Studies on Preservation of Germ Cells in Hanwoo I. Effects of Frozen Thawed Viability and Characteristics in Bovine Semen (한우의 생식세포 보존에 관한 연구 I. 한우 정액의 일반성상 및 동결후 생존성에 미치는 영향)

  • This study was carried out to cryopreserve and to investigate characteristics of semen in Hanwoo. Semen was obtained from bulls selected by Daekwanryeong Branch station. Semen was collected each morning of the experiment, placed in water jacketed tubes at 37$^{\circ}C$, and trans-ported to the research laboratory within 10 minutes. Semen was extended with Egg yolk-glycerol extender to contain 50${\times}$10$^{6}$ sperm/ml. Semen was cooled over a 6h period in water jacketed tubes from about 25 to 5$^{\circ}C$, Egg yolk-glycerol extender was added in one step at 5$^{\circ}C$. Semen was aspirated into 0.5ml straws, which were sealed with powder. Egg yolk-glycerol extender, which is used in Hanwoo sperm frozen and stored, semen from 13 Hanwoo bulls collected, the postthawed percentages of motile sperm were 65.7%. In semen characteristics of Hanwoo bulls, number of bulls volume are 5.7 ml and total cell count are 975${\times}$10$^{6}$ m1 ejaculate.

An Experimental Study on Filling Material for Bored Pile Using High Calcium Ash (고칼슘 연소재를 이용한 매입말뚝의 주면고정액에 관한 실험적 연구)

  • In this study, laboratory tests were performed to evaluate for new filling materials (ZA-Soil) for bored pile that were developed using by high calcium ash. As a result of laboratory test, the uniaxial compression strength of 2 types of ZA-Soil are shown 68.0% and 64.6% compared to ordinary portland cement. And it have a suitable flowability and environmental stability. Also, after 28days, uniaxial compression strength of material mixed with soil and high strength filling material (ZA-Soil) for bored pile is 1.10-1.23 times bigger than material mixed with ordinary portland cement.

  • Experiments were performed to find out the physico-chemical properties of milt and the sperm motilities in various conditions using the grey mullet, Mugil cephalus. The average concentration of sperm in the milt was $1,11 \pm0.36\times10^{10}/ml$. Spermatocrit was 96.7$\pm$2.6. pH and osmolality of seminal fluid were 7.8$\pm$0.1, 370$\pm$6 mOsm/kg, respectively, Total protein concentration of sperm was higher than that of seminal fluid, but total lipid concentration of seminal fluid was higher than that of sperm. The sperm motility was high in the diluent of milt : artificial seawater (1:10, by volume) and in 822 mOsm/kg and 983 mOsm/kg similar to seawater osmolality, but it decreased after 20 minutes. But activity of sperm was highly maintained in 482 mOsm/kg which was a little higher than osmolality of seminal fluid, and was high in pH 7$\~$9.

Analysis of the Applicability of Ground Stabilizer Using Recycled Resources as Prebored Piles (매입말뚝 주면고정액으로 순환자원을 재활용한 지반안정재의 활용 가능성 분석)

  • In this study, tests were performed to analyze the feasibility of using the ground stabilizer from recycled resources such as blast furnace slag powder as filling material of prebored piles. For this, specimens were prepared by applying 70% and 83% of the general water/binder ratio of the filling material of prebored piles. And compression test, model test, and shaking table test were performed to determine the compressive strength, skin friction on the surface between prebored pile and filling material, and seismic performance of ground stabilizer. As a result of the tests, the compressive strength exceeded the relevant domestic standards, and the skin friction was equivalent to that of ordinary portland cement. In addition, the amount of vertical and horizontal displacement caused by earthquakes was found to be much smaller than the domestic standard. Therefore, when considering the test results comprehensively, it is judged that the feasibility of using a ground stabilizer from recycled resources as filling material for prebored pile is sufficient.

Oxidative Stress in Spermatozoa during Boar Semen Storage (돼지 정액을 저장하는 동안 정자에 미치는 산화스트레스)

  • Oxidative stress is a critical factor affecting the quality and viability of sperm during boar semen storage. Oxidative stress is also a significant concern during the process of freezing semen. The process of semen storage involves exposing the sperm to various stressors, including temperature changes, cryoprotectants, and extended periods of incubation. In addition, oxidative stress can lead to the production of reactive oxygen species (ROS) within the sperm, resulting in oxidative damage to cellular components, such as lipids, proteins, and DNA. Striking a balance between ROS production and the antioxidant defense system is crucial for maintaining sperm viability and functionality during semen storage. Moreover, the prolonged storage of boar semen leads to an increase in ROS levels, which can impair sperm motility, membrane integrity, and DNA integrity. ROS-induced lipid peroxidation affects the fluidity and stability of sperm membranes, leading to decreased sperm motility. Moreover, oxidative damage to the DNA can result in DNA fragmentation, compromising the genetic integrity of the sperm. In conclusion, oxidative stress is a significant challenge in maintaining sperm quality during boar semen storage. Understanding the mechanisms underlying oxidative stress and their impacts on sperm function is crucial for developing effective strategies to minimize oxidative damage and improve sperm storage outcomes.

