배경: 많은 실험적인 연구에서 종양 조직 내의 아포프토시스와 미세 혈관의 생성은 서로 반비례한다고 보고된다. 비소세포 폐암 조직내에서 두 수치의 관계를 조사하여 보았다. 대상 및 방법:조직내의 아포프토시스의 정도는 deoxynucleotidyl trasferase방법으로(Apop Tag In Situ Apoptosis Detection Kit, ONCOR) 측정하였고, 종양내 미세 혈관 밀도는 항 CD 31 항체를 이용하였다. 결과:아포프토시스 지수와 종양내 미세 혈관 밀도 사이에는 통계적으로 유의하게 역 상관관계가 있었다(p = 0.047). 결론: 비소세포 폐암종에서 아포프토시스와 미세 혈관 생성의 정도는 서로 연관이 있다고에 할 수있다. 그리고 종양내의 신생 혈관의 생성이 종양내 아포프토시스의 억제에 기여한다고 유추 할 수 있다.

A numerical study of a hemodynamical model for the tumor angiogenesis is carried out. The tumor angiogenesis process is comprised of a sequence of events; secretion of tumor angiogenesis factor(TAF) from the solid tumor, degradation of the basement membrane of nearby blood vessels, migration and proliferation of the endothelial cells. The model takes into account the effect of TAF concentration and endothelial cell density, and their conservation equations are represented as a set of one-dimensional initial boundary value problems. These equations are discretized by using a finite difference method in which the second order schemes both in time and in space are used. The effects of the parameters contained in the model are Investigated extensively through the numerical simulation of the discretized model. The result for the typical case compares very well with the known result.
Cancer angiogenesis is simulated using a two dimensional model. Governing equation of angiogenesis is a TAE (Tumor angiogenesis factor) conservation equation in time and space. A stochastic process model is utilized to simulate vessel formation, proliferation, and migration to a cancer pellet. Numerical results are presented especially in case of growing cancer.

Hypoxia-inducible factor 1(HIF-1)는 당원 수송체, 당분해 효소, 혈관내피세포 성장인자 등의 유전자의 전사를 활성화한다고 알려져 있다. 그리고 HIF-1의 전사 활성도는 HIF-1 a subunit의 표현이 조절되는 정도에 의존한다. 그러나 식도암에서 HIF-1의 발현과 혈관 생성능 및 종양세포 증식능과의 관계 및 예후에 관한 연구는 전무하다. 대상 및 방법: 고신대학교 의과대학 흉부외과학교실에서 1995년부터 2000년까지 수술치험한 77예의 식도 편평세포암 환자의 조직에서 채취한 정상 편평상피와 암조직에서 면역조직화학검사를 이용하여 HIF-1 a의 발현을 조사하고 혈관생성인자, 증식지수, p53 단백과의 상관관계, 임상-병리학적인 인자 및 생존율과의 상관관계를 분석하였다. 결과: HIF-1 a의 고발현율은 42.9% (33예/77예)였다. HIF-1 a의 고발현은 조직학적 등급(p=0.032), 병리학적 병기(p=0.002), 종양 침윤의 깊이(p=0.022), 주위 림프절 전이(p=0.002), 원격전이(p=0.049), 림프관 침윤(p=0.004)과 관련이 있었다. HIF-1 a의 고발현은 혈관내피세포 성장인자의 발현, Ki-67 증식지수와 관련이 있었으나, 미세혈관수와는 관련이 없었고, p53의 발현과는 관련이 있는 경향을 보였다. 단변량분석과 다변량분석에서 HIF-1 a의 고발현은 불량한 예후를 나타내는 인자로 보였다. 결론: 식도 편평세포암 조직에서 HIF-1 a의 발현은 종양조직내 신생혈관의 생성과 관련이 있는 것으로 나타났고, 고발현 된 경우는 림프절 전이와 수술 후 불량한 예후를 나타내었으므로 보다 강화된 치료전략이 필요할 것으로 사료된다.
Objectives: Angiogenesis is the process of new blood vessel development from preexisting vessel. The growth of tumor is dependent on angiogenesis. This study was aimed to determine the role of the VEGF expression and MVD(microvessel density) in salivary gland tumors and to analyze the correlation between the expression of VEGF/MVD, and clinical manifestation. Material and Methods: We analyzed the expression of VEGF and average MVD and neo-MVD in the 57 cases of salivary gland tumors by immunohistochemistry. Results: VEGF expression was higher in malignant tumors than in benign tumors. Average and neo-MVD were not different in benign tumor and malignant tumor. There was a tendency of positive correlation between VEGF expression and MVD but it was not statistically significant. Stage, T stage and recurrance rate were not correlated with VEGF expression, average-MVD and neo-MVD. Expression of VEGF, average-MVD and neo-MVD were significantly increased in Warthin's tumor compared with pleomorphic adenoma. Conclusion: Overexpression of VEGF has an important role in malignant salivary gland tumors.

