction by the seed and hangs in cluster farm, 350 individuals are germinated at the Namsan the best environment, and 50 individuals at Toegyewon Rotary These seeds are dispersed by the effect of wind and geographical factor. Because the northwest wind is blowing mainly from late autumn to early spring, it is dispersed in the south. In Toegyewon Rotary, a slant site, is dispersed mainly by down the site. Compared with width of crown, the seedling of a seed germination are distributed outside more than inside the crown and the seed is spreaded until the maximum 7.5 times of crown width at the investigated area. 1 year seedlings appeared as a group at the place of 2-4 times of the crown (in 20m from the crown, 75% distribution). Considering the distribution by the seed scattering of the tree of heaven, there is a possibility of sudden individual increase even in the deteriorated city environment. So this possibility shows that we need to consider the protection of the city soil, and the species variety decrease resulted from the successful competition to the other species.

Effects of Seed Treatments for Promoting Seedling Emergence of Codonopsis lanceolata Trautv. (입묘율 향상을 위한 더덕 종자의 처리 효과)

  • Reasonable seedling emergence is a prerequisite for successful crop cultivation especially in medicinal crops. The study was carried out to model the pretreatment or seed treatment of Codonopsis lanceolata seeds by evaluating the treatment effects of priming, $GA_3$, drying and water imbibition after drying on their germination and then their successive seed treatment on the basis of its seedling emergence. The priming using $Ca(NO_3)_2$ and $GA_3$ treatment under their different concentrations and light quality illuminated for 12 hours a day were separately done and the two best results from priming and $GA_3$ treatments were compared to determine the better one. The drying of imbibed seeds using the above best result and water imbibition of the dried seeds were successively done to measure the rates of germination and emergence. On the greatest germination rate of the first two individual treatments, priming was obtained at $Ca(NO_3)_2$ 150 mM under blue, red light or darkness for 2 days but $GA_3$ was done at 0.1 mM under blue light imposed for 3 days. However, the result of the latter $GA_3$ treatment was better than that of priming. $GA_3$ treated seeds were best desiccated under $35^{\circ}C$ and 4 hour red light illumination. A day water imbibition immediately before sowing increased the germination rate of seeds dried after $GA_3$ treatment. Seedling emergence tested after all 3 successive treatments, $GA_3$, drying and water imbibition before sowing was greater than the two others, only $GA_3$ treatment and the combination of $GA_3$ and drying, meaning that its pretreatment of seeds or seed treatment must follow the successive procedure of the above 3 ones.

Varietal Differences and Drying Storage Effects for Some Treatment Conditions of EMS and MMS on Rice Seeds (수도종자에 EMS와 MMS의 처리조건에 따른 건조저장효과와 품종간차이)

  • In order to study the biolgiocal effects of dry back following EMS (Ethyl methane sulfonate) treatment on seeds of 'Dunghan Shali' that belongs to indica-type rice with various presoaking(24, 36, and 40 hrs respectivily) and treatment time(l, 2 and 3 hrs respectivily), this experiment was done. Seedling heights of wet seeds presoaked for 36 hrs was only greatly decreased with EMS treatment compared with other periods. In germination rate of wet seeds, all of these damage was not changed. For the drying back seeds, otherwise, seedling height and germination rate were greatly reduced depend upon the time of treatment and presoaking as well as storage effects after drying back. Reduction patterns, both seedling height and germination rate, between 1 week and 8 weeks after dry back were quite similar. The other experiment was carried out in order to study for the influences on the seedling growth as biological damage with EMS and MMS(Methyl methane sulfonate) treatments among different varieties. The indica-type rice varieties; Taichung Native No.1, Dunghan Shali and IR-8, were used as the experimental materials. From this trial some results were obtained as follows; 1. Both seedling growth and germination rate on all varieties, used, MMS, treatment showed more toxic effects than those of EMS treatment. 2. Seeding growth injury of rice seeds dried back was increased gradually, and then was approached the maximum at 6-8 days after storing. In IR-s variety, otherwise, growth damage was appeared a little. Germination rate of these were quite smilar, even though chemical used is different and storage period was prolonged. 3. It was showed clearly that varietal differences of chemical mutagene treatments were recognized.

Production of Dry-Type Artificial Seeds Using Alginate-Encapsulated Rice Somatic Embryos (벼 체세포배를 알긴산 캡슐에 넣어 제작한 건조형 인공종자)

  • Dry-type artificial seeds were produced by dehydrating alginate-encapsulated somatic embryos of lire. When placed on half- strength MS solid medium 20% of the artificial seeds dehydrated to the level of 80% water loss were capable of germination. Addition of 0.1 mg/L ABA to alginate solution before encapsulation enhanced the germination percentage of those dehydrated to the level of nil to 90% water loss by up to 1.7-folds. The results suggest that ABA may enable somatic embryos to overcome physical and/or physiological restraint by encapsulation and dehydration.