Studies on the Viability of Frozen Removed Seminal Plasma by Saline(RSP-S) and Tris-buffer(RSP-T) Semen of Small Spcies Dogs (소형 개 RSP-S와 RSP-T 정액의 동결 융해후의 생존성에 관한 연구)

  • This study was carried out to investigate the general characteristics such as volume, sperm concentration, sperm motility, sperm abnormality on whole semen, RSP-S and RSP-T semen and fractional semen of small size dogs, and the effect of temperature and preservation time and cryoproservation on motility of whole and RSP-S and RSP-T semen. Multiple ejaculates were collected from small dogs by the digital manipulation of penis. 1. The volume per ejaculate semen, sperm of concentration and motility and abnormal sperm rate of 1st fractional semen were 0.65$\pm$0.09 $m\ell$, 4.52$\pm$0.35$\times$10$^{6}$ cells/$m\ell$, 15.64$\pm$3.85% and 5.50$\pm$0.62%. Also, 2nd fractional semen were 1.25$\pm$0.20$m\ell$, 3.35$\pm$0.48$\times$10$^{6}$ cells/$m\ell$, 96.25$\pm$4.65% and 4.24$\pm$0.46%. And 3rd fractional semen were 1.45$\pm$0.21$m\ell$, 3.55$\pm$0.52$\times$10$^{6}$ cells/$m\ell$, 92.82$\pm$4.24% and 4.66 $\pm$0.58%, respectively. 2. The sperm of concentration and motility and abnormal sperm rates of whole, RSP-S and RSP-T semen were 5.45$\pm$0.82$\times$10$^{6}$ cells/$m\ell$, 95.55 $\pm$4.65%, 4.58$\pm$0.45% and 4.82$\pm$0.36$\times$10$^{6}$ cells/$m\ell$, 90.10$\pm$3.42%, 6.48$\pm$0.68% and 4.55$\pm$0.45$\times$10$^{6}$ cells/$m\ell$, 93.25$\pm$3.85%, 4.82$\pm$0.58%, respectively. 3. The motility of whole, RSP-S and RSP-T semen were higher at 4$^{\circ}C$ than at 38$^{\circ}C$. When preservation temperature was at 4$^{\circ}C$, survival rates of RSP-S and RSP-T sperm were 97.54~6.25% at 1~72 hrs, 97.40~5.62% at 1~100 hrs, respectively. 4. The survival rates of slow and rapid frozen 2nd fraction, RSP-S and RSP-T semen were 67.3$\pm$4.45%, 88.8$\pm$4.46% and 46.4$\pm$3.84%, 74.4$\pm$4.20%, respectively. Survival rates was significantly higher in frozen RSP-S and RSP-T semen than that in control group(8.5$\pm$2.12%).

The Effect of the Preoperative Semen Parameters for a Patient with Varicolcele on its Operative Results (정계정맥류 환자에서 수술 전 정액지표가 수술 결과에 미치는 영향)

  • Purpose: The purpose of this study is to determine the effect of preoperative semen parameters on both seminal improvement and pregnancy rates following varicocelectomy. Methods: This survey was done in 278 patients who underwent microsurgical inguinal varicocelectomy from January 2001 until October 2006. By the total motile sperm counts (TMSC) before operation, the patients were stratified into three groups. Group A (mild oligoasthenospermia) was defined as above 20 million, group B (moderate oligoasthenospermia) was defined as between 5 and 20 million, and group C (severe oligoasthenospermia) was defined as below 5 million. Improvement rates of TMSC and pregnancy rates following varicocelectomy of each groups were compared. Results: The average TMSC of all the patients was 25.75 million before operation and after operation, it was 80.24 million, showing an average increase of 54.49 million (211.6%). To take a look at mean absolute increase (mean relative increase proportion), group A showed 67.90 million (131.2%), group B 62.20 million (482.5%) and group C 26.33 million (1841.2%). The patients with varicocele whose semen parameter is in bad condition show relatively a low mean absolute increase but high mean relative increase proportion. There was no significant difference in natural pregnancy rate among each groups (p=0.119, p=0.059). Conclusions: Even in the varicocele patient whose semen parameter was in bad condition before surgical operation. varicocelectomy could be chosen as the first treatment to male infertility.