ABC(avidin-biotin complex) 방법에 기초한 면역조직화학검사를 이용하여 암조직과 정상조직에서 HIF-$1{\alpha}$, VEGF(vascular endothelial growth factor), p53 단백의 발현을 조사하고, Ki-67의 발현을 이용한 증식지수를 측정하였다. HIF-$1{\alpha}$ 발현과 환자의 생존기간을 포함한 임상적-병리학적 변수들과의 상관관계, VEGF, p53의 발현과 증식지수와의 상관관계를 분석하였다. 결과: HIF-$1{\alpha}$의 과발현은 40.7%(24예/59예)였다. HIF-$1{\alpha}$의 과발현은 병리학적 TNM병기(p=0.004), T병기(p=0.020), N병기(p=0.004), 림프관/혈관 침범(p=0.019) 등과 관련이 있었다. 또 혈관내피세포 성장인자의 발현(p<0.001) 및 이상형의 p53의 발현(p=0.040)과 관련성이 있었다. Kaplan-Meier 생존분석에서 HIF-$1{\alpha}$의 과발현이 있는 환자의 5년 생존울은 22%로 HIF-$1{\alpha}$의 저발현 환자의 5년 생존율 61%에 비해 불량한 생존율을 보였고, 단변량분석과 다변량분석에서 HIF-$1{\alpha}$의 발현은 불량한 예후를 나타내는 인자로 관찰되었다. 결론: 이상의 결과로 비소세포 폐암 환자에서 HIF-$1{\alpha}$의 과발현은 종양내 신생혈관의 생성과 림프절 전이와 관련이 있는 표지자로 여겨지며, 수술 후 불량한 예후를 나타내었다.
Objectives: The purpose of this study was to evaluate for prognostic significance of VEGF expression and tumor angiogenesis in papillary carcinomas of the thyroid. Materials and Methods: The materials were 79 cases of papillary thyroid carcinomas, and age, sex, tumor size, multiplicity of tumor, capsular invasion, lymph node metastasis, recurrence, TNM stage, DeGroot stage and AMES scale were evaluated. An immunohistochemical stains for CD 34 to estimate microvessel density (MVD), and VEGF were done. MVD was defined as an average count of vessels per ${\times}400$ power field in the most vascularized area. VEGF expression was interpreted as 1+ and 2+ according to staining intensity and percentages of positive cells. Results: Mean score of MVD was $39.7{\pm}16.9.$ MVD were significantly higher in cases with capsular invasion (p=0.0001), lymph node metastasis (p=0.0001), TNM stage III (p=0.0022), DeGroot stage III (p=0.0163) and high risk group by AMES scale (p=0.0001). VEGF 2+ expression rate was significantly increased in cases with capsular invasion and lymph node metastasis (p=0.0006, p=0.0013), and in cases with TNM stage III, DeGroot stage III and high risk group by AMES scale (p=0.0236, p=0.0003, p=0.0293). In VEGF 2 + expression group, MVD was significantly higher than in VEGF 1 + group (p=0.0008), and MVD showed positive relation to VEGF 2 + expression (r=0.4616). Conclusion: VEGF expression and high MVD were significantly correlated to capsular invasion, lymph node metastasis, TNM stage III, DeGroot stage III and high risk group by AMES scale. The expression of VEGF and high MVD could be considered to be one of prognostic factor in papillary thyroid carcinomas.

Background and Objectives: Angiogenesis is the process of new blood vessel development from preexisting vessel. Angiogenenesis has been considered to be essential for the growth and expansion of a solid tumor. Vascular endothelial growth factor (VEGF), known as one of the most important vascular permeability factors, induces proliferation of endothelial cells, stiumulates angiogenesis, and increases vascular permeability. Several recents reports have documented that VEGF overexpression is associated with poor clinical outcomes in many maligmancies. The aims of this study were to determine whether microvessel density and VEGF expression are related to clinicopathologic factors such as age, sex, tumor size, tumor stage, and prognostic factors and to evaluate the relationship between VEGF expression and angiogenesis in benign and malignant thyroid tumors. Materials and Methods: The subjects were 65 patients (27 with papillary carcinoma, 27 with adenomatous hyperplasia, 11 with follicular adenoma) who underwent thyroidectomy from 1995 to 2001. Imuunohistochemistry was used to detect VEGF expression and microvessel density (MVD) in paraffin-embedded thryoid tumor specimens. Results: The intensity of the VEGF expression did not show stastically difference between benign and malignant thyroid tumors. There was no apparent correlation between VEGF expression and age, tumor size, T stage or scores of the AGES, AMES and MACIS systems. The neo-microvessel density was higher in the maligant tumor than the benign tumors. Also, higher neo-microvessel density was associated with metastases of the lymph nodes and scores of the AMES and AGES systems. Conclusion: Our results suggest that neo-microvessel vessel density may be a significant prognostic factor in the thyroid papillary carcinoma. But the VEGF expression does not appear to be an significant independent prognostic factor for thyroid papillary carcinoma.
