Studies on the Conditions of Seed Germination of Gastrodia elata (천마 종자의 발아 조건에 관한 연구)

  • Gastrodia elata has been cultivated using mycorrizal fungi including Armillaria mellea as an energy source (myco-hetrotrophy) because it is aphyllous and achlorophyllous archid. But the yields of G. elata have been recently decreased owing to the degeneration of spawn tuber arised from successive asexual reproduction. Therefore, this study was carried out to solve this degeneration by seed germination, namely sexual reproduction. The seed germination of G. elata was excellent on the fallen leaves medium of oak tree. The fruetification rate of the capsule of G. elata by artificial pollination was 94.1% and better than natural pollination. The living weight of capsules of G. elata suitable for seeds germination was above 31 mg. The middle-matured seeds and matured seeds of capules were largely germinated, while the immatured seeds was small germinated. The storage temperature of the cropped capsules suitable for a favorable seed germination was $0{\sim}5^{\circ}C$ and the storage period of it was one month.

  • 1. Almost complete death of pitch pine seeds resulted when presoaked in 0.5 per cent ethyleneimine solution without neutralization. When neutralized with 1 N HCL, quite high germination percentages were obtained even with 2 per cent solution. 2. When presoaked in 0.25 or 0.50 per cent ethyl methanesulphonate, there was a delay in germination, but the germination per cent rapidly increased from about the 10 th day from sowing. 3. Basing upon the time when the germinative energy of control seeds was reached, the comparative lethal doses (concentrations) among mutagenic chemicals and radiations were investigated (Table 1).

  • To establish the mass propagation method of Typha orientatis Presl, several factors influencing seed germination were investigated. The best seed germination of T. orientalis was achieved under the light and 25~3$0^{\circ}C$ temperature. GA$_3$ tended to enhance seed germination, and 500 mg.L$^{-1}$ GA$_3$ showed 90.7% germination, which was 1.2 times of control. Compared to control, BA treatment prohibit seed germination. Especially, seeds were not germinated under 500 mg.L$^{-1}$ BA treatment. In case of KNO$_3$ and Ca(NO$_3$)$_2$ treatment, the germination rate was decreased by all concentrations, except 100 mM Ca(NO$_3$)$_2$.

  • This study was conducted to determine an appropriate coating method for mechanized sowing of small and light Italian ryegrass seeds. In the case of manufacturing coated seeds, 2% PVA was superior as an adhesive compared to PVP and Tween80. Calcium carbonate + talc adhered to the seeds smoothly and well and the seed weight was higher than that of the other coating materials. The germination percentage of seeds coated with calcium carbonate + talc was 95.8%, which was not significantly different from 96.5% of the control group, but the mean daily germination was the highest. As the coverage ratio of calcium carbonate + talc increased, the strength tended to increase when coated at 1.5 times the seed weight. There was no difference in the germination rate and plant height compared to the control group according to the coverage ratio.

  • The influences of temperature, harvest time, sowing time, seeding media, moisture content of media and light on the seed germination of Lycoris are as follows, Both L. koreana and L. aurea had over 90% seed setting, and seed maturity came in September or early October. It took six months to have over 60% seed germination. The optimum germination temperature was $25^{\circ}C$, and the alternating treatment was effective at $20{\rightleftarrows}25^{\circ}C$. The optimum harvest-time was around Sep. 20, and the optimum seed-time was immediately after havest. The optimum seeding media was sand loam, and the optimum moisture content was PF 1. 9/400ml seeding media. The dark condition was more effective for the seed germination than the light condition.

  • As environmentally sound agriculture emphasize gradually as well as the importance of environment, many studies on weed and soil nematode control of new concept which does not affect adversely environment are in progress. For this purpose, plant seeds which are representative weed species in field were selected and treated with microwave having generating capacity of $17.73kw/m^3$, and investigated germination rates of weed seeds and the number of soil nematode according to soil moisture contents, soil depth and irradiation times. The microwave effect on the germination rates was extremely high in soil moisture content of 40% and irradiation time over 40 seconds for irradiation time, while soil depth did not affect germination rate. In view of the results so far achieved, it seems that universal dispersed microwave system is effective for the control of weed seeds.

