An invertase from Korean ginseng (Panax ginseng C. A. Mayer) was purified by means of DEAE-cellulose column chromatography and gel-filtration through Sephadex G-75. The homogeneity of the purified invertase was proved by polyacrylamide gel disc electrophoresis. The enzyme was separated into two subunits by SDS-polyacrylamide gel electrophoresis, showing its molecular weights as 48,000. The enzyme preparation showed a characteristic protein UV-spectra.

Conformal methods such as staircase approximation and effective medium theory have been used to model arbitrary dielectric surfaces in Cartesian FDTD(finite-difference time-domain) grids. However, the staircase approximation doesn't guarantee accurate results and the effective medium theory cannot be applied for frequency dispersive medium. In this paper, a simple conformal method is introduced for analyzing arbitrary dielectric surfaces. These arbitrarily shaped dielectric surfaces make nonuniformly filled FDTD cells. E(H)-field in a nonuniformly filled FDTD cell is calculated by considering the cell as the combination of two kinds of uniformly filled cells whose material boundaries are normal and parallel to the E(H)-field in the cell. TM scattering from a rotated 2-D dielectric and a ferrite square cylinder is analyzed to show the accuracy of the proposed method.
Adipocytes have been known to secrete a number of important proteins called adipokines with roles in energy metabolism, reproduction, cardiovascular function and immunity. In this study we have attempted to identify intensively secretory proteins from 3T3-L1 adipocytes. 3T3-L1 preadipocytes were differentiated into mature adipocytes and then the cells were left in serum-free medium. The supernatant was filtrated and dialyzed. Lyophilized secretome was fractionated by two different methods, 1-D SDS PAGE and RP-FPLC. The tryptic peptides from the gel slices and the FPLC fractions were analyzed by nanoLC/ESI-MS/MS. We identified a total of 303 identical proteins from two methods, 251 proteins from 1-D gel and 184 proteins from RP-FPLC. 86 of them were listed as a secretory protein Finally, we identified many known or unknown secreted proteins existed in the low level including adiponectin, angiotensinogen, bone morphogenetic protein-1 (BMP-1), macrophage migration inhibitory factor (MIF), insulin like growth factor-II (IGF-II), interleukin-6 (IL-6), follistatin-related protein-1, minecan, and resistin. The existence of some of secreted proteins has been confirmed in RNA level. This proteomic experiment is useful for the intensive screening of secretory proteins in many kinds of other cells.
A Potent inhibitor of trypsin and other various serine proteases including chymotrypsin, elastase, kallikrein, plasmin and subtilisin, has been purified to homogeneity from chick skeletal muscle by convendonal chromatographic procedures. The Inhibitor has an apparent molecular weight of 66, 000 dalton as determined by gel filtration. When the purified inhibitor was electrophoresed in the presence of sodium dodecyl sulfate, there appeared rwo protein bands having molecular weights of 66, 000 and 64, 000 dalton. The 64, 000 dalton protein seems to be the product of 66, 000 dalton protein by a lin'ited proteolysis during the purification procedure or in viuo. Thus, it seems to consist of a single polypeptide. The inhibitor appeared to be glycoprotein and have an isoelectric point of 7.4. It contains relatively large amount (8.33 mole%) of cysteine residues.
The non-Newtonian flow curves of polyurethane melts were obtained by using a Physica cone-plate rheometer at various temperatures. The rheological parameters were obtained by applying non-Newtonian flow equation to the flow curves for polyurethane samples. When the polyurethane samples are under increasing-decreasing shear rate modes, the hysteresis loop and thixotropic behavior were shown. Polyurethane melts behave as strong gels when they are subjected to shear flow, but when the applied stress surpasses the yield stress, they exhibit non-linear viscoelasticity. Upon decreasing shear rate, its shear stress remains smaller than the values measured in the increasing shear rate mode, because of broken of its structure.
The purpose of this study is to examine the influence of 5% tetracycline(Tc) gel on healing of gingival tissue and change of diseased root surface when used with nonsurgical procedure. 7 patients with advanced periodontitis were received thorough scaling and root planning, followed by saline irrigation on 10 randomly selected control teeth and Tc gel application for 5 minutes with specifically designed plastic instrument of 10 test teeth in contralateral side. At 0, 1, 7, 14, and 21 days after treatment, biopsy and extraction were carried out. At day 7, Tc group showed decreased inflammation and delayed proliferation of pocket epithelium in comparison with control group which was continued for all experimental days. Scanning electron microscopic finding revealed demineralized and cleaned root surface with exposed dentinal tubules in Tc gel group. In the present study, clinically successful result is expected with combined use of nonsurgical periodontal therapy and intrapocket application of Tc gel.
A Purification device with 30-channel free-flow electrophoresis was assembled to treat samples of 240m1 volume for purification of lentil lectin (LcH) from lentil seeds with no impurities in a silverstained PAGIEF gel. HEPES(50mM)-Ttis(50mM), Cycloserine(50mM)-urea(3M), Histidine(50mM)-urea(3M) were used as ampholytea among which Histidine(50mM)-urea(3M) (pI 7.65) was found best in resolution. LcH is known to be present in the form of LcH-A, LcH-B and the complex of the two. The complex, however, disappeared when urea was added in the ampholytes, which suggested that the complete purification of two isolectins is possible using the present multistep purificaton device.
This study was conducted to investigate the usefulness of a hot water extract of licorice root as a source for production of healthy food. The electron donating capacity of the hot water extract of licorice root was very strong. This activity decreased by $6.9\%$ after keeping it in 5 days of cold storage, but it was not significantly different. Ten types of licorice jellies were prepared, using agar, agar-pectin, agar-cellulose, 2 different proportions of agar-pectin-cellulose as a gelling agent, and 2 levels of sugar. Among the 5 jellies containing $10\%$ sugar, the elasticity and overall acceptance of the agar jelly obtained the highest sensory score, but the color and flavor were not affected by the type of gelling agents. As the sugar concentration increased to $15\%$, the elasticity of the agar jelly was also evaluated as being the best one among the 5 jellies, but the overall acceptance was not significantly different from the others.