Effect of Extender, Preservation Temperature and Period of liquid Boar Semen on Semen Characteristics and Reproductive Performance (돼지 액상정액의 보존액, 보존온도 및 기간이 정액성상과 번식성적에 미치는 영향)

  • This study was carried out to investigate the effects of extenders such as Beltsville thawing solution(BTS), Modena and Androhep, preservation temperature and period of liquid boar semen on semen characteristics and reproductive performance. Boars were raised at Swine Artificial Insemination Center in National Livestock Research Institute, Sunghwan, Chungnam, Korea. This experiment was carried out from 1995 to 2000. The results obtained were summarized as follows. 1. Sperm motility in the samples with Androhep and BTS reduced from day 5 and in the samples with Modena reduced from day 3 of storage. pH or 3 extenders varied from 6.24 to 7.04 during day 1 to 5 of storage. Farrowing rate of sows inseminated with liquid boar semen offended with BTs, Modena and Androhep extenders did not show any differences until day f after semen collection. Sows inseminated with Androhep extender had better farrowing rates (P<0.05) than those with Modena extender at day 1 or 5 after semen collection, but farrowing rates after AI using BTS did not differ compared to those Androhep and Modena. Litter size did not show any differences among the three extenders, but Androhep had the decreased litter size from day i of storage. 2. Motility and normal acrosome of the sperm preserved at 5$^{\circ}C$ did not show any differences until day 4 of storage, but those at 17$^{\circ}C$ changed from day 3 and 4, respectively. Farrowing rate of sows artificially inseminated with liquid boa. semen preserved at 17$^{\circ}C$ had higher, than at 5$^{\circ}C$ (p<0.05), but there was no significant differences in litter size. Farrowing rates and litter size were decreased from day 2 and day 3 of storage at 17$^{\circ}C$, respectively. Farrowing rate of sows inseminated with the preserved semen at 5$^{\circ}C$ did not changed until day 4, but the litter size at 5$^{\circ}C$ was lower than that at 17$^{\circ}C$.

Analysis on Artificial Insemination Failure and Characteristics of Frozen Semen Used for Reproduction of Hanwoo Cow in Gangwon East Area (강원 영동지방 암소 인공수정에 이용된 한우보증씨수소 정액의 인공수정 실패율 분석 및 동결정액성상 분석)

  • This study was carried out to investigate artificial insemination (AI) failure status and frozen semen characteristics in Korean proven bulls' number (KPN) semen used for AI of Hanwoo cows in Gangwon East region (Gangneung, Donghae, Taebaek, Samcheok, Sokcho, Yangyang, Goseong). Among semen used for AI, AI failure rate showed lowest at KPN506 (27.6%), whereas highest at KPN593 (77.2%). Correlations of AI failure in between Korean proven bulls semen and cows was 0.2941, which means that AI failure rate of Korean proven bulls semen may have respectable effect on reproduction of Hanwoo cow. In addition, present study was conducted to investigate spermatozoal viability rate, ruptured acrosome rate and active mitochondria in frozen Korean proven bulls semen with flow cytometry. The semen of KPN593 showed significantly ($p$<0.05) higher viability rate in KPN593 (30.49%) than that in KPN637 (37.34%). Furthermore, percentage of ruptured acrosome was lower in KPN637 as 21.37% than in KPN637 (21.37%), but it was not statistically significant. In conclusion, these results indicate that choice of Korean proven bulls semen may correlate positively with conception rate in Hanwoo cow. Therefore, KPN with high AI failure rate might be avoid to increase conception rate and characteristics of frozen semen might be evaluated before its use for AI.

돼지 인공수정시 1회 주입 정자농도가 번식성적에 미치는 영향

  • 본 연구는 돼지 인공수정 시 1 회 주입하는 정자농도가 번식성적에 미치는 영향을 구명하기 위하여 활력이 70%이상인 정자수 기준으로 80㎖ 병당 30.0×10/sup 8/80㎖, 25.0, 20.0, 17.5, 15.0, 12.5, 10.0 및 7.5로 농도를 각각 조절하여 12시간 간격으로 2회 인공수정하고 분만율 및 산자수를 조사하였다. 본 실험에 사용된 액상정액은 축산기술연구소 돼지 인공수정 센타의 종모돈중 통일품종 정액을 BTS(Beltsville thawing solution) 보존액으로 혼합하여 5개 농장의 암돼지에 인공수정 하였으며, 정자농도는 광전비색계(Spectronic-20, USA)를 이용하여 측정하였다. 분만율은 72.3%-89.3%로 농도별로 통계적인 유의차는 인정되지 않았다.