  • A seed pelleting technique was developed for easy handling of small sesame seeds (Chinbaeckkae) and for direct seeding in field. The mixture of pelleting material, binder(sodium alginate) and seeds were dropped in 100mM CaCl$_2$ solution. Charcoal was appeared as a best material among all to make pellets with the characters of hardness, breakdown, destruction rate and germination percentage. Zeolite formed the hardest pellet seeds among the materials and 1~3 seeds contained in a pellet on the treatment of 87g of sesame seeds mixed in 1$\ell$ CaCl$_2$ solution was estimated optimum rate of seed mixure with few appearance of seedless pellet. The moisture absorption rate was greater in the order of peat+charcoal > peat > charcoal > zeolite. Germination percentage of pelleted seeds with charcoal was the best and similar to that of common non-treated seeds. Germination percentage of pelleted seeds by sowing depth of exposing a quarter of pellet seeds on the surface of vermiculite germination test box and were 92% in charcoal treatment and was the best among all released materials.

  • Lettuce, a typical light-induced seed germination type, exhibits different germination responses according to cultivars, light quality, and plant growth regulator (PGR) treatments. Germination rates in most tested cultivars were over 85% under both white and red light, and were slightly decreased by blue light. Although photo-inhibition in germination was observed from most cultivars by far-red light, 'Cheongguangcheongchima', 'Okdol', and 'Manchudaecheongchima' could be classified as photo-insensitive lettuce cultivars by exhibiting the germination rates as 78,63, and 48% under for-red light, respectively. 6-Benzylamino purine (BAP) and kinetin promoted seed germination and normal seedling production under far-red light, but ethephon did not show any positive effects. Cytokinins such as BAP, kinetin, thidiazuron (TDZ), and zeatin overcame photo-inhibition of seed germination even the concentration of below $50\;mg{\cdot}L^{-1}$. However, auxins such as IBA, 2,4-D, and NAA failed to overcome the far-red light-induced photo-inhibition.

  • Effects of light on germination of major weeds in upland fields were examined. Most weeds showed light positive response to germination at above 25$^{\circ}C$, but germination percentage was very low at lower temperatures. Red light promoted germination of Amaranthus lividus, Portulaca oleracea, and A. retroflexus, but far-red light inhibited germination. Germination resposnse of A. retroflexus, A. lividus, and P. oleracea showed reversible reaction to red/far-red light, and thus it seemed that phytochrome was involoved in seed germination, Echinochloa crusgali and Eleusine indica showed higher germination percentage under the conditions of continuous illumination of red, white, and bule lights, and thus it was estimated that blue light pigment (HIR, high irradiation reaction) related to long time illumination was involved in germination in addition to phytochrome.

Characteristics of Seed Germination and Bulb Dormancy in Korean- Native Allium victorialis L. (韓國産 산마늘 種子의 發芽와 種球의 休眠 特性)

  • The experiments were conducted to investigate the characteristics of the seed germination and the bulb dormancy in Korean-native Allium victorialis L. by the physicochemical treatments. The characteristics of two Allium victorialis seeds was similar in shape, color and threshing but different in other characters. Weight of 1,000 grains in Ullrung-native was 14.8g corresponding to 2.1 times as compared with Odae-native. Seeding immediatly after seed collection of Ullrung-native only germinated and chemicals didn't affect in germination improvement. The germination rate, average days to germination and coefficient of germination of Ullrung-native was 86%, 42 days and 2.05 at the dark condition of control, respectively. The germination rate, average days to germination and coefficient of germination of Odae-native was 70%, 47 days and 1.5 at the dark condition of control and 82%, 47 days, 1.75 at the light condition after 5$^{\circ}C$ prechilling for 30 days, respectively. The bedding days to reduce the physiological dormancy in the bulb of Odae-native assumed to be appropriate up to 45 days after the first prechilling.
